Inhibition of N-nitrosodimethylamine Formation of Extracts from Citrus Seeds (감귤류 종자 추출물의 N-nitrosodimethylamine 생성 억제에 관한 연구)

  • The effect of inhibition on N-nitrosodimethylamine (NDMA) formation by methanol extracts from 6 kinds of citrus seed (Citrus sunki, Citrus natsudaidai, Citrus sulcata, Citrus tangerina, Citrus grandis and Citrus obovoidea) were investigated. The contents of moisture, crude protein, crude fat, carbohydrate, ash, flavonoid and total phenol in citrus seeds were 4∼6, 11∼15, 32∼46, 22∼45, 2∼4%, 12∼24mg% and 53∼133mg%, respectively. The solid contents and yield of citrus seed extracts were 0.8∼1.0 and 0.7 ∼ 1.1%, respectively. Nitrite-scavenging activity by methanol extracts from citrus seeds increased with increasing extract dosage. Furthermore, the nitrite scavenging activity was pH dependent being the highest at pH 1.2 (42.7∼96.9%) and the lowest at pH 6.0 (19.9-62.6%). Scavenging effects of nitrite by reaction time showed high effects under 3hr reaction time. The inhibition effect of NDMA formation by the citrus seed was strengthened as the amount of extract increased. The inhibition rate of methanol extracts from citrus seeds on NDMA formation showed 1.2∼39.8%, 21.3∼60.1% and 47.4∼94.0%, according to add 1, 3 and 5$m\ell$, respectively. Therefore, the inhibition effect of NDMA formation by the citrus seed was strengthened as the concentration of extract increased.

Anti-invasion Effects of Calystegia soldanella Solvent Extracts and Partitioned Fractions on PMA-stimulated Fibrosarcoma Cells (갯메꽃 추출물과 유기용매 분획물의 암전이 억제 효과)

  • Calystegia soldanella is distributed in coastal sand dunes and has high environmental adaptability; it is also known to be effective for anti-oxidant, anti-pyretic, anti-septic, and diuretic action. This study investigated the effect of crude extracts and organic solvent fractions of C. soldanella on MMP-2 and MMP-9 expression, MMP activity, and cell mobility in phorbol-12-myristate-13-acetate (PMA)-induced fibrosarcoma HT-1080 cells. C. soldanella was twice extracted, once with methylene chloride (MC) and once with methanol (MeOH). After the MC and MeOH extracts were combined, their suppressive effects on MMP-2 and MMP-9 expression, MMP enzymatic activity, and gene and protein expression were measured by gelatin zymography, enzyme-linked immunosorbent assay, reverse-transcription polymerase chain reaction, and western blot method. Cell mobility for the HT-1080 cells was observed by wound healing assay. The combined crude extracts showed a significant suppressive effects on MMP-2 and MMP-9 expression. To explore active inhibitory elements, the combined extracts were fractionated according to polarity into with n-hexane, 85% aqueous methanol, n-butanol, and water. Across these four solvent fractions, MMP-2 and MMP-9 activity and cell mobility in the HT-1080 cells were all strongly inhibited by the n-hexane fraction. These results suggest that C. soldanella extract and organic solvent fractions could be used as potent MMP inhibitors for effective anti-cancer treatments to suppress cancer invasion and metastasis.

Immunohistoehemical Observation on the Antigens Inducing IgG and IgM Antibodies against Sparganum (IgG와 IgM 항체를 유도하는 sparganum의 항원에 관한 면역조직화학적 및 전기영동에 의한 연구)

