홍해삼 추출물의 멜라닌 형성 억제를 통한 미백효과 및 피부 재생효과에 관한 연구 (Whitening Effect and Skin Regeneration Effect of Red Sea Cucumber Extract)

  • 홍해삼 또는 Apostichopus japonicas는 동남아시아에서 발견되는 stichopodiae의 한 종이다. 본 연구에서는 홍해삼의 화장품 소재로서 사용가능한지 알아보기 위해 홍해삼 추출물의 미백, 항주름에 관한 실험을 진행하였다. tyrosinase 활성 분석 결과, 홍해삼 추출물 $200{\mu}g/ml$에서 tyrosinase 활성을 억제하였다. 또한, 홍해삼은 tyrosinase, TRP-1, TRP-2, MITF 및 matrix metalloproteinase (MMPs)의 mRNA 발현을 억제하였다. 이어서 HaCaT과 human Fibroblast를 이용한 3차원 세포배양을 통해 홍해삼의 피부 턴오버 주기 개선효과를 검증하였다. 이러한 결과를 바탕으로 홍해삼이 높은 미백효과 및 주름개선효과를 가지는 화장품 소재로서 충분한 가치를 지닐 것으로 판단된다.

Antioxidant Properties of Water and Aqueous Ethanol Extracts and Their Crude Saponin Fractions from a Far-eastern Sea Cucumber, Stichopus japonicus

  • Water and 70% ethanol extracts obtained from a sea cucumber (Stichopus japonicus) body wall by heat reflux or pressurized solvent extraction showed 2,2-diphenyl-1-picrylhydrazyl ($DPPH^{\cdot}$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation ($ABTS^{{\cdot}+}$) scavenging activities comparable to those of fruits and vegetables. The highest activities were observed for the water extract from heat reflux extraction. Crude saponins exhibited higher radical scavenging activities than the soluble matters in the extracts. However, they were responsible for only about 3 to 15% of the scavenging activities of the extracts. Total phenolic contents showed a significant correlation with $DPPH^{\cdot}$ scavenging activities, suggesting a significant contribution of phenolic constituents to the antioxidant properties of the extracts. However, total flavonoid contents showed little correlation with the radical scavenging activities. The results suggest that the water or 70% ethanol extract obtained from sea cucumber body wall by simple heat reflux extraction could provide considerable antioxidant benefits.

Preventive effects of sea cucumber (Apostichopus japonicus) ethanol extract on palmitate-induced vascular injury in vivo

  • Cardiovascular diseases (CVDs) have posed serious public health problems, accounting for nearly 30% of mortality worldwide and their incidence is still increasing. Therefore, new treatment resources are necessary to prevent or manage the ever-increasing population of patients with CVDs. Sea cucumber is well known for its medical and health benefit effects, but it is not well known what/how effect it has on vascular disease. In the present study, we examined the protect effect of sea cucumber, Apostichopus japonicus 80% ethanol extract (AJE) on zebrafish embryo with the stimulation of free fatty acid, palmitate (PA). In vivo study showed that AJE can attenuate PA-induced toxicity through relieving the rapid heartbeat, increasing the survival rate and reducing the malformation in both wild type and Tg (fli1a:eGFP) transgenic zebrafish lines. Additionally, compare with PA treated embryos, the yolk sac area, body length, axial vascular segment (AVS) and intersegmental vessel (ISV) of the co-treatment group of AJE and PA were comparable to the control group. Moreover, AJE lowered the expression of inducible nitric oxide synthase (iNOS), nitric oxide (NO) and inflammation-related genes induced by PA, and inhibited PA-induced vascular development disorders. Our data preliminarily verify that AJE could be a candidate resource for the prevention or therapy of CVDs.

