Continuous Production of Sorbitol with Zymomonas mobilis in a Packed Bed Reactor (Zymomonas mobilis에 의한 Packed Bed Reactor를 이용한 연속적인 sorbitol의 형성)

  • The purpose of this study is to develop a continuous process for sorbitol production using Zymomonas mobilis immobilized in K-carra-geenan. The glutaraldehyde cross-linking of toluene-treated cells immobilized in alginate or chitin showed high enzyme stability for long period. However, loss of enzyme activity was observed at 23% during 210h. In order to investigate the stability of glucose-fructose oxidoreductase of cethyltrimethylammoniumbromide (CT AB) treated cells, the long term continuous process was carried out with Z. mobilis immobilized in K-carrageenan in the continuous stirred tank reactor(CSTR) and the packed bed reactor. The continuous production of sorbitol with the immobilized CT AB permeabilized cells in packed bed reactor was more stable than in CSTR. Two stage continuous process with CT AB treated cells of Z. mobilis immobilized in K-carrageenan was carried out at various dilution rates. At the first stage, the productivity was increased up to 15 g/ $\ell$ -h as dilution rate increased and decreased over 0.32$h^{-1}$ of dilution rate. Similarly, maximum productivity obtained at the second stage was 22g/$\ell$ -h at 0.32$h^{-1}$

Effects of arsenite and variation of microbial community on continuous bio-hydrogen production from molasses using a sequence batch reactor (SBR)

  • This study investigated the effects of various arsenite concentrations on bio-hydrogen production from molasses using a sequence batch reactor (SBR) operated in a series of three batch cycles. In the first batch cycle, hydrogen production was stimulated at arsenite concentrations lower than 2.0 mg/L, while inhibition occurred at arsenite concentration higher than 2.0 mg/L compared to the control. Hydrogen production decreased substantially during the second batch cycle, while no hydrogen was produced during the third batch cycle at all tested concentrations. The toxic density increased with respect to the increase in arsenite concentrations (6.0 > 1.6 > 1.0 > 0.5 mg/L) and operation cycles (third cycle > second cycle > first cycle). The presence of microorganisms such as Clostridium sp. MSTE9, Uncultured Dysgonomonas sp. clone MEC-4, Pseudomonas parafulva FS04, and Uncultured bacterium clone 584CL3e9 resulted in active stimulation of hydrogen production, however, it was unlikely that Enterobacter sp. sed221 was not related to hydrogen production. The tolerance of arsenite in hydrogen producing microorganisms decreased with the increase in induction time, which resulted in severing the inhibition of continuous hydrogen production.

  • Bacillus subtilis SH-1 screened from coastal sea water of South Korea was used to produce bacteriolytic enzyme. The production of bacteriolytic enzyme by immobilized cells was investigated. The optimum conditions for the continuous production of the bacteriolytic enzyme using immobilized cells were 2.4 mm diameter of 0.3% alginate beads, 20 ml/h of substrate feeding rate and 20 l/min of aeration rate. A productivity of 76.5 to 88.0 units/ml could be obtained for 25 days by continuous column reactor under the optimum conditions.

Cumulative Weighted Score Control Schemes for Controlling the Mean of a Continuous Production Process

  • Cumulative sum schemes based on a weighted score are considered for controlling the mean of a continuous production process; in which both the one-sided and two-sided schemes are proposed. The average run lengths and the run length distributions for the proposed schemes are obtained by the Markov chain approach. Comparisons by the average run length show that the proposed schemes perform nearly as well as the standard cumulative sum schemes in detecting changes in the process mean. Comparisons of the one-sided schemes by the run length distribution are also presented.

  • The papaer reviews four theoretical acceptance sampling plans as OC(Operating Characteristics) curve based inspection, rectifying inspection, switching inspection, and continuous inspection. In addition, the study presents practical limitations of theoretical acceptance sampling by attribute and by variable. Finally, following research also recommends the sampling inspection based on production technology. However, the inspection method requires quality expertise with various experience and implicit knowledge of the field.

  • The formation of methanol directly from methane and steam was observed over Cu ion-exchanged mordenite. Furthermore, the continuous production of methanol was achieved by co-feeding methane and steam over Cumordenite. The methanol production rate was comparable to that reported in the stepwise process in which activation, methane reaction, and extraction of methanol were carried out separately.

