자색옥수수 포엽과 속대 혼합 추출물의 영양성분, 항산화 활성 물질 함량분석 및 생리활성 평가 (Assessment of Nutritional Components, Antioxidant Contents and Physiological Activity of Purple Corn Husk and Cob Extracts)

  • 본 연구는 색소 1호 포엽과 속대 추출물의 영양성분을 분석하고 항산화 및 항당뇨 활성을 검정하여 향후 기능성 식품으로서의 활용을 위한 기초자료로 제공하고자 수행되었다. 색소 1호 포엽과 속대 추출물의 일반성분 분석결과, 수분, 조회분, 조지방, 조단백질은 각각 6.90%, 7.31%, 0.52%, 7.07%이었다. 색소 1호 포엽 및 속대 혼합추출물의 지방산은 palmitic acid, oleic acid, linoleic acid, linolenic acid인 것으로 나타났으며 불포화 지방산의 비율은 83.33%이었다. 색소 1호 포엽 및 속대 혼합추출물의 구성 아미노산 함량을 분석한 결과 glutamic acid, aspartic acid 등을 포함한 총 17종의 아미노산이 검출되었으며 이 중 glutamic acid의 함량이 736.08 mg / 100 g으로 가장 높았다. 색소 1호 포엽 및 속대 추출물의 DPPH 및 ABTS radical 소거활성은 각각 95.62% ($1,000{\mu}g/mL$), 92.00% ($10,000{\mu}g/mL$)이었으며 총 폴리페놀 및 플라보노이드 함량은 각각 99.87 mg/g, 25.02 mg/g이었다. 색소 1호 포엽 및 속대 추출물의 ${\alpha}-amylase$${\alpha}-glucosidase$ 저해활성은 추출물 10 mg/mL의 처리농도에서 각각 95.86%, 76.92%인 것으로 나타났다.

더덕, 도라지 표고균사발효물의 유용성분 및 생리활성 (Beneficial constituents and physiological activity of fermented Codonopsis lanceolata and Platycodon grandiflorus by Lentinula edodes mycelium)

  • 한약재로 사용되는 더덕과 도라지에 표고의 유용성분을 함유한 소재를 개발하고자 더덕표고균사발효물(FCLM)과 더덕도라지균사발효물(FPLM)을 제조하였다. 두 가지 발효물의 일반성분, 유리당, 유기산, 𝛽-glucan, ergothioneine, ergosterol, vitamin D2 함량 및 3T3-L1 지방전구세포의 분화에 미치는 영향을 연구하였다. 일반성분 분석결과, FCLM에서 조섬유와 조지방함량이 FPLM에 비하여 높게 나타났으며, FPLM은 조단백질과 가용성무질소물의 함량이 FCLM에 비하여 높게 나타났다. 유리당은 FCLM과 FPLM 모두 arabinose, glucose, sucrose 3종이 검출되었으며, 총 유리당 함량은 FPLM에서 높게 나타났다. 유기산 함량은 발효 전 더덕과 도라지에 비하여 발효 후 FCLM과 FPLM에서 낮게 나타났다. 𝛽-Glucan 함량은 FCLM과 FPLM 두 발효물에서 대조구로 사용한 표고보다 높은 함량을 보였다. 항산화 물질로 알려진 ergothioneine의 함량은 FCLM이 FPLM보다 높게 나타났다. Ergosterol 함량은 대조구로 사용한 표고에서 가장 높게 나타났으며, 발효물들은 비슷한 함량을 나타내었다. Vitamin D2는 발효물들에서만 검출되었으며, FPLM (0.58±0.01 mg%)가 FCLM (0.47±0.01 mg%)보다 높은 함량을 보였다. 더덕과 도라지 표고균사발효물인 FCLM과 FPLM는 3T3-L1 지방전구세포에 대하여 발효 전 더덕과 도라지에 비하여 높은 수준의 세포 생존율을 나타내었다. 또한 FCLM과 FPLM은 3T3-L1 지방전구세포 분화를 발효 전 더덕과 도라지에 비하여 높은 수준으로 억제하여, 비만 억제 효과가 있는 것으로 기대된다.

