토복령알코홀추출물이 토끼 적출 십이지장관운동에 미치는 영향 (Effect of Chinae Rhizoma on the Motility of Isolated Rabbit Duodenum)

  • Chinae Rhizoma is dried crude roots and rhizomes obtained from Smilax china Linne which belongs to Smilax Genus of Liliaceae Familia, and it has been in wife use as a folk remedy for a long time. But very little systemic studies on its pharmacological actions have been made until now. Furthermore, the effective components except saponin of Chinae Rhizoma have been not known yet. It came to the attention of the authors that Chinae Rhizoma are used as antidiarrhetics in Chinese herb medicine, therefore they attempted to observe the effects of Chinae Rhizoma Alcoholic Extracts (Chinae Rhizoma Ethanol Extract=CREE, Chinae Rhizoma Methanol Extract=CRME) from Smilax china Linne on motility of the isolated rabbit duodenum in this experiment. The results of the study were as follow: 1. The movement of the isolated rabbit duodenum was slowed down and its tonus was relaxed in proportion to the concentration of Chinae Rhizoma Alcoholic Extracts (CREE & CRME). 2. With a concentration of CREE $10^{-4}g/ml$, the contractile effects of acetylcholine $10^{-6}g/ml$, pilocarpine $10^{-5}g/ml$ and physostigmine $10^{-5}g/ml$ were remarkably blocked in the movement of isolated rabbit duodenum. 3. The concentration of the isolated rabbit duodenum induced by barium chloride $10^{-4}g/ml$ was significantly inhibited by CREE $10^{-4}g/ml$. 4. With the administrations of histamine $10^{-5}g/ml$, serotonin $10^{-6}g/ml$ and prostaglandin $A_2\;10^{-6}g/ml$ respectively, contractions were observed in the rabbit duodenum. But, treatment of CREE $10^{-4}g/ml$ inhibited markedly the tonus and motility of the duodenum pretreated with the above drugs. 5. The effects of CREE and CRME were very much alike in this experiment. From the above results, it is concluded that the mechanism of action of Chinae Rhizoma on motility of isolated rabbit duodenum was partially due to cholinergic blocking effect.

사마귀류 추출물의 생물학적 활성 비교 (Comparison of Biological Activities in Crude Extracts of Mantis)

  • 한의학에서 사마귀는 여러 치료의 목적으로 사용이 되는 한약재이다. 본 연구는 사마귀의 항산화활성과 산화스트레스에 대한 억제활성을 검증하기 위하여 실시하였다. 사마귀 3종에 대하여 항산화활성과 과산화수소에 의한 세포사멸 억제 효과, NO 활성 억제 효과를 실시하였다. DPPH, FRAP 활성은 사마귀와 왕사마귀 추출물에서 높게 나타났다. $H_2O_2$를 이용하여 산화스트레스를 유도한 후 이의 억제활성을 비교하여 본 결과 왕사마귀 methanol 추출물에서 $H_2O_2$에 의한 산화스트레스를 다소 억제하여 세포의 생존율을 높이는 것으로 나타났다. 염증반응을 유도하는 NO의 활성 정도는 좀사마귀와 왕사마귀의 DMSO 추출물에서 NO의 활성을 다소 억제하는 것으로 나타났다.

국내산 상지(桑枝)의 품질관리 연구 (Studies on quality control of domestic Morus alba Linne)

  • Objectives : Mori Ramulus is a young branch of Morus alba Linne, which has been used as traditional medicines for antidiarrhea, antibacteria, and antihypertension in Korea. With the basis on these medicinal activities, it is registered at the Korean Herbal Pharmacopoeia. The standard for quality control in use, however, is not yet validated.Methods : We experiment on heavy metals test, TLC, loss on drying, ash, acid-insoluble ash, contents of ethanolsoluble extracts and contents of oxyresveratrol by using the 15 samples of Mori Ramulus which are collected domestically. We tested items according to the General Test, processes and apparatus, Crude Drugs Test of KP(2011).Results : Purity test(Heavy metals) showed contents for Pb, As, Hg, and Cd range of 0.1~1.1 ppm, 0.0~0.2 ppm, 0.00~0.01 ppm, and 0.00~0.02 ppm. Identification test(Thin layer chromatography) was on comparing with morucine standard solution in Rf value, all samples showed morucine spot (dark blue, Rf : 0.3) as same as Rf value of morucine-standard solution. Loss on drying was ranged from 5.1 to 8.9 %, ash and acid-insoluble ash were between the range 3.0~6.3% and 0.3~0.8%. Contents of ethanol-soluble extracts was ranged from 6.2 to 12.0%. Content of oxyresveratrol was ranged from 0.01 to 0.16 % based on the oxyresveratrol standard curve.Conclusions : We sincerely hope that this study will be contributed to the standardization and quality control of Korean herbal medicines.