다기능 해양소재로서 홍해삼의 효능과 지역별 성분 비교를 통한 활용 가능성 연구 고찰 (Review on the Feasibility of Using Red Sea Cucumber (Stichopus japonicus) as a Multi-functional Marine Biomaterial and Comparing the Biochemical Components Collected from Different Regions in Korea)

  • This study aims to review the progress of biotechnological applications of sea cucumber and presents fundamental validation data on a range of methods and applications to determine the most promising fields for future exploitation and application. Sea cucumbers from different regions, subjected to numerous industrially applicable drying methods, were evaluated for their biochemical composition. The study highlights the nutritional excellence of Jeju red sea cucumber, particularly the high amino acid and fatty acid composition of its intestinal organs and the possibility of the industrial utilization of these organs. In addition, an efficient extraction method to extract collagen and chondroitin sulfate from red sea cucumber was established. The extracted collagen and chondroitin sulfate can be used as commercially valid biomaterials. Accordingly, the red sea cucumber was confirmed as a valuable source of raw food material of varied functionality and bioactivity. Upon conducting an analysis of patents thus far relating to the sea cucumber, it was found that the associated technology was limited to some parts of primary and secondary industries. Therefore, there is a clear need for the strategic development of technologies to produce specialized functional products from sea cucumber's raw materials, with a view to promote other related industries.

대황 및 황련 추출물을 처리한 딸기 및 오이의 저장중 품질 변화 (Changes in the Keeping Quality of Strawberry and Cucumber treated with Korean Medical Herb Extracts)

  • Strawberry and cucumber were dipped with Korean medical herb extracts such as Rheum palmatum L. and Coptis chinensis Franch. to maintain the keeping quality during storage at 5$^{\circ}$C and 10$^{\circ}$C, respectively. Treatment with 500 ppm solutions gor strawberry retarded weight loss, microbial load increase, and decay during storage. Decay ratio was shown 68.8% for control, 45.0% for Rheum palmatum L. and 42.5% for Coptis chinensis Franch. at 10 days. Strawberry treated with Coptis chinensis Franch. showed a higher hardness and soluble solid content than others. Cucumber treated with 1,000 ppm solutions revealed a different O$_2$ and CO$_2$ concentrations after 16 days. Korean medical herb extracts showed an antimicrobial activity for the quality retention of cucumber during storage. Ascorbic acid content was generally increased till 8 days, but decreased thereafter.

품종별, 생육단계별 시설재배 오이의 페놀화합물 함량과 항균효과 및 Tyrosinase 억제활성 (Phenolic Compounds, Antimicrobial Effects and Tyrosinase Inhibition Activities of Cucumber Grown Greenhouse According to Cultivars and Growth Stages)

  • 시설오이로 많이 재배되고 있는 4 품종을 선발하여 생육단계별로 총 페놀 및 플라보노이드 함량, 피부염증과 가려움증 관련 Staphylococcus aureus, Staphylococcus epidermidis, Malassezia furfur균에 대한 항균효과 그리고 tyrosinase 억제 활성에 대하여 비교 분석하였다. 총 페놀 함량은 장죽청장과 장형낙합 품종에서 늘푸른청장과 신조은백다다기 품종에 비해 상대적으로 다소 높게 나타났으며, 생육단계별로는 대부분 수확기에 이른 5단계에서 높은 함량을 보였다. 총 플라보노이드 함량은 조사된 오이 4품종 모두 생육단계별로 큰 차이를 나타내지는 않았다. 항균활성은 Staphylococcus aureus균에서는 장형낙합 품종이 모든 생육단계에서 상대적으로 높은 활성을 보였으며, Staphylococcus epidermidis균에서는 생육단계 및 품종에 따라 저해환이 8~12 mm로 항균성에 차이를 나타냈다. 또한 Malassezia furfur균에서는 장죽청장에서 항균활성이 높게 나타났으며, 특히 생육 3, 4단계에서는 저해환이 14 mm로 형성되어 가장 높은 항균효과를 보였다. 전체적으로 오이의 항균성은 Staphylococcus aureus균에서 비교적 높은 것을 확인할 수 있었다. 오이 추출물의 tyrosinase 억제 활성은 장축청장과 신조은백다다기 품종에서 상대적으로 높은 활성을 보였으며, 다음으로 장형낙합 품종에서 비교적 높게 나타났지만, 늘푸른청장의 경우는 상대적으로 낮은 활성을 보였다. 또한 생육초기보다 수확기에 이르렀을 때 모든 품종에서 tyrosinase 억제 활성이 다소 높게 나타났다. 본 실험의 결과, 피부상재균 및 비듬균에 대한 오이의 항균성과 미백효능을 확인함으로서 향후 고부가가치 향장소재로서의 이용성을 더욱 증대시킬 수 있을 것으로 기대된다.

