Studies on the Relation between Allatectomy (picking out of corpora allata) and abnormal colouring pupa, Bombyx mori L. (인후측선적출과 이상착색용과의 관계에 관한 연구)

  • In order to identify hormone activities which related to the changing of cutaneous tissue and pupal bodies and also to identify the interaction of corpora allata hormone and prothoracic glandular band hormone which act upon the metamorphosis of the insect. The item of treatment is carried out in 5 different times at the interval of about 12 hours after 4 weeks from the first feeding of 4th instar stage, and as far as 5th instar stage in concerned 6 times of item treatments have been carried out, at about 12 hours interval after 72 hours from the molting, Item treatments were, made in the stage of spring, autumn rearing, However, during the period of post mounting stage and pre pupation by picking out of corpora allata in 5 times at the interval of hours the following change of pupal colour is observed; 1) By an early pick out of 4th instar stage, the three molters are found in greater number, However, by a later pick out the four molters are found instead of others. 2) An abnormal colour is found in the three and four molters when comparing with the control molters. 3) The most of control molters have shown the normal colour except a few samples. 4) As shown in the table 1 and 2, the death number of item treatment is greater in the autumn rearing season. 5) The picking out of corpora allata of 5th instar stage, the post mounting stage and pre pupation period is affecting the change of colour. 6) As a final conclusion, the corpora allata hormone is closely related to the changing of pupal colour.

Immunolocalization of Allatotropin Neuropeptide in the Developing Brain of the Silk Moth Bombyx mori

  • Polyclonal antiserum against Manduca sexta allatotropin has been utilized to investigate the localization of allatotropin-immunoreactivity in the brain of the si1k moth Bombyx mori. Manduca sexta allatotropin-immunoreactive (Mas-AT-IR) neurons were found in all larval brains investigated, but not in prepupal, pupal and adult brains. In the larval stages, first appearance of Mas-AT-immunoreactivity w8s shown in the brain of first instar larvae, which contains four pairs of bilateral Mas-AT-IR cell bodies. Labeled neurons increased to six pairs in the second instar larval brain, including two pairs of median neurosecretory cells in the pars intercerebralis. In the third and fourth instar larvae, five pairs of labeled cell bodies were distributed throughout each brain. In the fifth instar, there were about ten pairs of bilateral cell bodies in the day-1 brain, about seven pairs in the day-3 brains, and five pairs in the day-5 brains, respectively. Mas-AT-labeling was observed in both axons within nervi corpora cavdiaci (NCC) 1+11 and corpora allata. This suggests that the Mas-AT produced from the brain neurons is transported via some axons of the NCC 1+11 and nervi corpora allati I to the corpora allata, which appears to be a main accumulation site for the Mas-AT neuropeptide in some brain neurons produced in B. mori.

Genetical and Physiological Mechanisms of Adult Diapause in Insects

  • Adult diapause in insects is characterized by suppression of reproductive development. It is induced by environmental cues such as photoperiod, temperature, food availability, and other conditions Diapause-inducing environment is recognized and analyzed by the brain of the insects. The interpreted information is conveyed via endocrine system to target tissues such as ovaries, fat body, and other tissues. From this signal hierarchy of a brain-endocrine-target tissue axis, several factors are involved to express a diapause trait in a quantitative mode, even though the insects show a binomial phenotye between being in diapause or not. Recent works estimated that the number of the factors is relatively small by a series of crossing trials between high and low diapause lines. Heritability of the diapause is quite high (ca. 70%) in some species. Epistasis, sex-linkage, pleiotropism, and other nongenetic components also affect diapause inheritance. Most physiological studies have been focused on control mechanisms of the juvenile hormone (JH) synthesis in corpora allata (CA) because JH level in hemolymph of teneral adults is critical to decide a later developmental mode. Allatostatin, an antagonizer of JH synthesis, has been believed to be a potent brain message to CA for adult diapause induction.

  • Allatotropin is a 13-residue amidated neuropeptide isolated from pharate adult heads of the tobacco hornworm, Manduca serta and strongly stimulates biosynthesis of juvenile hormones in adults, but not larval, lepidopteran corpora allata. From a Bombyx mori midgut cDNA library, a cDNA that encodes a 130-amino-acid polypeptide containing M. sexta allatotropin sequence was isolated. Hypertrophical Changes of the Corpus Allatum Caused by Ovariectomy in Blattella germanica (난소제거 바퀴에서 알라타체의 이상비대화에 관한 연구)

  • The present study is undertaken to investigate the hypertrophical changes of the corpus allatum (corpora allata, CA) after the ovariectomy in Blattella germanica. In particular, the ultrastructural aspects of the normal and ovariectomized conditions, and induced factors of the hypertrophic phenomenon are focused. Ultrastructure of the CA from an immediately emergent adult is similar to that of the last larval stage that has stopped secreting juvenile hormone. The CA is composed of undifferentiated cells, exhibiting an electron-lucent matrix, a few mitochondria and less smooth endoplasmic reticulum. The karyoplasm occupy most of the cytoplasm. Electron-dense materials are filled with the intercellular spaces and gap junctions are also found. Almost no ultrastructural changes have been noticed during seven days until the oviposition. However, considerable changes in structure have been detected soon after the oviposition. Mitochondria are increased dramatically in number and cristae, and changed to the filamentous form with a high electron density. In addition, Golgi complexes, microtubules, and polysomes are also increased. After an oviposition, the total volume of the CA are decreased again. The volume of the CA are increased continuously, hypertrophy, after the ovariectomy. Morphological aspects of the CA in an early stage after the removal are similar to the structure of the secondary egg maturation. Large and electron dense globules are observed in the ovariectomized CA cytoplasms and they are present in those cells for a long period of time. Yet such a hypertrophical phenomenon occur only in the specific cells. The hypertrophy are caused by hollowing the part of the CA cells and later filling such site with polysomes. In 42 days after the ovariectomy, the nuclear membranes disappear in the CA cells, thus, exhibiting the prokaryotic-like features. Some results of the current study will contribute to the establishment of the model that explain unusual changes accompanied by certain treatment in insects and/or further in animals.

