Monitoring Pathogen Infection of Freshwater Cyprinid Fish and Crustacean in Soyang Lake in 2016 (2016년 소양호에서 채집한 자연산 잉어과 어류와 갑각류의 법정전염병 및 기생충성 질병 모니터링)

  • The presence of pathogens in wild fish is a potential threat to the fish being raised nearby fish farm. Surveillance of these pathogens in the wild is, thus, highly important to keep fish in the farm safe from serious communicable diseases. Fish and crustacean were sampled 4 times at Soyang Lake in 2016. Pathogens for five reportable communicable diseases including KHVD (koi herpesvirus disease), SVC (spring viraemia of carp), EUS (epizootic ulcerative syndrome) and WSD (white spot disease), and parasites were investigated. In Soyang Lake, pale chub Zacco platypus was the most abundant. Forty seven pale chub, 8 Korea piscivorous chub Opsarichthys uncirostris, 3 oily shiner Sarcocheilichthys variegatus wakiyae and 2 crucian carp Cyprinus carassius were investigated for detection of target diseases of fish. Seventeen Caridina denticulata denticulata, 12 Oriental river prawn Macrobrachium nipponense and 1 Chinese mitten crab Eriocheir sinensis were used for detection of WSD. As the result, reportable communicable diseases were not detected in all the freshwater animals. Dactylogyrus was detected in Korea piscivorous chub. Copepoda was also detected in pale chub and Korea piscivorous chub. Metacercaria was detected in the pale chub sampled in September and October. Those were not metacercaria of liver fluke Clonorchis sinensis.

Effects of Eyestalk Ablation on the Embryogenesis of Spider Crab, Libinia emarginata

  • Spider crabs, Libinia emarginata, were eyestalk-ablated unilaterally and bilaterally to manipulate endogenous methyl farnesoate (MF) to increase during the embryogenesis. Endogenous MF were measured weekly over the embryogenesis of the crab, using HPLC with the aids of GC/MS and MS database (CAS 010485-70-8) for the identification of the hormone. Initial MF titers both in the hemolymph and embryos of intact control were at bottom levels and the hormone concentrations kept unchanged (p<0.05), reflecting physiological unnecessaries of the hormone in the embryogenesis. Eyestalk ablation significantly stimulated the crabs to increase endogenous MF in both tissues (p<0.0l). In the response of the embryos to the increased MF, no growth stimulations were observed, at least, in the first part of embryogenesis. The increased mortalities and immature sheddings of embryos resulted from the crabs under the influence of elevated MF in both tissues, instead, suggesting that the elevated MF against the crab's requirement blocked the normal developmental process of the crab embryos. These data can give crustacean endocrinologists some insights to understand the effects of the hormone on the crustacean reproduction studied previously in which JH analogs ambiguously affected the crustacean reproduction depending on the reproductive stages. The data also can give shrimp aquaculturists some implication of a possible generation of unfavorable shrimp seeds attributed to elevated egg MF originated from their eyestalk-ablated mother shrimp.

The Complete Mitochondrial Genome of Pollicipes mitella (Crustacea, Maxillopoda, Cirripedia): Non-Monophylies of Maxillopoda and Crustacea

  • The whole mitochondrial genome (14,915 nt) of Pollicipes mitella (Crustacea, Maxillopoda, Cirripedia, Thoracica) was sequenced and characterized. It is the shortest of the 31 completely sequenced crustacean mitochondrial genomes, with the exception of a copepod Tigriopus japonicus (14,628 nt). It consists of the usual 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 relatively short non-coding region (294 nt). The thoracican cirripeds apart from Megabalanus volcano have the same arrangement of protein-coding genes as Limulus polypemus, but there are frequent tRNA gene translocations (at least 8). Some interesting translocation features that may be specific to the thoracican cirriped lineage are as follows: 1) trnK-trnQ lies between the control region and trnI, 2) trnA-trnE lies between trnN and trnS1, 3) trnP lies between ND4L and trnT, and 4) trnY-trnC lies between trnS2 and ND1. In P. mitella there are two trnL genes (L1 and L2) in the typical crustacean positions (ND1-L1-LrRNA and CO1-L2-CO2). The present result is compared and discussed with the other three cirriped mitochondrial genomes from one pedunculate (Pollicipes polymerus) and two sessiles (Tetraclita japonica and M. volcano) published so far. Mitochondrial protein phylogenies reconstructed by the BI and ML algorithms show that the thoracican Cirripedia is monophyletic (BPP 100/BP 100) and associated with Remipedia (BPP 98/BP 35). In addition, Oligostraca, including Ostracoda, Branchiura, and Pentastomida, is a monophyletic group (BPP 99/BP 68), and is basal to all the other examined arthropods. Remipedia + Cirripedia appears as an independent lineage within Arthropoda, apart from Thoracopoda (Malacostraca, Branchiopda, and Cephalocarida). The Thoracopoda is paraphyletic to Hexapoda. The present result suggests that the monophylies of Crustacea and Maxillopoda should be reconsidered.