  • A chemical investigation of the polar extract of the sea cucumber Stichopus japonicus, collected from Jeju Island, Korea, has led to isolation of five new fatty acid derivatives (1, 4 - 7) along with known compounds (2 - 3, 8 - 14). Their structures were elucidated by a combination of MS and NMR spectroscopy.

Red Sea Cucumber (Stichopus japonicus) Suppresses Cancer Progression by Promoting the ROS-Me diated Inhibition of the MAPK Pathway

  • Stichopus japonicas (red sea cucumbers) inhabit the coastal sea surrounding Jeju Island, South Korea, and are thought to have various medicinal properties. In this study, we investigated the anticancer activity of a red sea cucumber (S. japonicus) collected from Jeju Island. We obtained the red sea cucumber extract (RSCE), and observed that it inhibited the tumor cell growth and increased reactive oxygen species (ROS) production associated with the induction of apoptosis through the mitogen-activated protein kinase (MAPK) pathway in murine colon carcinoma cells (CT-26). Treatment with RSCE and N-acetylcysteine, which is a ROS scavenger, increased ROS production and apoptosis via the regulation by the MAPK pathway on the ERK and JNK compared with the nontreated group. Therefore, RSCE promotes ROS-mediated suppression of the ERK and JNK activation, and subsequently inhibits cancer progression, suggesting that RSCE may be beneficial in treating colon carcinoma.

오이에 대(對)한 유산균(乳酸菌) 생육촉진(生育促進) 인자(因子)에 관(關)하여 (Factors stimulating the Growth of Lactic acid Bacteria in Cucumber Juice)

  • 오이 성분(成分)이 유산균(乳酸菌) 생육(生育)에 미치는 영향(影響)과 이로인(因)한 김치의 한숙현상(旱熟現象)을 연구(硏究)하여 다음 결과(結果)를 얻었다. 1) 김치의 조성재료(組成材料)로 1%의 오이를 첨가(添加)하므로서 김치 발효(醱酵)의 lagphase를 현저히 단축(短縮)시켜 발효(醱酵)를 촉진(促進)하는 한중현상(旱中現象)을 보였다. 2) Cucumber serum 중(中)에서 alcohol-soluble fraction은 Lactobacillus plantarum에 대(對)하여 생육촉진(生育促進) 효과(?果)를 보였으나 Streptococcus faecalis의 생육(生育)에는 영향(影響)이 없었다. 3) Alcohol-soluble fraction의 효과는 투석(透析) 및 $120^{\circ}C$, 10 분간(分間)의 열처리(熱處理)에 의(依)하여 실활(失活)되었다. 오이 조직(組織)의 물리적성질(物理的性質)에 따른 내용성분(內容成分)의 용출정도(溶出程度)를 관찰(觀察)하여 생육(生育) 및 생산촉진요인(生産促進要因)을 물리화학적면(物理化學的面)에서 고찰(考察)하였다.

오이의 Pectinesterase 에 관한 연구 (Characteristics of Pectinesterase (PE) in Cucumbers)

  • This study was attempted to investigate the occurrence and the partial characteristics of pectinesterase (PE) in cucumbers. And the involvement of endogenous cucumber PE with the formation of insoluble pectic acid, in the presence of endogenous or added calcium ions, was also studied. The results of this study are as follows: 1) PE activity was detected in whole cucumber homogenate. 2) The optimum pH and temperature of this enzyme were found to be 8.5 and $50^{\circ}C$, respectively 3) When PE extract was added to 0.25% pectin solution, pectin gel was formed. The time required for the formation of pectin gel was reduced when the pectin solution was adjusted to the optimum conditions for PE activity(pH 8.5, $50^{\circ}C$) and $CaCl_2$ was added. 4) Cucumber juice which was heated to $100^{\circ}C$ for 10 minutes fatted to form any insoluble pectate precipitate. The formation of precipitate in cucumber juice was accelerated by preheating to $50^{\circ}C$, adjusting to pH 8.5 and addition of $CaCl_2$. 5) These results can be interpreted that endogenous PE in cucumber juice demethylates pectin, allowing interlinking of pectin molecules via divalent cation $(Ca^{++})$ and thus forming insoluble Ca-pectate. Therefore additional firming effects of cucumber can be expected to be obtained through activation of PE in conjunction with calcium ions.