Influence of Quaternary Benzophenantridine and Protopine Alkaloids on Growth Performance, Dietary Energy, Carcass Traits, Visceral Mass, and Rumen Health in Finishing Ewes under Conditions of Severe Temperature-humidity Index

  • Twenty $Pelibuey{\times}Katahdin$ ewes ($35{\pm}2.3kg$) were used to determine the effects of the consumption of standardized plant extract containing a mixture of quaternary benzophenanthridine alkaloids and protopine alkaloids (QBA+PA) on growth performance, dietary energetics, visceral mass, and ruminal epithelial health in heat-stressed ewes fed with a high-energy corn-based diet. The basal diet (13.9% crude protein and 2.09 Mcal of net energy [NE] of maintenance/kg of dry matter) contained 49.7% starch and 15.3% neutral detergent fiber. Source of QBA+PA was Sangrovit RS (SANG) which contains 3 g of quaternary benzophenathridine and protopine alkaloids per kg of product. Treatments consisted of a daily consumption of 0 or 0.5 g SANG/ewe. Ewes were grouped by weight and assigned to 10 pens (5 pens/treatment), with two ewes per pen. The experimental period lasted 70 days. The mean temperature humidity index during the course of this experiment was $81.7{\pm}1.0$ (severe heat stress). There were no treatment effects on water intake. Dry matter intake was not affected (p = 0.70) by treatments, but the group fed SANG had a numerically (11.2%) higher gain in comparison to the control group, SANG improved gain efficiency (8.3%, p = 0.04), dietary NE (5.2%, p<0.01) and the observed-to-expected NE (5.9%, p<0.01). Supplemental SANG did not affect ($p{\geq}0.12$) carcass characteristics, chemical composition of shoulder, and organ weights (g/kg empty body weight) of stomach complex, intestines, and heart/lung. Supplemental SANG decreased liver weight (10.3%, p = 0.02) and increased visceral fat (16.9%, p = 0.02). Rumen epithelium of ewes fed SANG had lower scores for cellular dropsical degeneration (2.08 vs 2.34, p = 0.02), parakeratosis (1.30 vs 1.82, p = 0.03) and neutrophil infiltration (2.08 vs 2.86, p = 0.05) than controls. It is concluded that SANG supplementation helped ameliorate the negative effects of severe heat on growth performance of feedlot ewes fed high-energy corn-based diets. Improvement in energetic efficiency may have been mediated, in part, by anti-inflammatory effects of supplemental SANG and corresponding enhancement of nutrient uptake.

Effect of Dietary Fiber Level on the Performance and Carcass Traits of Mong Cai, F1 Crossbred (Mong Cai×Yorkshire) and Landrace×Yorkshire Pigs

  • The effects of feeding diets containing 20% (L) or 30% (H) neutral detergent fiber (NDF) (DM basis) on performance and carcass traits were studied in three breeds of pig, including pure Mong Cai (MC), crossbred Landrace$\times$Yorkshire (LY) and crossbred MC$\times$Yorkshire (F1). The experiment had a factorial design with two factors, breed and diet. Eighteen piglets of each breed ($60{\pm}3days$) were randomly allocated to three treatments: L-L, low fiber diet in both growing and finishing periods; L-H, low and high fiber diet in the growing and finishing period, respectively; and H-H, high fiber diet in both periods. The diets were iso-energetic and iso-nitrogenous within feeding period. The main fibrous ingredients of the diets were rice bran and cassava residue. There were no effects of fiber level on daily dry matter feed intake (DMI), expressed as g/kg metabolic body weight (BW0.75), in both feeding periods (p>0.05). DMI was highest for MC, followed by F1 and LY (p<0.001). Average daily gain (ADG) in L-L and L-H was higher than in H-H in the growing period (p<0.001) and overall (p<0.05), while feed conversion ratio (FCR) was higher in H-H than in L-L and L-H in the growing period (p<0.05) and overall, but no significant differences between treatments were found in the finishing period. In both periods, Landrace$\times$Yorkshire had the highest ADG and the lowest FCR, followed by F1 and Mong Cai (p<0.001). There were no interactions between breed and diet for performance and carcass traits. Carcass and dressing percentage was lower for L-H and H-H than for L-L (p<0.05). There were no significant differences among treatments in back fat thickness and lean meat percentage, or in crude protein and ether extract contents of lean meat. Carcass, dressing and lean meat percentage was highest for LY, lowest for MC and intermediate for F1 (p<0.001). It can be concluded that feeding a high fiber diet in the growing period reduced pig performance, but there was no effect in the finishing period. Pure Mong Cai pigs are not particularly suitable for meat purposes, although the F1 cross with Large White had reasonably good growth performance and carcass quality.

