  • 동물실험을 통하여 새로이 밝혀진 항암제(anti-initiator/anti-ptomotor)인 CLA는 grilled ground beef에서 처음 분리되었다. CLA는 grilled ground beef 외에도, cheese 및 이들 관련식품에 많이 존재한다. CLA는 반추동물의 위에 서식하는 혐기성 bacteria에 의해 linoleic acid로부터 생성되며, 식품 가공 중에서도 생성된다. 이것은 또한 in vivo에서 linoleic acid의 carbon centered free radical 형태의 산화에 의해 생성되기도 한다. CLA는 아주 강력한 항산화제임이 밝혀져, 지금까지 알려지 있지 않았는 free radical에 대응하여 membrane을 보호하는 in situ defense mechanism 역할을 한다. 이는 또한 cytochrome P450 isozyme의 활성을 저해하는 반면, ODC 효소 활성 역시 저해한다. 그래서, 적어도 CLA의 이 세가지 biological activity가 CLA 항암기작에 관여하는 것으로 생각된다.

양조간장에서 분리한 갈색물질의 항산화성 (Antioxidative Activity of Browning Products Fractionated from Fermented Soybean Sauce)

  • 동결건조한 양조간장의 분말로 부터 갈색물질을 Sephadex G-10으로 겔-여과 크로마토그라피법에 의하여 분획한 다음 이를 동결건조하여 분말로 만든 후, 동갈색물질의 항산화력을 다른 항산화제인 부틸히드록시아니졸(BHA) 및 ${\alpha}-토코패롤과$ 비교하여 보았다. 갈색물질 및 양조간장은 다 같이 지방산의 산화반응에 있어서 과산화물의 생성을 크게 저해시켰고, 갈색물질이 양조간장 자체보다 항산화 효과가 더 높았으며 농도에 비례하여 항산화성이 증대되었다. 그러나 갈색물질의 항산화력은 과산화물 생성억제 능력 그리고 conjugated dienoic acid 생성저해 능력 등에서 BHA 및 ${\alpha}-토코페롤$ 보다는 낮았다. 이와같은 양조간장의 갈색물질의 항산화성은 그 획분이 대량 섭취도고 있는 발효조미식품이라는 점에서 중요한 의의를 가질 것으로 판단된다.

Effects of Polyunsaturated Fatty Acids on Intestinal Cell Proliferation

  • The effect of the polyunsaterated fatty acids, linoleic acid(LA), arachidonic acid(AA) and conjugated dienoic linoleic acid(CLA) on IEC-6 cells (rat intestinal cell)proliferation and cell transduction have been determined in vitro. IEC-6 cells proliferation was assessed by cell growth and [3H]-thymidine incroporation analysis. At 10 μM concentration , the proliferationof cells supplemented with AA or LA was significantly higher than that of CLA. [3H]-thymidine uptake showed the same results. LA and AA increased [3H]-thymidine uptake more than CLA. The stimulatory effect of LA or AA was even more pronounced in the presence of IGF. Both cell number analysis and [3H]-thymidine incorporation revealed that IEC-6 cell proliferation was influenced differently by exogenous free fatty acids, in which AA or LA stimulated IEC-6 cell proliferation and CLA inhibited it. Tyorosine phosphorylation provides a key switch to regulate celluar acitivity in response to extracellular stimuli. At 20 μM and 10μM, AA with IGF-1 stimulated protein tyrosine phophorylation in IEC-6 cells, but LA's impact was less than that of AA. CLA and CLA with IGF-1 inhibited protein tyrosine phosphorylation in IEC-6 cells. These results suggest there is a possible correlation between cell proliferation and IGF receptor tyrosine knase activity driven by AA.

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a series of positional and gemotric isomers of linoleic acid which are found naturally in food, mainly dietary products and breef. We studied the effects of CLA on the growth of tumor cells and the production of interleukin-1(IL-1) and interleukin-2(IL-2). CLA treatment markedly inhibited the growth of Yac-1 cells and sarcoma-180 cells by 99 and 82% to that of control, respectively, after four days of incubation at 37$^{\circ}C$. To elucidate the immunological mechanism of antitumor activity of CLA, spleen cells of Balb/c mouse were exposed to 31, 63, 125, 250 $\mu\textrm{g}$ of CLA per ml for 24 hrs at 37$^{\circ}C$. The culture supernatants of CLA-exposed spleen cells reduced the production of IL-1 and IL-2 in all of the test conditions. These results indicate that the anticarcino-genic effect of CLA was mediated by the other actions rather than the production of the Il-1 or IL-2. We suggest that CLA might have an antiinflammatory effect in part due to its inhibitory action on the production of IL-1.

