Protein Patterns of Rice(Oryza sativa L.) Cultivars as Affected by Herbicide Thiobencarb (제초제 Thiobencarb 처리에 의한 수도품종간 단백질 유형의 변화)

  • This study was conducted to evaluate the susceptibility of rice(Oryza sativa L.) cultivars to herbicide thiobencarb through the determination of protein patterns using SDS-PAGE. In both greenhouse and laboratory screening tests, IR 10198-66-2 and IR 9660-50-3-1 showed relatively tolerant response to thiobencarb, while IR 22, IR 31802-48-2-2-2 and IR 20656-R-R-R-6-1 were susceptible to it. The total protein content of susceptible cultivars markedly decreased as the thiobencarb concentrations increased, but tolerant cultivars such as IR 10198-66-2 and IR 9660-50-3-1 showed very slight changes suggesting that protein synthesis of susceptible cultivars may be inhibited by thiobencarb. The protein profiles of the tolerant cultivars were not much affected by thiobencarb treatment. However, the protein spots with molecular weights of 14.4 kD and 55 kD in the susceptible cultivar of IR 22 disappeared with the treatment of 3 ppm thiobencarb, indicating that differential susceptibilities of rice cultivars against thiobencarb can be attributed to their difference in protein metabolism affected by thiobencarb.

The Influence of Pre- and Post-harvest Factors on the Shelf-life and Quality of Leaf Lettuce (수확 전후 요인이 잎상추의 저장수명 및 품질에 미치는 영향)

  • Weight-loss, general-appearante, yellowing, and compositions such as chlorophyll, carotenoid, vitamin C and nitrate, were studied to investigate the influence of pre-(cultivar and growing season) and post-(low-temperature and high-density polyethylene film packaging) harvest factors on the shelf-life and quality of leaf lettuce(Lactuca sativa L). Weight-loss was reduced by the treatment of high-density polyethylene film packaging, and more at $0{\sim}1^{\circ}C$ than $4{\sim}5^{\circ}C$. Leaf-lettuce grown in Winter lost less fresh weight than that grown in Autumn. In experiment on difference between cultivars, the reduction of weight-loss in 'Cheongchima' lettuce was somewhat significant than that in 'Jeokchukmyeon' lettuce. The tendency of changes in general appearance was similar to that of weight-loss. The yellowing of leaf lettuce during the storage was caused by chlorophyll degradation. Vitamin C content before the storage was 128.0 mg/100 g FW. in lettuce grown in Winter, 123.8 mg/100g FW. in that grown in Autumn, and more 'Cheongchima' lettuce than 'Jeokchukmyeon' lettuce. In all treatment, vitamin C content decreased rapidly, and this tendency was more obvious at $4{\sim}5^{\circ}C$ than $0{\sim}1^{\circ}C$. The nitrate content of 'Jeokchukmyeon' was 3812.6 ppm and that of 'Cheongchima' was 2855.6 ppm before the storage. Nitrate content decreased more at $4{\sim}5^{\circ}C$ than $0{\sim}1^{\circ}C$ during the storage.

  • This study investigated the changes in the nutraceutical lipid components of brown rice oil after germination. Four different high-yielding cultivars (Dasan1, Segyejinmi, Hanareum1 and Hanareum2) of Korean brown rice were selected and brown rice oil was extracted from each cultivar before and after germination. Free fatty acid, squalene, policosanols, and isomers of phytosterol were analyzed using GC, and isomers of tocols (tocopherol and tocotrienol) and ${\gamma}$-oryzanol were quantified using HPLC from both brown rice oil (BRO) and germinated brown rice oil (GBRO). The contents of phytosterol isomers, campesterol, stigmasterol, and ${\beta}$-sitosterol were increased by 8.3%, 31.6%, 3.3% in GBRO, respectively. Furthermore, the squalene content showed the highest increase of up to 2.4 fold in GBRO compared to BRO. In addition, linoleic and linolenic acid composition increased whereas oleic and palmitic acid decreased in the GRBO. However, the contents of tocols (tocopherol and tocotrienol) in GBRO were lower than those in BRO, and there was no significant difference in policosanol and ${\gamma}$-oryzanol between GBRO and BRO. These results suggest that GBRO has the potential as a healthy and functional source due to its lipid profile on improved lipid metabolism.

Comparison of General Ingredients, Chromaticity and Hardness according to Kernel Type of Korean Maize (국산 옥수수 배유특성에 따른 일반성분, 색도 및 경도 비교)

  • This study was conducted to secure basic information for corn processing by comparing the quality characteristics according to maize cultivars and kernel types (dent, intermediate, flint-like). As a result of analyzing 15 cultivars, a range of measurements were observed: 100-kernel weight, 22.89~35.63 g; moisture, 7.57~8.42%; crude protein, 8.46~11.45%; crude lipids, 3.26~4.83%; Hunter's L-value, 83.70~86.79; a-value, 2.61~5.49; b-value, 22.01~28.15; and total carotenoids, 6.74~17.07 ㎍/g. Significance among the cultivars was shown in all quality characteristics (p<0.001), but the significance among the kernel types was found only in crude protein (p<0.005), crude fat (p<0.001), and Hunter's L-value (p<0.05). The hardness of maize was decreased proportionally to the soaking time for all maize cultivars (p<0.001). In particular, with the same soaking time for different kernel types, the hardness difference was shown in the order of flint-like > dent ≒ intermediate. It was confirmed that the decrease in the hardness of flint-like kernel of close to that of hard-type starch was slowed compare dent and intermediate kernels. So it is expected the some characteristic of kernel types will contribute to the appropriate customized use of the developed cultivars.

