목 적:타우린은 술폰 기를 산기로 하는 황 아미노산의 일종이며 뇌, 망막, 심장, 근육에 많이 분포되어 있다. 최근 국소적 뇌허혈에 대한 타우린의 신경보호효과에 관한 연구들이 발표되고 있으나 대부분 연구가 성인의 뇌졸중의 치료에 대한 연구이며 신생아시기의 저산소성 손상에 대한 효과를 구체적으로 연구한 바가 없다. 본 연구에서는 타우린이 저산소 상태로 유발된 뇌세포 배양과 신생 백서의 저산소성 허혈성 뇌손상에서 항세포사멸사를 통한 뇌보호 효과가 있는지를 알아보고자 실험하였다. 방 법:재태기간 18일된 태아 흰쥐의 대뇌피질 세포를 배양하여 1% $O_2$ 배양기에서 저산소 상태로 뇌세포 손상을 유도하여 저산소군, 손상 전 후 타우린 투여군($30{\mu}g/mL$)으로 나누어 정상산소군과 비교하였다. 세포사멸사와 관련을 알아보기 위해 Bcl-2, Bax, caspase-3 primer와 항체로 실시간 중합효소연쇄반응과 western blotting을 하였다. 또한, 생후 7일된 백서의 좌측 총 경동맥을 결찰한 후 저산소(8% $O_2$) 상태로 2시간 노출시켜서, 저산소성 허혈성 뇌 손상을 유발하였고, 뇌손상 전 후 30분에 타우린을 체중 kg당 30 mg을 투여하였다. 저산소성 허혈성 뇌손상 후 1일, 3일, 1주, 2주, 4주 째 뇌를 적출하여 Bcl-2, Bax, caspase-3 primer를 이용하여 실시간 중합효소연쇄반응을 하였고, 동일 항체로 western blotting하였다. 결 과:저산소로 유발된 뇌세포 배양에서 정상군에 비해 저산소군에서 뇌세포 손상이 많았고 저산소 손상전 타우린 투여군에서 뇌세포 손상이 회복되었으며 저산소 손상 후 타우린 투여군에서는 저산소 손상 전 투여군보다 회복력이 떨어졌다. 실시간 중합효소연쇄반응과 western blotting을 이용한 저산소 상태의 태아 백서 뇌세포 배양 실험뿐만 아니라 저산소성 허혈성 뇌손상 동물 모델에서도 타우린을 투여한 경우 Bcl-2의 발현은 증가하고, Bax/Bcl-2의 비율, Bax와 caspase-3의 발현은 감소함을 보였다. 결 론:본 연구에서 타우린은 주산기 저산소성 허혈성 뇌손상에서 Bcl-2 발현 감소, Bax와 caspase-3 발현 증가를 유발시켜 항 세포사멸사 기전을 통한 신경보호 역할을 하는 것을 알 수 있었다. 그리고 이것은 저산소 손상 후 1주와 2주째에 가장 효과가 있었다.
Objective : The effects on neural proliferation and differentiation of neural stem cells (NSC) of basic fibroblast growth factor-2 (bFGF). insulin growth factor-I (IGF-I). brain-derived neurotrophic factor (BDNF). and nerve growth factor (NGF) were assessed. Also, following combinations of various factors were investigated : bFGF+IGF-I, bFGF+BDNF, bFGF+NGF, IGF-I+BDNF, IGF-I+NGF, and BDNF+NGF. Methods : Isolated NSC of Fisher 344 rats were cultured with individual growth factors, combinations of factors, and no growth factor (control) for 14 days. A proportion of neurons was analyzed using $\beta$-tubulin III and NeuN as neural markers. Results : Neural differentiations in the presence of individual growth factors for $\beta$-tubulin III-positive cells were : BDNF, 35.3%; IGF-I, 30.9%; bFGF, 18.1%; and NGF, 15.1%, and for NeuN-positive cells was : BDNF, 34.3%; bFGF, 32.2%; IGF-I, 26.6%; and NGF, 24.9%. However, neural differentiations in the absence of growth factor was only 2.6% for $\beta$-tubulin III and 3.1% for NeuN. For $\beta$-tubulin III-positive cells, neural differentiations were evident for the growth factor combinations as follows : bFGF+IGF-I, 73.1 %; bFGF+NGF, 65.4%; bFGF+BDNF, 58.7%; BDNF+IGF-I, 52.2%; NGF+IGF-I, 40.6%; and BDNF+NGF, 40.0%. For NeuN-positive cells : bFGF+IGF-I, 81.9%; bFGF+NGF, 63.5%; bFGF+BDNF, 62.8%; NGF+IGF-I, 62.3%; BDNF+NGF, 56.3%; and BDNF+IGF-I, 46.0%. Significant differences in neural differentiation were evident for single growth factor and combination of growth factors respectively (p<0.05). Conclusion : Combinations of growth factors have an additive effect on neural differentiation. The most prominent neural differentiation results from growth factor combinations involving bFGF and IGF-I. These findings suggest that the combination of a mitogenic action of bFGF and post-mitotic differentiation action of IGF-I synergistically affects neural proliferation and NSC differentiation.
