3-Methylthiopropionic Acid of Rhizoctonia solani AG-3 and Its Role in the Pathogenicity of the Fungus

  • Studies were conducted to determine the role of 3-methylthioproprionic acid (MTPA) in the pathogenicity of potato stem canker, Rhizoctonia solani, and the concentrations required to inhibit growth of R. solani under laboratory and plant house-based conditions. The experiments were laid out in a completely randomized design with five treatments and five replications. The treatments were 0, 1, 2, 4, and 8 mM concentrations of MTPA. The purified toxin exhibited maximal activity at pH 2.5 and $30^{\circ}C$. MTPA at 1, 2, 4, and 8 mM levels reduced plant height, chlorophyll content, haulm fresh weight, number of stolons, canopy development, and tuber weight of potato plants, as compared to the control. MTPA significantly affected mycelial growth with 8 mM causing the highest infection. The potato seedlings treated with MTPA concentrations of 1.0-8.0 mM induced necrosis of up to 80% of root system area. Cankers were resulted from the injection of potato seedling stems with 8.0 mM MTPA. The results showed the disappearance of cell membrane, rough mitochondrial and cell walls, change of the shape of chloroplasts, and swollen endoplasmic reticulum. Seventy-six (76) hours after toxin treatment, cell contents were completely broken, cytoplasm dissolved, and more chromatin were seen in the nucleus. The results suggested that high levels of the toxin concentration caused cell membrane and cytoplasm fracture. The integrity of cellular structure was destroyed by the phytotoxin. The concentrations of the phytotoxin were significantly correlated with pathogenicity and caused damage to the cell membrane of potato stem base tissue.

  • The experimental study on disposal cover through the performance test facility offers reliability in the safety of near surface disposal of low and intermediate level radioactive waste. To ensure the long-term safety of the repository, the impermeability, integrity, resistance to degradation and ease of maintenance might be considered as the basic performance requirement of the disposal cover. considering the difficulties to meet these performance requirement by using single layer, the disposal cover design which is composed of top layer, middle drainage layer and bottom low permeability layer is schemed for the test facility. The water balance of the cover was evaluated by using HELP code. For the long-term monitoring of the soil moisture content and matric potential, TDR probes and tensiometers will be installed in 6 test cells. Each test cell is dimensioned 3$\times$3$\times$3.3m.

THE EFFECTS OF CREEP AND HYDRIDE ON SPENT FUEL INTEGRITY DURING INTERIM DRY STORAGE

  • Recently, many utilities have considered interim dry storage of spent nuclear fuel as an option for increasing spent fuel storage capacity. Foreign nuclear regulatory committees have provided some regulatory and licensing requirements for relatively low- and medium-burned spent fuel with respect to the prevention of spent fuel degradation during transportation and interim dry storage. In the present study, the effect of cladding creep and hydride distribution on spent fuel degradation is reviewed and performance tests with high-burned Zircaloy-4 and advanced Zr alloy spent fuel are proposed to investigate the effect of burnup and cladding materials on the current regulatory and licensing requirements. Creep tests were also performed to investigate the effect of temperature and tensile hoop stress on hydride reorientation and subsequently to examine the temperature and stress limits against cladding material failure. It is found that the spent fuel failure is mainly caused by cladding creep rupture combined with mechanical strength degradation and hydride reorientation. Hydride reorientation from the circumferential to radial direction may reduce the critical stress intensity that accelerates radial crack propagation. The results of cladding creep tests at $400^{\circ}C$ and 130MPa hoop stress performed in this study indicate that hydride reorientation may occur between 2.6% to 7.0% strain in tube diameter with a hydrogen content range of 40-120ppm. Therefore, it is concluded that hydride re-orientation behaviour is strongly correlated with the cladding creep-induced strain, which varies as functions of temperature and stress acting on the cladding.

Surface Characteristics of Fouling Resistant Low-Pressure RO Membranes (상업용 내오염성 저압 RO막의 표면 특성 분석)