  • Localization and characterization of the antigenic components of sparganum which induced IgG and IsM antibodies in the host were studied by immunohistochemical techniques and SDS-PAGT and Western blotting. The antigen recognized by IgG antibody of rats or mice which were immunised by infection or injection of crude extracts of metacestodes of Spirometra erinacei, was located in the parenchyme of sparganum, especially at the cortex and around the calcareous corpuscles. The immunoreaction was demonstrated not only in the encysted fibrous wall of host but around the arterioles or venules in the connective tissue of host. The antigen recognized by IgM antibody of rats or mice was also observed in the parenchyme of sparganum and in the connective tissue of host. By 5∼20% gradient SDS-PAGE and EIBT, we detected antigenic components by IgG and 1gG antibodies of the rat or mouse immunized by infection or injection of crude extract of spargana. Twenty-three antigenic bands from crude extracts of spargana were recognized by IgG antibody and 15 components by IgM antibody of immunized rats. Out of the bands recognized by IgG and IgM antibodies, 15 were cross-reacted each other. Twenty components of eBlcretory-secretory proteins from spargana were recognized by IgG, and 5 components by IgM antibody of immunized rats. By IgG and IgM antibodies of immunized mice, 16 components of crude extracts were recognized by IgG antibody and 9 components by IgM antibody. Twenty components of excretory-secretory preparation were recognized by IgG antibody and 5 components by IgM antibody. Thirteen components of crude extracts were cross-reacted by IgG antibody of rats and mice.

Effects of Yucca Extracts and Protein Levels on Growth Performance and Nutrient Utilization in Growing Pigs

  • A total of 120 pigs were used to investigate the effects of yucca extracts on the growth performance, nutrient digestibility and excretion of growing pigs fed different levels of dietary protein. Pigs were allotted into $2{\times}3$ factorial design by the supplementation with yucca extract (YE, 0 and 120 mg/kg) and 3 levels of dietary protein (16, 18, 20%). During the whole experimental period (18 to 52 kg body weight), there were no significant differences in ADG, ADFI or F/G by YE addition or different protein levels among treatments (p>0.05). Overall, although addition of YE to the diet and elevation of protein level showed better ADG, there were no significant differences in growth performance among treatments. Pigs fed diets with YE showed significantly (p<0.05) higher dry matter (DM), crude ash (CA) and crude protein (CP) digestibility than did the others during the growing period. Concerning the levels of dietary protein, only the CP digestibility was significantly higher in pigs fed high protein diet. Pig fed the low protein diet without YE showed a significantly low CP digestibility (p<0.05). No significant differences were found in crude fat (CF), calcium (Ca) and phosphorus (P) digestibilities regardless of YE supplementation or dietary protein levels. Pigs fed YE supplemented diets showed significantly (p<0.05) higher amino acid digestibility. Also, high CP level diets showed a higher amino acid digestibility than low CP diets (p<0.05). DM and N excretion did not show any significant differences among treatments, there was a slightly lower excretion with increase in dietary protein level. Supplementation with YE significantly decreased the DM and N excretion. Interaction (YE$\times$protein) was found in P excretion. Pigs fed a medium protein diet without YE showed the lowest P excretion during the growing period. The NH3-N content in the feces tended to be increased by the increased dietary protein levels and with YE supplementation. During the whole experimental period, the cost for YE supplementation was similar to value of the improvements of performance obtained. The cost of feeding high level protein was significantly higher than that of medium level protein by 10% and low level protein by 9% (p<0.05). It could be concluded that the effects of dietary protein level and yucca extract on growth performance, nutrient digestibility and excretion might play a role to some extent in growing pigs from the aspect of pollution control.

Purification of the Yolk Protein, and Identification of the Synthetic Site of Its Precursor in Eriocheir japonicus (Decapoda, Brachiura) (동남참게(Eriocheir japonicus)의 난황단백질 정제와 그 전구체의 합성부위 구명)

