  • 많은 양봉업자들이 그들의 벌을 보호하기 위해 동물성의약품들을 사용한다. 우리나라에서 벌에 사용되는 동물성 의약품은 아미트라즈, 브로모프로필레이트, 쿠마포스, 시미아졸 등이다. 2006년 중반에 한국 정부에서는 아미트라즈와 쿠마포스의 최대잔류허용기준을 각각 0.2, 0.1 ppm으로 설정하였다. 해마다 양봉환경이 변하고 있으므로 꿀 속의 동물성 의약품들에 대한 지속적인 모니터링이 필요하다. 본 연구에서는 2011년에 지역시장이나 인터넷을 통해 모은 10개 벌꿀 시료들에 대하여 아미트라즈, 브로모프로필레이트, 쿠마포스, 시미아졸 잔존량을 HPLC-DAD로 조사하였다. 조사한 10종의 벌꿀에서는 살충제로 사용되는 이들 성분이 25 ppb 이상으로는 검출되지 않았다.

  • 본 연구에서는 동물세포에서 매우 높은 독성을 갖는 유기인 살충제 coumaphos를 생분해하기 위하여, 재조합 대장균으로부터 발현된 organophosphorus hydrolase (OPH)가 사용되었다. OPH 생산성을 향상시키고자, 균주의 배지에 비이온성계면활성제(Tween 20, PEG 1000)와 유기용매(glycerol, propanol, ethanol)에 대한 효과를 고찰하였다. 0.25% Tween 20과 0.5% glycerol를 배지에 첨가하였을 때, 최대의 OPH가 생산되었다. 초음파를 이용한 균주 파쇄 공정으로부터 생성된 OPH를 사용 하였을 때, 0.2, 0.5, 1.0, 2.0 mM coumaphos는 각각 100, 88, 84, 78%의 생분해효율을 나타내었다. 이 연구에서 얻어진 결과들은 coumaphos가 다양한 농도로 오염된 지역을 생물학적으로 처리할 수 있는 새로운 방법으로서 활용될 수 있을 것이다.

  • Recently, we reported an improved technology for the degradation of organophosphate nerve agents using whole cells of genetically engineered Escherichia coli that anchored and displayed the enzyme organophosphorus hydrolase on the cell surface. In this paper we report the immobilization of these cells on highly porous sintered glass beads and the subsequent application of the immobilized cell in a continuous-flow packed bed bioreactor for the biodetoxification of a widely used insecticide, coumaphos.

Simultaneous Determination of Amitraz, Bromopropylate, Coumaphos, Cymiazole and 2,4-Dimethylaniline in Korean Honey Samples by High-Performance Liquid Chromatography

  • Simultaneous determination of amitraz, bromopropylate, coumaphos, cymiazole and 2,4-dimethylaniline in 200 honey samples purchased in Korea was performed by reversed-phase high-performance liquid chromatography with multiple UV detection. 2% Acetone in hexane was used for a liquid-liquid extraction and 20-40% water in acetonitrile solutions were used as mobile phases. The LOD for the analytes varied between 0.4 and 1.5 $\mu$g/L and the recoveries were yielded between 64 and 94%. Relative standard deviation of the repeatability of the method is less than 15%. Amitraz was not present in amount above 10 $\mu$ g/L and one for coumaphos and cymiazole and two for bromopropylate, and three for 2,4-dimethylanilne were detected in amount above 10 $\mu$ g/ L. Levels of the acaricide residues found were less than 50 $\mu$ g/L.

농약 혼용에 따른 꿀벌유충 독성의 상승 및 상쇄 영향 (Synergistic and Antagonistic Interactions for Pesticide mixtures to Honeybee Larvae Toxicity)

  • BACKGROUND: Recently, the widespread distribution of pesticides in the hive has been of concern about pesticide exposure on honeybee (Apis mellifera L.) health. Larval toxicity was adapted to assess the synergistic and antagonistic interaction of cumulative mortality to the honeybee larvae of the four most common pesticides detected in pollen. METHODS AND RESULTS: Acetamiprid($3.0{\mu}l/L$), chlorothalonil ($803.0{\mu}l/L$), coumaphos ($128.0{\mu}l/L$), and tau-fluvalinate ($123.0{\mu}l/L$) were tested in combination; binary, ternary and four component mixture. Larvae were exposed to four pesticides mixed in diet at the average levels detected in pollen. As a result, synthetic toxicity was observed in the binary mixture of acetamiprid with coumaphos. The binary and ternary component mixtures of tested pesticides have mostly demonstrated additive effect in larval bees. The significant antagonistic effects were found in four parings of mixtures including chlorothalonil added to acetamiprid/tau-fluvalinate or acetamiprid/coumaphos/tau-fluvalinate, and tau-fluvalinate added to acetamiprid/chlorothalonil or acetamiprid/coumaphos/chlorothalonil. CONCLUSION: Interactions between combinations of four pesticides showed mostly additive or antagonistic effects in larval bees. Therefore, predicting the larval mortality of pesticides mixtures on the basis of the results of single pesticide may actually overestimate the risk. We suggest that pesticide mixture in pollen be evaluated by adding their toxicity together for complete data on interactions.

