Discrimination and Genetic Relationship of Adenophorae triphylla(Thunb) A.DC. var. japonica Hara and Codonopsis lanceolata Trauty using RAPD analysis (RAPD분석에 의한 잔대와 더덕의 유연관계 비교 및 감별)

  • Dried parts of the two species are difficult to distinguish morphologically, thus Codonopsis radix has been sold instead of Adenophorae radix in herbal medicine market. Therefore, this study was conducted to develop the genetic marker through the examination of the phylogenetic relationships between two Adenophora triphylla(Thunb.) A. DC. var. japonica Hara, two Adenophora radiatifolia Nakai, five Codonopsis lanceolata(Sieb. et Zucc)Trautv. using RAPD analysis. Fifty decarmer oligonucleotide primers were screened for the RAPD analysis, and four primers generated distinct RAPD markers specific to Adenophorae radix and Codonopsis radix. Based on the RAPD patterns, the genetic relationships between three herbal medicine were analyzed by UPGMA method. As a result, Adenophorae radix and Codonopsis radix were classified into two major subgroups on the basis of the genetic similarity coefficient. The specific RAPD patterns generated by the selected primers were reproducible from dried materials. Furthermore, the specific RAPD patterns were produced from the mixture of dried roots of A. triphylla and C. lanceolata. These results prone the usefulness of the RAPD analysis for the discrimination of pure materials from the mixtures of A. triphylla and C. lanceolata.

Preparation Method of Indigo Standard Solution and Variation of Indigo Contents in Blue Dye Extract from Breeding Lines of Persicaria tinctoria H. Gross (인디고 분석 표준액의 제조법 및 쪽 육성계통의 인디고 함량변이)

  • This study was carried out to find out the optimum method of preparation of indigo standard solution and its stability, and to investigate the indigo contents in Niram, blue dye extract, from a total of 7 indigo plants and 34 breeding lines of Persicaria tinctoria H. Gross. Proper solvent for indigo standard was dimethyl sulfoxide (DMSO), and appropriate concentration was 1 mg of indigo in 10 mL of DMSO. Absorbance value of UV/Vis Spectrophotometer at 620 nm of standard solution was changed decreasingly 12 hours after the preparation of standard solution irrespective of the storage conditions such as temperature and light. Average value of absorbance of 8-fold diluted standard solutions prepared daily during 16 days was $0.210{\pm}0.005$, indicating the powder of indigo compound was stable chemically. Calibration curve was made for quantitative analysis of indigo of 7 Niram samples, and indigo contents ranged from 0.69% to 18.76% showing relatively larger variation. Across all 34 breeding lines, the range of indigo content was from 7.9 mg to 56.4 mg per 100 g of fresh leaves, averaging 25.2 mg of indigo content and showing a 47.7% coefficient of variation.

Non-destructive Method for Selection of Soybean Lines Contained High Protein and Oil by Near Infrared Reflectance Spectroscopy

  • The applicability of non-destructive near infrared reflectance spectroscopic (NIRS) method was tested to determine the protein and oil contents of intact soybean [Glycine max (L.) Merr.] seeds. A total of 198 soybean calibration samples and 101 validation samples were used for NIRS equation development and validation, respectively. In the developed non-destructive NIRS equation for analysis of protein and oil contents, the most accurate equation was obtained at 2, 8, 6, 1(2nd derivative, 8 nm gap, 6 points smoothing, and 1 point second smoothing) and 2, 1, 20, 10 math treatment conditions with Standard Normal Variate and Detrend (SNVD) scatter correction method and entire spectrum (400-2500 nm) by using Modified Partial Least Squares (MPLS) regression, respectively. Validation of these non-destructive NIRS equations showed very low bias (protein: 0.060%, oil: -0.017%) and standard error of prediction (SEP, protein: 0.568 %, oil : 0.451 %) as well as high coefficient of determination ($R^2$, protein: 0.927, oil: 0.906). Therefore, these non-destructive NIRS equations can be applicable and reliable for determination of protein and oil content of intact soybean seeds, and non-destructive NIRS method could be used as a mass screening technique for selection of high protein and oil soybean in breeding programs.

