This study was conducted to develope identification method with major broad leaved weed seedlings. Characteristics of weed seedlings were investigated at two to four true leaf stage. Qualitative characteristics were more useful to identification of weed species than quantitative characteristics at the seedling stage. Many species were elliptic and broadly elliptic type in their cotyledon shape. Cotyledon shape of Cassia nomame and Calystegia sepium were broadly oblong. Four species were transverse broadly elliptic in their cotyledon shape. Rumex japonicus, Persicaria hydropiper, Euphorbia supina and Acalypha australis have three or more types in their cotyledon shape. Cotyledons of Vigna angularis var. nipponensis remained below ground after its germination. Many species were round in apex and were cuneate in base of cotyledon. Humulus japonicus and Polygonum aviculare were sessile in cotyledon base. The second true leaf shape of some species changed after first leaf stages. Glycine soja, V. angularis var. nipponensis and Kummerowia striata were broadly ovate, heart-shaped, orbicular or broadly obovate in their first true leaf, respectively, but the second true leaf shape of them were tripalmately compound. Aeschynomene indica and Cassia nomame were paripinnately compound leaf in their first and second true leaves. Margin of true leaves were entire in many species but six species including Xanthium occidentale were dentate in their true leaf margins. Margin of Euphorbia supina was entire in first true leaf and dentate in second true leaf. Margin of Chenopodium album and C. serotinum were entire in their first true leaf stage and erose in their second true leaf stage. The key for weed seedling identification of major weed species were made based on quantitative and qualitative morphological characteristics.
The experiment was conducted to determine if seed size and seed coat color of soybean might be effective in water uptake and cotyledon damage after imbibition, and hypocotyl elongation. Eight soybean cultivars were separated into two classes of large and small seed based on seed weight, and each class included two cultivars with yellow and black seed color, respectively. Small seed size group was superior in water uptake by seed for 24 hour in imbibition at $25^{\circ}C$ , but its differences decreased as soaking time increased. Small seed cultivars germinated faster and had better germination rate than large ones. However, cultivars with black seed coat showed more slow water uptake at initial time and faster germination than yellow seed, but in 24 hour after imbibition, cultivars with black seed coat had higher water uptake rate than yellow seeds. Small seed cultivar group showed no cotyledon damage in imbibition for 24 hour while large seed cultivars were damaged 78% of cotyledon, and black seed showed low cotyledon damage compared to yellow seed. Hypocotyl length was shorter in large seed rather than in small seed, but hypocotyl thickness in large seed was more thick than in small seed. In correlation coefficients, seed coat rate, embryo rate exhibited significantly negative association with seed weight, and the correlation of seed weight with water uptake in 3 hour after soaking was significantly negative, but in 24 hour showed positive correlation.

Cultivars or genetic resources with a black seed coat and green cotyledons are rich in lutein, which can promote eye health, and anthocyanin, known for its numerous health benefits. However, mature seeds also contain P34, 7S α' subunit, and lectin proteins, which are allergenic and degrade quality. Here, we report the breeding of a new soybean line with a black seed coat, green cotyledon, and free of P34, 7S α' subunit, and lectin proteins. A total of 157 F2 seeds with black seed coats and green cotyledons were selected by crossing a female parent with a brown seed coat, green cotyledon, and lacking the 7S α' subunit and lectin proteins with a male parent with a black seed coat, green cotyledon, and lacking the P34 and lectin proteins. The P34 and 7S α' subunit proteins were consistent with a ratio of 9:3:3:1, indicating that they are independent of each other. From 14 F2 seeds that were recessive (cgy1cgy1p34p34) for both proteins, one individual F2 plant (F3 seeds) with the desired traits-black seed coat, green cotyledon, and lacking P34, 7S α' subunit, and lectin proteins- was finally selected. The triple null genotype (absence for P34, 7S α' subunit, and lectin proteins) was confirmed in random F3 seeds. The selected line has a black seed coat and green cotyledons, and when sown on June 14 in the greenhouse, the maturity date was approximately October 3, the height was about 68 cm, and the 100-seed weight was about 26.5 g.
