Evaluation of Electrolyzed Oxidizing Water as a Control Agent of Cucumber Powdery Mildew

  • The effect of the electrolyzed oxidizing water on Sphaerotheca fuliginea was investigated with germination and sporulation of the fungal conidia. The sporulation was inhibited by the electrolyzed oxidizing water of pH 2.5, 3.5, and 4.5, but was not inhibited by the distilled water adjusted pH with 1N-HCL solution. However, the electrolyzed oxidizing water did not affect conidial germination. The oxidation-reduction potential at pH 2.5 and pH 3.5 of electrolyzed oxidizing water were 1130 mV and 1060 mV, respectively, but those of distilled water adjusted with HCL solution were 550 mV and 490 mV, respectively. When the electrolyzed oxidizing water of ORP over 1100 mV was sprayed on cucumplanting, the disease severities of powdery mildew were about 8.5% and 19.2%, respectively. Disease severity of a standard control (triflumizole 30% WP, $500\textrm{mg}\textrm{/L}$) was about 3.0%, while that of plants without electrolyzed oxidizing water was to 45.8%.

Application of Volatile Antifungal Plant Essential Oils for Controlling Pepper Fruit Anthracnose by Colletotrichum gloeosporioides

  • Anthracnose caused by Colletotrichum gloeosporioides has been destructive during pepper fruit production in outdoor fields in Korea. In vitro antifungal activities of 15 different plant essential oils or its components were evaluated during conidial germination and mycelial growth of C. gloeosporioides. In vitro conidial germination was most drastically inhibited by vapour treatments with carvacrol, cinnamon oil, trans-cinnamaldehyde, citral, p-cymene and linalool. Inhibition of the mycelial growth by indirect vapour treatment with essential oils was also demonstrated compared with untreated control. Carvacrol, cinnamon oil, trans-cinnamaldehyde, citral and eugenol were among the most inhibitory plant essential oils by the indirect antifungal efficacies. Plant protection efficacies of the plant essential oils were demonstrated by reduced lesion diameter on the C. gloeosporioides-inoculated immature green pepper fruits compared to the inoculated control fruits without any plant essential oil treatment. In planta test showed that all plant essential oils tested in this study demonstrated plant protection efficacies against pepper fruit anthracnose with similar levels. Thus, application of different plant essential oils can be used for ecofriendly disease management of anthracnose during pepper fruit production.

Analysis of the Involvement of Chitin-Binding Domain of ChiCW in Antifungal Activity, and Engineering a Novel Chimeric Chitinase with High Enzyme and Antifungal Activities

  • An antifungal chitinase, ChiCW, produced by Bacillus cereus 28-9 is effective against conidial germination of Botrytis elliptica, the causal agent of lily leaf blight. ChiCW as a modular enzyme consists of a signal peptide, a catalytic domain, a fibronectin type-III-like domain, and a chitin-binding domain. When two C-terminal domains of ChiCW were truncated, $ChiCW{\Delta}FC$ (lacking the chitin-binding domain and fibronectin type III-like domain) lost its antifungal activity. Since $ChiCW{\Delta}C$ (lacking the chitin-binding domain) could not be expressed in Escherichia coli as $ChiCW{\Delta}FC$ did, a different strategy based on protein engineering technology was designed to investigate the involvement of the chitin-binding domain of ChiCW ($ChBD_{ChiCW}$) in antifungal activity in this study. Because ChiA1 of Bacillus circulans WL-12 is a modular enzyme with a higher hydrolytic activity than ChiCW but not inhibitory to conidial germination of Bo. elliptica and the similar domain composition of ChiA1 and ChiCW, the C-terminal truncated derivatives of ChiA1 were generated and used to construct chimeric chitinases with $ChBD_{ChiCW}$. When the chitin-binding domain of ChiA1 was replaced with $ChBD_{ChiCW}$, the chimeric chitinase named ChiAAAW exhibited both high enzyme activity and antifungal activity. The results indicate that $ChBD_{ChiCW}$ may play an important role in the antifungal activity of ChiCW.

  • The purpose of this study is to evaluate the efficacy of metabolites produced form Streptomyces rimosus in controlling the growth of sapwood - inhabiting fungi. In order to carry out this task, the following specific fungi were tested : sapstain fungi - Ceratocystis pilifera, Ceratocystis piceae, and Aureobasidium pullulans ; mold fungi - Trichoderma hazianum, Trichoderma viride, Penicillium cirtrinum, and Aspergillus niger. Based on the tests, the following observations can be drawn. 1. The conidial germination of sapstain and mold fungi was completely inhibited leaving a clear zone around the paper disc treated with metabolites. The best inhibition was observed in A. pullulans plate and the least in T. viride. 2. Concentration of SB medium for the production of metabolites from St, rimosus affected antifungal activity of metabolites against sapwood - inhabiting fungi. Metabolites prepared from 1/3${\times}$SB medium showed the best activity and the least activity was observed in metabolites form 1/4${\times}$medium. 3. in vivo and in vitro test using wood blocks, treatment of pine sapwood blocks with metabolites also inhibited conidial germination and thus prevented discoloration. 4. Treatment with metabolites did not change the macroscopic structure of wood and did not cause the discoloration of the surface of wood by pigments produced form St. rimosus. In conclusion the results of this study indicate that antifungal metabloites of St, rimosus could be used for the biological control of sapstain and mold fungi.

