In order to fine antioxidant and whitening agent source from nature, the comparisons of antioxidative activity and tyrosinase inhibitory activity were carried out for various ethanol extract on Pueraria Radix, Poria Cocos, and Coptidis Rhizoma. Comparing for three ethanol extracts, the highest electron donating ability was found at Poria Cocos (81.2%), but, the highest SOD-like ability, at the Coptidis Rhizoma(58%). Xanthine oxidase experiment exhibited 23.3% of hindrance effect in Pueraria Radix, and 79.3% in Poria Cocos. To evaluate at the whitening effect, tyrosinase inhibitory activity was conducted. Tyrosinase inhibitory activity was detected at 17.9% in the Puerariae Radix, 5.2% in the Poria Cocos and 83.3% in the Coptidis Rhizoma. From these results, we suggest that the ethanol extracts from Pueraria Radix, Poria Cocos, and Coptidis Rhizoma can be used for cosmetic ingredients.
Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

발아 과정을 통한 식물 종자의 변화를 확인하고, 발아된 식물종자로부터 새로운 펩타이드를 개발하여 항노화 화장품 소재로서의 가능성을 확인하였다. 발아과정을 통한 식물 종자의 변화는 단백질 양 측정, 겔 투과 크로마토그램(GPC)과 SDS-PAGE를 통한 단백질 분자량 분포 변화, 아미노산 조성분석을 통해 발아 전과 비교하였다. 발아된 검은 쌀로부터 펩타이드를 생산하기 위해 단백질 분해효소를 처리하였으며, 생산된 발아 검은 쌀 펩타이드의 효과를 발아 검은 쌀 단백질과 비교하여 확인하였다. In vitro matrix-metalloprotease (MMP) 활성 저해효과는 형광 정량법을 이용하였으며, 인간 섬유아세포 활성화 효과를 확인하였고, 콜라겐 생합성 촉진효과와 자외선 조사에 의한 MMP 발현 저해효과는 효소면역분석법(ELISA)을 이용하여 확인하였다. 발아에 의해 검은 쌀의 단백질량, 단백질 분자량 분포 및 아미노산의 조성이 변화함을 확인하였으며, 검은 쌀의 발아와 단백질 분해효소중 파파인 처리를 병행함으로서, 상대적으로 낮은 분자량을 갖는 신규 펩타이드의 제조가 가능하였다. GPC를 이용하여 제조된 발아 검은 쌀 펩타이드의 분자량 분포를 확인한 결과, 대부분이 900dalton 이하의 분자량을 가지는 것을 확인하였으며, 또한, 이러한 발아 검은 쌀 펩다이드는 약 40% 정도의 인간 섬유아세포 활성화 효과를 가지며, 또한 농도 의존적으로 콜라겐 분해효소 활성 저해효과를 가지는 것을 확인하였다. 또한, 자외선 조사에 의해 유도되는 콜라겐 분해효소 발현을 약 50% 억제하며, 콜라겐 생합성을 약 20% 촉진하는 것을 확인할 수 있었다.
본 연구에서는 수입에 의존되는 아로마 에센셜 오일의 의존도를 낮추고 국내 수요 및 재배가 늘고 있는 허브 식물의 가치를 높일 수 있는 차별 방안을 찾고자 SPME 법으로 국내 재배 라벤더 꽃의 원물 손상 없이 향기 성분만을 분석 재현해내는 연구를 통해 수입 라벤더 에센셜 오일과의 향기 구성성분 차이를 확인하고 국내 재배 허브 식물의 향기 독창성을 찾고자 한다. 라벤더 에센셜 오일은 총 38 종, 라벤더 꽃은 총 27 종의 향기 성분이 분석되었으며 이들 중 공통성분은 myrcene, d-limonene, 1,8-cineol, ocimene, p-cymene, α-terpinolene, camphor, linalool, linalyl acetate, bornyl acetate, 4-terpineol 및 a-terpineol 으로 총 12 종이 유일하였다. 또한 두 시료의 향기 구성성분 중 국내 재배 라벤더 꽃의 알러젠 종류와 함량이 적음을 확인할 수 있었으며 15 명의 패널을 대상으로 관능평가를 시행한 결과 국내 재배 라벤더 꽃 재현향이 종래까지 산업에 활용되어져 온 라벤더 에센셜 오일보다 높은 호감도를 나타내어 알러지 유발성분의 함량을 낮춘 라벤더향으로 산업화에 확대 가능할 것으로 사료된다.
Atopic dermatitis is a chronic inflammatory skin condition including severe pruritus, xerosis, visible eczematous skin lesions that mainly begin early in life. Atopic dermatitis exerts a profound impact on the quality of life of patients and their families. The estimated lifetime prevalence of atopic dermatitis has increased 2~3 fold during over the past 30 years, especially in urban areas in industrialized countries, emphasizing the importance of life-style and environment in the pathogenesis of atopic diseases. While the interplay of individual genetic predisposition and environmental factors contribute to the development of atopic dermatitis, the recent increase in the prevalence of atopic dermatitis might be attributed to increased exposure to various environmental factors rather than alterations in human genome. In recent decades, there has been an increasing exposure to chemicals from a variety of sources. In this study, the effects of various environmental chemicals we face in everyday life - air pollutants, contact allergens and skin irritants, ingredients in cosmetics and personal care products, and food additives - on the prevalence and severity of atopic dermatitis are reviewed.

