Laboratory Culture Media-Dependent Biocontrol Ability of Burkholderia gladioli strain B543

  • Cultivation of a biocontrol agent on a certain medium often results in reduced biocontrol efficacy and alters physiological state. In our previous study, Burkholderia gladioli strain B543 with long-term subculture on tryptic soy agar resulted in significantly reduced biocontrol ability against cucumber damping-off caused by P. ultimum. Therefore, we investigated the influence of laboratory culturing media on biocontrol activity and physiological state of Burkholderia gladioli strain B543 by using long-term repeated culture on a certain medium. When isolate B543 were successionally cultured on King's B agar (KBA), tryptic soy agar, nutrient agar (NA), or soil extract agar more than 20 times, the isolate cultured on KBA or NA showed a significantly enhanced biocontrol efficacy and higher population density in the rhizosphere of cucumber compared to that of the others. However, the isolates cultured on KBA more than 20 times showed the lowest production of protease, siderophore, or antifungal substance(s), measured by skim milk agar, Chrome-Azurol-S agar, and potato dextrose agar amended with 10% of the culture filtrate, respectively. Our results suggest that adaptation to proper culturing medium can alter biocontrol ability and physiological state, and we must consider laboratory media in optimizing the use of biocontrol agents.

Effects of Aluminium on Vitellogenin and Its mRNA Induction by Estradiol-17$\beta$ in the Primary Culture of Hepatocytes in the Rainbow Trout Oncorhynchus mykiss

  • Effects of Al on vitellogenin (VTG) and VTG mRNA induction by estradiol-17 $\beta$($E_2$) were examined in primary hepatocyte culture of rainbow trout. Hepatocytes were precultured for 2 days and then E2 ($2{\times}10^{-6}$M) and Al ($10^{-6}-10^{-4}$M) were simultaneously added to the incubation medium. Hepatocytes were cultured for 5 more days. Media and hepatocytes were then analyzed by SDS-PAGE and Northern blotting for VTG and VTG mRNA, respectively. These metal had no appreciable effect on the viability of hepatocytes in culture. However, Al interfered with VTG production and VTG mRNA expression. Al reduced VTG production in a concentration-dependent way and a significant reduction accurred at Al concentrations greater than $5{\times}10^{-5}$M. VTG mRNA expression also decreased with a negative correlation with Al concentration (r=-0.98). These results suggest that Al inhibit VTG production at the transcriptional level to reduce VTG mRNA expression.

  • A glucose-limited “pH-stat” continuous culture study of Lactobacillus bulgaricus NLS-4 in an anaerobic condition showed the marked effects of environmental pH on end products, fermentation blances and bioenergetic aspects of the organism. Lactic acid was the major end product of fermentation with minor products, such as acetic acid, formic acid and ethanol throughout the pH range tested. In acidic conditions below pH 6.5, a typi-cal pattern of homofermentation was revealed whereas in alkaline conditions, the metabolic pattern was changed from homofermentation to heterofermentation and led to acquire much energy. This metabolic change was likely due to the pH-dependent lactate dehydrogenase activity. Molar growth yields (Yglc=35.5-44.4) and YATP, $18.5\pm2.5$ in average which was 80% higher than the value ever postulated seemed to be accounted for less requirement of maintenance energy of the organism in the culture conditions.

Optimization of Staphylokinase Production in Bacillus subtilis Using Inducible and Constitutive Promoters

  • Staphylokinase (SAK) was produced in B. subtilis using two different promoter systems, i.e. the P43 and sacB promoters. To maximize SAK expression in B. subtilis, fermentation control strategies for each promoter were examined. SAK, under P43, a vegetative promoter transcribed mainly by $\sigma$(sup)B containing RNA polymerase, was overexpressed at low dissolved oxygen (D.O.) levels, suggesting that the sigB operon is somewhat affected by the energy charge of the cells. The expression of SAK at the 10% D.O. level was three times higher than that at the 50% D.O. level. In the case of sacB, a sucrose-inducible promoter, sucrose feeding was used to control the induction period and induction strength. Since sucrose is hydrolyzed by two sucrose hydrolyzing enzymes in the cell and culture broth, the control strategy was based on replenishing the loss of sucrose in the culture. With continuous feeding of sucrose, WB700 (pSAKBQ), which contains the SAK gene under sacB promoter, yielded ca. 35% more SAK than the batch culture. These results present efficient promoter-dependent control strategies in B. subtilis host system for foreign protein expression.

Stimulatory Effect of Porcine Epididymal Fluid on In Vitro Maturation of Porcine Immature Oocytes

  • The aim of this study was to investigate whether addition of porcine epididymal fluid (pEF) into culture medium during in vitro maturation influences the nuclear maturation of porcine germinal vesicle (GV) oocytes. Porcine cumulus-oocyte complexes (COCs) from follicles were cultured in tissue culture medium 199 (TCM 199) containing pEF. After 48hr of culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. The proportion of oocytes reaching at metaphase II (M II) stage was significantly (p<0.05) increased in oocytes cultured in the media supplemented with 10% pEF during in vitro maturation than in those without pEF regardless of cumulus presence or absence (54.6% vs 22.5%, 51.7% vs 24.2%). The supplementation of pEF during maturation of oocyte enhanced oocytes maturation in a dose-dependent manner in vitro. Also significant differences (p<0.05) in the percentage of MII oocytes were observed according to exposure period in pEF. Present study suggests that pEF contains a enhancing component(s) for nuclear maturation of porcine immature oocytes in vitro.