  • In this study, five commercially available fouling resistant low-pressure RO membranes were investigated for the treatment of seasonally brackish surface water with high organic content (${\approx}24mg/L$). The membranes investigated are LFC-1 (Hydranautics), X20 (Trisep), BW30FR1 (FilmTec), SG (Osmonics), and BE-FR (Saehan). The results of surface characterization revealed that each of these membranes has one or two unique surface characteristics to minimize the adherence of the fouling materials to the membrane. Specifically, the LFC1 membrane features a neutral or low negative surface to minimize electrostatic interactions with charged foulants. The X20, on the other hand, shows a highly negatively charged surface, and thus, is expected to perform well with feed waters containing negatively charged organics and colloids. The BW30FR1 exhibits a relatively neutral and hydrophilic surface, which could be beneficial for lessening organic and/or biofouling. The SG membrane has a smooth surface that makes it quite resistant to fouling, particularly for colloidal deposition. Lastly, BE-FR membrane demonstrated a medium surface charge and a slightly higher hydrophobicity. In the pilot study, all of the four membranes experienced a gradual increase in MTC (water mass transfer coefficient or specific flux) over time, indicating no fouling occurred during the pilot study. The deterioration of permeate water quality such as TDS was also observed over time, suggesting that the integrity of the membranes was compromised by the monochloramine used for biofouling control.

  • Steel, as a reinforcing mechanism in concrete, provides the tensile strength that is lacking in concrete, alone, and the high alkaline environment (pH 12.5) in concrete offers satisfactory protection against most corrosion of the steel. However, the corrosion of reinforcing steel in concrete can occur by chloride attack or carbonation, and it can cause a loss of integrity a section and concrete failure through cracking and spalling. In this study, a transient electro magnetic method (TEM) of a nondestructive technique is adapted to study the measuring method of steel corrosion in mortar. The sensor was made of an enameled wire, with a diameter of 0.25mm and anacril. He sensor configuration used was a coincident loop type. The secondary electro motive force (2nd EMF) was measured with SIROTEMIII, which equipped the accelerator. The accelerator allowsthe transmitter to turn off approximately $10\sim15$ times faster than normal. The high-resolution time series, used for very shallow or a high resistivity investigation was selected. After steels were corroded by the salt spray, during 4, 8, 15 and 25 days, they were embedded in mortar. The content results acquired in this study are as follows. The variation of the secondary electro motive force (2nd EMF) was shown by the change of steel surface with different corrosion time steel. It was confirmed that measurement of steel corrosion in mortar by a transient electro-magnetic method (TEM) can be possible.

Semi-Fragile Image Watermarking for Authentication Using Wavelet Packet Transform Based on The Subband Energy (부대역 에너지 기반 웨이블릿 패킷 변환을 이용한 인증을 위한 세미 프레자일 영상 워터마킹)

  • A new method of Semi-fragile image watermarking which ensures the integrity of the contents of digital image is presented. Proposed watermarking scheme embeds watermark in the form of quantization noise on the wavelet transform coefficients in a specific mid frequency subbands selected from a wavelet packet decomposition based on energy distribution of wavelet transform coefficients. By controlling the strength of embedded watermark using HVS (Human Visual System) characteristic, it is imperceptible by a human viewer while robust against non-malicious attack such as compression for storage and/or transmission. When an attack is applied on the original image, it is highly probable that wavelet transform coefficients not only at the exact attack positions but also the neighboring ones are modified. Therefore, proposed authentication method utilizes whether both current coefficient and its neighbors are damaged. together. So it can efficiently detect and accurately localize attacks inflicted on the content of original image. Decision threshold for authentication can be user controlled for different application areas as needed.

A Study on the Integrity Assessment of Bare Concrete Bridge Deck based on the Attenuation of Radar Signals (레이더 신호의 감쇠특성을 고려한 일체식 콘크리트 교량 바닥판의 상태평가 방법 고찰)

  • The signal characteristic of radar wave on concrete decks is determined by the attenuation of the radar due to the conversion of EM(Electromagnetic) energy to thermal energy through electrical conduction, dielectric relaxation, scattering, and geometric spreading. In this study, it is found that the attenuation of radar signal received on top rebars in bare deck concrete with 2 way travel time shows a general decreasing linear trend because of its same relative permittivity and conductivity. The radar signal after depth-normalization, can then be interpreted as being principally influenced by the content of chlorides penetrating cover concrete, which caused corrosion of rebars in bridge decks.

