Changes of Antioxidant Activities on Cultured Ginseng with Mushroom Mycelia During Cultivation (배양기간에 따른 버섯균사체 인삼배양물의 항산화활성)

  • This study was conducted to investigate the antioxidant activities of the cultured ginseng with mushroom mycelia (Phellinus linteus (PL), Ganoderma lucidum (GL), and Hericium erinaceum (HE)) during cultivation periods of 10, 20, 30, 40, and 50 days. The lyophilized powder from the cultured ginseng with mycelia was extracted with 80% ethanol, and then evaluated for antioxidant activities. Total phenolic contents ranged from 149.63 to 205.91 mg/g, and the highest value was 80% EtOH extract from the cultured ginseng with GL at 30 days. The highest antioxidant activity ($IC_{50}$) for DPPH was 1.16 mg/mL in the cultured ginseng with HE at 40 days, and total antioxidant activity for ABTS was the highest value of 4.03 mg AA eq/g in PL cultivation at 30 days. $\alpha$-Glucosidase inhibitory activity was the highest value of 92.51% in EtOH extract from the cultured ginseng with PL at 50 days, and tyrosinase inhibitory activity was highest value of 13.21% in GL cultivation at 40 days. These results suggest that mushroom mycelium cultivation period for enhancement of antioxidant activity might be 40 days.

A Comparative Study on Hydrolase Activities in Acanthamoeba culbeytconi and A. roureba (Acanthamoebaculbertsoni와 A. royreba의 가수분해 효소 활성도의 비교 연구)

  • Specific or non-specific cytolytic processes of free-living amoebae causing meningoencephalitls have been emphasized and the cytolytic ability related to hydrolases in Entantoeba sp. and Naegleria sp. has also been reported since the latter half of 1970's. However, no information on hydrolase activities in Acanthamoeba sp. is available. Hydrolases in Acanthamoeba culbertsoni, a pathogenic species of free-living amoebae, were assayed and compared with those in a non-pathogenic species, A. royreba. Pathogenicity of these two species was confirmed through experimental infection to BALB/c mice. Hydrolase activities and cytotoxic effects between pathogenic and non.pathogenic species were compared in the trophozoites cultured in CGV media and in CHO cell line, respectively. The results are summarized as follows: 1. The mice infected with A. culbertseni were all dead 15 days after nasal inoculation, and the mean survival time was 8.5 days. Also the mice infected with this pathogenic species manifested typical meningoencephalitis, whereas the mice infected with A. royreba did not. 2. Hydrolases detected both in the cell extracts and culture media were acid phosphatase, ${\beta}-N-acetyl$ galactosaminidase, ${\beta}-N-acetyl$ glucosaminidase, ${\alpha}-mannosidase$, neutral proteinase and acid proteinase, all of which were detected with remarkably higher rate in A. culbertsoni than in A. royreba. 3. A. cuzbertsoni revealed strong cytotoxicity for the target CHO cells, whereas A. royreba did not show any specific cytotoxicity. About 80% of the target cells mixed with A. culbertsoni were dead 48 hours after cultivation, and more than 95% of the target cells were dead 72 hours after cultivation. 4. Hydrolase activities in A. culbertsoni cultured with the target cell line were assayed according to the culture time. The activities of acid phosphatase, ${\beta}-N-acetyl$ galactosaminidase, ${\beta}-N-acetyl$ glucosaminidase, ${\alpha}-mannosidase$ and acid proteinase in this pathogenic amoeba were detected higher in amoeba extracts than in culture media up to 120 hours after cultivation, but after 120 hours of cultivation those activities were detected higher in culture media than in the amoeba Iysates. Neutral proteinase activity in A. culbertsoni increased more in EBSS medium than in the Iysate specimens although the activity in the extracts was generally steady according to the cultivation time. Summarizing the above results, it is concluded that there were differences in hydrolase activities between Pathogenic A. culbertsoni and non-pathogenic A. royreba, and that some hydrolase activities were detected remarkably higher in A. culbertsoni which revealed strong cytotoxicity to the target CHO cell line.

