중생 복합내병성 고품질 벼 '삼광1호' ('Samkwang1', a Medium Maturing, Multiple Disease Resistant, and High-quality Rice)

  • '삼광1호'는 중부지역 적응하는 고품질 품종을 육성할 목적으로 2005년 하계에 중생이고 단간이며 도복에 강한 '수원152호'와 중만생이고 흰잎마름병에 저항성이며 도정특성이 좋은 최고품질 품종 '삼광'를 인공교배한 잡종1세대에 삼광을 여교배하였다. 삼광이 여교배된 $F_1$개체를 양성하며 조기에 유전특성을 고정하고자 꽃가루 배양을 실시하고 육성계통 세대 진전 후 SR30176-HB2946-B-32-1-1계통을 선발하여 수원566호로 계통명을 부여하였다. 2013년부터 2015년까지 지역적응시험 3년을 실시한 결과 그 우수성이 인정되어 2015년 12월 농촌진흥청 직무육성 신품종선정위원회에서 신품종으로 선정됨과 동시에 '삼광1호'으로 명명되었다. '삼광1호'의 출수기는 보통기 재배에서 평균 출수기가 8월 8일로 '화성'보다 2일 빠른 중생종이고, 벼 간장은 77 cm로 '화성'보다 7 cm 작은 준단간이며, 이삭길이는 20 cm, 주당 이삭수 13개로 화성과 같은 특성이나, 이삭당 입수는 128개로 '화성'보다 많았고. 등숙률은 87.4%로 '화성'보다 낮았으며, 현미 천립중이 21.7 g으로 중소립종이다. '삼광1호'는 도열병에 강한 저항성을 보였고, 흰잎마름병은 K1, K2, K3 균계에 저항성을 보였지만, 오갈병 및 멸구류에 대한 저항성은 없었다. '삼광1호'는 도복에는 중정도이며, 수발아는 화성보다 강한 반응을 보였고, 내냉성은 '화성'과 비슷한 수준을 보였다. '삼광1호'의 백미외관은 심백이 0으로 아주 맑고 투명하였다. 알칼리붕괴도는 6.2, 단백질함량은 6.2%, 아밀로스함량은 18.9%로 '화성'보다 낮아서 미질특성이 우수하였고, 관능검정 결과 밥맛은 0.35로 '화성'보다 매우 우수한 품종이었다. 제현율, 현백률 및 도정률은 '화성'과 비슷하였으나 도정된 쌀의 완전립률이 94.2%로 '화성'의 81.6%보다 높은 편이었고 완전미도정수율도 70.5%로 '화성'보다 우수하였다. '삼광1호'의 쌀수량은 중부평야지와 중서부해안지 등 11개 지역에서 보통기 보비재배 평균 5.62 MT/ha로 '화성'의 5.22 MT/ha 보다 8% 높았다. 최고품질 품종 '삼광1호'의 적응지역은 중부 평야지와 중서부해안지이다.

2002-2004년의 벼줄무늬잎마름병 발생상황 및 약제처리에 의한 애멸구의 화학적 방제 (Incidence of Rice stripe virus during 2002 to 2004 in Korea and Chemical Control of Small Brown Plant Hopper)

  • 2002년에서 2004년에 걸쳐 전국적으로 Rice stripe tenuivirus(RSV)에 의한 벼줄무늬잎마름병의 발생지역과 발생 포장율을 조사한 결과, RSV에 의한 발병은 점진적으로 감소하였다. RSV의 발생주율은 감수성 품종인 추청과 새추청, 일품에서 각각 45.8, 45.0 43.7%로 매우 높은 데 반해 저항성 품종인 화성에서는 4.4%로 낮았고, 좌지주율 역시 추청과 새추청, 일품 품종에서 각각 33.6, 33.2, 31.9%인데 비해 화성에서는 0.8%에 그쳤다. 두 가지 보독충 채집방법간의 효율성을 비교한 결과 기존의 포충망 채집에 비해 동력흡충기를 이용한 채집이 매우 효율적이었다. 2002년부터 2004년에 걸쳐 전국적으로 보독충 밀도를 조사한 결과, 연도별로 전국적인 평균 보독충율은 각각 3.6, 2.3, 1.3%로 시험기간에 걸쳐 보독충율은 전반적으로 감소하여 병 발생이 감소할 것으로 예측하였다. 이앙전 육묘상의 육묘상처리제 및 수면전개제 처리에 의한 애멸구 방제 효과를 조사한 결과, Imidacloprid 입제 등 4종 처리제의 약효가 인정되었다.

  • Clubroot disease of curcifer crops caused by Plasmodiophora brassicae had been first reported in 1920 in Korea, and maintained mild occurrence until 1980s. Since 1990s the disease has become severe in alpine areas of Kyonggi and Kangwon, gradually spread to plain fields throughout the country, and remains as the greatest limiting factor for its production. Researches on the disease has begun in late 1990s in our laboratory after experiencing severe epidemics. Survey of occurrence and etiological and ecological studies have been carried out, particularly, on the pathogen physiology, race identification, quantification of soil pathogen population, host spectrum of the pathogen, and control measures.(중략)

Characterization of a Septobasidium sp. Associated with Felt Disease of Schisandra chinensis

  • Extensive disease surveys performed during the summers of 2013 and 2014 in Schisandra chinensis orchards resulted in the finding of a Septobasidium sp. associated with felt disease. The fungus was characterized to be symbiotic with a scale insect (Pseudaulacaspis cockerelli). Morphological and molecular characteristics of the Septobasidium isolates were investigated. The isolates were morphologically and phylogenetically close to S. bogoriense. We tentatively describe this isolate as a Septobasidium sp., mainly because of the limited amount of information available on the internal transcribed spacer region of the ribosomal DNA of Septobasidium spp.

