DPPH Radical Scavenging Effect and in vitro Lipid Peroxidation Inhibition Activity of Portulaca oleracea and Its Active Principles (쇠비름(Portulaca oleracea) 추출물의 DPPH radical 소거능과 in vitro 지질과산화 억제 효과와 활성성분)

  • An antioxidative activity of Portulaca oleracea was tested by in vitro experimental models. The antioxidative activities were determined by evaluation the DPPH radical scavenging activity and by measuring lipid peroxide using 2-thiobarbituric acid (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. Among them, a remarkable antioxidative effect was observed in the fractions of EtOAc and n-BuOH. The DPPH radical scavenging effect $(IC_{50}=17.90{\mu}g/ml)$ of the n-BuOH soluble fraction was comparable with that of natural antioxidant, ${\alpha}-tocopherol(IC_{50}=\;6.99{\mu}g/ml)$ and the inhibitory effect of lipid peroxidation in mouse liver homogenate was similar to that of natural antioxidant, L-ascorbic acid at a concentration of 0.1mg/ml to 5mg/ml. From the BuOH soluble fraction yielded two biophenolic glycosides, 3-hydroxy-1-(2-hydroxyethyl)phenyl-4-O-${\beta}$-D-glucopyranoside(1) and 2-(3,4-dihydroxyphenyl)ethyl-O-${\beta}$-D-glucopyranoside(2) using column chromatography and revered-phase HPLC. In particular, the DPPH radical scavenging activity of 2 was comparable to that tocopherol$(IC_{50}=6.59{\mu}g/ml)$.

  • The effects of Semen Ziziphi Spinosae extract were tested for the anti-stress action. 100g of crude drug of Semen Ziziphi Spinosae was extracted with pure water and the total extractive was evaporated under reduced pressure to give 10.7g. ICR male mice($20{\pm}2g$) were fed orally with the dose of 100mg/kg/day for five days. Foot shock was given to make experimental environment of physiological stress. Foot shock mice were placed individually in the foot shock compartments and sociopsychological mice were placed nonfoot shock compartment-the side of foot shock compartments. Mice were exposed to sociopsychological stress by restraining and seeing foot shock stressed mice for one hour for five days. Semen Ziziphi Spinosae extract administered group showed a significant decrease of serum corticosterone secretion compared with control group. Noradrenaline secretions in the dorsal cortex of brain were increased but not significant. Lipid peroxidation of the liver of mice were tested by measuring malondialdehyde, and Semen Ziziphi Spinosae extract had tendency of decreasing lipid peroxidation but not significant. But Semen Ziziphi Spinosae extract administration had the effect of decreasing serum level of aspartate aminotransferase and malondialdehyde. These results suggest that Semen Ziziphi Spinosae extract can effectively rid the sociopsychological stress and stress concerned diseases.

Antioxidant Activity of Seaweed Extracts from Kongsfjorden, an Inlet in the Arctic's Svalbard Archipelago (북극 스발바드 군도 콩스피요르드에서 채집한 해조류 추출물의 항산화 활성)

  • We examined the intracellular antioxidative effects of 20 Arctic seaweed extracts in Raw 264.7 cells. Each seaweed species was subjected to extraction using acetone/dichloromethane and methanol, respectively, after which the extracts were combined and used as the test sample. The antioxidant ability of all 20 seaweeds extracts was evaluated using four different activity tests, including the degree of occurrence of intracellular reactive oxygen species (ROS), $ONOO^-$, and lipid peroxidation in Raw 264.7 cells, as well as the extent of oxidative damage of genomic DNA purified from Raw 264.7 cells. Crude extracts from Monostroma obscurum, Alaria esculnta, Laminaria digitata, Desmarestia aculeata, Chorda filum, Ptilota seriata, Phycidrys rubens, Devaleraea ramentacea and Palmaria palmata exhibited significant scavenging effects on the generation of intracellular ROS. Among them, Monostroma obscurum and Phycidrys rubens significantly inhibited membrane lipid peroxidation and DNA oxidation. Moreover, Phycidrys rubens exhibited scavenging effects on peroxynitrite generated from SIN-1.

Studies on the Lipid Rancidity and Rheology of Yackwa During Storage (약과 저장시의 산패정도와 물성에 관한 연구)

  • The contents of crude lipids of Yackwa was especially high of 24.4%. After 30 days of storage, the acid value, TAB and the carbonyl value were 2.0, 1.3, and 1.4 times higher than the initial storage, respectively. Soaking in syrup prevented moisture loss and the progression from lipid oxidation to quality deterioration, and addition of ginger juice showed the slightly lower in lipid rancidity. When compared with the unsoaked group, the L value of the soaked group were lower during the 30-day storage. As for the textural changes during storage, elasticity and chewiness decreased slightly. It was hypothesized that the textural changes were affected not by the ginger juice or the storage period, but by soaking in syrup.

