핵의학과 내 작업 환경에서 공기층 납치마의 피폭선량 감소 효과 평가 (Evaluation of Reductive Effect of Exposure Dose by Using Air Gap Apron in Nuclear Medicine Related Work Environment)

  • 본 연구는 핵의학과에서 가장 많이 사용되고 있는 $^{99m}Tc$에서 방출되는 140 keV 감마선을 기준으로, 세 가지 실험을 통해 기존에 사용되고 있는 납치마와 공기층 납치마를 이용해 Optically Stimulated Luminescence Dosimeter (OSLD) 선량계에 도달하는 선량을 측정하고, 공기층 납치마의 피폭선량 감소효과에 대해 연구했다. 그 결과 선량계와 납판 사이의 거리를 납치마의 공기층이라고 가정했을 때, 선량계와 0.2 mm 납판 사이의 거리가 없는 0 Cm일 때 측정된 10개의 선량계의 평균값은 0.515 mSv이고, 선량계와 납판 사이의 거리를 20 Cm로 떨어뜨린 경우 10개의 측정값의 평균이 0.138 mSv로 약 0.377 mSv의 선량 감소 효과를 보였다. 선량계와 0.5 mm 납판 사이의 거리가 없는 0 Cm일 때 10개의 선량계의 측정값의 평균은 0.296 mSv이고, 선량계와 납판 사이의 거리를 20 Cm로 떨어뜨린 경우 10개의 측정값의 평균이 0.075 mSv로 약 0.221 mSv의 선량 감소 효과를 보였다. 3일 간의 누적선량을 측정한 결과, 공기층이 없는 납치마의 경우 측정된 누적선량의 평균은 0.238 mSv, 공기층 납치마의 누적선량은 0.176 mSv로 0.062 mSv의 선량 감소 효과를 보였다. 한 달 간의 누적선량을 측정한 결과, 공기층이 없는 납치마의 누적선량의 평균은 0.59 mSv, 공기층 납치마의 누적선량은 0.54 mSv로 0.05 mSv의 선량 감소 효과를 보였다.

  • Objectives : To assess the availability of the glycophorin A (GPA) assay to detect the biological effect of ionizing radiation in workers exposed to low-doses of radiation. Methods : Information on confounding factors, such as age and cigarette smoking was obtained on 144 nuclear power plant workers and 32 hospital workers, by a self-administered questionnaire. Information on physical exposure levels was obtained from the registries of radiation exposure monitoring and control at each facility. The GPA mutant assay was performed using the BR6 method with modification by using a FACScan flow cytometer. Results : As confounders, age and cigarette smoking habits showed increasing trends with GPA variants, but these were of no statistical significance. Hospital workers showed a higher frequency of the GPA variant than nuclear power plant workers in terms of the NO variant. Significant dose-response relationships were obtained from in simple and multiple linear regression models. The slope of the regression equation for nuclear power plant workers was much smaller than that of hospital workers. These findings suggest that there may be apparent dose-rate effects. Conclusion : In population exposed to chronic low-dose radiation, the GPA assay has a potential to be used as an effective biologic marker for assessing the bone marrow cumulative exposure dose.

DIFFERENTIAL EXPRESSION OF RADIATION RESPONSE GENES IN SPLEEN, LUNG, AND LIVER OF RATS FOLLOWING ACUTE OR CHRONIC RADIATION EXPOSURE

  • We analyzed the differential effects of histopathology, apoptosis and expression of radiation response genes after chronic low dose rate (LDR) and acute high dose rate (HDR) radiation exposure in spleen, lung and liver of rats. Female 6-week-old Sprague-Dawley rats were used. For chronic low-dose whole body irradiation, rats were maintained for 14 days in a $^{60}Co$ gamma ray irradiated room and received a cumulative dose of 2 Gy or 5 Gy. Rats in the acute whole body exposure group were exposed to an equal dose of radiation delivered as a single pulse ($^{137}Cs$-gamma). At 24 hours after exposure, spleen, lung and liver tissues were extracted for histopathologic examination, western blotting and RT-PCR analysis. 1. The spleen showed the most dramatic differential response to acute and chronic exposure, with the induction of substantial tissue damage by HDR but not by LDR radiation. Effects of LDR radiation on the lung were only apparent at the higher dose (5 Gy), but not at lower dose (2 Gy). In the liver, HDR and LDR exposure induced a similar damage response at both doses. RT-PCR analysis identified cyclin G1 as a LDR-responsive gene in the spleen of rats exposed to 2 Gy and 5 Gy gamma radiation and in the lung of animals irradiated with 5 Gy. 2. The effects of LDR radiation differed among lung, liver, and spleen tissues. The spleen showed the greatest differential effect between HDR and LDR. The response to LDR radiation may involve expression of cyclin G1.