Investigation of chemically synthesized inositol 1,4,5-trisphosphate [$Ins(1,4,5)P_3$] analogs has led to the isolation of a novel binding protein with a molecular size of 130 kDa, characterized as a molecule with similar domain organization to phospholipase C-${\delta}1$ (PLC-${\delta}1$) but lacking the enzymatic activity. An isoform of the molecule was subsequently identified, and these molecules have been named PRIP (PLC-related, but catalytically inactive protein), with the two isoforms named PRIP-1 and -2. Regarding its ability to bind $Ins(1,4,5)P_3$ via the pleckstrin homology domain, the involvement of PRIP-1 in $Ins(1,4,5)P_3$-mediated $Ca^{2+}$ signaling was examined using COS-1 cells overexpressing PRIP-1 and cultured neurons prepared from PRIP-1 knock-out mice. Yeast two hybrid screening of a brain cDNA library using a unique N-terminus as bait identified GABARAP ($GABA_A$ receptor associated protein) and PP1 (protein phosphatase 1), which led us to examine the possible involvement of PRIP in $GABA_A$ receptor signaling. For this purpose PRIP knock-out mice were analyzed for $GABA_A$ receptor function in relation to the action of benzodiazepines from the electrophysiological and behavioral aspects. During the course of these experiments we found that PRIP also binds to the b-subunit of $GABA_A$ receptors and PP2A (protein phosphtase 2A). Here, we summarize how PRIP is involved in $Ins(1,4,5)P_3$-mediated $Ca^{2+}$ signaling and $GABA_A$ receptor signaling based on the characteristics of binding molecules.
연구목적: 알코올의존에 내재된 분자생물학적 기전을 이해하고 알코올리즘 치료 약물의 새로운 표적을 알아내기 위해서는, 알코올에 반응하는 유전자 혹은 반응 경로를 알아내는 것이 필요하다. DNA microarray 기법의 발달로 고전적 연구 방법과 달리 동시에 수천 수만개의 유전자의 표현을 검사하는 것이 가능하게 되었다. 본 연구에서는 알코올을 흰쥐의 신경아교종 세포에 처리했을 때 어떤 유전자의 발현을 조절하는지 DNA microarray를 이용하여 알아보고자 하였다. 방 법: 흰쥐 신경아교종 C6 세포주를 배양하여 에탄올 처리하고 총 RNA를 분리한 후 유전자 발현 양상을 조사하기 위해 cDNA microarray를 수행하였다. 결 과: 에탄올 처리군과 대조군간의 유전자 발현의 차이를 비교 분석한 결과 에탄올이 처리된 군에서 대조군에 비해 15개의 유전자가 발현이 증가하였고 12개의 유전자가 발현이 감소하였다. 발현이 증가한 유전자는 Orthodenticle(Drosophila) homolog 1, procollagen type II, adenosine A2a receptor, GATA-bindning protein2를 포함하고 있었고, 발현이 감소한 유전자는 diacylglycerol kinase beta, PRKC, Protein phosphatase 1, clathrin-associated protein 17, nucleoporin p58, proteasome를 포함하였다. 결 론: 흰쥐의 신경아교종 세포주에 알코올을 처치하였을 때 급성기에 알코올에 반응하여 발현이 증가하거나 감소한 유전자는 전반적으로 전사의 조절, 신호전달체계, 허혈성 뇌손상의 중재, 신경세포의 퇴행에 관여하는 것들이었다. 본 연구는 유전자 발현 시스템을 이용하여 에탄올에 반응하는 새로운 후보 유전자들을 관찰하였다는데 의의가 있다.
