We examined the effects of crude hemicellulose, alcohol-insoluble hemicellulose, high molecular weight soluble polysaccharide (HMS-P : MW>10 kDa) and low molecular weight souble polysaccharide (LMS-P : MW<10 kDa) fraction isolated from buckwheat (raw, roast and steam) on digestive enzyme activity in vitro. The enzyme activities were measured after the polysaccharides-enzyme mixtures were incubated at $37^{\circ}C$ for 5 min. Crude hemicellulose, alcohol-insoluble hemicellulose and residue lowered ${\alpha}-amylase$ activity, whereas HMS-P and LMS-P had no inhibitory effect. All polysaccharides except LMS-P lowered lipase activity. Crude hemicellulose, alcohol-insoluble hemicellulose, residue and HMS-P showed a marked decrease of trypsin and chymotrypsin activity but LMS-P showed a slight decrease of them.
This study was conducted to investigate the characteristics of nonstarchy polysaccharides in Korean ginseng, (Panax ginseng C.A. Meyer). The results obtained are as follows. 1. The total sugar content of ginseng roots were decreased with increasing the cultural period. On the other hand, the crude fiber content was increased with that of the ginseng leaves or stems. But the crude fiber in root was much less than that of leaves and stems. 2. The dietary fiber content of ginseng root on 5 years old was 14.20% as neutral detergent fiber, 9.08% as acid detergent fiber, hemicellulose 5.12%, cellulose 7.98% and lignin of 1.10%, respectively. 3. Much more pectin was found in ginseng roots which was cultivated for shooter Period. And it was contained much more in the root than in the leaves and stems. 4. ginseng hemicellulose content in root was 5% to 10%. It was decreased with increasing: cultivated period. Hemicellulose was constituted of xylose, arabinose, glucose, rhamnose and xylose of these sugars was the predominant. 5. X-ray diffraction Pattern of ginseng cellulose showed maximum intensity at tile interplanar angle of 4.1$^{\circ}$.
Wood chips of oak(Quercus mongolica) and larch(Larix leptolepis) were exploded with the optimum condition. Main components of exploded wood were separated with hot hot water and methanol. The hemicelluloses were purified from hot water extracts and alditol complexs were prepared from purified hemicellulose. And also, cellulose nitrate was prepared from extractive residue and characterized. The results can be summarized as follows. 1. Amounts of carbohydrate(72~79%) in the crude hemicellulose of larch wood was more than those of oak wood(55~66%). 2. The crude hemicelluloses were mainly composed of oligosaccharides in oak wood but those in larch wood contained about 50% monosaccharides. 3. Decolorization of hemicellulose was successful with activated charcoal and ion-exchange resin treatment. The alditol yields were 56.3~82.9%. 4. The degree of substitution(D.S.) of cellulose nitrate was 1.95~2.87 and it showed a good acetone solubility.
This study was conducted to investigate the possibility that oil seed rape could be used as a forage fodder crop and to select the most suitable variety of forage rape at the southern area of Korea, Two varieties of oil seed rape currently grown for oil production and six introduced varieties of forage rape with relatively high yield and high nutritional value were grown at the same condition and their nutritional value were observed in Spring. Generally, rape was considered as a useful forage fodder crop with high content of crude protein and low contents of NDF, ADF, hemicellulose, cellulose and lignin. Differences in mean values of the above characters between two groups of rape were not statistically significant. Velox showed significantly higher content of crude protein and significantly lower contents of NDF, ADF, hemicellulose, cellulose and lignin compared with other varieties of forage rape in spring. Rape was relatively high in IVDMD compared with other forage fodder crops, and forage rape was more or less in IVDMD and DDMW than oil seed rape. Velox was the highest in IVDMD and DDMW among the varieties of forage rape in Spring, in this experiment.
This study was conducted to extract and isolate the polysaccharides from buckwheat. Also the sugar composition of the polysaccharides was investigated. The soluble and indigestible polysaccharides were isolated from supernatant and residue after enzyme treatment of raw, roast and steam buckwheat. The yields of low molecular weight soluble polysaccharides(LMS-P: MW<10 Kda) and high molecular weight soluble polysaccharides(HMS-P: MW>10 Kda) were $74.9{\sim}84.2%$ and $5.5{\sim}9.4%$, respectively. The yields of indigestible polysaccharides were low molecular weight insoluble polysaccharides; $0.8{\sim}4.2%$, crude hemicellulose; $3.2{\sim}9.6%$, alcohol insoluble hemicellulose; $0.9{\sim}1.7%$, residue; $2.0{\sim}2.4%$, respectively. The free sugars were detected in the soluble polysaccharides and low molecular weight insoluble polysaccharides but were not detected in the crude hemicellulose, alcohol insoluble hemicellulose and residue. The protein of all fraction were detected and the content was $1.0{\sim}18.9%$. The main sugar of soluble polysaccharides was glucose and the indigestible polysaccharides were composed of glucose, arabinose, rhamnose, xylose, mannose and galactose.
