Quality Classification and Its Application Based on Certification Standards of Kentucky Bluegrass(Poa pratensis L.) Seed (켄터키 블루그래스(Poa pratensis L.) 종자의 보증 기준에 따른 품질 분류와 적용)

  • The purpose of seed certification is to preserve the genetic purity and identity of seed varieties. This study is to provide information concerning seed certification procedures and certification standards of Kentucky bluegrass especially used in golf courses. We analyzed data from the seed certification standards of three states (Washington, Idaho and Oregon) in U.S.A. The certification processes both field inspection and laboratory requirement satisfying the minimum seed quality standards. The seed harvesting field must be propagated with the specified class of seeds and requires an adequate isolated distance from other crops. Moreover, the field should be clean and free from the objectionable weeds. The seed analysis tests include a germination rate, a percentage of pure seed, contents of other crop seed, weed seed, and inert matter. The certification standards of the certified seed and the sod quality seed showed general similarity in all three states. The certification standards of the sod quality seed should have less than 0.02% of maximum weed seed. The certified seed should have less than 0.3% of maximum weed seeds. Those certification standards of seed quality should guaranty the quality of turfgrass establishment of golf course.

Application of Electrical Resistivity Measurement to an Evaluation of Saline Soil in Cropping Field (염류집적 농경지에서 전기비저항 탐사기법의 활용성)

  • Salinity of soil under the plastic film houses in Korea is known as a significant factor to lower the crop production and to hamper the sustainable agricultural land management. In this study we propose a field monitoring technique to examine the methods applied to minimize the adverse effect of salts in soil based on the relationship between soil electrical characteristics and soil properties. Field experiments for 4 different treatments (water only, fertilizer only, DTPA only, and DTPA and fertilizer together) were conducted on soils at the plastic film house built for cultivating a cucumber plant located at Chunan-si, Chungchungnam-do in Korea. The electrical resistivity was measured by both a dipole-dipole and wenner multi-electrodes array method. After the electrical resistivity measurement we also measured the soil water content, temperature, and electrical conductivity on surface soil. The resulted image of the interpreted resistivity by the inversion technique presented a unique spatial distribution depending on the treatment, implying the effect of the different chemical components. It was also highly suspected that resistivity response changed with the nutrients level, suggesting that our proposed technique could be the effective tool for the monitoring soil water as well as nutrient during the cropping period. Especially, subsoils under DTPA treatment at 40 to 60 cm depth typically presented lower soil water accumulation comparing to subsoils under non-DTPA treatment. It is considered that DTPA resulted in increase of a root water uptake. However, our demonstrated results were mainly based on qualitative comparison. Further experiments need to be conducted to monitor temporal changes of electrical resistivity using time lapse analysis, providing that a plant root activity difference based on changes of soil water and nutrients level in time.

Analysis of the Cloud Removal Effect of Sentinel-2A/B NDVI Monthly Composite Images for Rice Paddy and High-altitude Cabbage Fields (논과 고랭지 배추밭 대상 Sentinel-2A/B 정규식생지수 월 합성영상의 구름 제거 효과 분석)

  • Crops show sensitive spectral characteristics according to their species and growth conditions and although frequent observation is required especially in summer, it is difficult to utilize optical satellite images due to the rainy season. To solve this problem, Constrained Cloud-Maximum Normalized difference vegetation index Composite (CC-MNC) algorithm was developed to generate periodic composite images with minimal cloud effect. In thisstudy, using this method, monthly Sentinel-2A/B Normalized Difference Vegetation Index (NDVI) composite images were produced for paddies and high-latitude cabbage fields from 2019 to 2021. In August 2020, which received 200mm more precipitation than other periods, the effect of clouds, was also significant in MODIS NDVI 16-day composite product. Except for this period, the CC-MNC method was able to reduce the cloud ratio of 45.4% of the original daily image to 14.9%. In the case of rice paddy, there was no significant difference between Sentinel-2A/B and MODIS NDVI values. In addition, it was possible to monitor the rice growth cycle well even with a revisit cycle 5 days. In the case of high-latitude cabbage fields, Sentinel-2A/B showed the short growth cycle of cabbage well, but MODIS showed limitations in spatial resolution. In addition, the CC-MNC method showed that cloud pixels were used for compositing at the harvest time, suggesting that the View Zenith Angle (VZA) threshold needsto be adjusted according to the domestic region.