Difference of Hypocotyl and Growth Characters of Soybean-sprouts by Sowing Date in Sprout-soybean Cultivar (나물용 콩 파종기에 따른 콩나물의 하배축 및 생장특성 차이)

  • This study was conducted to investigate the effects of sowing date on soybean-sprout growth characters and for improving cultural techniques to produce high quality soybean-sprout. Six sprout-soybean cultivars were planted on three sowing dates in 2000 and 2001. The growth of hypocotyl in soybean sprouts was accelerated at higher temperature and the differences among cultivars became greater at later culture days. The longest of hypocotyl and root lengths of soybean sprouts was obtained from July 5 sowing, followed by May 25 and June 15 sowing. Doremikong and Hannamkong had the thickest and the thinness hypocotyls, respectively. Total length of soybean-sprouts was longer in seeds harvested from later sowing, respectively. Eunhakong produced the longest soybean-sprouts, while Tawonkong produced the shortest. Percentage of rotten seed and imperfectly germinated seed harvested at later sowing was lower. Somyeongkong and Tawonkong showed the highest and the lowest percentages of rotten seeds, respectively, while Pungsannamulkong and Hannamkong showed the lowest and the highest percentages of imperfectly germinated seeds. The soybean-sprouts yields were higher in seeds harvested in 2001 and later sowing. Although Eunhakong and Doremikong showed higher soybean-sprout yield, Pungsannamulkong and Somyeongkong showed stable soybean-sprout yield regardless of the sowing dates.

Response of Antioxidative Enzymes of Two Rice Cultivars to Ozone Exposure and Nutrient Supply

  • Ozone ($O_3$)-induced changes in chlorophyll content and specific activities of antioxidant enzymes, such as superoxide dismutase (SOD), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) were investigated in two rice cultivars (Oryza sativa L.) grown under variable nutrient treatments. For this study, two rice cultivars of Ilpumbyeo (IL) and Keumobyeo#l (KM), which were known as resistant and susceptible to $O_3$, respectively, were exposed to $O_3$at 0.15ppm for 30 days and investigated with 10 days interval. The available nutrient regimes were varied by doubling the supply of nitrogen (N), phosphorus (P) and potassium (K) Within a basic fertilizer status (N, P, K; 15, 12, 12kg/l0a$^{-1}$ ). In both cultivars and at all nutrient status, chlorophyll content in $O_3$-treated plants decreased with prolonged treatment period, although higher N, P and K supply with $O_3$ treatment alleviated the decrease in chlorophyll content. The activities of almost all enzymes investigated for this study were decreased during initial stages of $O_3$- exposure except GPX which maintained higher activity throughout the exposure period than the non-treated plant. However, the antioxidant enzymes in $O_3$-treated plants showed almost the same or higher activities on 30 days after $O_3$ - exposure. The most significant changes in activities were observed in GR of the $O_3$-treated leaves. With the prolonged treatment period, the activity of GR at 30 days was increased by 3-8 times compared to those in 10 days. Most of the investigated enzymes showed very similar tendency to $O_3$ treatment in all fertilizer status. There was no observed evidence for enhanced detoxification of $O_3$-derived activated oxygen species in plants grown under higher fertilizer status compared with that in plants grown under basic fertilizer status. The increase in the activities of SOD, APX and GR in rice leaves by relatively long-term treatment with $O_3$ at low concentration is considered to indicate that the plant became adapted to the $O_3$ stress and the protection system increased its capacity to scavenge toxic oxygen species. Our results in two rice cultivars indicated that there was little difference in the activities of antioxidant enzymes between IL and KM, which were known as resistant and susceptible cultivar to $O_3$

Study on the morphological Characteristics of Local Cultivars in Paeonia spp. (지방 수집 작약의 유망계통에 대한 형태학적 특성에 관한 연구)

  • The major morphological characteristics of 97 lines selected from 1,510 local collection lines of Paeonia spp. were studied and classified into some groups according to their Q correlation. The Coefficient of Variation of 8 characteristics in 97 lines varied greatly with the range from 21.3% to 127.1%. The 22 lines with useful morphological characteristics selected from 97 lines had 2n=10 chromosomes, in which there were no difference among the number of chromosomes of 22 lines. The 97 lines were classified into 14 groups based upon the results of Q correlation analysis obtained from data with 16 quantitative factors; Group III and group VI were divided into two sub-groups. Group I was large flowered lines with a single flower, Group IIIa, doubled flowered lines, Group VII, late maturing lines and Group IX, early maturing lines flowering in 17th May. Group VIII were dwarf type lines which has the shortest stem length and leaf length among 14 groups, which are suitable for potted plants. Group XIII had a lot of stems per plant and a large type leaf with leaf length and width. A needle-type leaf was found in Group X, whereas a wide and oval-type leaf in group XII.