By-product of Tropical Vermicelli Waste as a Novel Alternative Feedstuff in Broiler Diets

  • Two experiments were conducted to determine physical and chemical properties of vermicelli waste (VW) and effect of VW inclusion levels on growth performance of broilers. In experiment 1, VW samples were randomly collected from vermicelli industry in Thailand to analyze nutritional composition. Vermicelli waste contained 9.96% moisture, 12.06% CP, 32.30% crude fiber (CF), and 0.57% ether extract (EE), as DM basis. The ratio of insoluble:soluble non-starch polysaccharide (NSP) was 43.4:8.9. A total of 120 chicks (6 pens per treatment and 10 chicks per pen) were fed a corn-soybean meal-based diet or 20% VW substituted diet to determine the apparent metabolizable energy corrected for nitrogen retention ($AME_n$) of VW. The $AME_n$ of VW was $1,844.7{\pm}130.71$ kcal/kg. In experiment 2, a total of 1,200 chicks were randomly allotted to 1 of 4 dietary treatments for 42-d growth assay. There were 300 chicks with 6 pens per treatment and 50 chicks per pen. The dietary treatments contained 0%, 5%, 10%, or 15% VW, respectively. All diets were formulated to be isocaloric and isonitrogenous. From 0 to 18 d of age chicks fed VW diets had higher (p<0.001) feed conversion ratio (FCR) compared with those fed the control diet. No difference was observed during grower and finisher phase (19 to 42 d). Chicks fed VW diets had lower relative weight of abdominal fat (p<0.001) but higher relative weight of gizzard (p<0.05) than those of chicks fed the control diet. Increasing VW inclusion levels increased ileal digesta viscosity (p<0.05) and intestinal villus height of chicks (p<0.001). For apparent total tract digestibility assay, there were 4 metabolic cages of 6 chicks that were fed experimental treatment diets (the same as in the growth assay) in a 10-d total excreta collection. Increasing VW inclusion levels linearly decreased (p<0.05) apparent total tract digestibility of DM and CF.

Effects of alfalfa flavonoids on the production performance, immune system, and ruminal fermentation of dairy cows

  • Objective: The objective of this study was to examine the effects of alfalfa flavonoids on the production performance, immunity, and ruminal fermentation of dairy cows. Methods: The experiments employed four primiparous Holstein cows fitted with ruminal cannulas, and used a $4{\times}4$ Latin square design. Cattle were fed total mixed ration supplemented with 0 (control group, Con), 20, 60, or 100 mg of alfalfa flavonoids extract (AFE) per kg of dairy cow body weight (BW). Results: The feed intake of the group receiving 60 mg/kg BW of AFE were significantly higher (p<0.05) than that of the group receiving 100 mg/kg BW. Milk yields and the fat, protein and lactose of milk were unaffected by AFE, while the total solids content of milk reduced (p = 0.05) linearly as AFE supplementation was increased. The somatic cell count of milk in group receiving 60 mg/kg BW of AFE was significantly lower (p<0.05) than that of the control group. Apparent total-tract digestibility of neutral detergent fiber and crude protein showed a tendency to increase (0.05<$p{\leq}0.10$) with ingestion of AFE. Methane dicarboxylic aldehyde concentration decreased (p = 0.03) linearly, whereas superoxide dismutase activity showed a tendency to increase (p = 0.10) quadratically, with increasing levels of AFE supplementation. The lymphocyte count and the proportion of lymphocytes decreased (p = 0.03) linearly, whereas the proportion of neutrophil granulocytes increased (p = 0.01) linearly with increasing levels of dietary AFE supplementation. The valeric acid/total volatile fatty acid (TVFA) ratio was increased (p = 0.01) linearly with increasing of the level of AFE supplementation, the other ruminal fermentation parameters were not affected by AFE supplementation. Relative levels of the rumen microbe Ruminococcus flavefaciens tended to decrease (p = 0.09) quadratically, whereas those of Butyrivibrio fibrisolvens showed a tendency to increase (p = 0.07) quadratically in response to AFE supplementation. Conclusion: The results of this study demonstrate that AFE supplementation can alter composition of milk, and may also have an increase tendency of nutrient digestion by regulating populations of microbes in the rumen, improve antioxidant properties by increasing antioxidant enzyme activities, and affect immunity by altering the proportions of lymphocyte and neutrophil granulocytes in dairy cows. The addition of 60 mg/kg BW of AFE to the diet of dairy cows was shown to be beneficial in this study.