다양한 배지에서 느타리버섯 No. 42균주로부터 리그닌분해효소 생산 (Production of ligninolytic enzymes by Pleurotus ostreatus No. 42 in various culture media)

  • 느타리 버섯 No. 42균주를 5가지 배지에서 연구한 결과, 리그닌 분해효소인 Lac와 MnP는 생산되었으나 LiP는 생산되지 않았다. 실험한 본 균주의 경우 리그닌 분해효소인 Lac와 MnP의 생산에 밀기울이 관여하는 것을 확인 할 수 있었다. 액체배지에서는 GPYW에서 Lac(2.4 U/ml)와 MnP(3.6 U/ml)가 최대 생산됨을 알 수 있었으며 고체배지에서는 WMW에서 MnP(4.0 U/ml)가 최대 생산되었으며 W에서는 Lac(11.0 U/ml)가 최대 생산되었다.

  • To investigate the influence of age and dietary fat level on the immune function and the growing potential of the fibroblast cells, male rats of 2 month, 6 month and 30 month of age were fed either 6% or 30% fat diet for 16 weeks. The weight of thymus decreased linearly with increasing age. And this age-dependent degeneration of thymus was delayed in rats fed low fat diets. The blastogenesis of spleen lymphocytes to PHA, ConA, and PWM was decreased with increasing age, however, no effect of dietary fat level was observed. The age-related decline in ratios of PHA/ConA response may suggest that T suppressor cell activity increases with age. In cell culture system, lung fibroblast cells from 30M rats showed lower plating efficiency. longer doubling time. and shorter cumulative doubling potential than those from 2M or 6M animals. Also. the morphology of fibroblasts from 30M rats was tended to be rouned rather than flattened and more variable in size and being generally larger. wherease those from 2M and 6M rats were uniform in size and adhered tightly to the culture vessel in ordered arrays. Therefore fibroblast cell culture system tried in this study reflects the changes of cellular aging.

The Technology Readiness of Thai Governmental Agency

  • The paper aims to analyze the factors influencing the digital technology readiness of the governmental agency in Thailand, namely the Office of the Welfare Promotion Commission for Teachers and Educational Personnel (OTEP). This paper discusses challenges regarding the technology readiness of OTEP, which is taken as a case study for Thai governmental agencies. Data is collected through questionnaires distributed from October to December 2020. With a population of 777 OTEP staff, 534 employees are the respondents of this study. The study employs correlation, multiple linear regression, and structural equation modeling to analyze the data. The dependent variable is the digital technology readiness, while the independent variables are age, technology literacy, technology experience, attitude, organizational culture, leadership, and learning facilities. One of the principal findings is that the digital technology readiness of OTEP is at a moderate level. In addition, learning facilities, technology literacy, leadership, and organizational culture are found to be statistically significant for digital technology readiness. The findings highlight the issues and obstacles associated with encouraging human resource development, notably in the field of digital technology. Adopting digital technology to give better services to a large scale of customers is challenging for most large governmental enterprises, considering OTEP as a wonderful example for organizations under government oversight.

김치 발효에 관여하는 효모의 다양성 및 역할 (Diversity and Role of Yeast on Kimchi Fermentation)

  • This review summarizes the studies on a wide variety of yeast found in kimchi and the effects of yeast on kimchi fermentation, and discusses the direction for further research. Yeast belongs to the genera Trichosporon, Saccharomyces, Sporisorium, Pichia, Lodderomyces, Kluyveromyces, Candida, Debaryomyces, Geotrichum, Kazachstania, Brassica, Yarrowia, Hanseniaspora, Brettanomyces, Citeromyces, Rhodotorula, and Torulopsis have been identified using culture-dependent methods and metagenomics analysis. The application of yeast as a starter into kimchi has resulted in an extension of shelf life and improvement of sensory characteristics due to a decrease in the amount of lactic acid. On the other hand, some yeast cause kimchi spoilage, which typically appears as an off-odor, texture-softening, and white-colony or white-film formation on the surface of kimchi. In contrast to lactic acid bacteria, there are limited reports on yeast isolated from kimchi. In addition, it is unclear how yeast affects the fermentation of kimchi and the mechanism by which white colony forming yeast predominate in the later stage of kimchi fermentation. Therefore, more research will be needed to solve these issues.

발효식품의 마이크로바이옴 분석 기술 (Analysis techniques for fermented foods microbiome)

  • Human have eaten various traditional fermented foods for a numbers of million years for health benefit as well as survival. The beneficial effects of fermented foods have been resulted from complex microbial communications within the fermented foods. Therefore, the holistic approaches for individual identification and complete microbial profiling involved in their communications have been of interest to food microbiology fields. Microbiome is the ecological community of microorganisms that literally share our environments including foods as well as human body. However, due to the limitation of culture-dependent methods such as simple isolations of just culturable microorganisms, the culture-independent methods have been consistently developed, resulting in new light on the diverse non-culturable and hitherto unknown microorganisms, and even microbial communities in the fermented foods. For the culture-independent approaches, the food microbiome has been deciphered by employing various molecular analysis tools such as fluorescence in situ hybridization, quantitative PCR, and denaturing gradient gel-electrophoresis. More recently, next-generation-sequencing (NGS) platform-based microbiome analysis has been of interest, because NGS is a powerful analytical tool capable of resolving the microbiome in respect to community structures, dynamics, and activities. In this overview, the development status of analysis tools for the fermented food microbiome is covered and research trend for NGS-based food microbiome analysis is also discussed.