Strength Development and Carbonation Characteristics of Slag Cement/Class C Fly Ash blended CO2 Injection Well Sealant

  • CO2 injection well sealant is vulnerable to supercritical CO2 (scCO2) exposure. To develop an alternative to the conventional sealant system (class G cement/class F fly ash), the performance of slag cement (SPC) systems containing class F fly ash (FFA) or class C fly ash (CFA) was evaluated and compared with the conventional sealant under scCO2 conditions. All sealant systems showed an immediate increase in compressive strength upon scCO2 exposure and, at 37.6 MPa, SPC/CFA showed the highest compressive strength after 14 days, which was much higher than the 29.8 MPa of the conventional sealant system. Substantial decreases in porosity were observed in all sealant systems, which were partly responsible for the increase in strength. Carbonation reactions led to pH decreases in the tested sealants from 12.5 to 10~11.6. In particular, the greatest decrease in pH in slag cement/class C fly ash probably supported relatively sustainable alkali activation reactions and the integrity of cement hydrates in this system. XRD revealed the presence of CaCO3 and a decrease in the content of cement hydrates in the tested sealants upon scCO2 exposure. TGA demonstrated a greater increase of CaCO3 and calcium-silicate-hydrate phases in SPC/CFA than in the conventional sealant upon scCO2 exposure.

An Experimental Study on the Effect of Hypothermic Oxygenated Cardioplegic Solution on Myocardial Protection during Prolonged Aortic Cross-clamping (대동맥 차단시 저온 산소화 심정지액이 심근보호에 미치는 영향)

  • This study was experimentally undertaken to evaluate the effect of hypothermic oxygenated cardioplegic solution on myocardial protection during prolonged aortic cross clamping under cardiopulmonary bypass. Dogs were divided into two groups control group [received hypothermic unoxygenated cardioplegic solution] and experimental group [received hypothermic oxygenated cardioplegic solution]. Coronary sinus effluent was obtained at once and 30, 60, 90 minutes after cross-clamping for the determination of pH, PCO2,PO2 and lactate level during the infusion of cardioplegic solution and myocardial biopsies were obtained after cessation of 90 minutes of aortic cross-clamping. The results obtained were as follows: 1. There was no significant differences in the pH and PCO2 between the oxygenated and unoxygenated cardioplegic solution but the PO2 of the oxygenated solution was 4 times greater than unoxygenated solution, and also the oxygenated solution had a significantly greater oxygen content [2.020.05 ml 02/min] and had much more oxygen delivery than unoxygenated solution. 2. The myocardial oxygen consumption and the myocardial oxygen extraction in oxygenated group were 1.63 ml 02/100 ml and 67.32% respectively, which was greater than those in unoxygenated group. 3. Regarding to pH and PCO2 of coronary sinus effluent, there was no significant differences between two groups in early period of infusion of cardioplegic solution, but the pH shifted to acidosis from 60 minutes, PCO2 increased from 90 minutes of aortic cross-clamping, and PO2 markedly decreased from 90 minutes of aortic cross-clamping in unoxygenated group. 4. The lactate concentration of coronary sinus effluent revealed relatively normal in both groups, but showed slight increase up to 27.54.56 mg/100 ml at 90 minutes of aortic cross-clamping in unoxygenated group. 5. On electron microscopic study, the ultrastructural integrity of myocardial cells in oxygenated group was well preserved within 90 minutes. Slight swelling and deformity of mitochondria, interfibrillar widening, and disarrangement of myofibrils were observed at 90 minutes after aortic cross-clamping in unoxygenated group. From these results, the use of hypothermic oxygenated cardioplegic solution seemed to be effective and better method for the preservation of ischemic myocardium during the prolonged aortic cross-clamping.

  • Purpose: The aim of this study is to identify the elements of success in curriculum reformation using focus group interview and faculty survey. Methods: We analyzed the 105 questionnaires that were collected horn 450 faculties in Yonsei University College of Medicine from September 1 to September 12, 2005. The interview of focus group was conducted to 105 faculty and students using 10 interview questions from July 25 to August 29, 2005. The data of focus group interview and faculty survey were analyzed by content analysis. frequency and percentile. Results : 66.7% of faculty have pointed out that it is right a direction of new curriculum but there are some problems in curriculum management. The most frequently addressed problem in the new curriculum is as follows : ability of self-directed learning(15.3%). deficiency of faculty understanding(10.5%). The faculty comments on the improvement areas of new curriculum such as curriculum revision (8.6%). efficiency of curriculum management (6.7%), reward and incentive system of education(6.7%). The focus groups pointed out the important factors of curriculum reformation such as horizontal and vertical integrity between subjects, improvement of student achievement test, enhancement of self-directed learning. responsibility and authorities of course director. Conclusions : It should be concluded. to succeed curriculum reformation, which the purpose of curriculum reformation announces obviously and management group should effort continuously so that professors may understand the new curriculum. The course directors have to responsibility and authorities operating the subjects and the university should develop the students' evaluation system and faculty performance appraisal system.
