The recycling method of enokitake cultural waste and the potentiality of second flush for enokitake were determined, because this fungus is not as prolific as the more commonly cultivated white rot fungi in the conversion of sawdust to mycelial mass. The mycelial growth of F. velutipes on several substrates, variously treated with rice bran was promoted at ratios of 10-20% (w/w) on all substrates, but suppressed at above ratios, although some difference was there. The mycelial densities generally increased correlated to the supplementation contents of rice bran. It could be concluded that F. velutipes preferred mild acidic to acidic conditions for mycelial growth, considering that the mycelial growth rate was highest on waste of pH 6.01, treated with 0.1 % Ca(OH)$_2$ and on populus mixed waste of pH 6.02, non treated. The ranges of substrate bulk densities, which was pertinent for mycelial linear growth were from B.D. (g/cc) 0.17 to 0.23 on waste and populus mixed waste all. The pertinent contents of rice bran supplementation in bottle cultivation was from 20 to 30% on waste and 20% on populus mixed waste, considering the requried duration for pinheading and fruiting yields. Standard bulk density for filling and utilizing the waste and populus mixed waste for commercial f. velutipes cultivation were B.D.(g/cc) 0.19 ~ 0.23, and 0.23~ 0.25, which could be conversed to 510~ 540g/900m1 and 520~ 570g/900m1, respectively, The second flush of F. velutipes was tried and the re-inoculation by sawdust and liquid spawn showed somewhat good results, indicating the potentiality of second crop and suggesting further research for it.
The effect of pH on degradation of paper by some fungi, which able to degrade cellulose, was investigated. Trichoderma koningii, Aspergillus nigerand Penicillium nigulosum were cultured at $28^{\circ}C$ for 16 days in the selective medium (PH3, PH4, PH5, PH6, PH7, PH8, PH9, PH10, PHC) containing paper as substrate. Each paper was pretreated with each pH buffer (pH 3∼pH 10, D.W.)prior to addition to the selective medium. Enzyme activities in the each culture medium were measured spectroph to metrically using C.M.C., Avicel, PNPG as the substrates for endoglucanase, exoglucanase and $\beta$-glucosidase, respectively. In all experimental fungi, the enzyme activities of PH3 and PH9 medium were usually much higher than those of other experimental groups. However in the PH6medium, enzyme activity was lower than other groups. To analyze the concentration and pattern of protein in the each culture medium, the medium was concentrated by lyophilization. The protein concentration of PH3 and PH9 medium were relatively high (T.koningii; 6.31mg, 6,19mg, A.niger; 1.62mg, 1.96mg, P.nigulosum;2.50mg, 2.73mg, respectively), but that of PH6 was relatively low. The protein pattern of each medium was analyzed by using SDS-PAGE and VDS Image Master Analysis Program. The concentrations of bands in the each lane were usually high at lane2 (PH3) and lane8 (PH9) and low at lane5 (PH6). Therefore, the incresed cellulolytic activity of fungus against acidified paper could be result of structural change and deterioration of paper caused by being acidified.
This study was carried out to investigate the physiological charateristics of F. pinicola in sawdust media. The optimum temperature in sawdust media was $30^{\circ}C$ in of F. pinicola. The optimum pH was 5 in F. pinicola. Mycelial growth and density of F. pinicola was quite good when birch tree and oak sawdust, respectively were used as cultural substrates. The best mycelial growth in F. pinicola was observed when beer waste was added as supplement on sawdust substrates. The optimum supplement ratios of beer waste and a magnecium sulfate were 20%, and 0.1% respectively. However, optimun supplement ratios of a calcium oxide and a LVD were different as 0.1% in F. pinicola.
Microorganisms capable of utilizing diaminododecane containing amine groups diterminally were isolated from the soil by enrichment culture. One strain of these isolated strain, designated as EL-0112L, was selected for this study. The results of this study were as follows. 1. This isolated strain EL-0112L was identified as Microbacterium, from the results of morphological, cultural, and biochemical tests. This isolated strain was named temporarily Microbacterium sp. EL-0112L for convenience. 2. Microbacterium sp. EL-0112L was tested for ability to utilize different kinds of substitued alkanes containing cyan, amine, chloro, and thiol groups(monoterminally or diterminall substituted) as carbon source. Pentamethylenediamine, hexamethylenediamine, n-decane, laurylamine, and alkane derivatives containing cyan, chloro, and thiol groups were not utilized by Microbacterium sp. EL-0112L. 3. The alkane derivatives that did not serve as growth substrates were tested further in oxidation tests using resting cell preparation of Microbacterium sp. EL-0112 L. Alkane derivatives containing cyan, chloro, thiol groups, and n-decane were oxidized by Microbacterium sp. EL-0112 L. It is possible that this isolated strain is also able to degrade their substituted counterparts since they are structually similar to diaminododecane. The remarkable substrates that were being oxidized were dichlorodecane, and 1-dodecanethiol. Microbacterium sp. EL- 0112L could not oxidize pentamethylenediamine, and hexamethylenediamine. 4. The metabolic products formed from diaminododecane by Microbacterium sp. EL-0112 L were acid compound containing carboxyl group and not containing amine group. On the thin layer chromatography, Rf values of these metabolic products were different from that of the product formed by Corynebacterium sp. EL-0112L. These results suggested the specificity of diaminododecane as carbon source.
This study was carried out to select the suitable substrates for bag cultivation of Lentinula edodes. We investigated the optimal additive materials and its mixing ratio in bag cultivation of L. edodes, Sanjo 701 ho. The suitable substrates for L. edodes bag cultivation were oak sawdust as new material plus deffatted corn flour, and corn husk as an additive at the ratio 8:1:1(v/v), as the result of shorter mycelial growth, higher biological efficient, and a higher yield than any other substrates.