조미배합 조건과 과일농축물 첨가에 의한 노령 사슴육포의 품질특성 (Effect of Seasoning Condition and Fruit Extracts Level on the Quality Characteristics of Venison Jerky)

  • The principal objectives of the experiments in this study were to assess the quality of venison jerky made from Elk deer (female, weight; 380 kg, age; 15 old years) in accordance with various seasoning sauce and mixture fruit extract level. Salinity, saccharinity and acidity values of the seasoning sauces were all high(VJ-1; 3.72%), (VJ-3; 3.95%), (VJ-1; 0.35%). The salinity range of the venison jerky products was 5.65~5.92%, the saccharinity range was 3.75~4.18%, and the seasoning sauce samples resulted in a significant difference in acidity values(p<0.05). Crude protein, fat and ash contents of the venison jerky products satisfied the Korean jerky standard value, but the moisture content range was 36.18~38.59%, and this was also in excess of the jerky standard level. The manufacturing yield was 42.24~44.48%, with VJ-1 at lowest levels followed by VJ-2 and VJ-3. The pH value of VJ-1 was high, but not significantly higher than the other samples(p>0.05). The water activity value range was measured at 0.68~0.71, and the lightness(L) and redness(a) value evidenced marked increases with increased amounts of added sodium nitrite(p<0.05), and the yellowness(b) values did not significantly differ(p>0.05). The overall sensory acceptance scores ranged between 5.53~7.49, in the order VJ-3, VJ-2, VJ-1(p<0.05).

Feeding Value of Sugarcane Stalk for Cattle

  • A metabolism trial with four castrated male Brahman cattle, average body weight 320 kg, was conducted in order to determine the nutritive value of chopped sugarcane stalk (CSS) for the establishment of feeding strategy in the dry season in Northeast Thailand. Animals were subjected to the following four dietary treatments: Treatment 1; 100% of CSS, Treatment 2; 70% of CSS and 30% of commercial complete feed (TMR), Treatment 3; 40% of CSS and 60% of TMR, and Treatment 4; 100% of TMR. The average CP, ether extracts, nitrogen free extracts, crude fiber and ash contents of CSS were 2.0, 0.9, 79.0, 16.1 and 2.2%, respectively. Although the amount of feed given was approximately at maintenance level, animals in treatments 1 and 2 refused a part of feed. The metabolism trial revealed that total digestible nutrient and metabolizable energy contents of CSS were 61.5% and 9.04 MJ/kgDM, respectively, when it was properly supplemented with protein sources. Nutritive value of CSS was lowered when animals were given CSS solely. This was due to the large loss of energy into urine and methane. Voluntary intake of CSS in cattle was not enough to satisfy energy requirement for maintenance. The CSS can be used as a roughage for feeding cattle in the dry season with proper supplementation of protein and energy.

Wild Mushrooms: A Potential Source of Nutritional and Antioxidant Attributes with Acceptable Toxicity

  • This paper describes in detail proximate composition, nutritional profile, phytochemical constituents, antioxidant activities, antimicrobial potential, and antihemolytic activity (towards human erythrocytes) of various fractions of wild Ganoderma lucidum. Proximate analysis established that wild G. lucidum comprises about $87.02{\pm}5.45%$ of moisture, and the remaining part is a rich source of proteins ($8.59{\pm}0.37%$), crude fiber ($54.21{\pm}1.2%$), and carbohydrate (35.16%) with smaller fat content (3.33 %). Similarly, phytochemical screening revealed the presence of flavonoids ($217.51{\pm}0.30mg/g$), ascorbic acid ($116{\pm}7.32mg/g$), phenolics ($360.72{\pm}34.07mg/g$), ${\beta}$-carotenes ($0.42{\pm}0.04{\mu}g/g$), and lycopene ($0.05{\pm}0.00{\mu}g/g$). Extracts of wild G. lucidum in various solvents provided first line protection against Escherichia coli and Pasteurella multocida in the order of ethyl acetate> ethanol> methanol> n-hexane> water. Furthermore, aqueous and methanolic extracts of wild G. lucidum were found to be safe towards human erythrocytes. Overall, wild mushroom (G. lucidum) was found to be a good source of dietary supplements, antimicrobial and antioxidant agents in the pursuance of its commercial utilization in food and pharmaceutical industries.

비늘버섯류의 추출수율 및 영양 약리학적 특성 (Extraction yield and nutraceuticals of mushroom Pholiota species)

  • 본 연구에서는 비늘버섯을 이용하여 비교적 가격이 저렴하고 부작용이 없는 우수한 건강 기능성 제품을 개발하기 위한 기초 자료를 얻고자 17종의 비늘버섯 자실체 분말을 각종 유기용매로 추출한 후 이들의 수율을 조사하였고 여러 가지 영양학적, 생리기능적 성분 함량을 조사하였다. 17종의 비늘버섯 자실체들의 물과 메탄올 추출물을 제조하여 이들의 추출 수율을 측정한 결과 Pholiota adiposa ASI 24027의 물 추출물이 58.7%로 가장 높았다. 비늘버섯 자실체의 조단백질 함량은 대체로 0.2%-25.0% 이었고 이중 Pholiota adiposa ASI 24015가 가장 많은 단백질을 함유하고 있었다. 무기물 중에는 K를 많이 함유하고 있었고 철과 인은 Pholiota sp. ASI 24024의 자실체에서 각각 100g 건조 자실체량 19.6 mg, 859.1 mg 을 함유하고 있었다. 항암성분으로 알려진 ${\beta}$-glucan은 Pholiota adiposa ASI 24005에서 약 0.66%로 비교적 많이 함유하고 있었으나 비타민과 아미노산 및 지방산 등은 비늘버섯 시료간에 큰 차이가 없었다.