Distribution of Indicator Organisms in Commercial Fish and Shellfish and Influence of Storage Temperature and Period (시판 어패류에서의 오염지표세균의 분포와 저장 온도 및 저장 기간이 오염지표세균에 미치는 영향)

  • This paper intends to investigate commercial fish and shellfish 25 species (fish 8 species, shellfish 7 species, crustacean 3 species, moluse 4 species and echinodermata) for the distribution of sanitary indicator organisms (total viable counts, coliforms, staphylococci, vibrios, and enterococci) and distributional change of indicator organisms accordion to storage temperature and period. The logarithmic mean of total viable counts for total commercial fish and shellfish 25 species was 5.41$\pm$.026 CFU/g, and in accordance with fish and shellfishes, crustacean 6.76$\pm$0.67 CFU/g, shellfish 5.67$\pm$0.56 CFU/g, echinodermata 5.47$\pm$1.50 CFU/g, fish 5.02$\pm$0.38 CFU/g, and mollusc 5.03$\pm$0.65 CFU/g. The logarithmic mean of enterococci was 2.36$\pm$0.37 CFU/g, and in accordane with fish and shellfish, crustacean 3.44$\pm$2.12 CFU/g, shellfish 3.87$\pm$0.45 CFU/g, echinodermata 3.38$\pm$0.0 CFU/g, fish 2.16$\pm$0.41 CFU/g and mollusc 0.01$\pm$0.0 CFU/g. The logarithmic mean of vibrios was 1.60$\pm$1.59 CFU/g, and in accordance with fish and shellfish, crustacean 4.23$\pm$1.11 CFU/g, shellfish 3.58$\pm$0.90 CFU/g, echinodermata 1.64$\pm$0.34 CFU/g, fish 1.79$\pm$0.67 CFU/g and mollusc 1.07$\pm$0.61 CFU/g. The logarithmic mean of staphylococci was 1.60$\pm$1.59 CFU/g, and in accordance with fish and shellfish, shellfish 0.01$\pm$0.00 CFU/g, echinodermata 3.51$\pm$0.60 CFU/g, fish 1.68$\pm$0.64 CFU/g, crustacean 0.34$\pm$0.33 and mollusc 3.90$\pm$0.11 CFU/g. The logarithmic mean of coliformas was 2.52$\pm$0.32 CFU/g, and in accordance with fish and shellfish, echinodermata 3.58$\pm$1.89 CFU/g was highest, shellfish 3.25$\pm$0.30 CFU/g, crustacean 3.23$\pm$0.49 CFU/g, fish 2.18$\pm$0.63 CFU/g, peeled shellfish 1.80$\pm$0.51 CFU/g and mollusc 1.55$\pm$0.95 CFU/g. As the results of research of the change of the contaminate indicator microflora in working with storage period at 1$0^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$, total viable counts was increased without storage temperature and enterococci were decreased slowly at 1$0^{\circ}C$, but increased at 20 and 3$0^{\circ}C$. Vibrios were decreased slowly at 1$0^{\circ}C$, decreased at 2$0^{\circ}C$ and 3$0^{\circ}C$in 2 days after increased rapidly. Staphylococci were increased promptly without storage temperature in 2 days, then the total viable counts were maintained. Coliforms were increased at 1$0^{\circ}C$ by 7 days, then decreased or maintained after 14 days, changed at 2$0^{\circ}C$ in accordance with fish species in 2 days, then returned to the initial total viable count, and decreased rapidly at 3$0^{\circ}C$ on 2 days. By the way, there were no difference among the species.

Anti-CHH Antibody Causes Impaired Hyperglycemia in Penaeus monodon

  • Crustacean hyperglycemic hormone (CHH) plays a major role in controlling glucose level in the haemolymph and also triggers important events during molting and reproductive cycles. In Penaeus monodon, three types of CHH, namely Pem-CHH1, Pem-CHH2 and Pem-CHH3, have been previously characterized. In this study, mouse polyclonal antibody was raised against recombinant Pem-CHH1 that was expressed in Escherichia coli. The anti-Pem-CHH1 antibody recognized all three types of Pem-CHHs but did not cross-react with either related hormone, molt-inhibiting hormone of P. monodon, or unrelated human growth hormone. The hyperglycemic activity in the extract from the eyestalk neural tissues was significantly depleted after incubating with anti-Pem-CHH antibody. Direct injection of the antibody into shrimp caused about 30-50% reduction in the haemolymph glucose level. The result demonstrates the ability of anti-Pem-CHH1 antibody to deplete the activity of CHH in vivo, and thus provides a possibility of using anti-Pem-CHH1 antibody to inhibit the hormone activity as a strategy to modulate growth and reproduction in this species.