Comparative Analysis of Rice Lodging Area Using a UAV-based Multispectral Imagery (무인기 기반 다중분광 영상을 이용한 벼 쓰러짐 영역의 특성 분석)

  • Lodging rice is one of critical agro-meteorological disasters. In this study, the UAV-based multispectral imageries before and after rice lodging in rice paddy field of Jeollanamdo agricultural research and extension servicesin 2020 was analyzed. The UAV imagery on 14th Aug. includesthe paddy rice without any damage. However, 4th and 19th Sep. showed the area of rice lodging. Multispectral camera of 10 bands from 444 nm to 842 nm was used. At the area of restoration work against lodging rice, the reflectance from 531 nm to 842 nm were decreased in comparison to un-lodging rice. At the area of lodging rice, the reflectance of around 668 nm had small increases. Further, the blue and NIR (Near-Infrared) wavelength had larger. However, according to the types of lodging, the change of reflectance was different. The NDVI (Normalized Difference Vegetation Index) and NDRE (Normalized Difference Red Edge) shows dome sensitivities to lodging rice, but they were different to types of lodging. These results will be useful to make algorithm to detect the area of lodging rice using a UAV.

Application of Reverse Transcription Droplet Digital PCR for Detection and Quantification of Tomato Spotted Wilt Virus (Reverse Transcription Droplet Digital PCR을 활용한 Tomato Spotted Wilt Virus 검출 및 정량)

  • Plant viruses cause significant yield losses, continuously compromising crop production and thus representing a serious threat to global food security. Tomato spotted wilt virus (TSWV) is the most harmful plant virus that mainly infects horticultural crops and has a wide host range. Reverse-transcription quantitative real-time PCR (RT-qPCR) has been widely used for detecting TSWV with high sensitivity, but its application is limited owing to the lack of standardization. Therefore, in this study, a sensitive and accurate reverse transcription droplet digital polymerase chain reaction (RT-ddPCR) method was established for TSWV detection. Additionally, we compared the sensitivities of RT-qPCR and RT-ddPCR for TSWV detection. Specificity analysis of RT-ddPCR for TSWV showed no amplification for main pepper viruses and negative control. TSWV transcripts levels measured by RT-ddPCR and RT-qPCR showed a high degree of linearity; however, the former yielded results that were at least 10-fold more sensitive and detected lower TSWV copy numbers than the latter. Collectively, our findings show that RT-ddPCR provides improved analytical sensitivity and specificity for TSWV detection, making it suitable for identifying low TSWV concentrations in field samples.

  • Plant molecular farming has attracted a lot of attention lately in the field of mass production of industrially valuable materials by extending application of the plant as a kind of factory concept. Among them, protein expression system using rice(Oryza sativa L.) callus is a technology capable of mass culture and industrialization because of a high expression rate of a target protein. This study was carried out to develop an Agrobacterium-mediated transformation system to increase the utilization of rice callus. The transformation efficiency was improved by using the hand when seeds were de-husked for callus induction. Furthermore, we were possible induction of callus from 6 years old seed smoothly. Selection of the callus contained the target gene was required a cultivation period of at least 3 weeks, and the most efficient selection period was after 6 weeks of culture including one passage. This selection was confirmed that the gene was stably inserted into the genomic DNA of the plant cell by the southern blot analysis and progeny test. Such an efficient selection system of rice callus that can be cultured in the long term will be contribute to the industrialization of useful recombinant proteins using rice.

Assessment of gene flow from insect-resistant genetically modified rice (Agb0101) to non-GM rice (해충저항성 유전자변형 벼(Agb0101) 유전자 이동성 평가)

  • Genetically modified (GM) crops have been developed worldwide through the recombinant DNA technology and commercialized by global agricultural companies. Until now, GM crops have not been cultivated commercially in Korea. Commercialization of GM crops requires a compulsory assessment of environmental risk associated with the release of GM crops. This study was conducted to evaluate the frequency of pollen mediated gene flow from Bt transgenic rice (Agb0101) to japonica non-GM rice (Nakdongbyeo), indica non-GM rice (IR36), and weedy rice (R55). A total of 729,917, 596,318 and 230,635 seeds were collected from Nakdongbyeo, IR36, and R55, respectively, which were planted around Agb0101. Selection of the hybrids was determined by repeated spraying of herbicide and Cry1Ac1 immunostrip assay. Finally, the hybrids were confirmed by PCR analysis using specific primer. The hybrids were found in all non-GM rice and out-crossing ranged from 0.0005% at IR36 to 0.0027% at Nakdongbyeo. All of hybrids were located within 1.2 m distance from the Agb0101 rice plot. The meteorological elements including rainfall and temperature during rice flowering time were found to be important factors to determine rice out-crossing rate. Consideration should be taken for many factors like the meteorological elements of field and physiological condition of crop to set up the safety management guideline to prevention of GM crops gene flow.