Effects of Planting Densities of Two Growth Type of Soybean Cultivars on the Vegetative Growth and Yield after Barley (맥후작(麥後作)에 있어서 신육형(伸育型)이 다른 대두(大豆)의 재식밀도(栽植密度)가 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • Two soybean cultivars different in plant type, Hwanggeumkong and Suweon-125, were planted under two different planting densities to investigate the proper planting densities after barley crops. The results obtained are summarized as follow: 1. No apparent varietal differences due to the planting densities were observed in flowering time and maturity. 2. The number of nodes of main stem showed significant differences between plant types, while they were not significantly different between planting densities. The number and length of branches per plant were apparently decreased as the planting densities were higher. 3. The number of pods per plant, and number and weight of grain per plant decreased as the planting densities increased and they showed also significant differences between planting densities. However, the number of pods, and number and weight of grain per unit land area were increased as the planting densities increased. 4. The grain yield per 10a showed significant cultivar difference under different planting densities. The Hwanggeumkong, determinate type, showed highest grain yield at the planting densities of $60{\times}7.5cm$, while Suweon-125, indeterminate type, highest at the planting densities of $60{\times}5.0cm$.

Effect of Variety and Shading Material on Growth Characteristics and Ginsenoside Contents of 2-Year-Old Ginseng (Panax ginseng C. A. Meyer) Grown in Imperfectly Drained Paddy Soil (배수약간불량 논토양에서 품종 및 해가림 피복물 종류가 2년생 인삼의 생육과 진세노사이드 함량에 미치는 영향)

  • To selection of optimal shading material, two-year-old ginseng (Panax ginseng C. A. Meyer) of new cultivar, 'Cheonpoong' (CP), and native species 'Hwangsookjong' (HS) were cultured under three kinds of shading materials such as three-layered blue and a one-layered black PE (polyethylene) net (TBSB), blue PE sheet (BS), and aluminium coated PE sheet (AS) in imperfectly drained paddy soil. Growth characteristics, yield and ginsenoside contents were investigated under three shading materials. Yield and ginsenoside contents of ginseng were distinctly affected by intensity and quality of sunlight penetrated through shading materials. Light transmission ratio, air and soil temperature according to shading materials were higher in order of BS, AS, and TBSB. However, ratio of aerial phase and porosity of the soil were higher in order of AS, BS, and TBSB, respectively. There was no significantly difference in the ratio of rusty colored root by shading materials. CP showed higher stem length, leaf area, and root weight than that of HS, while the former showed distinctly lower discolored leaf ratio than that the other. Eight kinds of ginsenosides content of CP were higher than that of HS in $Rg_1$, Re, Rf, $Rb_1$ and Rc except $Rg_2$, $Rb_2$, and $Rb_3$. Total ginsenoside contents of CP was distinctly higher than that of HS. Total ginsenoside contents as affected by shading materials was higher in order of BS, TBSB, and AS in CP, while TBSB, BS, and AS in HS.

Analysis of Aroma Pattern of Panax Species by Potable Handheld Gas Chromatograph (Potable handheld gas chromatograph(PHGC)를 이용한 인삼속(Panax species) 식물들의 향기패턴 분석)

  • This study was performed to analyze aroma pattern of Panax species (Korean Panax ginseng C.A. Meyer, Chinese Panax ginseng C.A. Meyer, Panax quinquefolium L, and Panax notoginseng F.H. Chen) by the PHGC (potable handheld gas chromatograph). Ratios of several peak areas in chromatogram of derivative parrtern were as follows. If ratio of Korean Panax ginseng was 1, Panax notoginseng was $0.030{\sim}0.674$, Chinese Panax ginseng was $0.005{\sim}0.212$ and panax quinquefolium was $0.241{\sim}0.871$. Ratios of peak area at $Rt_{20.02}$ were that if Korean panax ginseng was 1, Chinese Panax ginseng was 0.212, Panax quinquefolium was 0.343 and Panax notoginseng was 0.065. Ratios also of peak area at $Rt_{21.70}\;and\;Rt_{24.90}$ showed clear difference among aroma patterns of Panax specie cultivars. Flavor component at $Rt_{26.15}$ was not detected in Panax quinquefolium and Panax notoginseng but in Korean Panax ginseng and Chinese Panax ginseng. Ratios of peak area at $Rt_{26.15}$ were that if Korean Panax ginseng was 1, Chinese Panax ginseng was 0.185. And so habitat of Panax species cultivars was discriminated. Cultivar and habitat of dried panax species was remarkably distinguised by the chromatogram of frequency pattern, derivative pattern and visual pattern using olfactory images known as Vapor $print^{TM}$.