Nutritional composition and antioxidant activity of pink oyster mushrooms (Pleurotus djamor var. roseus) grown on a paddy straw substrate

  • Pleurotus djamor var. roseus is an edible mushroom isolated from the wild and cultivated on paddy straw substrates. The present study was carried out to compare the nutritional composition and antioxidant properties of P. djamor var. roseus at different growth stages (primordia, basidiomata, and mycelia). The protein content was is in the range of 31.48 to 35.50 g/100g dw. The crude fiber content ranged from 8.0 to 14.60 g, and that of total carbohydrates ranged from 44.75 to 48.90 g. Sodium, magnesium, and calcium reached the maximum levels in basidiomata, and selenium was detected in basidiomata and mycelia (0.47 - 0.22 mg/Kg). The amino acid profile showed that all essential and nonessential amino acids and glycine showed maximum levels in basidiomata and 15.98 ± 0.01 g/100g. The fatty acid profile showed the presence of saturated and unsaturated fatty acids; the unsaturated fatty acid content was maximum in all of the samples, ranging from 76 - 40.41%. The total phenol and flavonoid contents as well as the scavenging (DPPH), ferric thiocyanate (FTC), and thiobarbituric acid (TBA) activities in the context of methanol and water extracts from primordia, basidiomata, and mycelium were determined. Among them, basidiomata and mycelial methanol extracts exhibited significant antioxidant activity. Overall, these findings show that P. djamor var. roseus can be used as a functional food for daily consumption.

해양심층수로 배양한 해양미세조류 Tetraselmis sp. JK-46의 성분 조성 및 생리활성 (Physiological Properties of Extracts and the Chemical Composition of Tetraselmis sp. JK-46 Cultured with Deep Seawater)

  • This study examined Tetraselmis sp. JK-46 isolated from seawater from the East Sea. Deep seawater (DSW) had a greater effect on the growth of Tetraselmis sp. JK-46 than surface seawater (SSW). The crude protein, lipid, carbohydrate and ash contents of Tetraselmis sp. JK-46 cultured with DSW were 27.2, 37.1, 13.2 and 26.3 %, respectively, and these values were similar to the results for samples cultured with SSW. The contents of Mg, Ca, Fe and K in the DSW cultured samples were 7080.3, 1009.6, 251.2, and 2749.7 mg/100 g, respectively. The fatty acid compositions of Tetraselmis sp. JK-46 cultured with DSW and SSW were 53.7 and 49.0 % polyunsaturated fatty acids (PUFA) and 25.7 and 30.7 % saturated fatty acids (SFA), respectively. The total amino acid contents of the samples cultured with DSW and SSW were 7392.6 and 6376.0 mg/100 g respectively. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of Tetraselmis sp. JK-46 extracts increased with the concentration of the chloroform and ethyl acetate fractions. The half maximal inhibitiory concentrations ($IC_{50}$) of the chloroform and ethyl acetate fractions of DSW and SSW cultured samples were 1.2 and 2.6 mg/mL, and 3.1 and 3.3 mg/mL, respectively. The ethyl acetate fractions of DSW and SSW cultured samples has anticoagulant activity and the activated partial thromboplastin times (APTT) were 93.4 and 89.3 sec., respectively. The chloroform and ethyl acetate fractions showed antimicrobial activity against Bacillus subtilis, Escherichia coli and Candida albicans.

Antimicrobial Activity of Mulberry Leaf against Mutans Streptococci and Periodontopathogens

  • This study investigated the antimicrobial activity of methanol extract of mulberry leaf against 16 strains of mutans streptococci and four species of periodontopathogens: Porphyromonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum, and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of the crude extracts or silica gel chromatography fractions of methanol-extracted mulberry leaf were evaluated by determining minimal inhibitory concentrations using an established microdilution method. The cytotoxicity of the extracts of mulberry leaf on KB cells was tested by the methyl thiazolyl tetrazolium assay. Chromatography fraction 12 displayed the most potent antimicrobial activity against all 16 strains of mutans streptococci, P. gingivalis, and P. intermedia. No KB cell cytotoxicity was evident up to $128{\mu}g/ml$ of fraction 12. The methanol extract had no antimicrobial activity against F. nucleatum and A. actinomycetemcomitans. These results suggest chromatography fraction 12 methanol extract of mulberry leaf could be useful in the development of oral hygiene products, such as dentifrice and oral hygiene solution, for the prevention of dental caries.