Morphological Study of the Digestive Tract of the Mud Crab (Hemigrapsus Penicillatus De Haan) and the Symbiotic Crab (Pinnotheres cyclinus Shen)

  • The influence of eating habits and food type on the ultrastructural characteristics of the digestive tracts was studied under the scanning and transmission electron microscopes in two crustacean decapods (Hemigrapsus penicillatus De Haan; mud crab, Pinnotheres cyclinus Shen; symbiotic crab). The relative ratio of the length of midgut versus hindgut was 1:1 in the mud crab, but 4:1 in the symbiotic crab. Observation through the scanning electron microscope revealed that the midguts of both species have densely-arranged longitudinal mucosal folds with a smooth surface. In the hindgut of the mud crab, mucosal folds were longitudinally oriented, clusters of two to five spines were observed on the cuticular surface, and the length of the spine in the distal hindgut was longer than that in the proximal portion. In the symbiotic crab, the mucosal folds were irregulary arranged, and numerous rudimentary spinal structures were noted on the cuticular surface. Through observation of a transmission electron microscope, the epithelial cells of the midgut in both species had numerous microvilli, but the length of the microvilli was slightly longer in the mud crab than in the symbiotic crab. The central layer of the basement membrane and the muscular layer of the midgut were more developed in the mud crab than in the symbiotic crab. The thickness of the cuticular layer over the hindgut surface in the mud crab was about 4 times than that of the symbiotic crab.

Community Structure and Spatial Variation of Meiobenthos Associated with an Artificial Structure (퇴적촉진 구조물 설치에 따른 중형저서동물 군집구조 및 변동)

  • We investigated the spring and summer community structure of meiobenthos in a tidal-falt near Iwon, Korea, in 2002 and 2003. In total, 12 meiofaunal groups were found in the study area among which nematodes were the most dominant. Benthic foraminiferans, harpacticoid copepods, polychaetes, and crustacean nauplli were also dominant groups at all sites. The total density of meiobenthos at each station was be 246-2,177 ind./$10cm^2$. As the depth of sediment increased, the density of meiobenthos at each station gradually decreased. Changes in the vertical distribution of meiobenthos in the study area occurred mainly near the sediment surface (0-1 cm). Generally, between spring and summer the density of nematodes increased, and the density of other dominant meiofaunal groups (benthic harpacticoids, crustacean nauplii, benthic foraminiferans) decreased near the sediment trap the control site of sediment traps compared to that at the control site. The results of cluster and multidimensional scaling plots indicate that the meiofaunal community changed following construction of a low artificial wood groin structure.

Temporal Variation of the Macro-crustacean Assemblages on Jangbong Tidal Flat, Incheon, Korea (장봉도 갯벌을 이용하는 대형 갑각류 군집의 계절 변화)

  • This study examined the community structure and temporal variation of the macro-crustaceans on Jangbong tidal flat, Incheon, Korea. Macro-crustaceans on this tidal flat were collected using a modified otter trawl from November 1999 to January 2001. The macro-crustaceans comprised 32 species, 7,741 individuals, and 29,211.3 gWWt during the study period. The Jangbong tidal flat was dominated numerically by three shrimp (Palaemon carinicauda, Crangon affinis, and Metapenaeus joyneri) and two portunid crabs (Charybdis japonica and Portunus trituberculatus). The macro-crustacean assemblages were divided into two different season and species groups based on the q-mode and r-mode clustering methods. In terms of q-mode, they consisted of a winter species group from December through April and a summer species group from May through November. The r-mode analysis showed that the species group visiting the tidal flat in winter was dominated numerically by palaemonid and crangonid shrimps, while a species group characterized by an abundance of portunid crabs and penaeid shrimps was more frequent in spring, summer, and autumn. In conclusion, our study revealed distinct temporal variation in the tidal flat use by macro-crustaceans around Jangbong Island, Incheon, Korea.

Feeding Habits of 6 Shark Species in the Southern Sea of Korea (남해안에 출현하는 상어류 6종의 식성)

  • The feeding habits of six shark species, Scyliorhinus torazame, Mustelus manazo, Squalus megalops, Isurus oxyrinchus, Alopias pelagicus, and Carcharhinus plumbeus were studied using the stomach contents of 463 specimens collected between January and February 2007 in the southern sea of Korea. They consumed different prey items. S. torazame preyed mainly crustaceans, and M. manazo on crustaceans and fishes. S. megalops, I. oxyrinchus, A. pelagicus, and C. plumbeus consumed predominately fishes. Non-metric multi-dimensional scaling (NMDS) ordination plots emphasized that dietary similarities separated thesix shark species into three trophic groups based on similarities in percentage Index of Relative Importance (%IRI): Group I (crustacean feeders), Group II (fish and crustacean feeders), and Group III (fish feeders).