Differences in isolates of Tomato yellow leaf curl virus in tomato fields located in Daejeon and Chungcheongnam-do between 2017 and 2018

  • To follow up on a 2017 survey of tomato virus diseases, samples with virus-like symptoms were collected from the same areas (Buyeo-gun, Chungchungnam-Do and Daejeon, Korea) in 2018. While in 2017 mixed infections of Tomato mosaic virus with either Tomato yellow leaf curl virus (TYLCV) or Tomato chlorosis virus were detected, only TYLCV was detected in symptomatic samples in 2018. TYLCV amplicons of c.777 bp representing the coat protein (CP) coding region were cloned from the TYLCV positive samples, and the sequence data showed a 97.17% to 98.84% nucleotide and 98.45% to 99.22% amino acid identity with the 2017 Buyeo-gun isolate (MG787542), which had the highest amino acid (aa) sequence identity of up to 99.2% with four 2018 Buyeo-gun sequences (MK521830, MK521833, MK521834, and MK521835). The lowest aa sequence identity of 98.45% was found in a 2018 Daejeon isolate (MK521836); the distance between Buyeo-gun and Daejeon is about 45 km. Phylogenetic analysis indicated that the currently reported CP sequences are most closely related to Korean sequences from Masan (HM130912), Goseong (JN680149), Busan (GQ141873), Boseong (GU325634), and the 2017 isolate TYLCV-N (MG787543) in the 'Japan' cluster of TYLCV isolates and distinct from the 'China' cluster isolates from Nonsan (GU325632), Jeonju (HM130913) and Jeju (GU325633, HM130914). Our survey data from 2017 and 2018 suggest that TYLCV has become established in Korea and may be spread by whitefly vectors from weed reservoirs within the farm environment.

SHRIMP Zircon U-Pb Age and Geochemistry of Granites in the Gudambong-Sainam Geosites, Danyang Geopark (단양 지질공원 구담봉-사인암 지질명소 화강암의 SHRIMP 저어콘 U-Pb 연령과 지구화학)

  • We carried out the sensitive high resolution ion microprobe zircon U-Pb age dating and whole rock geochemical analysis of granites in the Gudambong and Sainam geosites, Danyang Geopark. The granites crop out in the western and southern parts of Danyang County, and intruded sedimentary successions of the Yeongweol and Taebaek Groups, respectively. The U-Pb isotopic compositions of zircon from the Gudambong and Sainam granite samples yielded the Cretaceous intrusion ages of $90.4{\pm}0.5Ma(t{\sigma})$ and $90.0{\pm}1.5Ma(t{\sigma})$, respectively. The major and trace elements compositions of the samples showed an affinity of typical A-type granite, indicating their petrogenesis during the late stage of the Bulguksa orogeny or a tectonic dormancy. The geochronologic and geochemical results are identical to those of granites previously reported from the Cretaceous Muamsa and Wolaksan suites.

Selection of Entomopathogenic Fungus Isaria javanica FT333 for Dual Control of Thrips and Anthracnose (총채벌레 및 고추탄저병의 동시 방제를 위한 곤충병원성 곰팡이 Isaria javanica FT333 선발)

  • Red pepper is seriously damaged by thrips (Thrips palmi) and anthracnose caused by Colletotrichum acutatum throughout its development. Because of biotic constraints, producers often depend on chemicals that are expensive and have adverse effects on the environment, operator, and beneficial insects. In addition, resistance is developed because of the repeated use of chemicals. In recent decades, the use of microorganisms in crop protection has become a credible alternative because it is eco-friendly. In this study, we aimed to select isolates with insecticidal and fungicidal activities against the pathogens that cause anthracnose and thrips. We treated T. palmi adults and juveniles with 13 strains of entomopathogenic fungi (isolated from the soil by using the insect-bait method), and 6 strains showed excellent insecticidal activity (70-100%) 5 days after the treatment. The selected isolates were cultured with C. acutatum to screen for the strain with excellent anti-fungal activities, among which an isolate FT333 showed more than 95% control efficacy against C. acutatum in vitro. The isolate was identified as Isaria javanica through its morphological characteristics and phylogenetic analysis of the ITS and ${\beta}-tubulin$ nucleotide sequences. The Isaria javanica FT333 isolate could be used effectively for dual bio-control of thrips and anthracnose during red pepper cultivation.
