기내 환경에 따른 거베라 'Beauty' 배양묘 잎의 구조적 특성과 탄수화물 함량의 차이 (Structural Characteristics of Leaves and Carbohydrate Content of Propagules Grown at Different Culture Conditions in Gerbera hybrida 'Beauty')

  • 배양환경을 달리하여 생산한 3가지의 유묘 (타가영양묘, 혼합영양묘, 자가영양묘)를 대상으로 잎의 형태적 특성 및 탄수화물 함량 등을 비교 분석한 결과, 온실묘, 자가영양묘와 혼합영양묘는 하표피층에 왁스의 결정형이 관찰되었으나 타가영양묘에서도 관찰되지 않았다. 기공수와 크기에 있어서는 타가영양묘가 자가영양묘에 비하여 기공이 크고 많았다. 특히, 자가영양묘의 기공형태와 크기는 온실에서 자란 유묘와 거의 유사하였다. 식물체의 탄수화물함량은 자가영양묘가 혼합영양과 타가영양묘에 비하여 많았으며 유리당도 역시 자가영양묘가 가장 많았고 모든 배양묘에서 glucose의 함량이 가장 많았다.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

  • The weight average molecular weight of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) synthesized by Alcaligenes sp. SH-69 was altered between $3.2$\times$10^5$ and $1.1$\times$10^6$ depending upon various culture conditions. It appeared that culture conditions favorable for the efficient production of copolyesters promoted the formation of higher molecular weight copolyesters. Polydispersity indices of isolated copolyesters were in the range of 1.5 to 2.5.

  • Over the past decades, in vitro culture media have been developed to successfully support IVF embryo growth in a variety of species. Advanced reproductive technologies, such as somatic cell nuclear transfer (SCNT), challenge us with a new type of embryo, with special nutritional requirements and altered physiology under in vitro conditions. Numerous studies have successfully reconstructed cloned embryos of domestic animals for biomedical research and livestock production. However, studies evaluating suitable culture conditions for SCNT embryos in wildlife species are scarce (for both intra- and interspecies SCNT). Most of the existing studies derive from previous IVF work done in conventional domestic species. Extrapolation to non-domestic species presents significant challenges since we lack information on reproductive processes and embryo development in most wildlife species. Given the challenges in adapting culture media and conditions from IVF to SCNT embryos, developmental competence of SCNT embryos remains low. This review summarizes research efforts to tailor culture media to SCNT embryos and explore the different outcomes in diverse species. It will also consider how these culture media protocols have been extrapolated to wildlife species, most particularly using SCNT as a cutting-edge technical resource to assist in the preservation of endangered species.

청각, Codium fragile (Suringar) Hariot의 분리수사 재생에 의한 종묘생산과 가이식 조건 (Artificial Seed Production and Nursery Culture Conditions Using Regeneration of Isolated Utricles and Medullary Filaments of Codium fragile (Suringar) Hariot)

  • Codium fragile is commercially farmed in Korea by natural blooming zygote attachment. Experiments found optimum conditions for artificial seed production and nursery culture of C. fragile by asexual reproduction. Isolated utricles and medullary filaments were regenerated to erect thalli using both indoor and outdoor culture experiments. Under the indoor culture conditions, irradiance was an important factor to control the development of erect thalli. Formation of erect thallus from the isolated medullary filaments in the indoor culture was induced after 30 days under $20^{\circ}C$ and $60{\mu}mol/m^2/sec$. The detachment of isolated utricles and medullary filaments from the substrates of seed strings was reduced by exposure to the air during 2 hrs before the indoor culture of seed strings. The maximum growth and development of erect thalli in the nursery culture was induced at a water depth of 0.5 m. Depending on the substrates of the seed strings the growth of erect thalli was not significantly different (p>0.05).

왕겨 소각로 배연가스 이용을 위한 미세죠류 배양 조건 확립 (Microalgal Culture Conditions for Utilization of Flue Gas from Rice Husk Incinerator)

  • This study was performed to investigate the optimum microalgal culture conditions using flask culture and to find the feasibility of using the flue gas of the rice husk incinerator for cultivating the microalgae. The optimum initial pH of media was 4.5 for the microalgae culture, and the intermittently illuminated culture was more effective than the continuous illuminated culture. Thus, the balance between photosynthesis and formative metabolism must be considered thoroughly to cultivate microalgal cells. The optimum CO2 concentrations were in the range of 7 to 10%, and the optimum temperature was about 35$^{\circ}C$ in both the daytime and the nighttime for the culture. When flue gas of the rice husk incinerator was applied to the microalgae culture using stirred photobioreactor, the dry cell weight was 0.026 g dry biomass/hr$.$l. The results obtained in experiments indicated that the flue gas was effective for microalgae culture without any limitations.

