Effects of Chrysanthemum Extract Drinks on Sociopsychological Stress (사회.심리적 스트레스에 미치는 국화 추출 음료의 영향)

  • The effects of anti-stress chrysanthemum flower (ASCF) extract drink (ASCF-0.1% and ASCF-0.5% concentrations) were tested for the anti-stress action. ICR male mice (20$\pm$2 g) were fed with basic experimental diets and given free access to water containing these ingredients for 18 days. Psychological stress/sociopsychologcal stress exposed by foot-shock for 1 hour for 3 days. Both ASCF-0.1 and ASCF-0.5 groups in the sociopsychological stress resulted in a significant decrease of 28.1% and 27.3% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 49.7% and 53.9%, respectively, in ASCF-0.1 and ASCF-0.5 groups compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxyphenylethy-leneglycol sulfate) levels in the brain resulted in a marked decreases of 12.9% and 16.6%, respectively in ASCF-0.1 and ASCF-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASCF-0.1 and ASCF-0.5 groups resulted in a significantly increase of 71.3% and 81.0%, respectively, compared with control group. These results suggest that anti-stress chrysanthemum flower (ASCF) drink can effectively ridded the sociopsychological stress.

  • The effects of anti-stress mind tonic (ASMT) drink (ASMT-0.1% and ASMT-0.5% concentration) were tested for the anti-stress action. ICR male mice (20$\pm$2 g) were fed with basic experimental diets free access to water containing these ingredients for 18 days. Psychological stress/sociopsychological stress were exposed by foot-shock for 1 hour for 3 days. Both ASMT-0.1 and ASMT-0.5 groups in the sociopsychological stress resulted in a significant decrease of 19.1% and 41.9% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 23.4% and 35.9%, respectively, in ASMT-0.1 and ASMT-0.5 groups compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxyphenylethyleneglycol sulfate) levels in the brain resulted in a marked decreases of 27.1% and 19.6%, respectively in ASMT-0.1 and ASMT-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASMT-0.1 and ASMT-0.5 groups resulted in a significantly increase of about 70% compared with control group. These suggest that mind tonic anti-stress drink can effectively ridded the sociopsychological stress.

  • The effects of anti-stress rose fruit (ASRF) extract drink (ASRF-0.1% and ASRF-0.5% concentrations) were tested for the anti-stress action. ICR male mice (20$\pm$2 g) were fed with basic experimental diets and given free access to water containing these ingredients for 18 days. Psychological stress/sociopsychological stress exposed by foot-shock for 1 hour for 3 days. Both ASRF-0.1 and ASRF-0.5 groups in the sociopsychological stress resulted in a significant decrease of 24.3% and 38.0% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretion in the brain were significantly increased 35.9% in ASRF-0.5 group, but NA secretion in ASRF-0.1 group could not be obtained significant result compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxy-phenylethy leneglycol sulfate) levels in the brain resulted in a marked decreases of 16.5% and 18.9% respectively in ASRF-0.1 and ASRF-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASRF-0.1 and ASRF-0.5 groups resulted in a significantly increase of 16.5% and 67.0%, respectively, compared with control group. These results suggest that anti-stress rose fruit (ASRF) drink also can effectively ridded the sociopsychological stress.

Effect of Coicis Semen on Starvation Stress in Mice (억이인이 생쥐의 기아 Stress에 미치는 영향)

  • Objective : In 2001, the rate of obesity in Korea reached 30.6%. There are many therapeutic ways to reduce body weight, such as low and very low calorie diet, exercise therapy, behavior modification therapy, etc. However, in many cases the patients feel stress under obesity treatment because of starvation. This study was aimed to evaluate the anti-starvation stress effect of Coicis Semen on mice. Methods : First, the mice were divided into 6 groups : Normal (group with no starvation), Control (administrated normal saline 6 times before starting 36 hours starvation), and Samples A, B, C, and D (administrated 0.25, 0.5, 1.0, 3.0 g/kg Coicis Semen respectively 6 times before starting 36 hours starvation). Then the plasma corticosterone level and rectal temperature were measured. The norepinephrine, dopamine, DOPAC (dihydroxy-phenylacetic acid), 5-HT (5-hydroxytryptamine) and 5- HIAA (5-hydroxy-indole-acetic acid) in the hypothalamus were measured by the HPLC method. Result : I. The rectal temperature in Sample group D showed a significant difference (P<0.05) compared with the Control group. 2. The DOPAC in Sample groups A, C and D showed the significant difference (P<0.05) compared with the Control group. Conclusion : It might be recognized that Coicis Semen has an anti-starvation stress effect.