Strategies to make finished products more stable and efficacious can take advantage of formulation technologies- ingredients and techniques - to improve their delivery into the skin by both enhanced penetration and delayed release. Nanometer range particles and emulsion droplets can be formed with a selection of either silicone copolyol or conventional organic surfactants and dispersion shear rates sufficient to generate stable submicron droplets. By incorporating these into systems with biomimetic liquid crystalline gel networks in either simple or multiple phase emulsions the skin delivery can be enhanced as shown in tape stripping experiments. Such systems can stabilize labile actives, such as Vitamin C and retinol, and aid delivery. Laboratories in U.S and Europe investigated actives including green tea polyphenols, salicylic acid and methyl and benzyl nicotinate.

Herbal medicines have been used as a basic means of clinical trial throughout history, and traditional medicines are targeted to seek functional components. To discover new cosmetic or food ingredients among numerous natural resources from Jeju island, we screened for inhibition activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Although NO formation plays an important role to relax vascular muscles or eliminate tumors, NO produced excessively in inflammatory condition can cause metabolic diseases or inflammatory dysfunctions. Among 52 natural resources ethanol extracts, 5 extracts inhibited NO production over 25% compared to only LPS-treated control at the concentration of $100{\mu}g/mL$. In further study, we try to investigate other bio-activities and the phytochemicals of 5 different extracts as useful ingredients for cosmetics or functional foods.
The matrix metalloproteinases (MMPs) are a family of enzymes responsible for degrading connective tissue. MMPs catalyze the breakdown of collagen from the extracellular matrix, leading to wrinkle formation and accelerated skin aging. Furthermore, ultraviolet irradiation causes increased expression of certain MMPs. In the extracellular matrix turnover, MMPs are interacting with endogenous regulators named tissue inhibitors of metalloproteinases (TIMPs). Using peptide substrate assays, it has been demonstrated that TIMP-MMP complexes interact highly specifically with $K_{i}$ values of 10$^{-9}$ -10$^{-16}$ M. Therefore applications for TIMP as inhibitor of collagen degradation are suggested for cosmetic anti-aging products to prevent wrinkle formation and loss of elasticity. To date four TIMP proteins (TIMP-1, TIMP-2, TIMP-3 and TIMP-4) have been identified which show a high degree in sequence similarity. The production of human TIMP-2, a 194-residue nonglycosylated protein, was performed by fed-batch culture of Escherichia coli. TIMP-2 accumulated in the bacterial cells in an insoluble form as inclusion bodies. The inclusion bodies were solubilized and the protein refolded to yield the native TIMP-2 in the active form. The integrity of the protein was confirmed by mass analysis, Edman sequencing and gel shift experiments with authentic samples. The inhibitory activity of the refolded and purified TIMP-2 was demonstrated with MMP-1 and MMP-2 assays using synthetic fluorogenic peptide substrates.s.
Vitamin C or L-ascorbic acid, an essential nutrient for humans with antioxidative property, was coated with natural wax to be used as functional additives for cosmetics. Coating of vitamin C was performed using carnauba wax and olive wax in self-designed reaction chamber for 60 min. The yield of coated vitamin C with carnawuba wax and olive wax were up to $94.6{\pm}1.2%$ and $82.5{\pm}1.4%$, respectively. The stability of the carnauba wax-coated vitamin C in the lotion was improved 17.8% than that of uncoated vitamin C during 30 day storage at $25^{\circ}C$. Color and the layer separation of the lotion added with the coated vitamin C showed better property than with the uncoated vitamin C. The coated vitamin C can be applied to cosmetic ingredients as a stable additive.

In this study, we have investigated the potent anti-aging effect of DA-3711, a cosmetic ingredient derived from artificial skin culture. The artificial skin was originally developed as a skin replacement for the treatment of chronic skin wounds. To produce DA-3711, neonatal human fibroblasts were seeded into biocompatible collagen/chitosan/glycosaminoglycan (GAG) scaffolds and cultured in Dulbecco's modified Eagle's medium (DMEM) supplemented with fetal bovine serum and nonessential amino acids. Analysis of the culture broth (DA-3711) showed that growth factors such as VEGF, TGF-$\beta$, KGF were present at significantly higher levels that in the culture broth of fibroblasts cultured in monolayer. The biological activity of DA-3711 was assessed by measuring in vitro cell proliferation and collagen synthesis of normal human fibroblasts. Fibroblasts treated with 10% DA-3711 showed a 2-fold higher proliferation and 2 to 4-fold higher collagen synthesis than untreated cells. DA-3711 also exhibited anti-oxidative effects, since cells under peroxide-induced oxidative stress showed a 30% higher viability in DA-3711-containing medium than in medium without DA-3711 addition. The results suggest that DA-3711 may have anti-aging effects by stimulating skin regeneration and protecting against oxidative stress.