가모만에서의 기초생산력 향상방안에 관한 생태계모델링 (The ecosystem modelling for enhancement of primary productivity in Kamak Bay)

  • From the environmental aspects, primary productivity of phytoplankton plays the most improtant role in enhancement of marine culture oyster production. This study may be divided into two branches; one is estimation of maximum oyster meat production per unit facility(Carrying Capacity) under the present enviromental conditions in Kamak Bay, the other is improvement of carrying from increase of primary productivity by changing the environmental conditions that cause not ot form an unfavorable environment such as the formation of oxygen deficient water mass using the eco-hydrodynamic model. By simulation of three-dimensional hydrdynamic model and ecosystem model, the comparison between observed and computed data showed good agreement. The results of sensitivity analysis showed that phytoplankton maximum growth rate was the most important parameter for phytoplankton and dissolved oxygen. The estimation of mean primary productivity of Wonpo, Kamak, Pyongsa, and Kunnae culture grounds in Kamak Bay during culturing period were 3.73gC/$m^2$/d, 2.12gC/$m^2$/d, 1.98gC/$m^2$/d, and 1.26gC/$m^2$/d, respectively. Under condition not ot form the oxygen deficient water mass, four times increasing of pollutants loading as much as the present loading from river increased mean primary productivity of whole culture grounds to 4.02gC/$m^2$/d. Sediment N, P fluxes that allowed for 35% increasing from the present conditions increased mean primary productivity of whole culture grounds to 3.65gC/$m^2$/d. Finally, ten times increasing of boundary loadings from the present conditions increased mean primary productivity of whole culture grounds to 3.95gC/$m^2$/d. The maximum oyster meat production per year and that of unit facility in actual oyster culture grounds under the present conditions were 6,929ton and 0.93ton, respectively. This 0.93ton/unit facility is considered to be the carrying capacity in study area, and if the primary productivity is increased by changing the environmental conditions, oyster production can be increased.

Optimazation of Submerged Culture Conditions for Exo-Biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica ws studied. Maltose, yeast extract, and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in a flask culture were pH 5.0, $25^{\circ}C$, and 150 rpm in a medium containing (as in g/l) 30 maltose, 6 yeast extruct, 2 polypeptone, $0.5{\;}K_3HPO_4,{\;}0.2{\;}KH_2PO_4,{\;}0.2{\;}MnSO_4{\cdot}5H_2O,{\;}0.2{\;}MgSO_4{\cdot}7H_2O$. Exo-biopolymer production and mycelial growth in the above suggested medium were significantly increased in a 2.5-1 jar fermentor, where the maximum biopolymer concentration was 8 g/l. The morphological changes of the mycelium in the submerged culture were observed within pH ranges from 4.0 to 9.0; i.e., growth of the filamentous form was optimal at culture pHs of 5.0 and 6.0, whereas pellet was formed at other pHs.

Improvement of Ergone Production from Mycelial Culture of Polyporus umbellatus

  • Ergone, a fungal metabolite derived from ergosterol, was previously isolated and identified from Polyporus umbellatus. Ergone is a major component of P. umbellatus known to have anti-aldosteronic diuretic effect and also displays cytotoxic activities. Most of mushroom's fruit bodies used for test contained less than 10 ${\mu}g/g$ of ergone. But P. umbellatus have larger amount of ergone than any other mushrooms. In order to improve the ergone production from the submerged culture of P. umbellatus, several factors including medium composition, culture conditions (temperature and pH) and different combinations of co-cultivation with various mycelia were studied. Among various carbon sources examined, starch proved to be most effective for the production of mycelia. The optimum pH and temperature for a flask culture of P. umbellatus mycelia were found to be 4.5 and $25^{\circ}C$, respectively. Under the optimized culture conditions, both the ergone production (86.9 ${\mu}g/g$) and mycelial growth (3.5 g/l) increased when P. umbellatus was cultured with Armillariella mellea. When the optimized conditions were applied, both mycelium and ergone production were significantly enhanced.

Culture Conditions for Mycelial Growth and Anti-Cancer Properties of Termitomyces

  • Termitomyces sp. that grow in symbiosis with fungus-farming Termites have medicinal properties. However, they are rare in nature, and their artificial culture is challenging. The expression of AXL receptor tyrosine kinase and immune checkpoint molecules favor the growth of cancer cells. The study evaluated the optimal conditions for the artificial culture of Termitomyces and their inhibitory activity on AXL and immune checkpoint molecules in lung adenocarcinoma and melanoma cell lines. The culture of 45 strains of Termitomyces was compared. Five strains with marked growth rates were selected. Four of the selected strains form a single cluster by sequence analysis. The mycelium of 4 selected strains produces more fungal mass in potato dextrose broth than in a mixed media. The bark was the most appropriate solid substrate for Termitomyces mycelia culture. The mycelium of all five selected strains showed a higher growth rate under normal CO2 conditions. The culture broth, methanol, and ethyl acetate of one selected strain (T-120) inhibited the mRNA relative expression of AXL receptor tyrosine kinase and immune checkpoint molecules in cancer cell lines. Overall, these results suggest the potential usefulness of Termitomyces extracts as a coadjuvant therapy in malignant diseases.