Conjugated Linoleic Acid Changes fatty Acid Composition by Decreasing Monounsaturated fatty Acids in Rabbits and Hep G2 Cells

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a mixture of positional and geometric isomers of linoleic acid(LA). We previously found that CLA changes the fatty acid profile in chicken eggs and serum by decreasing monounsaturated fatty acids. Studies were conducted to explore the effects of CLA on fatty acid composition. Rabbits were fed a semisynthetic diet with or without CLA(0.5g CLA/rabbit/day) for 22 weeks. Compared to the control, rabbits fed CLA had significantly lower monounsaturated fatty acid levels(palmitoleic acid Cl6 : 1 by 50% and oleic acid Cl8 : 1, by 20%) in plasma lipids. We found similar differences in fatty acid composition in the liver and the aorta. The inhibitory effect of CLA on $\Delta$9 desaturation was confirmed in a human hepatoma cell line, Hep G2. CLA significantly decreased $\Delta$9 desaturation in 4-5 hours as shown by an increase in the ratio of Cl6 : 0 to C 16 1, This is apparently due to a decrease in $\Delta$9 desaturase(stearoyl-CoA desaturase, SCD) activity ; it was decreased more than 50%. These results, along with our previous findings, indicate that CLA is an inhibitor of $\Delta$9 desaturase in the liver.

Effects of Conjugated Linoleic Acid on Adipocyte Secreted Proteins in vitro

  • Conjugated linoleic acid (CLA) is a class of positional, geometric conjugated dienoic isomers of linoleic acid (LA). CLA activates the immune system, protects against tumorigenesis, and reduces the incidence of atherosclerosis. Trans-10, cis-12 CLA has specific effects on lipid metabolism, it has been shown to reduce body fat gain and regulates some adipocyte secreted proteins in vivo and in vitro. Here we report that a CLA mixture affects cytokine secretion from rat primary adipocytes. Rat primary adipocytes were treated with 1 mM, 100 $\mu$M, 1 $\mu$M or 100 nM CLA mixture doses; and leptin, tumor necrosis factor alpha (TNF a ), interleukin-6 (IL-6) and glycerol levels in the medium were measured. Leptin secretion was lower, TNF $\alpha$ secretion higher and IL-6 secretion did not change in response to the CLA mixture. Leptin and TNF $\alpha$ secretions did not change with CLA mixture treatment in a dose-dependent manner. In addition, the CLA mixture did not appear to enhance lipolysis in rat primary adipocytes. In conclusion, our study demonstrates that the decrease in leptin and increase in TNF $\alpha$ secretion in adipocytes treated with CLA mixture may be due to the apoptotic effect and to a reduction in peroxisome proliferator-activated receptor gamma (PPAR ${\gamma}$ ) ligands.

Anti-diabetic Effect of Fermented Milk Containing Conjugated Linoleic Acid on Type II Diabetes Mellitus

  • Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of conjugated dienoic derivatives of linoleic acid. CLA has been reported to be able to reduce body fat. In this study, we investigated the antidiabetic effect of fermented milk (FM) containing CLA on type II diabetes db/db mice. Mice were treated with 0.2% low FM, 0.6% high FM, or Glimepiride (GLM) for 6 wk. Our results revealed that the body weight and the levels of fasting blood glucose, serum insulin, and leptin were significantly decreased in FM fed mice compared to db/db mice. Oral glucose tolerance and insulin tolerance were significantly ameliorated in FM fed mice compared to db/db mice. Consistent with these results, the concentrations of serum total cholesterol, triglycerides, and LDL cholesterol were also significantly decreased in FM fed mice compared to db/db mice. However, the concentration of HDL cholesterol was significantly higher in FM fed mice compared to db/db mice. These results were similar to those of GLM, a commercial anti-diabetic drug. Therefore, our results suggest that FM has anti-diabetic effect as a functional food to treat type II diabetes mellitus.