Physicochemical properties, intestinal microbial growth, and inhibitory effects of alcohol-insoluble polysaccharide (AIP) extracted from cucumber peel were investigated. AIP was composed of 14.54% crude protein, 1.04% crude lipid, 13.74 % crude ash, 9.1% soluble dietary fiber, and 41.2% insoluble dietary fiber. AIP showed low bulk density (0.18 g/mL) and water-holding capacity (6.39 g/g), and high oil-holding capacity (3.96 g/g). Pectic substance fractions [water-soluble pectic substance (WSP), ethylenediaminetetraacetic acid-soluble pectic substance (ESP), and alkali-soluble pectic substances (ASP)] and hemicellulose fractions [1 M KOH-soluble hemicellulose (KHP1) and 4 M KOH-soluble hemicellulose (KHP4)] were obtained from sequential chemical fractionation of AIP. WSP showed higher total sugar contents than total uronic acid contents, whereas opposite results were observed in ESP and ASP. Molecular weight distributions of three pectic substance fractions were in order of ASP>ESP>WSP. Ion exchange chromatogram pattern of WSP was different from those of ESP and ASP. Major component of WSP was fraction eluted by 0.05 M ammonium acetate buffer, whereas that of ESP and ASP was fraction eluted by 0.2 M NaOH. WSP and ASP showed growth-promoting activities against Lactobacillus brevis, Bifidobacterium bifidum, and B. longum, whereas B. bifidum and B. longum for ESP. KHP1 and KHP4 fractions had significant growth-promoting activities against B. bifidum.

$HF_1$(1H KOH soluble hemicellulose fraction), $HF_2$(2H KOH soluble hemicellulose fraction) $HF_3$(3H KOH soluble hemicellulose fraction) and $HF_4$(4H KOH soluble hemicellulose fraction) were fractionated from Fuji crude cell wall and purified using Sephacryl S-500 to determine the effects of these hemicellulosic fractions on the Ca- pectate gel formation. By increasing of KOH concentration, from 1 to 4N, hexose peas became higher in led, and molecula weights, especially pentose peaks in high molecular weight. Hemicellulose fractions using gel filtration were composed of $8{\sim}10$ peaks which were $10^4{\sim}143{\times}10^4$ molecular weight. Higher values of hardness, adhesiveness and gumminess were found in low molecular weight than in high molecular weight, also in hexose and uronic acid contained than in hexose contained.
The objective of this study was to characterize organic mater decomposition with application of livestock manure compost in upland soil. Six different composts, which were chicken (CHM), pig (PIM), and cow (COM) manure compost added and chicken (CHMS), pig (PIMS), cow (COMS) manure compost with sawdust added, were prepared for this study. These composts have different composition of crude carbohydrate (hemicellulose, cellulose, and lignin). The buried-bag method was used to determine the rate of organic mater decomposition and the changes of crude carbohydrate content during 36 months in the field. In all treatment, hemicellulose content was sharply decreased within 8 months, but considerable amount of lignin was remained after 36 months. After 40 months, the rates of carbon decreasing were 81, 80, 72, 69, 67, and 64 % for CHM, PIM, COM, CHMS, PIMS, and COMS, respectively. The estimated equation of carbon decreasing rate (D), $D=aT^b$, was fit to the carbon decreasing rate vs. elapsed time (T) using a non-linear regression procedure. After 40 months, significant difference of carbon decreasing rate between observed and estimated was not found. The relationship between constant a, b and hemicellulose content in the compost was not observed in this experiment. The cellulose and lignin content in the compost were positively correlated to the constant b and negatively correlated to the constant a.
Change on the structural carbohydrate(several fiberous components) was determined by vegetables(crown daisy and butterbur)-cultivated in Ulsan, Kyungnam, Korea-as its stage of maturity developed. Samples were separated into leaf and stem, which were dried at 7$0^{\circ}C$ for 24hr, and ground to pass a 1mm screen. They were subjected to moisture, crude protein, crude fat and several dietary fiber-DF(dietary fiber, include unavaible components), NDF(neutral detergent fiber), ADF(acid detergent fiber), lignin, hemicellulose, cellulose and protein corrected NDF(c-NDF), IDF(indigestible fiber, include lignin, hemicellulose and cellulose). In general, structural carbohytrate(several dietary fiber) of vegetable was affected by the growth stage. In case of crown daisy and butterbur, dietary fiber in leaf was higher than DF in stem.
Black liquor (BL) is a by-product of rice straw pulping process. It is a low costs raw material for production value-adding proteins and enzymes, which has been paid more and more attention to reduce its environmental pollution. Mixed cultures of micelial fungi, Trichoderma reesei Northern Regional Research Laboratory (NRRL)11236, Trichoderma reesei NRRL 6165 and Aspergillus niger strains NRC 5A, NRC 7A, and NRC 9A were evaluated for their ability to produce xylanase using crude hemicellulose (CHC) prepared from BL and peat moss as an inert support under solid state fermentation (SSF). The most potent strains, A. niger NRC 9A (818.26 U/g CHC) and T. reesei NRRL 6165 ($100.9{\pm}57.14$ U/g CHC), were used in a mixed culture to enhance xylanase production by co-culturing under SSF. In the mixed culture, xylanase production ($1070.52{\pm}12.57$ U/g CHC) was nearly1.3 and 10.6-fold increases over the activities attained in their monocultures, A. niger NRC 9A and T. reesei NRRL 6165, respectively. Optimization of the culture parameters of the mixed culture SSF process, concentration of ammonium sulfate and corn steep liquor, CHC/peat moss ratio, inoculum size and ratios of the two strains, initial pH value, initial moisture content and incubation time, exhibited a significant increase ($2414.98{\pm}84.02$ U/g CHC) in xylanase production than before optimization.