클로렐라 균주의 분리 동정 및 열수 추출물의 이화학적 조성 (Isolation and Identification of Chlorella sp. CMS-1 and the Chemical Composition of Its Hot Water Extract)

  • 클로렐라의 생리활성 물질을 활용하여 기능성 식품을 개발하기 위한 기초 자료를 얻기 위하여 클로렐라 배양장에서 생육이 빠른 클로렐라를 분리하여 동정하고 이화학적 특성을 조사하였다. 분리된 클로렐라는 전형적인 둥근 모양과 $3{\mu}m$ 크기로 주름진 형태를 취하고 있었다. 분리된 클로렐라의 동정을 위하여 18S rRNA sequences를 실시한 결과 Chlorella sp. IFRPD 1018와 99%의 높은 상동성을 보여 Chlorella sp.와 거의 일치하여 Chlorella sp. CMS-1로 명명하였다. 클로렐라 열수 추출물에 조단백질 함량이 59.21%로 많이 함유되어 있었고, 조지방 함량은 0.01%로 미량 함유되어 있었다. 클로렐라 CMS-1의 무기질 농도는 K 2.52%, P 2.25%, Mg 0.63%, Ca 0.63% 순으로 함유되어 있었다. 클로렐라 CMS-1의 주요 구성 아미노산은 glutamic acid 6.21%, alanine 5.76%, aspartic acid 5.44% 순이였으며, 특히 유리아미노산 중 ${\gamma}$-aminobutyric acid (GABA)의 함량은 7.13%로 전체 유리아미노산의 63.8% 비율로 차지하였고 그 다음으로 L-alanine 1.44%, L-glutamic acid 0.90%, L-leucine 0.26%, L-glycine 0.20% 순으로 함유되어 있었다.

자색(紫色) 고구마의 영양성분에 관한 연구 (Studies on the Nutritional Components of Purple Sweet Potato(Ipomoea batatas))

  • 고안토시아닌을 함유한 자색고구마의 식량자원으로서의 활용도를 높히고자 엷은 황색고구마와 동시에 영양성분을 분석하였다. 자색고구마의 일반성분 중 단백질 함량이 2.01%, 무질소물이 25.73%였으며, 조지방이 0.25%로 엷은 황색 고구마보다 높았다. 총아미노산의 구성은 2품종 모두 aspartic acid, serine, alanine 및 valine이 주성분이었으며, 자색고구마일 경우 함유황 아미노산이 제1제한 아미노산이었다. 지방산의 조성은 2품종 모두 palmitic acid, linoleic acid가 80% 이상으로 주종을 이루었으며, 불포화지방산 함량이 포화지방산 함량보다 더 높았다. 2품종의 고구마 모두 탄수화물 함량은 75%이상이었고 자색고구마의 중성당 함량은 67.22%로 이중 대부분은 glucose였다. 그리고 uronic acid는 11.88%, starch 59.42%였으며, 특히 자색고구마의 유리당은 엷은 황색고구마의 1/3 정도로서 고구마 고유의 단맛이 적었다. 비타민 $B_1$, 비타민 $B_2$ niacin 함량은 2품종 모두 큰 차이는 없었으나 비타민 C의 함량은 자색고구마가 63.4mg%, 엷은 황색고구마가 48.7mg%였다. 다음 무기질 구성원소로는 자색고구마일 경우 K, Mg, Ca, Na 등이 주성분이었고, 그외 Fe, Mn, Zn, Cu, Al 등이 확인되었다. 총식이섬유는 자색고구마가 13.43%로서, 이중 수용성식이섬유가 7.74%, 불용성식이섬유는 5.69%였으며 또 엷은 황색 고구마는 총식이섬유 함량이 9.79%로 수용성, 불용성 식이섬유 함량이 비슷하였다.