병원내 방사선작업종사자들의 염색체이상빈도 (Chromosome Aberration in Peripheral Lymphocyte of Radiation Workers in Hospital)

  • 병원내 방사선 작업 종사자 51명(진단방사선과 18명, 치료방사선과 17명, 핵의학과 16명)에서 림프구의 염색체이상 빈도를 대조군 49명과 비교하여 저선량 만성피폭시 인체에 미치는 영향을 조사하였다. 종사자의 축적선량은 0.30-40.07 mSv 였으며 총축적선량 5 mSv 이하가 46%로 가장 많았다. 염색체 이상이 있는 세포수는 종사자군에서 1.275%로, 대조군에서의 0.677%에 비해 더 높았다(p<0.001). 염색분체형 이상은 종사자군이 약간 높은 경향은 있지만 통계적인 유의성은 없었으며, 염색체형 이상은 대조군 $0.344{\times}10^{-2}$/cell에 비해 종사자군이 $0.706{\times}10^{-2}$/cell로 2.1배 높았다(p<0.05). 이중 교환은 대조군 $0.083{\times}10^{-2}$/cell에 비해 종사자군 $0.245{\times}10^{-2}$/cell로 3.0배 높았다(p<0.05). 총축적선량, 최근 5년 및 최근 2년간의 선량은 염색체이상과 선량-반응 관계가 없었으나, 최근 1년간 피폭선량과 염색체형 교환 빈도사이에 유의한 상관관계가 있었다(p<0.05). 근무년수와 염색체 이상빈도와 선량-반응관계는 없었으며, 각 작업부서 별 염색체이상 빈도 및 대조군과의 비교시, 염색체형 교환이 핵의학과 $0.313{\times}10^{-2}$/cell로 가장 높았으며 (p<0.05), 치료방사선과 근무자 $0.265{\times}10^{-2}$/cell (p=0.0622), 진단방사선과 근무자 $0.167{\times}10^{-2}$/cell (p=0.1107)순이었다. 색체이상이 유발될 수 있음을 확인하였으며, 따라서 림프구 염색체이상은 매우 낮은 양의 방사선 피폭에 의하여도 발생함을 알 수 있었다.

백금항암제 Cisplatin의 신장독성에 대한 Aspalactone의 경감작용 (Ameliorating Activity of Aspalactone on Cisplatin Induced Nephrotoxicity)

  • Cisplatin is an inorganic complex formed by a central atom of platinum surrounded by chlorine and ammonia atoms in the cis position in the horizontal plane, Cisplatin is one of the most effective anticancer drug, widely used against various tumor such as testicular tumor, brain tumor, ovary tumor, bladder carcinoma, colon cancer etc. However its clinical use has been limited by nephrotoxicity, ototoxicity , gastrointestinal disturbances, myeloscrppression and allergic reactions. In these toxicities, dose related and cumulative nephrotoxicity is the major dose limit factor. So, to evaluate the protective effect of aspalactone on cisplatin nephrotoxicity in rats, both compounds were given intraperitoneally, Protective effects of aspalactone against nephrotoxicity of cisplatin were observed when aspalactone was administered to rats 1hr beforecisplatin injection. Hepatotoxicity induced by combination treatment of cisplatin and aspalactone was not observed. The present results indicate that aspalactone may provide protection against cisplatin nephrotoxicity, when it is given 1hr before cisplatin injection.

실리콘 핀 포토다이오드를 이용한 전자 선량계의 설계 및 구현 (A Study on the Development of Electronic Personal Dosimeter with Silicon PIN Photodiode)

  • Recently, electronic personal dosimeters based upon silicon PIN photodiode or miniature GM tube were developed and have attracted a lot of attention because of the advantages of their nature such as indication of dose rate and the cumulative dose, and facilitation of record keeping. In this paper, we have developed a high-sensitivity electronic personal dosimeter with silicon PIN photodiode. The electronic personal dosimeter is constructed with silicon PIN photodiode, preamplifier, and shaping amplifier. To show the effectiveness of electronic personal dosimeter, we conducted nuclear radiation experiments using $\gamma$-ray Ba-133, Cs-137, and Co-60. The electronic personal dosimeter have a good linearity on $\gamma$-ray energy and activity.

Genetic radiation risks: a neglected topic in the low dose debate

  • Objectives To investigate the accuracy and scientific validity of the current very low risk factor for hereditary diseases in humans following exposures to ionizing radiation adopted by the United Nations Scientific Committee on the Effects of Atomic Radiation and the International Commission on Radiological Protection. The value is based on experiments on mice due to reportedly absent effects in the Japanese atomic bomb (A-bomb) survivors. Methods To review the published evidence for heritable effects after ionising radiation exposures particularly, but not restricted to, populations exposed to contamination from the Chernobyl accident and from atmospheric nuclear test fallout. To make a compilation of findings about early deaths, congenital malformations, Down's syndrome, cancer and other genetic effects observed in humans after the exposure of the parents. To also examine more closely the evidence from the Japanese A-bomb epidemiology and discuss its scientific validity. Results Nearly all types of hereditary defects were found at doses as low as one to 10 mSv. We discuss the clash between the current risk model and these observations on the basis of biological mechanism and assumptions about linear relationships between dose and effect in neonatal and foetal epidemiology. The evidence supports a dose response relationship which is non-linear and is either biphasic or supralinear (hogs-back) and largely either saturates or falls above 10 mSv. Conclusions We conclude that the current risk model for heritable effects of radiation is unsafe. The dose response relationship is non-linear with the greatest effects at the lowest doses. Using Chernobyl data we derive an excess relative risk for all malformations of 1.0 per 10 mSv cumulative dose. The safety of the Japanese A-bomb epidemiology is argued to be both scientifically and philosophically questionable owing to errors in the choice of control groups, omission of internal exposure effects and assumptions about linear dose response.

