Mapping of Quantitative Trait Loci Affecting Growth Traits in a Japanese Native Chicken Cross

  • The Hinai-dori is a breed of chicken native to Akita Prefecture, Japan. An $F_2$ resource population produced by crossing low- and high-growth lines of the Hinai-dori breed was analyzed to detect quantitative trait loci (QTL) for growth traits. Highly significant QTLs for body weight at 10 and 14 weeks of age and average daily gain between 4 and 10 weeks and between 10 and 14 weeks of age were accordingly mapped in a common region between ADL0198 and ABR0287 on chromosome 1 and between MCW0240 and ABR0622 on chromosome 4, respectively. A significant QTL for body weight at 4 weeks of age and a significant QTL for average daily gain between 0 and 4 weeks of age were mapped for the first time to the same region flanking ABR0204 and ABR0284 on chromosome 1. These QTLs are good candidates for application in the development of marker-assisted selection strategies for increasing growth efficiencies in the Hinai-dori breed and native breeds of chickens in Asia.

Development of a Segregating Population with Biological Efficiency in Agaricus bisporus

  • In this study, we made a population with high biological efficiency (BE) to investigate the complex genetic architecture of yield-related traits in Agaricus bisporus. MB-013 crossed between bisp 015-p2 and bisp 034-p2, had high BE. Additionally MB-013 was an intervarietal hybrid that intercrosses with A. bisporus var. burnettii, bisp 015, and A. bisporus var. bisporus, bisp 034. One hundred and seventy homokaryons were selected using the cleaved amplified polymorphic sequence (CAPS) markers (PIN primer/HaeIII) from 300 single spore isolates (SSIs). One hundred $BC_1F_1$ hybrids were obtained by crossing the homokaryons of MB-013 with bisp15-p1. The population of 100 BC1F1 hybrids is suitable for analyses of BE.

  • This study is to develop new wheat breeding material through wide hybridization with wild species harboring useful characteristics such as salt, heat, and drought tolerance. Leymus mollis, wild rye was used to improve wheat genetic quality. L. mollis, is a perennial plant harboring tolerance against salt, heat, and drought because L. mollis distributes on the seaside. The F1 hybrids were produced by crossing between common wheat (Triticum aestivum L., Chinese Spring) and L. mollis. Genomic in situ hybridization revealed that the F1 hybrids have L. mollis genome. For the evaluation of salt and drought tolerance, seeds from the F2 were used. Under 2% NaCl solution, the F3 wheat-Leymus addition plants with salt tolerance showed more tillering and longer roots than other F3 plants without salt tolerance. Also, the F3 plants with salt tolerance showed better shallow-rooted than other F3 plants without salt tolerance. Finally, the F3 plants with salt tolerance made seed-setting under 2% NaCl condition, but other F3 plants without salt tolerance were not. Under drought conditions, the F3 plants with drought tolerance showed longer culm and spike length than other F3 plants without drought tolerance and even those of Chinese Spring under well-water conditions. We evaluated and selected the F3 plants with salt or drought tolerance for generation advancement.

Phenotypic and Genetic Effects of Dwarfing Genes on Plant Height and Some Agronomic Traits in Wheat

  • Wheat is one of the most widely grown food crops worldwide. Extreme precipitation and wind disturbances increased due to the abnormal climate, which resulted in increased lodging. Introduction of dwarf genes in wheat significantly increased lodging resistance and productivity in wheat breeding. In this study, we performed the genotyping of dwarfing genes between 'Keumkang' and 'Komac 5' ('Keumkang' mutant). In addition, we investigated the relationship between plant height and several phenotypic characters using F2 segregation populations derived from crosses between the two varieties. There was no significant difference in phenotypic characters between the two varieties except for plant height. In the genotyping analysis using dwarfing genes, mutations of two dwarfing gene were found to be induced between the two varieties. The four genotypes of the F2 populations from a crossing between 'Keumkang' and 'Komac 5' were used to compare and evaluate the effects of two dwarfing genes. Plants with two single mutant dwarfing gene and double mutant dwarfing gene revealed reduced plant heights than control plants by 4.5%, 6.9%, and 33.2%, respectively. The phenotype analysis showed that double mutant dwarfing gene affected wheat stem growth as the length decreases from the second node, resulting in decreased plant height. However, there were no significant differences in the agronomic traits between mutant plants and control plant. These results may provide novel information about the effect of double mutant dwarfing gene on plant height, and may help improve lodging tolerance and wheat yield.