  • To identify the histological site of synthesis of yolk protein precursor, vitellogenin, by immunocytochemical method in the freshwater crab Eriocheir japonicus, we purified the yolk protein, vitellin, from crude egg extracts, and prepared the anti-rabbit serum against vitellin. Then, the site of vitellogenin synthesis was demonstrated by immunotytochemical method with PAP(peroxidase-antiperoxidase) reaction using the rabbit antiserum aganist vitellin. Female specific serum protein was identified in female serum by immunoelectrophoresis and Ouchterlony's immunodiffusion test for mature male and female sera. Based on the immunoelectrophoresis and Ouchterlony's diffusion test for mature male and female sera and crude egg extracts using antiserum against vitellogenic female serum absorbed with male serum, the female specific serum protein was identified as vitellogenin, detected in female serum only. The major yolk protein, vitellin, was purified from the crude egg extracts by DEAE-cellulose ion exchange chromatography, followed by sepharose CL-4B gel filteration chromatography. The molecular weight of vitellin was estimated to be about 245,000 dalton by sepharose CL-4B gel filteration chromatography. from the results of immunological analysis for vitellin, it was found that the vitellin antiserum contained the antibody against vitellogenin. In the results of immunocytochemical reaction by PAP method with the rabbit antiserum against vitellin, the vitellogenic oocytes and the hepatopancreas of mature female showed positive PAP reaction, but not in follicle cells and previtellogenic oocytes nf ovary, muscle of female and mature male hepatopancreas. Therefore, it showed that the hepatopancreas of mature female is the site of vitellogenic synthesis.

Physicochemical Properties and Antioxidant Activities of Steam-Dried Allium hookeri Root (증포 처리한 삼채 뿌리의 이화학적 특성 및 항산화 활성)

  • This study was carried out to investigate the effect of steam-drying method on physicochemical properties and antioxidant activities of Alliun hookeri root (AHR). Moisture, crude protein, crude fat, and crude ash contents of raw and steam-dried AHRs were 10.91~14.15%, 11.14~13.49%, 0.83~3.02%, and 7.55~8.98%, respectively. Sulfur contents of steam-dried AHRs were 2.0 and 2.2 times lower than that of raw AHR (0.51%), respectively. pH and total sugar contents of AHRs were reduced by steam-drying, whereas titrate acidity and browning intensity were increased. The L and b values of AHRs in Hunter's value were also reduced, but a value was increased by steam-drying. Among hot water extracts from raw and steam-dried AHRs, four times steam-drying showed the lowest $EC_{50}$ values (0.44, 9.01, and 0.48 mg/mL, respectively) in DPPH radical assay, ABTS radical assay, and reducing power, whereas four times steam-drying had the highest total phenolic content ($34.47{\mu}g/mg$) and browning intensity (2.05 and 0.20 at 280 and 420 nm, respectively). The antioxidant activities of hot water extracts from raw and steam-dried AHRs were closely correlated with their total phenolic contents and browning intensity, showing coefficient of determination ($R^2$) values higher than 0.87. From the results, we suggest that steam-drying method could be used as an effective process for increasing the antioxidant activity of AHR.

Analysis of Nutritional Components and Evaluation of Functional Activities of Sasa borealis Leaf Tea (조릿대 잎차의 영양성분 분석 및 기능성 평가)

  • In this study, the nutritional components and functional activities of Sasa borealis leaf tea were evaluated. The proximate compositions were as follows; moisture 5.68%, crude protein 16.38%, crude fat 4.68%, nitrogen free extracts 32.37%, crude fiber 32.36%, and ash 8.53%, respectively. The mineral elements were as follows: K 2,133.83, Ca 1,144.09 and P 543.00 mg%, respectively. The amino acid contents of the Sasa borealis leaf tea were very rich in proline (1,275.26 mg/100 g) and deficient in cystine (71.49 mg/100 g). The major fatty acid components were linoleic acid (50.52%), palmitic acid (18.52%), and oleic acid (14.16%). Finally, based on our sensory evaluations, the $80^{\circ}C$ extracted Sasa borealis leaf tea evidenced the best overall quality. The contents of total phenol and total flavonoids of the 80% methanol and hot water extracts were 15.09, 7.69 mg/g and 12.03, 6.12 mg/g, respectively. The DPPH and $ABTS^+$ radical scavenging activities of the 80% methanol extract from Sasa borealis leaf tea were 86.87% and 83.85% at a concentration of 1.25 mg/mL. The 80% methanol and hot water extracts evidenced reducing power and inhibitory effects against acetylcholinesterase in a dose-dependent manner.

