Studies on Antioxidant, Anti-inflammation and Moisturizing Activities of Gastrodia elata Flower Extract (천마꽃 추출물의 항산화, 항염, 보습 활성에 대한 연구)

  • Gastrodia elata has a very low pollination rate in natural state, and even in artificial cultivation, there are very few individuals that bloom due to the degeneration, so little studies have been conducted. This study confirmed that the potential as a cosmetic ingredient by evaluating the antioxidant activity through the evaluation of DPPH radical scavenging activity, anti-inflammatory activity through the inhibitory effect on nitric oxide production, and the moisturizing activity through the effect on promoting hyaluronic acid production by artificially flowering G. elata flower. It was also confirmed that the appearance rate and flowering rate of G. elata harvested in spring were high, and the content of gastrodin was 0.36%. The IC50 value of G. elata flower extract was 0.045% and it was confirmed that G. elata flower extract had higher radical scavenging activity than G. elata root extract. The NO production inhibitory activity against the flower extract showed a significant inhibitory effect from 1% to 83.2%. Hyaluronic acid production promotion efficacy was not confirmed in the G. elata root extract, but the production rate increased with concentration dependence in the flower extract, and it was the highest at 46.9% when 0.02% treatment was performed. Based on the above research results, it is judged that G. elata flower extract has high potential for use as an antioxidant, anti-inflammatory, and skin moisturizing cosmetic ingredient.

Development of Whitenin Agents by Synthesis of Polyhydroxy Aromatic Compounds

  • Some natural polyhydroxy aromatic compounds have inhibitory activity against tyrosinase, key enzyme for formation of melanin pigment. We examined the structure-activity relationship of the natural polyhydrowy aromatic compounds and synthesized a number of new derivetives through various methods. Skin lightening effects of these compounds were examined through inhibition of mushroom tyrosinase and inhibitory of melanogenesis on B-16 melanoma cells. These new compounds showed strong inhibitory activity against tyrosinase. Good lightening effects sue to inhibition of melanogenesis were observed from several resorcinol and pyrogallol derivatives. In toxicological tests such as skin primary irritation and sensitization, the above compounds were sufficiently safe for cosmetic use.

  • In eastern Asian region, cosmetic preparations containing the molecues possessing the whitening activity are promising. In this study, 100 plant extracts were screened in order to elucidate the whitening effects using in vitro tyrosinase assay and DOPA autooxidation. Several plant extracts such as Morus alba, Glycyrrhiza glabra showed the inhibitory activity (>50%) against mushroom tyrosinase at the concentration 333$mu extrm{m}$/ml. And plant extracts including Morus alba and Bupleurum falcatum showed the inhibitory activity (>50%) at the concentration of 500($\mu\textrm{g}$/ml against DOPA autooxidation. The IC50 values of the several extracts were also found.

A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative (인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구)

  • Here, we conducted the study on the possibility of using FK-13, a short analog of human-derived antibacterial peptide LL-37, as a cosmetic preservative to discover a natural cosmetic preservative that is safe for human body. For the purpose, FK-13 composed of 13 amino acids was synthesized by solid-phase peptide synthesis, and purified using reversed phase-high performance liquid chromatography (RP-HPLC). The purity and molecular weight were confirmed by liquid chromatography-mass spectrometry (LC-MS) analysis. FK-13 showed high antimicrobial activity on the three gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, and Staphylococcus epidermidis), the three gram-negative bacteria (Escherichia coli, Salmonella typhimurium, and Pseudomonas aeruginosa), and also even the fungus Candida glabrata. FK-13 had a broad spectrum of antibacterial activity, showing a suitability as a cosmetic preservative. In addition, FK-13 showed high thermostability and higher antibacterial activity in a comparative test with existing natural herbal cosmetic and chemical preservatives. Therefore, as FK-13 is a safe material and has high antibacterial activity at a low concentration, it is likely to be applied as a peptide natural cosmetic preservative that can replace existing chemical preservatives.

Component Analysis of Persicaria hydropiper L. Extracts (여뀌 추출물의 성분 분석)

  • In the previous study, we reported the antioxidative activity, antiaging activity, antibacterial activity and moisturizing effect of cream containing Persicaria hydropiper L. extract. In this study, the components of Persicaria hydropiper L. extract were analyzed by TLC and HPLC. Aglycone fractions obtained from the deglycosylation reaction of ethyl acetate fraction among the Persicaria hydropiper L. extract, showed 2 bands and 2 peaks in TLC and HPLC experiments, respectively. Two components were identified as quercetin and kaempferol. TLC chromatogram of ethyl acetate fraction of Persicaria hydropiper L. extract revealed 6 bands and HPLC chromatogram showed 7 peaks, which were identified as quercetin, hyperin, isoquercitrin, quercitrin, kaempferol. In conclusion, with the antioxidative activity, antiaging activity, antibacterial activity and moisturizing effect reported previously, component analysis of Persicaria hydropiper L. extracts could be applicable to new cosmeceuticals.