IDENTIFICATION OF GENES EXPRESSED IN LOW-DOSE-RATE γ-IRRADIATED MOUSE WHOLE BRAIN

  • While high-dose ionizing radiation results in long term cellular cytotoxicity, chronic low-dose (<0.2 Gy) of X- or ${\gamma}$-ray irradiation can be beneficial to living organisms by inducing radiation hormesis, stimulating immune function, and adaptive responses. During chronic low-dose-rate radiation (LDR) exposure, whole body of mice is exposed to radiation, however, it remains unclear if LDR causes changes in gene expression of the whole brain. Therefore, we aim to investigate expressed genes (EGs) and signaling pathways specifically regulated by LDR-irradiation ($^{137}Cs$, a cumulative dose of 1.7 Gy for total 100 days) in the whole brain. Using microarray analysis of whole brain RNA extracts harvested from ICR and AKR/J mice after LDR-irradiation, we discovered that two mice strains displayed distinct gene regulation patterns upon LDR-irradiation. In ICR mice, genes involved in ion transport, transition metal ion transport, and developmental cell growth were turned on while, in AKR/J mice, genes involved in sensory perception, cognition, olfactory transduction, G-protein coupled receptor pathways, inflammatory response, proteolysis, and base excision repair were found to be affected by LDR. We validated LDR-sensitive EGs by qPCR and confirmed specific upregulation of S100a7a, Olfr624, and Gm4868 genes in AKR/J mice whole brain. Therefore, our data provide the first report of genetic changes regulated by LDR in the mouse whole brain, which may affect several aspects of brain function.

Dose metrology: TLD/OSL dose accuracy and energy response performance

  • An essential step in evaluating and comparing the performance of two passive radiation dosimeter types, thermosluminescent (TLD) and optically stimulated luminescence (OSL), used by workers in environments with ionizing radiation for individual radiological monitoring and control of external exposure at various times (cumulative dose for 1 month), is to compare the measured dose accuracy, energy response, and coefficient of variation. In fact this performance study consists in determining the accuracy of both R(10) and R(0.07) which are considered as the ratios of the measured dose (Hp(10) or Hp(0.07)) to the delivered dose (Hp(10) or Hp(0.07)) for each photon energy. The validity of the results of this test is based on the acceptance limits of the ICRP and the international standard IEC-62387. The relative energy response used is normalized to the 137Cs 662 keV energy to find which energy response is closest to the ideal case, and the coefficient of variation that allows to determine the statistical fluctuation of the Hp(10) and Hp(0.07) doses. The results of the accuracy test for the OSL and TLD dosimeters are acceptable because they fall within the ICRP limits. For the energy response, the OSL performs better than the TLD for Hp(10) and Hp(0.07), and for the coefficient of variation, the OSL satisfies the requirements of ISO 62387 for both Hp(10) and Hp(0.07), while the TLD satisfies these requirements only for the measurement of Hp (0.07).

시스플라틴 항암제에 의해 유발되는 식욕부진과 신경병증성 통증이 쥐의 뒷다리근에 미치는 영향 (Effect of Anorexia and Neuropathic Pain Induced by Cisplatin on Hindlimb Muscles of Rat)

  • Purpose: The purpose of this study was to examine the effect of anorexia and neuropathic pain induced by cisplatin on hindlimb muscles of rats. Methods: Adult male Sprague-Dawley rats were divided into two groups, a cisplatin-treated group (n=10) and a control group (n=10). In the cisplatin-treated group, cisplatin at a dose of 2 mg/kg was injected intraperitoneally two times a week up to a cumulative dose of 20 mg/kg over 5 weeks, and in the control group saline (0.9% NaCl) was injected intraperitoneally at the same dose and duration as the cisplatin-treated group. At 34 days all rats were anesthetized, after which the soleus and plantaris muscles were dissected. Withdrawal threshold, body weight, food intake, activity, muscle weight, Type I and II fiber cross-sectional areas and myofibrillar protein content of the dissected muscles were determined. Results: Compared with the control group, the cisplatin-treated group showed significant decreases (p<.05) in withdrawal threshold, activity, food intake, body weight, Type I and II fiber cross-sectional areas, myofibrillar protein content and weight of the soleus and plantaris muscles. Conclusion: Muscular atrophy in hindlimb occurs due to anorexia and neuropathic pain induced by the cisplatin treatment.