A total of 179 isolates of Trichoderma spp. were collected from oyster mushroom substrates in Korea. On the basis of morphological and cultural characteristics, Trichoderma isolates were divided into seven groups, namely T. atroviride, T. citrinoviride, T. harzianum, T. longibrachiatum, T. virens, and two unidentified species, referred to as Trichoderma sp. 1 and 2. The predominant species was Trichoderma sp. 2 (n=86) followed by Trichoderma sp. 1 (n=52). Trichoderma sp. 1 and 2 were morphologically distinct not only from the other species of Trichoderma reported but also from each other in the characteristics such as mycelial growth rate, colony appearance, shape of conidia and conidiophores and branching pattern of phialides, although branching pattern of phialides of Trichoderma sp. 1 was similar to that of T. harzianum. In virulence test, the degree for compost colonization of Trichoderma sp. 2 was significantly greater than that of the other Trichoderma species. Trichoderma sp. 2 was found to be the main cause of green mold disease in oyster mushroom production. More work including molecular characterization is needed to confirm the species of Trichoderma sp. 1 and 2.

The metabolic behavior of Debaryomyces hansenii was investigated in terms of substrate utilization and by product formation under different cultural conditions. Debaryomyces hansenii exhibited best growth and most tolerant of increased NaCl, sucrose and potassium sorbate at their optimum pH (5.0). A combination of two or more environmental stresses had stronger inhibitory effects on their growth kinetics, utilization of carbohydrate substrates and the production of organic acids, volatile compounds and other metabolites. Significant amounts of glycerol (0.35-4.4 g/L) and arabitol (0.08-9.8 g/L) were produced by D. hansenii. The main organic acids produced were citric (0.6-1.4 g/L), acetic (0.3-2.8 g/L), fumaric (0.2-1.0 g/L) and malic acids (1.1-1.7 g/L). A range of other compounds such as ethyl acetate, n-propanol, isoamyl alcohol, 2-phenylethanol and acetoin were also produced. The concentration of these compounds varied with the cultural conditions. Such compounds would have specific impacts on food quality in which D. hansenii is found.

Laser cleaning have been found to be a useful cleaning tool to remove contaminants without inducing damage to the substrate and making secondary pollutant. In this study, the effect of Nd:YAG laser cleaning system, emitting at 1064 nm and 532 nm, on acrylic resin applied onto copper coupons and pieces of bronze was investigated. The samples after laser cleaning tests were examined using microscopy, FT-IR, SEM-EDS. As a result, the acrylic resin could be removed from most of the samples at low laser energy density. Laser wavelength 532 nm was more effective than 1064 nm because of using lower laser energy density, which could reduce heat damage to substrates. Although the acrylic resin was easily removed, it revealed melted surfaces and removed bronze patina which must remain. The problems should be solved by future studies using different laser system or laser wavelengths.

In the midst of increasing importance of modern cultural assets, especially, most modern bronze objects are exposed to outdoor environment, and as the objects are corroded steadily due to environmental factors the objects lost their original colors on the surface. We performed artificial patinas on the bronze sample per each color of red, black and green and checked cuprite and tenorite which are detected from actual bronze corrosion by analyzing the components. In addition, we applied the existing corrosion removal methods of grinder and sand blaster on a similar sample of bronze mirror per injection pressure and performed comparative analysis on the result with Nd:YAG laser. As a result of Nd:YAG laser cleaning artificial patina from bronze samples, all of the patinas were removed by laser wavelength 1064 nm better than 532 nm. Upon applying to a similar sample of bronze mirror, the artificial patina could be selectively removed from substrates without surface damage when Nd:YAG laser was conducted other than the existing removal method, and so it showed the possibility of application.
The standardization of test procedures and reproducibility of the toxicity data are prerequisite for the toxicity testing with the earthworm culturing in the laboratory. No in-depth study on culturing conditions of earthworms has been conducted in Korea, even of massive cultural practice is common for composting and production of biochemicals. The earthworms, Lumbricus rubellus and Eisenia foetida, were cultured in three kinds of artificial soil substrates(I, II and III) based on the OECD Guideline, which consist of different ratios of components (sand, sphagnum peat and kaolinite), and fed with a mixture of grain powders. During the period of culturing, the body weight and reproduction parameters were measured. L. rubellus showed the best results for increasing body weight and cocoon production in the artificial soil substrate(I) compared with E. foetida. The cocoon production was significantly high in both species cultured in the artificial soil substrate(I) among the three kinds of soil substrates, but the cumulative cocoon production of L. rubellus was 11 cocoon per worm compared with 3.7 cocoons per worm of E.foetida. L. rubellus, therefore, was more prolific than E. foetida in these culture schemes. The cumulative mortality in both species was less than 10%, and the number of juvenile worms per cocoon ranged from 1.5 to 2.3 and thus did not show any relationships with soil substrates or species. From these data, the culture of L. rubellus in the laboratory could be standardized, but for E. foetida, further study would be necessary to establish the optimal growth conditions in the laboratory.
