호알카리성 Alcaligenes sp.의 배양에 의한 ${\gamma}$-PGA의 생산 (${\gamma}$-PGA Production by Cultures of Alkalophilic Alcaligenes sp.)

  • 호알카리성 Alcaligenes속의 한 균종응 이용하여 peptide계 생분해성 고분자인 ${\gamma}$-PGA( p이yglutarnic a acid}의 회분 맺 연속배양어l 의한 대량 생산성을 조사하였다. 화분배양시 ${\gamma}$-PGA의 생생량 벚 생산생은 포도당 농도 50g/P 까지 포도당의 농도증가에 따라 증가하였으나 최대 생산수율은 10gjP 의 포도당 농도 에서 얻어졌으며, 이때의 수율은 63% 이였다. 또, 비 증식속도는 포도당 농도 50g/l 에셔 약 $0.25hr^{-1}$로 최대값을 나타냈으나, 이 이상의 농도에서는 고농도 기섣에 의한 균체생육의 저해륜 나타내였다. 온도, pH 및 교반속도(rpm)를 독랩변수로 한 반응표면 분 석에 의한 ${\gamma}$-PGA 생산의 최적 희분배양 조건은 배 양온도 $31^{\circ}C$, pH 10 및 회전속도 87rpm의 정상점에 서 얻어졌으며, 이 조건에서 ${\gamma}$-PGA의 생산량은 약 11g/l 로 추정되었다. 연속배양의 경우 wash out은 희석률 0.5hr-1에서 일어났으며, 희석률 $0.40hr^{-1}$일 때 최대 생산성을 나타내였고, 최대 생산생은 2.80g/l/h로 회분배양보다 약 9배 높았다.

Effects of Food Waste Mixed Organic Fertilizer Treatment on Growth and Yield of Capsicum annuum

  • The global population is increasing every year, and the amount of food waste is also increasing. Direct landfilling of food waste has been prohibited since 2005, and in accordance with the London Convention in 2013, the discharge of livestock manure, sewage sludge, and food waste into the sea is prohibited. In the case of incineration to treat the discharged food waste, the heat point is lowered due to the moisture in the food waste itself, so fuel must be added. Therefore, this study was conducted to get basic data for setting the limit of application by investigating the growth and yield of crops after treating food waste dry powder mixed fertilizer (MF) on red pepper. In the experiment, continuous cultivation was carried out for two years in 2021 (1st year) and 2022 (2nd year). The treatment groups were set as Not Treatment (NT), Chemical Fertilizer (CF), Mixed Fertilizer (MF), Mixed Fertilizer×2 (MF×2). After harvest, crop growth and yield were investigated. As a result of the 1st years of growth survey, CF, MF, MF×2 show significant difference in shoot length compared to NT. About fresh weight and dry weight, CF show significant difference compared to NT. The 2nd years of growth survey, the shoot and root length, fresh weight did not show significant difference with NT. In case of dry weight, MF is significant increased compared to NT. As a result of the yield survey of the 1st year, all treatment groups did not show a significance in yield compared to the NT. In case of 2nd year, all treatment groups show significantly increased value compared to NT. The yield of MF was highest among the treatment groups. In the future, it is thought that it is necessary to quantitatively evaluate the effect of food waste dry powder mixed fertilizer through additional experiments and continuous cultivation, and to establish an appropriate amount of use and establishment of a manual based on this.

미사양토에서 벼-콩 윤작재배가 토양화학성과 작물생육에 미치는 영향 (Influence of Rice-soybean Rotation on Soil Chemical Properties and Crop Growth in Silt Loam Soil)

  • 본 연구는 미사양토인 호남평야지 논토양에서 벼와 콩의 윤작재배에 의한 토양화학성 변화와 작물생육에 미치는 영향을 구명하였다. 벼 연작, 콩-벼 1년 윤작, 콩 2년-벼 1년 윤작, 콩 3년-벼 1년 윤작의 처리를 두었다. 논 토양에서 벼를 연작 후 토양 중 pH, 토양 유기물, $Ca^{2+}$$K^+$함량은 별 변화가 없었으나 유효 인산함량은 점차 감소하였다. 한편 벼와 콩의 윤작으로 토양 중 유기물 함량은 감소하고, 유효인산과 $Ca^{2+}$$K^+$함량은 증가하였으며, 벼 생육기간 중 토양 중 암모니아태 질소함량도 높아졌다. 이러한 토양 중 양분함량의 변화로 인해 벼의 $m^2$당 수수와 수당립수가 많아져서, 쌀 수량은 1~4%증가하였다. 콩은 연작 연수가 길어질수록 수량이 낮아지는 경향이어서, 콩 논 재배 시에는 벼와 매년 윤작하는 것이 수량 저하를 방지하였다.

