The Effect of Department Satisfaction and Ego-Identity on College Life Adaptation (학과만족도와 자아정체감이 대학생활 적응에 미치는 영향)

  • The purpose of this study is to examines the factors influencing college students' adaptation to College life adaptation, and to investigate the relationship between relationship between variables and College life adaptation. Structured questionnaire was given on 210 freshman, and data was analyzed using SPSS/WIN 22.0. There was a significant correlation between College life adaptation and Department satisfaction (r = .216, p <.001) and Ego identity (r = 395, p <.001). As a result of the statistical significance of the model predicted to College life adaptation, the F statistic value is 12.222 and the significance probability is .000. Variables have significant explanations for college adjustment at a significance level of 0.1. College life adaptation is also explained by 44% variable. If we develop the adaptation program for college life considering variables, it is expected that it will have a positive effect on the improvement of adaptation ability of college students.

Partial Characterization and Induction of Ferulic Acid Esterase and Xylanase from Pseudomonas sp. LG2 (Pseudomonas sp. LG2의 Ferulic acid esterase 및 Xylanase 유도와 부분적 특성)

  • Lignin degrading bacterium Pseudomonas sp. LG2 was able to degrade lignin substrate to a lot of APPL compound. APPL compound was detected in culture supernatants from Pseudomonas sp. LG2 grown with BSC(brewer's spent grain). FAE(ferulic acid esterase) and xylanase are induced from Pseudomonas sp. LG2 in the presence of carbon sources such as oat spelt xylan, HBSG I, II(hydrolyzed brewer's spent grain I, II) and AFBSG(autoclaved fraction from brewer's spent grain). However, xylanase and FAE are not induced by growth of Pseudomonas sp. LG2 on xylose and arabinose. Pseudomonas sp. LG2 is grown on medium containing oat spelt xylan, HBSG I, II and AFBSG and the induction of FAE and xylanase activities of extracellular proteins determined during 14 days. Maximum level of xylanase activity(5.3 U/mg) found at 6 days in culture contained oat spelt xylan as carbon source, whereas maximum level of FAE activity(15.4 mU/mg) was found at 8 days in culture contained AFBSG as carbon source. Most ferulic acid was released in culture supernatants when Pseudomonas sp. LG2 grown on oat spelt xylan, HBSG I, II and AFBSG. FAE of extracellular enzymes was also specific activity on methyl ferulic acid, methyl caffeic acid and methyl p-coumaric acid respectively, but not methyl sinapinic acid, methyl vanillic acid and methyl gallic acid.

The Effect of Leisure Activities on School Life Satisfaction in Teenagers -The Intermediation Effect of Happiness and Life Satisfaction-

  • This study analyzed the impact of leisure activities on school life satisfaction in the cultural rights and self-determination of teenagers who spend most of their time in school in the college entrance exam-oriented climate, and examined the effects of happiness and life satisfaction on the school life satisfaction through a process macro applying Bias Corrected Bootstrapping in these relationships. For research purposes, 2,590 cohorts in the first year of the 2018 Korea Children and Youth Panel Survey were analyzed through process macros. The result was that leisure life had a significant effect on school life satisfaction. It was also found that happiness and satisfaction in life affect satisfaction, and happiness and satisfaction in life affect satisfaction in school life and serve as a medium. This study confirmed that teenagers' participation in leisure activities greatly affects school life satisfaction and that happiness and life satisfaction have a mediating effect on school life satisfaction. In addition, we could recognize the importance of leisure activities to the satisfaction of school life of teenagers, and find it necessary to develop and provide various program graphs of schools and communities for happiness and satisfaction in life. This study verified that the participation of leisure activities of adolescents has a significant effect on school life satisfaction, and that happiness and life satisfaction have a mediating effect on school life satisfaction. In addition, we could recognize the importance of leisure activities to the satisfaction of school life of teenagers, and find it necessary to develop and provide various program graphs of schools and communities for happiness and satisfaction in life. Therefore, this research is believed to be possible to contribute to academic and practical programs for youth leisure activities and school life relationships, and to make policy suggestions for the development and participation of various leisure activities in order to fully enjoy and protect youth's rights to culture and self-determination of leisure activities.