오렌지색 중.소륜 절화용 다수성 거베라 신품종 '스위티' 육성 (Breeding of New, Orange Mini/Medium Type, High Yielding Cut Flower Gerbera hybrida 'Sweety')

  • 국내 재배환경에 적합한 다수성인 거베라 소륜계통의 우수 신품종 육성을 위하여 경남농업기술원 화훼육종연구소에서 1999년부터 교배조합 육성을 위하여 국내의 재배농가와 종묘업체로부터 품종을 수집한 후, 2001년 3월에 핑크색 대륜계의 'Grandeur'를 모본, 오렌지색 중 대륜계의 'Songsongee'를 부본으로 인공교배를 실시하여 교배조합으로부터 436개의 종자를 획득하였다. 교배 후 결실이 된 조합은 채종 즉시 파종하였으며, 파종 1개월 후 발아된 378개의 유묘는 본엽이 4~5매 전개되었을 때 화훼육종연구소 유리온실 내의 선발 포장에 정식하였다. 그 후 교잡 1세대를 육묘하여 화색이 선명하고 수량이 많은 우수 5개체(01129-028, 105, 188, 325, 335)를 1차로 선발하였다. 그 후 특성이 우수한 01129-335(경남 G-24)개체를 생장점배양에 의해 개체증식 후 화훼육종연구소 유리온실내의 특성검정 포장에 정식하여 2003년 6월부터 2004년 10월까지 3회에 걸쳐 생육 및 수량특성 검정을 실시하였다. 특성검정을 실시한 결과 화색, 화형, 수확량 등 품질과 수량이 우수하다고 판단되어 2005년 농작물 직무육성 신품종선정위원회의 심의를 거쳐 'Sweety'로 명명하였다. 'Sweety'는 화색이 선명한 오렌지색의 반겹꽃 절화용으로 수량이 많고 안정된 중 소륜화로서 화색과 화심색이 잘 조화된 기호도가 높은 품종이다. 본당 수량은 연간 3회 검정 결과 평균 51.2개이며, 평균 절화수명은 10.4일이다.

  • '핑키' 품종은 경남농업기술원 화훼육종연구소에서 2006년도에 육성한 절화용 스프레이국화 신품종이다. 이 품종은 '02년 10월에 분홍색 홑꽃 품종인 'Rima'를 모본, 백색 홑꽃 'Yen'을 부본으로 인공교배를 실시하여 497개의 종자를 획득하였다. 채종 즉시 파종하였고 파종 10일 후 발아된 유묘는 플러그 트레이(200공) 에 옮겨 심고 1개월 정도 육묘하여 화훼육종연구소 비닐온실내의 선발 포장에 정식하였으며, 그 후 초세가 좋고 화색이 선명하며, 조기 개화하는 우수 5개체(RIY02-89, 106, 208, 294, 413)를 1차('03년)로 예비선발 하였다. 2004년부터 2006년도에 걸쳐 매년 3.15(촉성), 7.27(자연단일), 9.11(억제) 에 정식하여 각각 3회에 걸친 특성검정과 품평회를 통해 생산자 및 소비자 기호성 평가를 받은 결과 기호성이 좋고 화색 및 화형 등 절화특성이 우수한 RIY02 -208(경남CS-3호)계통을 최종 선발하여 2006년 농작물 직무육성 신품종선정위원회의 심의를 거쳐 'Pinky'로 명명하였다. 품종은 선명한 분홍색 홑꽃의 스프레이국화로서 개화 소요기간이 6.5주로 아주 짧으며, 주당 평균 착화수도 26.8개로 다화성이다. 꽃 크기는 3.1 cm로 소륜이며, 초장이 93.8 cm인 산티니계 스프레이국화로서 평균 절화수명은 20.9일이다.

흰가루병에 강하고 다수성인 장미 적색 품종 '해도지' 육성 (Breeding of New Rose Cultivar 'Haedoji' with Red Color Flower, Resistance to the Powderly Mildew and High Productivity)

  • 절화장미 '해도지' 품종은 전라남도농업기술원에서 육성한 적색 스탠다드 신품종으로 절화수명이 우수한 'Vital'을 모본으로, 화색이 선명한 적색 'Cardinal'를 부본으로 하여 2003년에 인공교배 하였다. 화형과 화색이 예쁘고 꽃잎수가 적합한 2계통을 2004년 1차 선발 하였다. 2005년부터 2007년까지 특성 및 생산력검정을 실시하여 적색이며 묘 양성이 쉽고 줄기가 곧으며 비교적 흰가루병에 강한 특성을 가지고 있으며 수량성도 우수한 '해도지' 품종을 육성하였다. 육성된 품종은 스탠다드 중형계 품종으로 꽃크기가 9.2 cm, 절화장이 62 cm, 절화수명이 9.5일로 줄기가 곧고 흰가루병에 강하고 절화수명이 우수하며 절화수량은 3년 동안의 검정결과 167본/$m^2$/년으로 대비품종 '티아모' (164본/$m^2$/년)에 비해 2% 증가하였다.

Breeding of New Productive Bivoltine Hybrid, CSR12 $\times$ CSR6 of Silkworm Bombyx mori L.