Mass Culture of Ultra-small Rotifer, Synchaeta kitina at the Exchange Rate of Culture Water and Initial Inoculation Density (환수율 및 접종밀도에 따른 초소형 rotifer, Synchaeta kitina의 대량배양)

  • The productivity of ultra-small rotifer, Synchaeta kitina was investigated at the exchange rate of culture water (10, 20, 30, 40 and 50%) and initial inoculation densities (250, 600 and 900 inds. per mL) in semi-continuous culture. Also, the possibility of mass culture was investigated in a 100 L culture tank. Tetraselmis suecica was used as the feed for S. kitina in all experiments. The production of S. kitina increased with an increase in exchange rate of culture water. The highest production ($82.0{\times}10^5$ inds.) was achieved at 40% exchange rate of culture water. The production of S. kitina increased with an increase of initial inoculation density during the first week and the highest total production ($17.4{\times}10^6$ inds.) was achieved at 900 inds. per mL of initial inoculation density. However, on the second week, all treatments were not significantly different in total production (P>0.05). During the two weeks, total production of S. kitina at 900 inds. per mL of initial inoculation density was higher than at 600 inds. of initial inoculation density, but there was no significant difference (P>0.05). In the 100 L culture tank, density of S. kitina was kept from 516 to 890 inds. per mL and S. kitina was daily harvested $15.5{\times}10^6$ to $26.7{\times}10^6$ during the experimental period. The production cost for 100 million rotifers in semi-continuous culture was 63,656 won. The results from this study indicate that the optimal exchange rate of culture water and initial inoculation density for the semi-continuous culture of ultra-small rotifer, S. kitina are 40% and 600 inds. per mL, respectively.

고정화균체 반응기에서 L-Sorbose 연속생산

  • The conversion of D-sorbitol to L-sorbose by Gluconobater suboxydans was analyzed, and continuous production of L-sorbose was carried out in immobilized cell reactors. L-Sorbose production by high densities of resting cells was more effective than by conventional batch fermentations. Sorbitol dehydrogenase, an enzyme converting D-sorbitol to L-sorbose, did not suffer from substrate inhibition, but from product inhibition. When L-sorbose production was carried out with Ca-alginate-immobilized cells, about 60 g/l of L-sorbose was obtained. On the other hand, when the corn steep liquor (CSL) concentration of medium was reduced to 0.08%, 80 g/l of L-sorbose was obtained. Outgrowth inside the immobilized carriers was thought to block the pores of the carriers so that substrate could not easily diffuse through the carriers. Continuous production of L-sorbose was well accomplished in a bubble column reactor, and 6. 5 g/l.h of productivity and 81.2% of yield were obtained at a substrate feeding rate of 0.08h$^{-1}$ under the optimum conditions with carrier volume of 55% and aeration rate of 3 vvm.

  • In batch culture of A. pullulans HP-2001, maximal production of pullulan was 57.90 g/l at 96 hr. The maximal production of pullulan with 76.55 g/l occurred when substituted solution was 10% (w/v) sucrose, 0.25% (w/v) yeast extract and mineral salts. The maximal production of pullulan was 75.00 g/l with a dilution rate of $0.015\;h^{-1}$. Productivity of pullulan in the continuous culture fed with the fresh medium was higher than that fed with the fresh medium without nitrogen source.

  • A marine bacterium, Pseudomonas aeruginosa BYK-2 KCTC 18012P was immobilized in modified polyvinyl alcohol for the continuous production of rhamnolipids. The stability of rhamnolipids production, the mechanical strength of beads and the scanning electron microscope of immobilized cell were determined in a repeated batch culture. The rhamnolipids production was maintained $80{\sim}90%$ stability of initial production, and the mechanical strength also was stable during the repeated batch culture more than 14 cycles. In the case of SEM studies, the internal distribution pattern of the cell entrapped in modified PVA beads was observed. On the basis of optimal conditions, the continuous culture was investigated in 1.8L air lift bioreactor. The result suggested 0.1g/h rhamnolipids was obtained from 1%(v/v) fish oil continuously in conditions of 1.2L working volume, 0.5vvm and 20ml/h flow rate.