시설수박 연작지 토양특성에 대한 후작물 재배의 영향 (Post Harvest Cropping Impacts on Soil Properties in Continuous Watermelon (Citrullus lanatus Thunb.) Cultivation Plots)

  • 수박을 7년간 연작한 시설재배지 토양을 개선하기 위해 수박후작물로 옥수수, 배추, 무, 총각무, 상추, 시금치, 및 양파를 재배한 후 토양특성과 수박의 생산성 및 품질의 변화를 조사하였다. 토양 pH는 무를 제외한 모든 후작물 재배지 토양에서 증가하였고, 토양 중 유기물 함량은 후작물 재배구에서 감소하는 경향을 보였으며 특히 상추 재배구에서 현저하게 감소하였고, 유효인산 함량은 시금치, 양파 재배구에서 증가하는 경향을 보였다. 치환성 Ca 및 Mg은 대부분의 후작물 재배구에서 증가하는 경향을 보였으나, 치환성 K은 후작물 재배 후 50% 이상 현저히 감소하였다. 또한 수용성 음이온($Cl^-$, $NO_3{^-}$, $SO{_4}^{2-}$)과 EC는 모든 후작물 재배구에서 감소하였다. 특히 EC의 감소는K와 Ca+Mg의 비율의 감소에 의한 영향으로 보여진다. 모든 후작물 재배구에서 Bacteria/Fungi 비율이 재배전보다 증가하였으며, 수박의 고사주율은 모든 후작물 재배구에서 대조구보다 낮게 나타났으나, 수박의 생산성 및 품질에 있어 후작물 재배의 영향은 뚜렷하게 나타나지 않았다. 그러므로 시설수박 연작지에서 단기윤작의 적용은 토양의 문제점을 개선하는데 있어 효과적인 방법이라고 생각된다.

하중 제어법을 이용한 파이프 줄기초의 현장 인발저항 특성 (Characteristics of Field Uplift Tests of Continuous Greenhouse using the Load Control Method)

  • Institutional inertia anti-disaster standard was presented mainly on the upper surface, it is necessary to improve to the soil type standard and uplift the resistance standard greenhouse that are vulnerable to strong winds. In this study, we carried out a field test using the load control method in order to evaluate the uplift resistance of continuous foundation of greenhouse with different depths of the rafters. Institutional inertia anti-disaster standard of greenhouse foundation did not protect the greenhouse structure from the damages caused by strong winds and heavy snow. Therefore, field tests for behavior characteristics of continuous greenhouse foundation were carried out to ensure stable facility cultivation. The field test condition was evaluated using different embedded depth as follows: 30cm, 40cm, 50cm and spacing 50cm, 60cm, 70 cm. As a result of the uplift resistance field tests using the load control method, the minimum uplift resistance was found to be over 90kg and uplift resistance displacement was 9.4mm. Uplift resistance of the continuous greenhouse foundation was in the range of 90-180 kg according to embedded depth and spacing. Using the test condition, there was no constant trend in the uplift resistance.

Aspergillus sojae B-10에 의한 반응성 염료의 탈색조건 (Continuous Biodegradation of Reactive Dyes by Aspergillus sojae B-10)

  • Dyes are released into the environment from industrial wasterwater. They are considered to be a pollution problem because of the wide spread into environment with a variety of colors. In order to biological treatment of industrial dyes waste water, biodegradation of reactive dyes such as Rifacion Red H-3EB, Rifazol Blue BT, Rifacion Yellow P-4G and Rifacion Brown RT were carried out decolorized by Aspergillus sojae B-10. Aspergillus sojae B-10 showed the almost completely biodegradation ability when it was cultivated in a 2.0% glucose, 0.06% sodium nitrite, 0.1% $KH_2PO_4$, 0.5% $MgSO_4\cdot 7H_2O$ containing each reactive dyes (500 ppm) under the optimal conditions of 32$\circ$C and pH 5. The mycelium of Aspergillus sojae B-10 was produced extracellular enzyme which has concerned responsible for dyes biodegradation. Under optimal conditions, reactive dyes started being decolorized within 24 hr and its was almost decolorized c ompletely after 5 days incubation. Rifazol blue RT was not completely decolorized until 5 days of cultivation. Rafacion Red BT, Rifasol blue BT and Rifacion yellow P-4G were completely decolorized after 5 days cultivation.