Anti-oxidant and Whitening Effects of Oryza sativa cv. Heugjinmi Extracts (흑진미(Oryza sativa cv. Heugjinmi) 추출물의 항산화 및 미백 효능 연구)

  • In this study, the efficacy of antioxidant and whitening factors was analyzed in order to verify the possibility of use as functional cosmetic materials related to antioxidant and whitening by using the extract of Oryza sativa cv. Heugjinmi. As a result, S OD-like activity, FRAP, reducing power, xanthine oxidase inhibitory activity, and elastase inhibitory activity were similar to those of the control group. Tyrosinase inhibitory activity had no effect in hydrothermal extract and 59% inhibitory activity in ethanol extract. Ethanol extract was found to inhibit cellular tyrosinase inhibitory activity and melanin biosynthesis at concentrations of 25, 50, and 100 ㎍/mL, which will not affect survival in the B16F10 cell line. In addition, the results of confirming the mRNA expression of tyrosinase, TRP-1, and TRP-2 showed inhibition rates of 37%, 51%, and 34%, respectively, at the highest concentration of 100 ㎍/mL. Therefore, it is believed that O. sativa extract has potential to be utilized as a functional cosmetic material related to antioxidant and whitening.

Brazilin as a new sunless tanning agent

  • To develop an active material for skin darkening, we examined the effect of 300 plants on tyrosinase activity and found only Caesalpinia sappan has an ability to increase tyrosinase activity highly and melanin contents in B-16 melanoma cells. A compound increasing tyrosinase activity and melanin production was isolated from Caesalpinia sappan Lignum. Brazilin was identified as a new active agent. Brazilin increases the tyrosinase activity and malanin production of B-16 melanoma cells. In conclusion, it seems that brazilin cal be used as a new sunless tanning agent.

  • A tyrosinase inhibitor, Lonjapoin, was isolated from the ethanolic extracts of Flos Lonicera by activity-guided fractionation. Lonjapoin ($IC_{50}$/; 2.07 $\mu\textrm{g}$/mL) was found to be potentially as effective inhibitor of production of melamin. Flos Lonicera (Jinyinhua) is the dry flower buds of Lonicera japonica. Thunb. It is used as an antibacterial and antiphlogistic agent in the treatment of abscess, laryngeal catarrh, erysipelas, dysentery, cold and fever. In our continuing search for natural source as cosmetic ingredients from Chinese medicinal herbs and found the extracts of Flos Lonicera showed the significant tyrosinase inhibition activity. In this study the active constituent of Lonicera japonica. obtained with the process of tyrosinase activity assay and column chromatography.

Antimicrobial Activity of Biosurfactants

  • Biosurfactant has antibiological activities. Due to its low toxicity, biosurfactant can be applied to human health care products. Posseing the antibiological activities as well as the surfactant properties implies that biosurfactant can be widely used in the cosmetic industry. In this presentation, applications of biosurfactant have been reviewed including the antimicrobial activities of sophorolipid. Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm as MIC (minimum inhibitory concentration). Also 100 ppm of sophorolipid inhibited 50% of cell growth of plant pathogenic fungus, B. cinera. However, sophorolipid showed no effect on the Escherichia coli., indicating its selective antimicrobial activity depending on the cell wall structure. Treatment of B. subtilis with sophorolipid increased the leakage of intracellular enzyme, malate dehydrogenase, indicating the possible interaction of sophorolipid with cellular membrane. Between lactone-type and acid-type sophorolipid, the former showed higher antimicrobial activity.

A Study on Peanut Spouts Extract as the Anti-oxidant Activity and the Skin Whitening Cosmetic Ingredients (항산화 및 미백화장품 원료로서의 땅콩새싹 추출물에 관한 연구)

  • To investigate the effect of peanut sprout extract on skin care, we measured anti-oxidant activity and whitening action. As a result of measuring DPPH radical scavenging activity to examine independent anti-oxidation of peanut sprout extract, there was strongly scavenging activity. Fluorescent material DCF-DA was used to measure hydrogen peroxide created in RAW 264.7 cells, and all concentration dependently decreased ROS production. As a result of measuring nitric oxide to examine anti-inflammation of peanut sprout extract, there was strongly inhibited nitric oxide production in RAW 264.7 cells. Tyrosinase activation was found to inhibited dose-dependant. Melanin production was also prevented dose-dependant. Therefore, it is expected to be used effectively in development of functional cosmetic materials.