  • With an objective of evolving quantitatively and qualitatively superior bivoltine breeds/hybrids of silkworm Bombyx mori L. for tropical conditions, breeding work was initiated in Central Sericultural Research and Training Institute, Mysore during 1992 by utilizing two Japanese hybrids namely BNl8$\times$BCS25 and Shunrei$\times$Shogetsu along with Indian evolved breed, KA. The breed CSR12 which is characterized with sex-limited larval marking and white oval cocoons was evolved from the Japanese hybrid BNl8 ${\times}$ BCS25 by crossing with KA, while the breed CSR6 which is characterized with normal marking (marked larvae) and white dumbbell cocoons was extracted from the Japanese commercial hybrid Shunrei${\times}$Shogetsu through continuous inbreeding coupled with selection. After fixation, these breeds along with other newly evolved breeds were subjected to hybrid study under optimum environmental conditions in the laboratory for expression of full potential of the genotypes. These hybrids were evaluated by Multiple Trait Evaluation Index (Mano et al., 1993). The hybrid CSR12${\times}$CSR6 was selected based multiple trait evaluation index value. The hybrid CSR12$\times$CSR6 recorded survival of 96.0%, shell weight of 50.0 cg, shell ratio of 24.3%, raw silk percentage of 19.6, filament length of 1,216 m, boil off loss of 22.4% and renditta of 5.1. On the other hand, the control hybrid (KA ${\times}$ NB4D2) has recorded survival of 90.6%, shell weight of 42.1 cg, shell ratio of 20.4%, raw silk percentage of 15.9, filament length of 999 m, boil off loss of 24.8% and renditta of 6.3. The hybrid CSR12$\times$CSR6 was authorized during 1997 by Central Silk Board, Government of India for commercial exploitation during favourable months based on national level race authorization test.

좀비비추[Hosta minor (Baker) Nakai] '늘푸름 1' 육성 (New Cultivar Breeding of Hosta minor 'Neulpureum 1')

  • '늘푸름 1 (Neulpureum 1)'은 국립수목원에서 육종한 품종으로, 좀비비추와 원예품종인 Hosta 'Krossa Regal'을 교잡 후 얻은 개체를 특성에 따라 선발하여 육종하였다. 선발은 잎의 색이 진하고 잎에 존재하는 굴곡의 크기가 작은 개체를 기준으로 하여 선발하였다. 그 결과 대조품종에 비해 짙은 녹색을 유지하는 기간이 길고, 초장이 짧으며, 풍부한 잎을 가지고 있는 개체가 선발되었다. 이렇게 선발된 개체는 무성증식방법으로 개체수를 늘렸으며, 3년간 재배하며 특성을 고정 및 조사하였다. 이렇게 육종된 '늘푸름 1 (Neulpureum 1)'은 가지고 있는 특징 때문에 반음지의 지피용 뿐만 아니라 분화용으로도 이용될 수 있는 만큼 활용적 가치가 매우 높은 신품종이다.

Inheritance of Cgy1 gene and Ti gene in Mature Soybean Seed

  • Soybean proteins are widely used for human and animal feed in the world. ${\beta}$-conglycinin protein exhibits poor nutritional and food processing properties and Kunitz trypsin inhibitor (KTI) protein is a main anti-nutritional factor in soybean seed. The objective of this research was to identify the inheritance of $cgy_1$ gene and ti gene for the improvement of soybean cultivar with no KTI proteins and low amount of ${\beta}$-conglycinin. $F_2$ population was made by crossing between "Gaechuck2ho" (${\alpha}^{\prime}$-subunit present $Cgy_1Cgy_1$, KTI protein absent titi) and PI506876 (${\alpha}^{\prime}$-subunit absent $cgy_1cgy_1$, KTI protein present TiTi) parent. A total of 434 $F_2$ seeds were obtained and analyzed for the segregation of ${\alpha}^{\prime}$-subunit protein and KTI protein using SDS-PAGE. The segregation ratio of 3 : 1 for $Cgy_1$ locus (310 $Cgy_1$_ : 124 $cgy_1cgy_1$) and Ti locus (339 Ti_ : 95 titi) were observed. Segregation ratios of 9 : 3 : 3 : 1 (241 $Cgy_1$_Ti_: 69 $Cgy_1$_titi: 98 $cgy_1cgy_1$Ti_: 26 $cgy_1cgy_1titi$) between $Cgy_1$ gene and Ti gene in $F_2$ seeds were also observed (${\chi}^2= 5.367$, P = 0.10 - 0.20). This data showed that $Cgy_1$ gene was inherited independently with the Ti gene in soybean. These results will be useful in breeding program for selecting the line that does not exhibit or lacks both ${\alpha}^{\prime}$-subunit protein and KTI protein in soybean.