Dietary Salt Modulates the Adrenocortical Expression of P450 11Beta-hydroxylase in Mice

  • This study was conducted to determine the effect of dietary salt on the synthesis of glucocorticoids in the adrenal cortex of mice. Mice had ad libitum access to 3% sodium chloride as the only drinking fluid (high salt diet) for either 4 days or 4 weeks. Adrenocortical expression of cytochrome P450 11beta-hydroxylase, a major regulatory enzyme in the biosynthesis of glucocorticoids, was examined by immunohistochemistry and western blot analysis. Ultrastructure of adrenocortical cell and plasma level of corticosterone were analyzed as well. Size and density of lipid droplets in the cortical cell were increased by high salt diet. Four days of high salt diet decreased P450 11beta-hydroxylase in the adrenal cortex, but 4 weeks increased it. Plasma level of corticosterone changed in parallel with the Cortical level of P450 11 beta-hydroxylase. These results suggest that high salt diet may modulate the biosynthesis of glucocorticoids, at least partly, via regulating the expression of P450 11beta-hydroxylase in adrenocortical cells.

A Study on Immunomodulation and Serotonin Metabolism Changes by Immobilization Stress in Mice Fed Tryptophan Supplemented Diet. (고 트립토판 식이를 섭취한 마우스에서 Immobilization 스트레스로 인한 면역변조와 Serotonin 대사의 변화에 대한 연구)

  • We fed high trypotophan diet(3.5% tryptophan/diet(w/w) to mice for 7 days and treated then with 3 hour immobilization(IMMB) stress to investigate tryptophan metabolism and immunomodulation. The levels of serum tryptophan, brain tryptophan, serotonin(5HT) and 5-hydroxyindoleacetic acid(5HIAA) in the tryptophan diet fed animals were higher than those of the normal diet fed animals. Feeding tryptophan supplemented diet to stressed animal significantly decreased the levels of serum and brain tryptophan and 5HT levels. However, the amount of 5HIAA which is the metabolite of serotonin was increased in brain. Plasma corticosterone level was increased by the stress in both groups but the degree of this increase was smaller in high tryptophan fed animals. The relative numbers of CD8+ T cells, CD4+ T cells and B cells in spleen were decreased in high tryptophan diet fed and stressed animals compared to control diet fed and no stressed animals. CD8+ T cells decreased more than CD4+ T cells. The decrease of CD8+ T cells in high tryptophan fed and stressed animals was similar to that in high tryptophan fed animals or normal diet fed and stressed animals. Stress and tryptophan supplement acted synergistically to decrease the number of B cells. This study suggests that stress and tryptophan supplement could modify the number of lymphocyte cells, and indicates that the interaction of stress and tryptophan supplement on immune fuction depends on the types of immune cells.

Comparative Study of Hwangnyeonhaedok-tang and Geongangbuja-tang on the Plasma Hormones Level in Mice Exposed to Cold Stress (황련해독탕(黃連解毒湯)과 건강부자탕(乾薑附子湯)의 Cold Stress로 유발된 생쥐의 혈중(血中) 호르몬 농도변화에 대한 비교연구)

  • Objectives : The aim of this study was to evaluate the efficacy of Hwangnyeonhaedok-tang (HH) and Geongangbuja-tang (GB) on the plasma hormone level in mice exposed to cold stress. HH and GB are the representative prescriptions of cold and hot property, respectively. Methods : We established cold condition by confining ICR mice to a $4^{\circ}C$ cage for 24 hours, ICR mice were given a HH (100, 300, 1000 mg/kg) or GB (100, 300, 1000 mg/kg) extract orally twice a day for three consecutive days. From the second day, they were given cold stress ($4^{\circ}C$) for twenty four hours. To measure the plasma corticosterone, insulin, thyroxine, epinephrine and norepinephrine levels of mice, their blood samples were collected from cardiac puncture, immediately centrifuged at $4^{\circ}C$. The protein level of HSP70 and JNK was examined using western blot analysis in cortex and hypothalamus. Results : Oral administration of GB more significantly reduced plasma corticosterone level raised by cold stress than HH. Gardeniae Fructus (CJ), the constituent of HH, significantly increased the thyroxine level. Western blot analysis showed that cold stress-induced Heat shock protein 70 (HSP70) expression was increased by HH and GB, HH decreased JNK expression and GB increased JNK expression dose-depently in hypothalamus. Scutellariae Radix (HG), Zingiberis Rhizoma (GG) and Aconiti Tuber (BJ) decreased HSP70 in hypothalamus and GG, BJ decreased HSP70 in cortex as well. Conclusions : These results suggest Geongangbuja-tang (GB) is more effective for ameliorating the stress response caused by cold stress.