  • The optimum conditions of the production of exo-polymer by Ganoderma lucidum WK-003 and it's hepatoprotective effect was studied. Optimum conditions for the production of exo-polymer (3.18 g/$\ell$) by using shaken flask culture of G. lucidum WK-003 were pH 4.5, 30$^{\circ}C$, 120 rpm for 18 days cultivation. Also exo-polymer productio (7.15 g/$\ell$) was optimized by 5$\ell$ jar fermenter cultivation with condition of pH 4.5, 30$^{\circ}C$, 200 rpm, 1.0 vvm for 6 days cultivation. Glutamic pyruvic transaminase(GPT) activities in serum of intoxicated Sprague-Dawley rat were decreased from 704 IU/L to 330 IU/L by oral administration of the exo-polymer (20mg/kg$.$day)f or 4 consecutive days.

  • Effect of allylisothiocyanate on the enzyme activites including malate degydrogenase, isocitrate dehydrogenase, NADPH and acetyl CoA which were related to aflatoxin production of Aspergillus parasticus R-716 were invetigated. The activities of malate dehydrogenase (EC.1.1.1.37), isocitrate dehydrogenase (E.C.1.1.1.42) and NADPH oxidase (E.C.1.6.99.1) indicated relatively high in the 50 ppm allylisothiocyanate-added-culture. In contrast, the activity of acetyl CoA in the 50 ppm allylisothiocyanate-added-culture showed rather lower level through the cultivation.

Activities of Antioxidative Enzymes in Photobleaching of Leaves from Panax ginseng C. A. Meyer (인삼엽의 Photobleaching과 항산화효소 활성)

  • This study investigated the relations between the photosynthetic rate and the activities of antioxidatile enzymes, glutathione reductase, ascorbate free radical reductase, ascorbate peroxidase, glutathione peroxidase, and ascorbate oxidase, in the leaves of Panax ginseng. Under the normal cultivation condition, Panax in showed lower g1utathione reductase and ascorbate free radical reductase activities the Glycine max. But P ginseng showed higher 91utathione Peroxidase, ascorbate Peroxidase, and ascorbate oxidase activities than C. Panax. Therefore, P. ginseng showed weak activities of reductases for the reduction of the oxidized antioxidants. Under the light intensity of 25 KLux, the reductases showed a decrease of over 75% after 24 hours. But the peroyoxidases decreased about 40%. These results showed that the decrease of reductases acitivities was consistent with the decrease of photosynthetic rate. Therefore, we consider that the regulation of antioxidative enzymes or the application of exogenous antioxidants will be effective means for the protection of photodamage in p. ginseng.

The Growth of Ginseng Industry and the Activities of Ginseng Cultivators in the 1930s: Focusing on Non-Government-Contract Cultivation Areas (1930년대 인삼업의 성장과 삼포민의 활동 -특별경작구역 이외 지역을 중심으로-)

  • This article focuses on changes in the ginseng industry in the 1930s in areas other than the Government Contract Cultivation (GCC) zones. A major characteristic of the ginseng industry in the 1930s was the rapid increase in the area covered by ginseng gardens: the area was about 212 ha in 1929 and 252 ha in 1930 and 1931 but soared to around 441 ha in 1938. This occurred because the non-GCC areas increased significantly during this period. Until the early 1930s, the ratio of GCC to non-GCC areas was 70:30. By the late 1930s, however, the ratio had changed to 53:47. The reason for this change was that the area of the newly established ginseng gardens in the GCC zones had decreased, while that of the non-GCC ginseng gardens had steadily increased. Due to the Japanese invasion of China, China boycotted red ginseng, and exports were sluggish, so the GCC areas were reduced. On the other hand, the non-GCC ginseng gardens were not affected, and the area they covered steadily increased. As a result, in the 1930s, the ginseng industry outside of the GCC areas grew rapidly. The region that led the growth of the ginseng industry outside of the GCC zone was Jeonbuk. By the late 1930s, Jeonbuk dominated the other provinces and accounted for more than 50% of the non-GCC farming zone. Gyeongbuk and Gangwon-do followed Jeonbuk in terms of ginseng cultivation areas. While Gyeonggi-do, Gyeongnam, and Chungbuk were also active in ginseng cultivation, Jeonnam and Chungnam were not active. In the 1930s, the growth of the ginseng industry outside of the GCC zones was driven by the efforts of ginseng farmers and the support of local governments. An examination of Yecheon-gun in Gyeongbuk, Ganghwa-gun in Gyeonggi, and Jecheon-gun in Chungcheongbuk-do showed that ginseng farmers organized cooperatives as the ginseng industry steadily developed in these regions, and these cooperatives worked systematically to cultivate and sell ginseng. In the case of Ganghwa-gun, activities were carried out to incorporate the GCC zone. The Deoksan Ginseng Association in Jecheon-gun determined that financing for cultivation was key and requested subsidies from the provincial government. Administrative authorities also supported the activities of the ginseng farmers. The activities of the farmers and the support of the administrative authorities together led to the growth of the ginseng industry during this period.