In order to investigate the effects of seed weight, size (length and thickness), and maturation period (early maturing seed or late maturing seed) on growth of first-year seedlings in Pinus koraiensis, pot cultivation experiment with seeds collected from Gangweon University Forest was carried out under relatively controlled environment condition. At the end of one growing season, not only fresh weight, length, diameter (at underside of cotyledon) and root length of seedlings, but also number and length of cotyledon were measured. Results are as follows; 1. Germination percentage is independent of weight and size of seeds, but it was influenced by maturation period, that is, late maturing seeds surpass early maturing one in germination percentage. 2. Germination percentage is affected by maturation period of seeds in 18.3% of contribution rate. 3. Each growth of first-year seedlings is influenced considerably by the differences of weight, length and thickness of seeds, that is, heavy and large seeds produce larger seedlings than do light and small seeds. Particularly, fresh weight, diameter, length of cotyledon are significantly affected by differences of seed weight and size (length and thickness) in 50-90% of contribution rate. 4. Differences of seed weight and thickness have a tendency to affect more on growth of first-year seedlings than do differences of seed length. 5. Seedlings from late maturing seeds are superior than those from early maturing one in fresh weight, diameter of seedlings and length of cotyledon, but it's influence is a little.
To obtain the basic information about the translocation of seed reserves of soybean[Glycine max(L.) Merrill] cultivars, 'Hill', 'Paldalkong' and 'Jangyeobkong' , the dry matter changes of emerging organs during the germination and seedling development were observed at the research farm of College of Natural Resources, Korea University, on May 26, June 5 and June 14. Mean fresh weight, dry weight and moisture content of three soybean cultivars were increased until the V2 stage in all seedling parts except cotyledons. Cotyledon dry weight was continuously decreased and the decreased amount at the V2 stage was about 80% of the cotyledon dry weight at the germination stage. Structural component of cotyledons was 22.2% of the cotyledon dry weight at the germination stage. Metabolic components of cotyledons were markedly decreased until V2 stage, and about 91% of cotyledon metabolic components at germination stage was utilized. However, those of whole seedling were increased after the V1 stage. Therefore, it appeared that role of cotyledons as nutrient supplier for germination and seedling growth was important until the V1 stage especially.
This study was carried out in order to establish plant regeneration via somatic embryogenesis from leaf explant of Ostericum koreanum Kitagawa and to elucidate the effects of NAA and cytokinins (kinetin, BA) on the abnormalities of somatic embryo and the relationship between thecotyledon numberand germinability. Calli were formed on leaf explants cultured on MS agar medium supplemented with various concentrations (0, 0.1, 0.5, 1, 2 mg/L) of NAA and cytokinins. The calli were white, watery and soft, became browning during cultures. Somatic embryos were formed from pale yellowish calli derived browning calli. High frequency somatic embryos were observed on MS medium containing 1 mg/L NAA and 0.1 mg/L BA after 60 days of culture. The mature somatic embryos germinated into plantlets without subculture after 2 weeks. The frequency of normal somatic embryo with two cotyledons was 39.8%. On the other hand, cotyledonary abnormalities of somatic embryos were observed at considerable frequency: 33.6% of somatic embryo with one cotyledon, 15.3% cotyledons with three, 8.2% four cotyledons and 3.1% jar shaped cotyledon. Germination frequency of somatic embryos with two cotyledons was 97.4%, and that of the embryos with abnormal cotyledon was almost similar to that of embryos with two cotyledons, except jar shaped somatic embryos (33.3%).
Culture condition for callus formation and plant regeneration were optimized by the selection of explants and the manipulation of hormonal combination in the culture medium. The calli induced from seed, cotyledon, hypocotyl and mesophyll segments were more vigorously proliferated under dark condition than those under continuous light condition. Hypocotyl-and cotyledon-derived calli were more regenerative as compared with those of seed and mesophyll. Callus formation from hypocotyl and cotyledonary explants was enhanced on MS medium with 1.0 mg/${\ell}$ 2, 4-D and 0.1 to 0.5 mg/${\ell}$ kinetin or BAP. The combination of 0.1 mg/${\ell}$ NAA and 2.0 to 4.0 mg/${\ell}$ kinetin was the most effective for shoot regeneration from the callus. The maximum frequency (24.0%) of shoot regeneration was obtained from the hypocotyl-derived callus transferred to MS medium supplemented with 0.1mg/${\ell}$ NAA and 4.0 mg/${\ell}$ kinetin. The capacities for callus, root and shoot formation from cotyledon and hypocotyl explants were remarkably different among cultivars of B. napus tested. The calli induced from hypocotyl produced more shoots than those from cotyledon.