호남지역의 시설원예재배지 지하수중 화학성분 변동요인 조사 (Changes of Chemical Contents in Groundwater at Controlled Horticulture in Honam Area)

  • This study was carried out to investigate factors influencing on the groundwater quality at controlled horticulture in Honam area in 1995. The deeper groundwater sampling, the lower concentrations of $NH_4-N$ and $NO_3-N$ were observed. There was no difference in concentrations of $SO_4\;^{2-}$ and $Na^+$ in the groundwater below 15m. Contents of $NH_4-N$, $NO_3-N$ and $PO_4\;^{3-}$ in groundwater were the highest at rice transplanting season(the late May) and that of $SO_4\;^{2-}$, $Na^+$ and $Cl^-$ were the highest at dry season(the mid-February). Continuous cultivation of horticultural crops showed higher concentration of $NH_4-N$, $NO_3-N$, $PO_4\;^{3-}$, $Na^+$ and $Cl^-$ in groundwater than rotational culture with rice. The longer cultivation years with horticultural crops, the higher concentrations of $NO_3-N$, $PO_4\;^{3-}$, $SO_4\;^{2-}$ and $Cl^-$ were shown, and constitutional ratio of $NO_3-N$ among the anions increased gradually. Nitrate-N level, exceeded 20mg/l, the critical level for agricultural usage, frequently at Yongjinmyeon Wanju and Janglockdong Kwangju, and $PO_4\;^{3-}$ levels were higher at Seogtandong Iksan than the other places.

경사지에 대한 가축분뇨 시용시 옥수수의 생산성과 양분유실에 관한 연구 (Study on Productivity of Corn and Nutrient Runoff of the Soil by Application of Swine Manure in the Slopes)

  • This study was conducted to investigate the effects of application levels of swine manure fermented with sawdust(SMFS) and degree of the slope on productivity of corn, and nitrogen and organic matter runoff in corn cultivation soil. Main plots were consisted of the degree of slope, such as 0, 5 and 10, and the subplots consisted of application levels of SMFS, such as control, 200kgNha and 400kgNha. Lysimeter was constructed with 0.33m width, 3m length and 0.4m height. Dry matter(DM) yields of corn decreased as the slope increased but there was no significant differences between the degree of the slope. DM yields increased significantly as application levels of SMFS increased (pc0.05). DM yields decreased according to continuous cultivation of corn. Total nitrogen(TN) contents of the whole corn decreased as the slope increased(p<0.05). TN content in SMFS treatments was higher than that of non-SMFS treatment but there was no significantly different between SMFS treatments. TN yields decreased significantly as the slope and the application level of SMFS increased(p<0.05). Nitrogen and organic matter yields in the runoff waters increased as the slope and application levels of SMFS increased. The runoff of nitrogen and organic matter was more affected by the increase of the slope than application levels of SMFS. (Key words : Corn, Runoff, Swine manure, Degree of slope, Lysimeter)

Complement-mediated tail degradation of Neodiplostomum seoulense cercariae

  • The furcocercus cercariae of Neodiplostomum seoulense (Digenea: Neodiplostomidae) penetrate the skins of tadpoles and shed their tails. The speculated mechanism of this tail loss was physical efforts required to produce a vigorous zigzag motion during skin penetration; no other mechanism has been proposed. We examined the relationship between the host serum and cercarial tail loss. Cercariae of N. seoulense were collected from experimentally infected Segmentina hemisphaerula, and lots of 300 cercariae were cultured in medium 199 contained several types of sera. Cercarial tail degradation was induced in all media, but all the cercariae cultured except those cultured in media containing fetal bovine serum (FBS) died within 48 hr. After 72 hr cultivation in media containing FBS, cercarial tail degradation was induced in 67.0%; in continuous cultivation 13.3% of larvae survived for 7 days. Tail degradation did not occur in the absence of serum and when serum was heat inactivated at $56^{\circ}C$ for 30 min. The addition of 20 mM ethylenediaminetetraacetic acid (EDTA) blocked cercarial tail degradation completely. Moreover, the addition of 20 mM $MgCl_2$ restored tail degradation blocked by EDTA. These results suggest that the alternative complement pathway is related with the N. seoulense cercarial tail degradation induced by serum.

양파농가의 공동체의식이 참여만족에 미치는 영향 분석 (The Effect of Sense of Community on Participation Satisfaction in Onion Farms)

  • This study analyzed the effect of the farm's sense of community on participation satisfaction by participating in the contract cultivation of onions. To this end, we investigated the participation satisfaction with the sense of community's emotional connection, fulfillment of need, membership, and influence. According to the analysis of the sense of community of onion farmers, the definition of need, membership, and influenced participation satisfaction and were statistically significant. It was also shown to affect sense of community in the order of membership, Influence, fulfillment of need, and emotional connection. Therefore, it is necessary to form a continuous relationship with the leader and Nonghyup, to lead to improved farm income, induce pride as a member of participating farmers, and form a positive perception among onion farmers.