벌꿀 중 동물용의약품 잔류량 모니터링 (Monitoring of Veterinary Medicine Residues in Honey)

  • 기준 설정 전인 2006년 식품의약품안전청에서 실시한 연구과제의 결과보고서에 의하면 200건 벌꿀 중 네오마이신은 검출되지 않았고, 스트렙토마이신의 경우 23건이 검출되었으며, 아미트라즈(그 대사산물 포함), 코마포스의 경우 각각 10건, 9건의 검출되었다(8). 그러나 기준 설정 후인 본 연구의 모니터링 결과 해당 동물용의약품의 검출율이 현저하게 떨어진 것으로 미루어 볼때 07년 기준 설정 후 설정된 잔류허용기준에 맞춰 관리되고 있음을 알 수 있었다.

Determination of Acaricides in Honey by Solid-Phase Extraction and Gas Chromatography / Mass Spectrometry

  • An analytical method based on solid-phase extraction and gas chromatography / mass spectrometry has been developed for measurement of acaricides (amitraz, bromopropylate, coumaphos, cymiazole, and tetradifon) in honey sample. In the stability test of acaricides in honey, amitraz underwent a rapid degradation into 2,4-dimethylaniline (DMA), 2,4-dimethylphenylformamide (DMPF), and N-(2,4-dimethylphenyl)-N'-methylformamidine (DMPMF), whileas other acaricides were found to be stable even for over three months. Extraction of five acaricides from 5g of honey sample was carried out by liquid-liquid extraction using 20mL of ethylacetate. For purification, Florisil-SPE cartridge with elution of 5mL of n-hexane/ acetone (55:45, v/v) was found to remove interferences effectively. Quantification was performed using gas chromatography / mass spectrometry in the selected ion monitoring mode. Spiking experiments were carried out to determine the recovery, precision, and limits of detection (LODs) of the method. The overall recovery values from honey spiked at 0.02 and 0.20 ${\mu}g/g$ levels, respectively, were found to be greater than 75% for all acaricides. The method detection limits for acaricides were ranged from 0.1 to 3 ppb. The developed method in this study was applied for the monitoring of acaricides in honey products collected from urban markets in Korea.

Biodegradation of Diazinon by Serratia marcescens DI101 and its Use in Bioremediation of Contaminated Environment

  • Four diazinon-degrading bacteria were isolated from agricultural soil by using an enrichment technique. The biochemical analysis and molecular method including RFLP indicated that these isolates were identical, and one strain designated DI101 was selected for further study. Phylogenetic analysis based on 16S rDNA sequencing indicated that the strain DI101 clearly belongs to the Serratia marcescens group. The ability of the strain to utilize diazinon as a source of carbon and phosphorus was investigated under different culture conditions. The DI101 strain was able to completely degrade 50 mg/l diazinon in MSM within 11 days with a degradation rate of 0.226 $day^{-1}$. The inoculation of sterilized soil treated with 100 mg/kg of diazinon with $10^6$ CFU/g DI101 resulted in a faster degradation rate than was recorded in non-sterilized soil. The diazinon degradation rate by DI101 was efficient at temperatures from 25 to $30^{\circ}C$ and at pHs from 7.0 to 8.0. The degradation rate of diazinon was not affected by the absence of a phosphorus supplement, and addition of other carbon sources (glucose or succinate) resulted in the slowing down of the degradation rate. The maximum degradation rate ($V_{max}$) of diazinon was 0.292 $day^{-1}$ and its saturation constant ($K_s$) was 11 mg/l, as determined by a Michaelis-Menten curve. The strain was able to degrade diethylthiophosphate-containing organophosphates such as chlorpyrifos, coumaphos, parathion, and isazofos when provided as a source of carbon and phosphorus, but not ethoprophos, cadusafos, and fenamiphos. These results propose useful information for the potential application of the DI101 strain in bioremediation of pesticide-contaminated environments.

HPLC-MS/MS를 이용한 벌꿀 중 동물용의약품 동시분석방법 연구 (Simultaneous Analysis for Veterinary Drug Residues in Honey by HPLC/MS/MS)

  • HPLC-MS/MS를 이용하여 벌꿀의 동물용의약품 동시분석을 위한 분석법을 정립하고자 식품공전 중 벌꿀에 규격이 설정된 11종의 동물용의약품을 2개 그룹 Group 1(streptomycine, dihydrostreptomycine, neomycine)과 Group 2 (oxytetracycline, enrofloxacin, ciprofloxacin, cymiazole, chloramphenicol, amitraz, coumaphos, fluvalinate)으로 나눠서 동시에 분석 할 수 있는 방법을 연구 하였다. 두 그룹 모두 RT는 15분 이내였고, 검출한계는 $0.0056{\sim}0.0643{\mu}g/g$, 정량한계는 $0.0169{\sim}0.1948{\mu}g/g$으로 나타났으며 Group 1 ($0.05{\sim}1.0{\mu}g/g$의 농도범위)과 Group 2 ($0.01{\sim}1.0{\mu}g/mL$)의 검량선을 작성한 결과 각 동물용의약품의 직선상관계수($R^2$)는 0.9917~0.9987, 0.9923~1.000으로 매우 양호한 상태를 보였고, 최종 농도가 $0.25{\mu}g/g$, $1.0{\mu}g/g$에서의 회수율은 Group 1 65.1~80.6%, Group 2 64.2~90.3%를 나타났다, 또한 area 및 RT 에 대한 inter (n = 3), intra day (n = 6) RSD (%) 분석결과는 area는 10.92%이하, RT은 1.57% 이하로 나타나 양호한 수준을 보여 벌꿀 중 동물용의약품 동시분석 방법으로 적합하다고 판단된다.