Effect of Dietary Composition with Different Feeding Regime on Compensatory Growth of Juvenile Olive Flounder Paralichthys olivaceus

  • Compensatory growth of juvenile olive flounder Paralichthys olivaceus fed different diets with different feeding regime was compared. Four hundred fifty fish (twenty five fish per tank) were randomly distributed into 18 of 180 L flow-through tanks. Six treatments were prepared: fish were hand-fed with the control (C) diet to satiation twice a day, six days a week, for 8 weeks (C-8W treatment); and other groups of fish were starved for 2 weeks and then fed with the C, high protein (HP), high carbohydrate (HC), high lipid (HL), and combined protein, carbohydrate and lipid (CPCL) diets to satiation twice a day, six days a week, for 6 weeks, referred to as C-6W, HP-6W, HC-6W, HL-6W, and CPCL-6W treatments, respectively. Final body weight of fish in HP-6W treatment was higher than that of fish in C-6W, but not different from that of fish in C-8W, HC-6W, HL-6W and CPCL-6W treatments. Specific growth rate of fish in HP-6W treatment was higher than that of fish in all other treatments except for fish in CPCL-6W treatment. Feeding rate of fish in C-8W treatment was higher than that of fish in HP-6W, HC-6W, HL-6W and CPCL-6W treatments, but not different from that of fish in C-6W treatment. In addition, feeding rate of fish in C-6W treatment was higher than that of fish in HP-6W, HL-6W and CPCL-6W treatments. Feed and protein efficiency ratios of fish in HP-6W, HC-6W, HL-6W and CPCL-6W treatments were higher than those of fish in C-6W treatment. None of moisture, crude protein and ash content of the whole body of fish excluding the liver was different among treatments. Dietary supplementation of protein, carbohydrate, lipid and their combination could improve compensatory growth of fish when fish were fed for 6 weeks after 2-week feed deprivation; especially, supplementation of dietary protein was the most effective to improve compensatory growth of fish.

Cytoprotective and Antioxidative Effects of Crude Drug Preparation (E-kong-san) (이공산(異功散)의 세포보호 및 항산화 작용)

  • In the previous report, E-kong-san, which is usually used for recovering health in traditional medicine, has been shown to decrease cisplatin induced nephrotoxicity in vivo and in vitro. The significant reduction of E-kong-san on the cisplatin induced nephrotoxicity led us to investigate whether the effect of this water extract was a result of triggering antioxidation. In monkey kidney Vero cells, E-kong-san at $5{\sim}10\;mg/ml$ was able to attenuate 2mM cisplatin-stimulated cell death by 46.8% and 31.8%, respectively. E-kong-san showed strong free radical scavengering activities on 1,1-diphenyl-2-picrylhydrazil (DPPH) radical and xanthine/xanthine oxidase (XOD) generated superoxide anion radical $(O_2^{-.})$. We further studied the effects of E-kong-san on lipid peroxidation in rat liver microsomes induced by enzymatic and nonenzymatic methods. Moreover, E-kong-san exhibited significant inhibition on both ascorbic $acid/Fe^{2+}$ and $ADP/NADPH/Fe^{3+}$ induced lipid peroxidation in rat liver microsomes. Based on these results, we suggest that E-kong-san protects the cisplatin induced cytotoxicity by its antioxidative mechanism.

Influence of Dietary Beef Tallow and Dextrin on the Growth and Body Composition of Juvenile Far Eastern Catfish Silurus asotus (배합사료의 우지와 덱스트린이 메기(Silurus asotus)의 성장과 체조성에 미치는 영향)

  • This study investigated the utilization of beef tallow and dextrin as non-protein energy sources in the diet of juvenile far eastern catfish Silurus asotus. Three replicated groups of juveniles (initial mean weight of 3.6 g) were fed diets containing high levels of beef tallow or dextrin for six weeks. The final mean weight of fish fed on a 17% lipid diet with a high level of beef tallow was significantly higher than that of fish fed on a 9% lipid diet with a high level of dextrin (P=0.04). No significant differences were observed in the feed efficiency and protein efficiency ratio of fish fed on the experimental diets. Crude lipid content and the C18:0, C18:1n-9 and C18:2n-6 contents of whole-body fish were significantly affected by diet (P<0.05). The results of this study suggest that beef tallow is better than dextrin as a non-protein energy source for improving the growth of juvenile far eastern catfish, and that beef tallow could be economically used as an ingredient in practical feed.