국민건강영양조사 식품섭취빈도조사지 개발: 국민건강영양조사 제4기 (2007-2009) 자료 이용 (Development of a Food Frequency Questionnaire for the Korea National Health and Nutrition Examination Survey: Data from the Fourth Korea National Health and Nutrition Examination Survey (KNHANES IV))

  • The objective of this study was to develop a semi-quantitative food frequency questionnaire (FFQ) for an adult population for use in the Korea National Health and Nutrition Examination Survey (KNHANES). The 24-hour recall data for 2,939 subjects aged 19 years and over from the fourth KNHANES first year (2007) were used to extract the items included in the questionnaire. The FFQ items were developed by selection of major dishes based on the cumulative contribution rate, between-person variability based on the cumulative R2 of energy and 14 nutrients (carbohydrate, protein, fat, crude fiber, calcium, phosphorous, iron, sodium, potassium, vitamin A, thiamin, riboflavin, niacin, and vitamin C) and the number of consumers of each dish. In addition, the FFQ items were revised with analysis of 24-hour recall data of the fourth KNHANES second and third year (2008, 2009). Finally, 112 items were included in the FFQ and grouped as follows: rice (5 items), noodles and dumplings (6), breads and rice cakes (8), soups and stews (12), soybeans, eggs, meat and fish (23), vegetables, seaweed and potatoes (27), milk and dairy products (4), fruits (13), beverages (5), snacks (6) and alcoholic beverages (3). The food items of FFQ accounted for an average of 87.0% of energy and 14 nutrient intakes and also accounted for 81.7% of the between-person variability. The frequency range of the FFQ items was classified into nine categories (never or seldom, once per month, 2-3 times per month, once per week, 2-4 times per week, 5-6 times per week, once per day, twice per day and three times per day) and the portion size was divided into three categories (small, medium and large). We expect that this developed dish-based FFQ could be used in assessment of longterm dietary intakes of Korean adults.

Essential oils as growth-promoting additives on performance, nutrient digestibility, cecal microbes, and serum metabolites of broiler chickens: a meta-analysis

  • Objective: The purpose of this meta-analysis was to evaluate the effect of dietary essential oils (EOs) on productive performance, nutrient digestibility, and serum metabolite profiles of broiler chickens and to compare their effectiveness as growth-promoting additives against antibiotics. Methods: Peer-reviewed articles were retrieved from Web of Science, Science Direct, PubMed, and Google scholar and selected based on pre-determined criteria. A total of 41 articles containing 55 experiments with 163 treatment units were eligible for analyses. Data were subjected to a meta-analysis based on mixed model methodology considering the doses of EOs as fixed effects and the different studies as random effects. Results: Results showed a linear increase (p<0.001) on body weight gain (BWG) where Antibiotics (FCR) and average daily feed intake decreased (p<0.001) linearly with an increasing dose of EOs. Positive effects were observed on the increased (p<0.01) digestibility of dry matter, crude protein, ether extract, and cecal Lactobacillus while Escherichia coli (E. coli) population in the cecum decreased (p<0.001) linearly. There was a quadratic effect on the weight of gizzard (p<0.01), spleen (p<0.05), bursa of fabricius (p<0.001), and liver (p<0.10) while carcass, abdominal fat, and pancreas increased (p<0.01) linearly. The dose of EOs linearly increased high density lipoprotein, glucose, protein, and globulin concentrations (p<0.01). In comparison to control and antibiotics, all type of EOs significantly reduced (p<0.001) FCR and tended to increase (p<0.1) BWG and final body weight. Cinnamaldehyde-compound was the only EOs type showing a tendency to increase (p<0.1) carcass weight, albumin, and protein of serum metabolites while this EOs together with EOs-Blend 1 decreased (p<0.01) E. coli population. Low density lipoprotein concentration decreased (p<0.05) with antibiotics and carvacrol-based compound when compared to the control group. Conclusion: This evidence confirms that EOs are suitable to be used as growth promoters and their economical benefit appears to be promising.