  • Vitellogenesis is the process by which yolk accumulates in developing oocytes. The initiation of vitellogenesis represents an important control point in oogenesis. When females of the model insect Drosophila melanogaster molt to become adults, their ovaries lack mature vitellogenic oocytes, only producing them after reproductive maturation. After maturation, vitellogenesis stops until a mating signal re-activates it. Juvenile hormone (JH) from the endocrine organ known as the corpora allata (CA) is the major insect gonadotropin that stimulates vitellogenesis, and the seminal protein sex peptide (SP) has long been implicated as a mating signal that stimulates JH biosynthesis. In this review, we discuss our new findings that explain how the nervous system gates JH biosynthesis and vitellogenesis associated with reproductive maturation and the SP-induced post-mating response. Mated females exhibit diurnal rhythmicity in oogenesis. A subset of brain circadian pacemaker neurons produce Allatostatin C (AstC) to generate a circadian oogenesis rhythm by indirectly regulating JH and vitellogenesis through the brain insulin-producing cells. We also discuss genetic evidence that supports this model and future research directions.

Synergism of Several Synergists of Antii-Juvenile Hormone Analog, 7-Ethoxy Precocene II to Milkweed Bug Oncopeltus fasciatus Dallas (7-Ethoxy precocene II 항유약호르몬유사물에 대한 몇가지 협력제의 Oncopeltus fasciatus Dallas에 있어서 협력작용)

  • The second instar milkweed bugs, Oncopeltus fasciatus were exposed to the residue of 6-methoxy-7-ethoxy-2, 2- dimethylchromene(or 7-ethoxy precocene 11, 7-EP-11) with one, two, or four different kinds of synergist(s) deposited as a residue on 9 cm diameter petri dishes. The anti juvenile hormone analog, 7-EP-I1 yielded a 50% effectiveness concentration at 1.18 wg/cmZ. The concentration of 7-EP-I1 and R020-9747 combined gave 0.084 ~*g/cm', as much 14 times more potent than with 7-EP-I1 alone. Most of the other synergists, including geraniol, 4-chalcone oxide, isosafrole, and piperonyl butoxide however, showed comparatively low level synergism with 1.0-4.3 times, depending upon the combinations of the synergist(s). The obtained results were considered that R020-9747, an analog of benzopyran with similar structure to that of 7-EP-I1 inhibited selectively the insect monooxygenase oxidative defense mechanism, in the body except for corpora allata because general antioxidants of piperonyl butoxide, isosafrole, and 4-phenyl chalcone oxide showed relatively mild synergism.

Structural Correlates of Hormone Production by the Corpora Allata in the Pine Moth, Dendrolimus spectablis Butler, during Larval-Pupal-Adult Transformations (松蟲變態에 따른 알라타體의 호르몬 生産과 그 構造的變化의 相關)

  • Ultrastructural changes in the cells of the corpora allata of the pine moth, Dendrolimus spectabilis Butler, were studied by electron microscope to know the structural correlates of hormone production by the gland during the larval-pupal-adult transformations. Mitochondria are in active phases from the overwintered to the last instar larvae and from the pupae just after pupation to the 20-day old pupae, while they are in inactive phases from the making cocoon stage to the prepupae just before pupation. The peripheral allatum cells have electron dense granules in the intracellular vacuoles of smooth-surfaced endoplasmic reticulum in the larval life, particularly in the overwintered larvae and in the early adults but the swollen smooth-surfaced intracytoplasmic vacuoles made by expansion of an end of the tubular rough endoplasmic reticulum, some of which contain fibrous proteins, are observed in addition to the vacuoles in the intercellular spaces in which the vacuoles grow by fusing each other from the mature larvae to the prepupae, both of them disappearing during just before pupation. After pupation the cytolasmic vacuoles develop again in the allatum cells so that they seem to begin the secretory activity. The fact that the neurosecretory granules stored within the axons terminated in the corpus allatum are visible only from the 20-day old pupa about two days before abult emergence to the 5-day old adult means that the secretion from the allatum cells is under the control of the brain from the late pupal stage, while the secretion during from the larval to the early pupal life has no relation with the brain, because such granules are not observed within the axons. It is, therefore, suggested that at least two kinds of hormone are released with the ages as far as concerned with the production and secretion mechanisms of the allatum hormone: juvenile hormone is released until the last instar larvae without any direct stimlation of the brain and gonadotropic hormone is secreted from the late pupa to the adult by getting brain's stimulation and that the secretory phases observed from the mature larvae to prepupae are presumably concerned with the biosynthesis of protein owing to the ecdysone and those from the early pupal stage in uncontrolled condition of the brain with the prothoracotropic activity.