Studies on Immunomodulatory and Antioxidant Activities of Astragali membranacei Radix according to the Cultivated Years (황기(黃芪)의 재배 년수에 따른 면역 및 항산화 활성 연구)

  • Purpose: Contents of astragaloside I, II and IV, cytotoxicity, anticancer activity, immunomodulatory activity and antioxidant capacity were to be compared as a function of the cultivated years as one, three, five and seven years. Method: Major components of Astragali membranacei Radix were separated as astragaloside I, astragaloside II, astragaloside IV by HPLC analysis. Cytotoxicity and anticancer activities were measured by MTT and SRB assay. For immunomodulatory activity, the secretion of IL -6 and $TNF-{\alpha}$, NK cell activation and macrophage activation were observed as well as kinetics of responding to human T cells by a microphysiometer. In vitro antioxidant activities were measured by several radical scavenging activities of superoxide anion radican, DPPH, LDL and linoleic acid. For in vivo activity, the activation of SOD, GSH-px, catalase, ALDH and ADH was measured as well the relative weight of liver. Result : 1. For HPLC analysis, the contents of all of astragaloside I, astragaloside II, astragaloside IV were in order of three, five, one and seven years. 2. The cytotoxicity of normal human lung cell line, HEL299 showed lower than 18% in adding 0.25 mg/ml, and 28.9% in adding 1.0 mg/ml of water extract of seven year root. For methanol extracts, three year root showed highest cytotoxicity as 35.2 % and there was no difference between the cultivated years. 3. For anticancer activities, methanol extracts of one and three year roots showed relatively high inhibition of human stomach cancer cells, AGS, breast cancer cells, MCF-7, lung cancer cells, A549 and liver cancer cell, Hep3B as well as high selectivities. 4. The water extract of seven year root could yield high secretion of IL-6 from both human Band T cells while the methanol extracts of three and five year roots secreted high amounts of IL-6 and $TNF-{\alpha}$ from both Band T cells. 5. As a result of in vitro antioxidant activities, both water and methanol extracts from five and seven year roots showed high activities for superoxide anion radical scavenging activity, inhibiting linoleic acid peroxide and contents of total phenols. 6. For in vivo tests, Mn-SOD and GSH-px activities and weight of liver were better in adding seven year root. For ALDH activity one year root was better and for ADH activity five year root. Overall speaking, seven year root showed relatively better antioxidant activities. Conclusion:There was difference of the contents of astragaloside I, astragaloside II, astragaloside IV according to cultivation year. Methanol extract showed better activities of anticancer and immune activation rather than water extract Interestingly enough, for methanol extracts, overall activities were improved as the cultivation year increased. There might be further investigation required for the clinical uses of the results as several biological activities varied according to the cultivated year of Astragali membranacei Radix.