갓에서 Chlorophyll 유도체 및 $\beta$-Carotene의 분리와 이들의 항산화 작용 (Isolation of Chlorophyll Derivatives and $\beta$-Carotene from Mustard Leaf and Their Antioxidative Activities on the Lipid Autoxidation)

  • Chlorophyll a and b, pheophytin a and b and $\beta$-carotene, crude chlorophylls and carotenoids (CCC) extracts of mustard leaf kimchi were isolated by DEAE-sepharose CL-6B and Sepharose CL-6B colume and TLC. The effects of chlorophyll a and b, pheophytin a and b and $\beta$-carotene on linoleic acid autoxidation were examined by the determination of peroxide value and conjugated dienoic acid content. Among them, chlorophyll a showed greater antioxidative activity than others, followed by chlorophyll b, pheophytin a, pheophytin b and $\beta$-carotene. Degradation of pheophytin b was observed to be slower than others and $\beta$-carotene showed highest degradation level during the autoxidation reaction of linoleic acid.

인간 전립선 암세포인 TSU-Pr1에서 trans-10,cis-12 Conjugated Linoleic Acid에 의한 Apoptosis 유발과 Bcl-2 단백질의 발현억제 (Induction of Apoptotic Cell Death and Depression of Bcl-2 Protein Levels by Trans-10,cis-12 Conjugated Linoleic Acid in Human Prostate Cancer)

  • CLA 이성체 mixture와 trans-10,cis-12 CLA는 살아있는 TSU-Prl 세포 수를 감소시키는 반면 LA와 cis-9,trans-11 CLA는 세포 수에 아무런 효과가 없었다. 세포 수를 감소시키는 CLA 이성체 mixture와 tran-10, cis-12 CLA는 apoptotic cell death를 유도하는 것을 관찰하였고, 특히 trans-10,cis-12 CLA는 Bcl-2/Bax 비율을 감소시켰다. Trans-10,cis-12 CLA에 의해 Bax와 Bcl-2 mRNA의 수준이 변하지 않았으므로 이 지방산에 의한 Bcl-2의 변화는 mRNA 전사 후에 일어나는 단계에서 일어난다고 결론지을 수 있다. 이로써 CLA에 의한 세포증식 억제는 부분적으로 TSU-Prl 세포의 apoptosis를 유도함으로서 이루어지는 것이라 할 수 있다.

가열산화가 냉압착 들기름의 품질특성에 미치는 영향 (Influence of Thermal Treatment on Chemical Changes in Cold-Pressed Perilla Seed Oil)

  • 냉압착 들기름과 볶음 압착 들기름 및 볶음 가열 압착 들기름의 가열산화에 의한 색도 및 갈변도, 총 페놀 함량, 과산화물가, 산가, 이중공액산가, 전자공여능 및 지방산 함량 변화를 측정하여 각 들기름의 특성 및 가열산화에 의한 품질을 비교하였다. 냉압착 들기름은 Hunter L 값은 76.62로서 볶음 압착 들기름의 39.77에 비하여 2배 이상 높게 나타났으며, 갈색도의 경우에도 각각 0.027과 0.244을 나타내어 냉압착유의 갈색화 정도가 낮은 것으로 나타났다. 산가와 이중공액산가는 냉압착 들기름이 가장 낮은 값을 나타내었고, 총 페놀 함량의 경우 볶음 압착 들기름이 7.27 mg/100 g oil로 냉압착 들기름에 비하여 1.60배 이상인 것으로 나타났으며, 전자공여능도 볶음 압착 들기름이 냉압착 들기름에 비하여 높게 나타내었다. 지방산 함량의 경우에도 냉압착 들기름의 oleic acid와 linoleic acid 함량이 가장 높았으며 포화지방산에 대한 불포화지방산의 비율이 12.10으로 가장 높게 나타났다. 또한 가열산화 처리에 의해 들기름의 Hunter L 값, 산가, 이중 공액산가 및 과산화물가는 증가하는 경향을 나타낸 반면, Hunter a 및 b 값, 갈변도, 총 페놀 함량은 감소하는 경향을 나타내었다.