Co-Occurrence of Two Phylogenetic Clades of Pseudoperonospora cubensis, the Causal Agent of Downy Mildew Disease, on Oriental Pickling Melon

  • The genus Pseudoperonospora, an obligate biotrophic group of Oomycota, causes the most destructive foliar downy mildew disease on many economically important crops and wild plants. A previously unreported disease by Pseudoperonospora was found on oriental pickling melon (Cucumis melo var. conomon) in Korea, which is a minor crop cultivated in the temperate climate zone of East Asia, including China, Korea, and Japan. Based on molecular phylogenetic and morphological analyses, the causal agent was identified as Pseudoperonospora cubensis, and its pathogenicity has been proven. Importantly, two phylogenetic clades of P. cubensis, harboring probably two distinct species, were detected within the same plots, suggesting simultaneous coexistence of the two clades. This is the first report of P. cubensis causing downy mildew on oriental pickling melon in Korea, and the confirmation of presence of two phylogenetic clades of this pathogen in Korea. Given the high incidence of P. cubensis and high susceptibility of oriental pickling melon to this disease, phytosanitary measures, including rapid diagnosis and effective control management, are urgently required.

White Rot of Korean Wild Chive Caused by Stromatinia cepivora

  • In May 2020, we surveyed disease occurrence on vegetables grown in Seosan area, Korea. During the disease survey, white rot symptoms were observed in Korean wild chive (Allium monanthum) plants growing in fields. The symptoms occurred mainly in the seed bulb-producing fields of the crop. The above ground parts of the diseased plants displayed premature yellowing and dying of older leaves and stunting of the plants. The bulbs and roots of the diseased plants turned black and rotted. The disease occurred in a range of 1-60% in four of the eight fields surveyed. Three isolates of Sclerotium sp. were obtained from the bulb lesions of diseased plants. All isolates were identified as Stromatinia cepivora based on the morphological characteristics and phylogenetic analysis. Pathogenicity of the isolates on Korean wild chive was confirmed by artificial inoculation test. The lesions induced by the inoculation test were similar to those observed in the investigated fields. This is the first report of S. cepivora causing white rot in Korean wild chive.

Overexpression of rice premnaspirodiene oxygenase reduces the infection rate of Xanthomonas oryzae pv. oryzae

  • Plants utilize cytochrome P450, a large superfamily of heme-containing mono-oxygenases, in the synthesis of lignins, UV protectants, pigments, defense compounds, fatty acids, hormones, and signaling molecules. Despite the overwhelming assortment of rice P450 accession numbers in the database, their functional studies are lacking. So far, there is no evidence involving rice P450 in disease immunity. Most of our understanding has been based on other plant systems that are mostly dicot. In this study, we isolated the cytochrome P450 (OsCYP71) in rice, and screened the gene using gain-of-function technique. The full-length cDNA of OsCYP71 was constitutively overexpressed using the 35S promoter. We then explored the functions of OsCYP71 in the rice - Xanthomonas oryzae pv. oryzae pathosystem. Using the gene expression assays, we demonstrate the interesting correlation of PR gene activation and the magnitude of resistance in P450-mediated immunity.

First Report of Fusarium Wilt of Fallopia multiflora Caused by Fusarium oxysporum in Korea

  • In April 2014, seedlings of Fallopia multiflora showing wilt symptom were first found at a greenhouse in Punggi-eup, Yeongju-si, Korea. A Fusarium-like fungus was isolated from the wilted plant and it was identified as Fusarium oxysporum based on morphological characteristics and nucleotide sequence data of translation elongation factor 1-${\alpha}$. The fungus isolated from the diseased plant was revealed to be pathogenic to the host plant through pathogenicity tests, and the reisolation of the pathogen confirmed Koch's postulates. This is the first report of Fusarium wilt occurring on Fallopia multiflora in the world.

  • Barley Yellow Dwarf Virus (BYDV), an aphid-borne luteovirus, is a major plant pathogenic disease causing a huge economic loss in the grain production of a wide range of Gramineae species throughout the world. It has been recently reported that BYDV also occurred frequently in wheat field of Korea. Here, we performed to develop the detection and classification methods of BYDV strains that were accomplished by reverse transcription-polymerase chain reaction (RT-PCR). Since there are high variations among BYDV strains, three pairs of primers were designed to detect BYDV strains such as PAV (Vic-PAV and CN-PAV) and MAV (primer A) simultaneously, specifically Vic-PAV(primer B), and MAV (primer C) based on the genomic RNA sequences of BYDV strains previously published. The validity of the primers was confirmed using several BYDV strains obtained from CIMMYT. Though three BYDV strains were able to be detected using primer A, PCR products were not distinguished between two PAV strains. It was possible to separate them with a restriction enzyme, EcoRI, whose restriction site was present in the amplified DNA fragment from Vic-PAV, but not from CN-PAV.

Microbial Community Dysbiosis and Functional Gene Content Changes in Apple Flowers due to Fire Blight

  • Despite the plant microbiota plays an important role in plant health, little is known about the potential interactions of the flower microbiota with pathogens. In this study, we investigated the microbial community of apple blossoms when infected with Erwinia amylovora. The long-read sequencing technology, which significantly increased the genome sequence resolution, thus enabling the characterization of fire blight-induced changes in the flower microbial community. Each sample showed a unique microbial community at the species level. Pantoea agglomerans and P. allii were the most predominant bacteria in healthy flowers, whereas E. amylovora comprised more than 90% of the microbial population in diseased flowers. Furthermore, gene function analysis revealed that glucose and xylose metabolism were enriched in diseased flowers. Overall, our results showed that the microbiome of apple blossoms is rich in specific bacteria, and the nutritional composition of flowers is important for the incidence and spread of bacterial disease.