Embryogenic calli from Codonopsis lanceolata L. were cultured in MS liquid media and supplemented with various concentrations of primary carbon sources to study the effects of carbohydrates and osmoticum on somatic embryogenesis and somatic embryo morphology. Sucrose, glucose, and a combination of 3% sucrose and various concentrations of sorbitol or mannitol as osmoticum were used as carbon supplements. The maximum number of somatic embryos per flask was greater in media exclusively supplemented with 3% sucrose (128.29) than exclusively glucose-supplemented media (47.67) and either supplement combination of 3% sucrose and osmoticum (95.67 with mannitol and 114.00 with sorbitol). The number of somatic embryos gradually decreased in media with increasing concentrations of combined osmoticum supplement. Decreases also occurred in the highest concentrations of sucrose- and glucose-supplemented media. The total frequency of somatic embryos with two cotyledons was slightly higher in medium with 3% + mannitol (24.09%) compared with exclusively sucrose (21.52%), glucose (21.22%), or 3% sucrose + sorbitol (22.13%). As concentrations of sucrose and glucose increased, the occurrence of two cotyledons and trumpet cotyledons gradually decreased and the occurrence of polycotyledon and globular stage embryos increased. Furthermore, as concentrations of 3% sucrose and osmoticum increased, the occurrence of trumpet cotyledon and globular stage embryos increased and the occurrence of polycotyledon gradually decreased. These results demonstrated that the somatic embryogenesis and occurrence of cotyledon morphology were influenced by the concentration of carbohydrates and combinations of 3% sucrose and osmoticum supplements.
Several physiological responses were investigated in plants treated with TOPE as a preliminary step to know its action site. Unlike photo-dependent diphenylethers, herbicidal activity of TOPE appeared slowly and its typical symptoms were both burning of leaf blades and abnormal division of meristem in grasses, Similarly, both leakage of cell electrolytes and the curling of cotyledon margin were also shown in cucumber(Cucumis sativus L.). Biosynthesis of chlorophyll in etiolated cucumber cotyledon was not inhibited directly by treatment of TOPE at low light intensity(5.5${\mu}$ mol $m^{-2}s^{-1}$ PAR) and protoporphyrin IX was not also accumulated. The contents of phytoene, phytofluene and ${\beta}$-carotene were abnormaly increased. Photosynthesis was inhibited only at high concentration. Mitochondrial respiration was inhibited at high concentration but rather increased significantly at 10${\mu}$M of TOPE. However, respiration inhibitors did not alleviate the two symptoms of TOPE in cucumber cotyledon. In the same experiments, using inhibitors of protein or nucleic acid biosynthesis, only one of the two symptoms was alleviated by chloramphenicol and cycloheximide. In contrast, both symptoms were alleviated by actinomycin-D and hydroxyurea, suggesting that nucleic acid metabolism might be preferentially related to the mode of action of TOPE. DNA, RNA and protein contents were accumulated in both cucumber cotyledon and rice (Oryza sativa L.) routs treated with TOPE, and the DNA of them was increased at first. Thus, it is conjectured that TOPE increase nucleic acid metabolism directly or indirectly, and then disturb various metabolic pathways causing abnormal physiological and morphological effects followed by final death.

This study was conducted to investigate the effect of grafting methods on plug seedling quality, growth after transplanting, and yield of oriental melon that those seedlings were raised in the 32 hole plug tray for protected cultivation. Number of leaf and leaf area in approach grafting were higher until 15 days after grafting but lower on 20 days after grafting. Plant height, stem diameter and content of chlorophyll were nearly the same of each grafting methods on 20 days after grafting. Growth of top part, that is, plant height, leaf number, and leaf area on root removed single cotyledon ordinary splice grafting was the highest among the grafting methods. In the underground part, dry weight of root was high and T/R ratio was the lowest. Seedling quality of root removed single cotyledon ordinary splice grafting was the highest among grafting methods before transplanting. Plant height, number of leaf, leaf area and dry weight of root removed single cotyledon ordinary splice grafting in the growth of field were the highest on 30 days after grafting among grafting methods and days required for first flowering was also shorter, 38.4 days among grafting methods. But Fruit weight, content of soluble solids, fruit hardness, and color characteristics in fruit quality were insignificant among each grafting methods. Consequently, grafting methods influenced on the seedling quality, of oriental melon early growth after transplanting and yields. It was concluded that root removed single cotyledon ordinary splice grafting was the best methods in present study. It will be needed to convert grafting methods. Thus new grafting method should be applied.