Effect of the DSSE(Defatted Sesame Seed Extracts) on Quality Chracteristis of Soybean Sprouts (콩나물의 품질에 미치는 탈지참깨박 추출물의 영향)

  • Quality improvements of soybean sprouts on DSSE(Defatted Sesame Seed Extracts)-treated cultivation were determined in terms of growth rate, nutrient compositions value and the total bacteria. The length, diameter and weight of treated soybean sprouts were increased by 6.2%, 10.0% and 9.1%, respectively, compared with control. The contents of moisture, crude ash, crude fat of the treated soybean sprouts were similar to the control soybean sprouts, whereas crude protein content of the treated sprouts was 17% higher than the control. Both soybean sprouts contained 17 amino acids but the treated sprouts contained the higher contents of glutamic acid, threonine, methionine, tyrosine and valine. Little difference in fatty acid composition was noted between the two soybean sprout samples. Vitamin C contents of the treated soybean sprouts were 18.2 mg%, which corresponds to about 50% more vitamin C than the control. The treated soybean sprouts contained higher contents in minerals such as K, P and Ca than those of the control. The total bacteria of soybean sprouts in the control soybean sprouts increased rapidly after 2-day storage, but treated soybean sprouts increased immediately after 4-day storage.

Antioxidant activities of Rhus verniciflua seed extract and quality characteristics of fermented milk containing Rhus verniciflua seed extract (옻씨 추출물의 항산화 활성과 옻씨 추출물을 첨가한 발효유의 품질특성)

  • This study was carried out to investigate the quality characteristics of fermented milk containing Rhus verniciflua seed (RVS) extracts (0.0%, 0.1%, 0.3% and 0.5%) and antioxidant activity of RVS extracts. The proximate composition of RVS was moisture (4.76%), crude ash (1.40%), crude fat (5.33%) and crude protein (11.10%). A total polyphenol content of 70% ethanol extract of RVS ($554{\pm}2.64mg/g$) was higher than that of RVS water extract ($145{\pm}3.47mg/g$). 70% ethanol extract ($1103{\pm}6.42mg/g$) of RVS showed higher content in the total flavonoids ($37{\pm}2.30mg/g$) and activities on DPPH free radical scavenging (73.23%) and ABTS free radical scavenging (83.47%) when compared with water extracted of RVS. The quality characteristics, such as pH, titratable acidity, and the number of lactic acid producing bacteria were not remarkably different between the fermented milk samples subjected to treatments with and without the addition of RVS extracts.

Accumulation of Crude Lipids, Phenolic Compounds and Iron in Rusty Ginseng Root Epidermis (적변삼 외피에서 지질, 페놀성물질 및 철 성분의 축적에 관한 연구)

  • The study was conducted to investigate the tissue and chemical characteristics of rusty root epidermal cells. In histological study, the rusty symptoms were frequently observed in the epidermis of ginseng root and to be yellow under microscopic observation. Disks of the epidermal cell tissue of the rusty root were usually 2 and 3 times greater in the number of cell layer and thickness of cell wall than the healthy root, respectively. The color degree of methanol extracts from the rusty root epidermis was 5.5 times higher than that of the healthy root. And the extracts of rust matter in the root epidermis were easily dissolved in polar solvents compared to nonpolar solvents. UV-absorption spectra of methanol extracts in various fractions of phenolics showed a maximum peak between 275∼280 nm. The crude lipids and phenolic compounds such as acid insoluble bound phenolics, acid insoluble esterified phenolics, acid insoluble condensed phenolics, insoluble bound phenolics and free phenolics were also more in the rusty root epidermis than in the healthy one. Fe content in the rusty root epidermis was 2.7 times higher than that of healthy one. It was presumed that the phenolic compounds(precursor of the rusty) in association with lipid and iron in the root epidermis might defence the root when ginseng root was depressed by the unfavorable conditions in soil and/or portions of a root system were subjected to anoxic conditions.