Development of an Enrichment Culture Growing at Low Temperature used for Ensiling Rice Straw

  • To speed up the conversion of rice straw into feeds in a low-temperature region, a start culture used for ensiling rice straw at low temperature was selected by continuous enrichment cultivation. During the selection, the microbial source for enrichment was rice straw and soil from two places in Northeast China. Lab-scale rice straw fermentation at $10^{\circ}C$ verified, compared with the commercial inoculant, that the selected start culture lowered the pH of the fermented rice straw more rapidly and produced more lactic acid. The results from denatured gradient gel eletrophoresis showed that the selected start culture could colonize into the rice straw fermentation system. To analyze the composition of the culture, a 16S rRNA gene clone library was constructed. Sequencing results showed that the culture mainly consisted of two bacterial species. One (A) belonged to Lactobacillus and another (B) belonged to Leuconostoc. To make clear the roles of composition microbes in the fermented system, quantitative PCR was used. For species A, the DNA mass increased continuously until sixteen days of the fermentation, which occupied 65%. For species B, the DNA mass amounted to 5.5% at six days of the fermentation, which was the maximum relative value during the fermentation. To the authors' best knowledge, this is the first report on ensiling rice straw with a selected starter at low temperature and investigation of the fermented characteristics.

High Productivity of t-PA in CHO Cells Using Hypoxia Response Element

  • The dissolved oxygen level of any cell culture environment has a critical effect on cellular metabolism. Specifically, hypoxia condition decreases cell viability and recombinant protein productivity. In this work, to develop CHO cells producing recombinant protein with high productivity, mammalian expression vectors containing a human tissue-type plasminogen activator (t-PA) gene with hypoxia response element (HRE) were constructed and stably transfected into CHO cells. CHO/2HRE-t-PA cells produced 2-folds higher recombinant t-PA production than CHO/t-PA cells in a $Ba^{2+}-alginate$ immobilized culture, and 16.8-folds in a repeated batch culture. In a non-aerated batch culture of suspension-adapted cells, t-PA productivity of CHO/2HRE/t-PA cells was 4.2-folds higher than that of CHO/t-PA cells. Our results indicate that HRE is a useful tool for the enhancement of protein productivity in mammalian cell cultures.

The Effects of Quality of Life and Occupational Performance on Quality of Sleep in College Students

  • The purpose of this study is investigate the effects of quality of life and occupational performance on quality of sleep in college students. Data had been collected from Sep. 1 to Sep. 30, 2021. The subjects of this study were students majoring in occupational therapy at C College situated in C region. For analysis, 103 copies of questionnaire were used. As study instruments, a structured questionnaire incorporating questions about general characteristics, and measurement scales for quality of life, quality of sleep, and occupational performance evaluation were applied. Subjective sleep quality and sleep latency of study participants had lower scores than other components. According to the analysis on different quality of sleep depending on general characteristics, women had more sleep disturbances as they didn't have any part-time job. In terms of the correlation between quality of sleep and quality of life, physical health and total score (quality of life) had correlations with daytime dysfunction. With regard to influential factors on quality of sleep, rest & sleep as a domain of occupational performance and work affected quality of sleep. Students' quality of sleep will be improved if they keep regular habits of living, have rest appropriately for alleviating their stress, and do their jobs with interest and successfully according to a procedure. It is expected that the results of this study will be helpful to care for the health of would-be experts in charge of national health care.

Efficient Culture of Porcine Fetal Fibroblasts (돼지 태아 섬유아 세포의 효과적인 배양)

  • Culture method of somatic cells is one of the important factors in the production of transgenic pigs by somatic cell nuclear transfer. In this study, we established an efficient culture method of porcine fetal fibroblasts. Porcine fetal fibroblasts were isolated from 33-day-old fetuses. The proliferation of porcine fetal fibroblasts was analyzed by different serum types and culture media. The cultures in medium supplied 15% ES screened FBS showed faster increase in cell number than 15% FBS. Also, fetal fibroblasts have been propagated continuously for $7{\sim}8$ passages in ES modified DMEM and DMEM medium. We transfected $PGK-neo^r$ vector (pKJ2) into porcine fetal fibroblasts to estimate colony formation in this culture condition. The formation of colonies was confirmed in the medium containing $300\;{\mu}g/ml$ G418 at 12 day. These data show that this culture system can be used screening of porcine somatic cells transfected transgene.

University Students' Thoughts on Artifical Abortion

  • This study is a phenomenological qualitative study that confirms the structure of college students' thoughts on artificial abortion. The data collection period was from 5 March to 10 April 2022. To this end, a total of three interviews were conducted on seven college students aged 20 to 25. Interview data were conducted through analysis and interpretation using the phenomenological research method, the Giorgi method, and as a result, 40 semantic units were derived, grouped into six sub-components, and divided into three categories. As a result of the analysis, college students' thoughts on artificial abortion consisted of fetal rights, respect for women's rights, and choices for a healthy life. Based on the above meaning, college students' thoughts on artificial abortion were, in conclusion, that considering the happiness of the baby and the quality of life of the woman, consideration for non-marriage mothers was more urgent than legal sanctions, and that abortion was not irresponsible. Accordingly, this study suggests that understanding and consideration for pregnant women should be prioritized over legal sanctions.