Quality Characteristics of Vegetable Soybeans at Different Harvesting Time (풋콩의 수확시기에 따른 품질관련특성의 변화)

  • The changes in major characteristics of vegetable soybean at different stages were investigated. The 100-seed weight of vegetable soybean had a tendency to increase before 43 days after flowering, but decrease slightly after then. After 37 days after flowering, the sucrose content of Douya had a tendency to decline and in case of Shirofusa, total sugar content was sharply decreased at 40 days after flowering. The total vitamin C content of Douya and Shirofusa was dramatically increased at 37 days after flowering and then got the plateau, otherwise the total vitamin C content of Shirofumi and Hwaeomputkong was the highest level at 43 days after flowering. The hardness changes of vegetable soybeans were tend to increase according days after flowering. In sensory evaluation, preference score was significantly associated with harvesting stages. Correlation coefficient between sweetness and beany taste was significantly negative.

Effect of Planting Density and Nitrogen Level on Growth and Yield in Heavy Panicle Weight Type of Japonica Rice

  • To investigate the effects of planting density and nitrogen level on growth and yield potential of newly bred heavy panicle japonica rice with large grain (Iksan 435 and Iksan 438) or many spikelets per panicle(HR14022-21-8-4 and HR14022-21-8-6), four heavy panicle type rices and two many panicle type rices(Dongjinbyeo and Donganbyeo) as the checks were planted under standard planting density (30$\times$15 cm) and dense planting density (15$\times$15 cm) with two nitrogen levels of standard nitrogen level(110 kg h $a^{-1}$) and heavy nitrogen level(165 kg h $a^{-1}$). Effective tiller rate decreased in dense planting or heavy nitrogen, when compared to standard nitrogen and planting, while leaf area index and to dry weight increased in dense planting or heavy nitrogen. Tiller numbers and panicle numbers were more increased by dense planting than heavy nitrogen, whereas spikelet numbers were more increased by heavy nitrogen than dense planting. Ripened grain ratio was slightly lower only in dense planting. 1,000 grain weight in brown rice was not significantly different in dense planting or heavy nitrogen. Milled rice yield was highest in heavy nitrogen with standard planting for heavy panicle type rice, while yield for many panicle type rice was highest in heavy nitrogen with dense planting, suggesting that many panicle type rice possesses higher adapt-ability for dense planting than heavy panicle type rice. Path coefficient analysis revealed that top dry weight, spikelet number and grain weight were the greatest positive contributors to yield, whereas tiller number was negative to yield.d.

  • To estimate the total dry matter(TDM) of rice plant by non-destructive method, spectral reflectance from rice plant canopy was measured by using the spectroradiometer (LI-1800, LICOR Inc.) with one week interval during the rice growing season at Suwon paddy field in 1993. Two medium late maturing rices, Daechung-byeo Ilpum-byeo, and one early maturing variety, Jinbu-byeo, were cultured to observe TDM, then they were compared with those estimated by vegetation index together. Vegetation index determined by the reflectance of visible against near infrared wavelength showed high correlation with TDM. Vegetation index derived from narrow band(10nm interval) ratio, R910/R460, has the highest correlation coefficient with TDM. TDM estimated from R910/R460 from transplanting to heading stage corresponded well to measured values (Y=21.2428X-212.734 ; $R^2$=0.87). But another vegetation index, NIR(720~1,100nm) /Red(600~700nm) showed higher correlation with TDM than NIR(720~1,100nm) /Blue(400~500nm) did from heading stage to maturity.

The Effect of Phosphate Fertilizers on Soybean Growth and Lime Application in the Korean Hilly Land Soil (양산개간지 토양에서 인산비종에 따른 대두생육 및 석회시용효과에 관한 연구)

  • A soybean variety KWANG KYO was planted on the hilly land and fertile soil, and concentrated superphosphate and fused phosphate were applied on calcium hydroxide treated soil at level of 2.5%, 5% and 7.5% of phosphate absorbtion coefficient of soil. Application of phosphate fertilizer and lime caused higher grain yield in hilly land soil than fertile soil. The effect of fused phosphate on soybean yield was superior to concentrated superphosphate, and the effect of lime on fused phosphate availability was not significant. When the two phosphate sources were compared at equal $P_2O_5$ basis, fused phosphate required higher amount than concentrated super-phosphate. Negative correlation was obtained between potassium content and Ca + Mg in soybean leaf and in soil respectively.