The Effects of Yunjogyosim-tang on Stress and Sleep-related Substance of Rats Induced by Chronic Mild Stress (Chronic Mild Stress 유발 흰쥐에서 윤조교심탕(潤燥交心湯)이 스트레스 및 수면 관련 호르몬에 미치는 영향)

  • Objectives: The study aims were to evaluate the Anti-stress and Sleep-inductive effects of Yunjogyosim-tang (YJGS) in rats induced by Chronic Mild Stress (CMS). Methods: 25 healthy Rats were randomly divided into 5 groups; Normal, CMS (Control), YJGS 50, YJGS 100, and YJGS 200. All rats except the normal group were exposed to random stress conditions like water deprivation, empty bottles, forced runs on the treadmill, etc. according to the timetable of CSM for 3 weeks. After a week starting the experiment, Rats in YJGS 50, YJGS 100, and YJGS 200 groups were fed orally once a day for 2 weeks. Blood samples were taken afterward from each rat for analysis of Complete blood count, AST, ALT, and glucose levels. Corticosterone, serotonin, and melatonin were measured by ELISA kit. BDNF and $TNF-{\alpha}$ were measured by RT-PCT. Results: 1. Corticosterone levels were lower in YJGS 50, and YJGS 200 groups compared to the control group. 2. Serotonin levels were lower in the YJGS 100 and YJGS 200 groups compared to the control group. 3. Melatonin levels were higher in the YJGS 50 group compared to the control group. 4. Activity levels were higher in the BDNF, and YJGS 50 groups compared to the control group. 5. In the Activity of $TNF-{\alpha}$, YJGS 50 group showed a significant decrease compared to the control group. 6. In the Glucose content, YJGS 50, YJGS 100, and YJGS 200 groups showed a significant decrease compared to the control group. 7. When analyzing leukocytes, monocytes were found to be increased in YJGS 100 and YJGS 200 groups compared to the control group. 8. When analyzing red blood cells, more changes occurred to erythrocytes and thrombocytes in the YJGS 50 and YJGS 200 groups than in the control group. Hemoglobin was significantly increased in the YJGS 100 group and platelet levels were significantly increased in all experimental groups compared with the control group. Conclusions: These results suggest that Yunjogyosim-tang has anti-stress and sleep-inductive effects on rats induced by CMS.

Effects of an Unexpected Change in Housing Environment on Stress in Poultry (급격한 사육환경의 변화가 가금의 생산성과 스트레스에 미치는 영향)

  • The objective of this study was to evaluate the effects of an unexpected change in housing environment on stress in poultry. Laying hens (Hy-line Brown), adapted to a free-range housing system for more than a month, were randomly divided into two groups to be subjected to an unexpected change of their housing environment: one half of them were individually housed into conventional battery cages and the other continually left in the same housing system throughout the entire period of the study. The sudden change resulted in an increase in egg production and albumen height (P<0.05), but decrease in eggshell thickness (P<0.05). The change had tendency to increase Haugh unit ($P{\leq}0.061$) and to decrease eggshell color ($P{\leq}0.074$), but did not affect body weight for 5 days thereafter. No significant changes were detected in liver color. Plasma concentrations of corticosterone was acutely increased one day after the change (P<0.05), then returned to similar control levels. The results of the present study indicate that unexpected changes in housing environment cause acute increase in stress hormone concentrations, but interpretation of the results should be cautious due to the experimental conditions.

Anti-stress Effect of Scutellatia baicalensis in SD Rats and ICR Mice

  • The aim of this study is to investigate anti-stress effect of Scutellaria baicalensis(SB). The experiments were performed with the use of young (9 weeks of age) male rats of SD strain and the male ICR mice (20-25 g) at the time of first treatment with SB. Animals of the normal group were not exposed to any stress and the control group were exposed to stress. The rats of the Ginseng, Diazepam(BZ) and SB supplementary group were orally administered once a day 100 mg of red ginseng extract, 5 mg of BZ or 100 mg of SB extract/kg body weight and they were exposed to stress. The mice of the Ginseng, BZ and SB supplementary group were given water containing 200 mg of red ginseng extract, 10 mg of BZ or SB extract/100 ml potable water and exposed to stress. Animals were given supplements for 7 days without stress, and then were given supplement for 5 days with restraining and electroshock stress. We recorded stress related behavioral changes of the experimental animals by stressing them using the Etho-vision system and measured levels of blood corticosterone and IL-2. SB supplementation partially blocked the stress effect on locomotion in the rats and mice, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, grooming, smelling, and rearing behavior in the rats and smelling, grooming, tailing, and rearing in the mice. in elevated plus maze test, the staying time of the stressed rats and mice in the open area decreased while it increased in the closed area. But these changes also partially were blocked by SB-supplementation. SB-supplementation decreased levels of the blood corticosterone which was increased by stress in the rats but did not significantly increase levels of blood interleukin 2 which was decreased by stress in mice.