Successful start-up of pilot-scale single-stage ANAMMOX reactor through cultivation of ammonia oxidizing and ANAMMOX bacteria (암모니아 산화균 및 아나목스균의 배양을 통한 파일롯 규모 단일 아나목스 반응기의 성공적인 시운전)

  • The lack of seed sludges for Ammonium Oxidizing Bacteria (AOB) and slow-growing ANaerobic AMMonium OXidation (ANAMMOX) bacteria is one of the major problem for large-scale application. In this study, $24m^3$ of single-stage SBR (Sequencing Batch Reactor) was operated to remove nitrogen from reject water using AOB and ANAMMOX bacteria cultivated from activated sludge in the field. The ANAMMOX activity was found after 44 days of cultivation in the ANAMMOX cultivation reactor, and then $0.66kg\;N/m^3/d$ of the nitrogen removal rate was achieved at $0.78kg\;N/m^3/d$ of the nitrogen loading rate at 153 days of cultivation. The AOB cultivation reactor showed $0.2kg\;N/m^3/d$ of nitrite production rate at $0.4kg\;N/m^3/d$ of nitrogen loading rate after 36 days of operation. The cultivated ANAMMOX bacteria and AOB was mixed into the single-stage SBR. The feed distribution was applied to remove total nitrogen stably in the single-stage SBR. The nitrogen removal rate in the single-stage SBR was gradually enhanced with an increase of specific activities of both AOB and ANAMMOX bacteria by showing $0.49kg\;N/m^3/d$ of the nitrogen removal rate at $0.56kg\;N/m^3/d$ of the nitrogen loading rate at 54 days of operation.

Changes of Antioxidant Activity as affected by cultivation period in Buckwheat (Fagopyrum species) Sprouts (재배기간에 따른 쓴메밀(Fagopyrum tataricum Gaertner)싹의 항산화 활성 및 생리활성 평가)

  • Buckwheat (Fagopyrum species) has long been an excellent functional food. Besides, buckwheat sprouts contain various functional substances. In this study, we investigated the antioxidant activity of buckwheat sprouts in the context of cultivars harvested after different cultivation periods (0, 3, 5, 7, 9, 13, and 15 days after planting). Buckwheat sprouts were cultivated at 25℃ for up to 15 days and then extracted with ethanol. Antioxidant components were then extracted from sprouts and leaves using a freeze dryer. The total polyphenolic content, flavonoid content, and antioxidant activity were then analyzed. The total polyphenol content increased from 32.26 mg GA eq/100 g for raw buckwheat to 114.75 mg GA eq/100 g after 7 days of cultivation. Also, the flavonoid content increased from 20.61 mg catechin eq/100 g (0 days) to 56.54 mg/g after 9 days of cultivation. The DPPH radical scavenging activity (concentration of extract at 0.25 mg/mL) increased from 7.89% at day 0 to 53.48% after 9 days of cultivation. Additionally, the ABTS radical scavenging activity increased from 10.26% at day 0 to 32.89% after 7 days of cultivation; of note, the activity decreased afterward. These results suggest that the best buckwheat sprouts with higher biological activities are those cultivated for 7-9 days. For a complete understanding of the potential of buckwheat sprouts as functional foods, we plan to further analyze their antioxidant activity in the future.

Change and Continuity in Traditional Timugon Rice Cultivation Beliefs and Practices

  • Before the start of the North Borneo Company administration in North Borneo (now Sabah, Malaysia) in 1882, the Timugon Murut of today's interior Tenom District lived in longhouses, and practiced head-hunting during wars with other Murutic ethnic groups. Their economy revolved around swidden agriculture of hill rice, sago, and cassava. Wet rice cultivation and water buffaloes were introduced just before 1885. Wet rice was planted on the alluvial plains around the Pegalan and Padas Rivers, while dry rice was planted on hillside swiddens that had been cleared by slash-and-burn methods. Today, wet rice cultivation and cash-cropping on the plains are the main Timugon socioeconomic activities, while some families also plant dry rice on the hills as a back-up. The Timugon believe that the physical world is surrounded by the spiritual world, and everything was made by the creator Aki Kapuuno'. The focus of this field research paper is on the beliefs and ritual practices of the Timugon connected to their traditional rice agriculture. This study found that for generations, the Timugon believed that since animals were created by Aki Kapuuno' for the wellbeing of humans, various types of animals and birds convey omens to guide people. Thus, the older Timugon rice cultivation is strongly influenced by good and bad omens and taboos, and also involves symbolic practices and ritual offerings to guardian spirits of the rice. After the 1930s and especially since the 1960s, most Timugon became Roman Catholic Christians. Hence, this paper also examines changes in the traditional Timugon rice cultivation related beliefs and practices due to religious conversion and other factors.