Discrimination of Korean Domestic and Foreign Soybeans using Near Infrared Reflectance Spectroscopy (근적외선분광광도계(NIRS)를 이용한 국내산 콩과 수입콩의 판별분석)

  • Discrimination of geographic origin of agricultural products is a important issue in Korea because the price difference between Korean domestic and imported cereals is a key among some reasons. NIRS (Near Infrared Reflectance Spectroscopy) has been applied to classify the geographical origin of soybeans. Total 135 samples (Korean domestic 92 and foreign 43) were used to obtain calibration equation through 400~2,500 nm wavelength. The math treatment with 1st derivative and 4 nm gap and the modified partial least squares(MPLS)regression was outstanding for calibration equation. The standard error of calibration and determination coefficient in calibration set(n=115) was 6.65 and 0.98, respectively. And it showed that the extra 20 samples for validation equation were identified their authentication correctly. This study describes that the application of NIRS would be possible for discrimination of geographical origin between Korean domestic and imported soybeans.

Variation of Antifreeze Proteins during Cold Acclimation among Winter Cereals and Their Relationship with Freezing Resistance

  • Freezing-resistant plants can survive subzero temperatures by withstanding extracellular ice formation. During cold acclimation, their leaves accumulate antifreeze proteins (AFPs) that are secreted into the apoplast and have the ability to modify the normal growth of ice crystals. Three barley, two wheat and two rye cultivars were grown under two different temperature regimes (20/16$^{\circ}C$ and 5/2$^{\circ}C$, day/night). Apoplastic proteins from winter cereals were separated by SDS-PAGE and detected with antisera to AFPs from winter rye. Apoplastic proteins accumulated to much higher levels in cold-acclimated (CA) leaves compared with nonacclimated (NA) ones in winter cereals. After cold acclimation, the protein concentration of apoplastic extracts increased significantly from 0.088 $mgmL^{-1}$ to 0.448 $mgmL^{-1}$, with about 5-fold increment. Also, the apoplastic protein content per gram leaf fresh weight in CA leaves ranged from 31 $\mu\textrm{g}$ $(gFW)^{-1}$ to 120 $\mu\textrm{g}$ $(gFW)^{-1}$ with an averaged value of 77 $\mu\textrm{g}$ $(gFW)^{-1}$, and coefficients of variation of 54.9%. The CA leaves in Musketeer (a Canadian winter rye cultivar) showed the greatest AFPs and antifreeze activity followed by 'Geurumil' (a Korean winter wheat cultivar), and 'Dongbori l' (Korean facultative barley cultivar). The proteins secreted into the wheat leaf apoplast at CA condition were more numerous than those observed in winter rye, where two $\beta$-1,3-glucanase-like proteins (GLPs), two chitinase-like proteins (CLPs) and two thaumatin-like proteins (TLPs) accumulated during cold acclimation. The proteins in barley leaf apoplast at CA conditions were a little different from those in wheat leaves. The AFPs were various among and within species. More freezing-resistant cultivars had more clear and numerous bands than less freezing-resistant ones. The high determination coefficient ($R^2$ =91 %) between freezing resistance and AFPs per gram leaf fresh weight indicated that the amount of AFPs was highly related to freezing resistance in winter cereal crops.

  • According with variations in principal agronomic traits of fourteen malting barley cultivars as affected by different seeding methods and seeding rates. the analysis and interpretation of correlation coefficients and path coefficients was intended. From all treatments (seeding methods and rates), the positive significant relations detected between flowering days and maturing days, however the negative between maturing days and culm length, number of grains per spike, yields, and between 1000 grain weight and one liter weight, respectively. Direct effects on the yields were greatest by number of grains per spike in case of narrow-spaced row seeding and number of spikes per m$^2$ and number of grains per spike among others in broadcast seeding method On the other hand, indirect effects were not recognized in any treatments.