Compositions of Fatty Acid, Free Amino Acid and Total Amino Acid of Lespedeza x chiisanensis T. LEE (지리산 싸리의 지질 구성 지방산과 유리 아미노산 및 구성 총 아미노산 조성에 관하여)

  • The compositions of amino acid in the protein and fatty acid in the lipid of Lespedeza x chiisanensis T. LEE were analyzed by HPLC and GC, respectively. The contents of crude oil and protein from the extracts were 11.13% and 5.18%, respectively. The amount of free anino acids in the protein was 443.14mg/100g, and 94.84mg/100g of essential amino acid were contained in the free amino acid. The amount of total amino acids in the protein was 3159.85mg/100g, and 1068.18mg/100g of essential amino acid were contained in the total amino acid. The compositions of fatty acid in the lipid were $C_{18\;:\;2}=45.05%,\;C_{18\;:\;3}=18.71%,\;C_{19}=14.70%,C_{18\;:\;1}=6.81%,\;C_{16}=4.35%,\;C_{16\;:\;1}=1.59%$ in order, respectively. 72.44% of unsaturated fatty acids were contained in the lipid.

Effects of prolonged photoperiod on growth performance, serum lipids and meat quality of Jinjiang cattle in winter

  • Objective: This study was conducted to investigate the potential effects of prolonged photoperiod on the serum lipids, carcass traits, and meat quality of Jinjiang cattle during winter. Methods: Thirty-four Jinjiang bulls aged between 14 and 16 months were randomly assigned to two groups that were alternatively subjected to either natural daylight +4 h supplemental light (long photoperiod, LP) or natural daylight (natural photoperiod, NP) for 96 days. The potential effects on the levels of serum lipids, carcass traits, meat quality, and genes regulating lipid metabolism in the intramuscular fat (IMF) of the cattle were evaluated. Results: Jinjiang cattle kept under LP showed significant increase in both dry matter intake and backfat thickness. the serum glucose and the plasma leptin levels were significantly reduced, while that of melatonin and insulin were observed to be increased. The crude fat contents of biceps femoris muscle and longissimus dorsi muscle were higher in LP than in NP group. In longissimus dorsi muscle, the proportions of C17:0 and C18:0 were significantly higher but that of the C16:1 was found to be significantly lower in LP group. The relative mRNA expressions in IMF of longissimus dorsi muscle, the lipid synthesis genes (proliferator-activated receptor gamma, fatty acid-binding protein) and the fatty acid synthesis genes (acetyl-coa carboxylase, fatty acid synthetase, 1-acylglycerol-3-phosphate acyltransferase) were significantly up-regulated in LP group (p<0.05); whereas the hormone-sensitive lipase and stearoyl-CoA desaturase 1 were significantly down-regulated in LP than in NP group. Conclusion: Prolonged photoperiod significantly altered the growth performance, hormonal levels, gene expression and fat deposition in Jinjiang cattle. It suggested that the LP improved the fat deposition by regulating the levels of different hormones and genes related to lipid metabolism, thereby improving the fattening of Jinjiang cattle during winter.

Effects of maternal dietary energy restriction on laying performance, embryonic development, and lipid metabolism in broilers

  • Objective: The objective of this study was to investigate the effects of different degrees of maternal dietary energy restriction on lipid deposition in embryonic tissues during the medium laying period (37 to 39 weeks) in Arbor Acres (AA) broiler breeders. Methods: A single factor design was adopted, and 400 AA broiler breeders (20 weeks of age) with a similar weight were randomly allocated into four groups. The birds in the control group were fed a corn-soybean meal based diet, and those in trial groups were fed diets with 80%, 70%, and 50% energy levels of the basal diet. Incubated eggs from the medium laying period were collected. Samples of developing embryos at various stages were prepared for composition analysis. Results: The embryo weight in the 80% energy group was higher than those of the other groups on embryonic day (E) 13, but at 21 E, they were significantly decreased with decreasing energy intake of the broiler breeders (p<0.05). Additionally, the levels of crude fat in tissues in the restriction groups were significantly decreased (p<0.05). The long axis and area of adipocytes in breast muscle, thigh muscle and the liver were significantly decreased (p<0.05) at 21 E in the 80%, 70%, and 50% energy groups. Conclusion: The effects of the 80% maternal dietary energy restriction energy affects egg production performance, egg quality, and nutrient deposition in egg weights, which then directly impacts on the developmental process of embryos, especially on fat utilization and deposition.