Roles of Forkhead-box Transcription Factors in Controlling Development, Pathogenicity, and Stress Response in Magnaporthe oryzae

  • Although multiple transcription factors (TFs) have been characterized via mutagenesis to understand their roles in controlling pathogenicity and infection-related development in Magnaporthe oryzae, the causal agent of rice blast, if and how forkhead-box (FOX) TFs contribute to these processes remain to be characterized. Four putative FOX TF genes were identified in the genome of M. oryzae, and phylogenetic analysis suggested that two of them (MoFKH1 and MoHCM1) correspond to Ascomycota-specific members of the FOX TF family while the others (MoFOX1 and MoFOX2) are Pezizomycotina-specific members. Deletion of MoFKH1 (${\Delta}Mofkh1$) resulted in reduced mycelial growth and conidial germination, abnormal septation and stress response, and reduced virulence. Similarly, ${\Delta}Mohcm1$ exhibited reduced mycelial growth and conidial germination. Conidia of ${\Delta}Mofkh1$ and ${\Delta}Mohcm1$ were more sensitive to one or both of the cell cycle inhibitors hydroxyurea and benomyl, suggesting their role in cell cycle control. On the other hand, loss of MoFOX1 (${\Delta}Mofox1$) did not show any noticeable changes in development, pathogenicity, and stress response. Deletion of MoFOX2 was not successful even after repeated attempts. Taken together, these results suggested that MoFKH1 and MoHCM1 are important in fungal development and that MoFKH1 is further implicated in pathogenicity and stress response in M. oryzae.

Biocontrol Activity of Volatile-Producing Bacillus megaterium and Pseudomonas protegens Against Aspergillus and Penicillium spp. Predominant in Stored Rice Grains: Study II

  • In our previous studies, Bacillus megaterium KU143, Microbacterium testaceum KU313, and Pseudomonas protegens AS15 have been shown to be antagonistic to Aspergillus flavus in stored rice grains. In this study, the biocontrol activities of these strains were evaluated against Aspergillus candidus, Aspergillus fumigatus, Penicillium fellutanum, and Penicillium islandicum, which are predominant in stored rice grains. In vitro and in vivo antifungal activities of the bacterial strains were evaluated against the fungi on media and rice grains, respectively. The antifungal activities of the volatiles produced by the strains against fungal development and population were also tested using I-plates. In in vitro tests, the strains produced secondary metabolites capable of reducing conidial germination, germ-tube elongation, and mycelial growth of all the tested fungi. In in vivo tests, the strains significantly inhibited the fungal growth in rice grains. Additionally, in I-plate tests, strains KU143 and AS15 produced volatiles that significantly inhibited not only mycelial growth, sporulation, and conidial germination of the fungi on media but also fungal populations on rice grains. GC-MS analysis of the volatiles by strains KU143 and AS15 identified 12 and 17 compounds, respectively. Among these, the antifungal compound, 5-methyl-2-phenyl-1H-indole, was produced by strain KU143 and the antimicrobial compounds, 2-butyl 1-octanal, dimethyl disulfide, 2-isopropyl-5-methyl-1-heptanol, and 4-trifluoroacetoxyhexadecane, were produced by strain AS15. These results suggest that the tested strains producing extracellular metabolites and/or volatiles may have a broad spectrum of antifungal activities against the grain fungi. In particular, B. megaterium KU143 and P. protegens AS15 may be potential biocontrol agents against Aspergillus and Penicillium spp. during rice grain storage.

Foliar Application of Plant Growth-Promoting Rhizobacteria Increases Antifungal Compounds in Pea (pisum sativum) Against Erysiphe pisi

  • Systemic effect of two plant growth-promoting rhizobacterial (PGPR) strains, viz., Pseudomonas fluorescens (Pf4) and P. aeruginosa (Pag), was evaluated on pea (Pisum sativum) against the powdery mildew pathogen Erysiphe pisi. Foliar spray of the two PGPR strains was done on specific nodal leaves of pea and conidial germination of E. pisi was observed on other nodal leaves, distal to the treated ones. Conidial germination was reduced on distant leaves and at the same time, specific as well as total phenolic compounds increased in the leaves distal to those applied with PGPR strains, thereby indicating a positive correlation. The strains induced accumulation of phenolic compounds in pea leaves and the amount increased when such leaves were get inoculated with E. pisi conidia. Between the two strains, Pag was found to be more effective than Pf4 as its effect was more persistent in pea leaves. Foliar application of PGPR strains for the control of powdery mildew of pea is demonstrated in vitro while correlating it with the increased accumulation of plant phenolics.

인삼 탄저병에 대한 항균성식물의 탐색과 이용 (Screening and Utilization of Antifungal Plant against Ginseng Anthracnose (Colletotrichum gloeosporioides))

  • 16과 20종의 공시 식물체 중 양파(Allium cepa), 대황(Rheum undulatum) 및 황련(Coptis japonica) 등의 추출액이 인삼 탄저병균(C. gloeosporioides)에 대해 균사생장 억제 효과가 우수하였다. 양파와 대황은 배지내의 추출액 농도가 1.0% 이상일 때 균사 생장억제 효과가 인정 되었고, 황련의 추출액은 배지 내의 농도가 0.5% 이상일 때에도 균사 생장 억제 효과가 50% 이상으로 나타나 공시된 식물체 추출액 중에서 항균 활성이 가장 우수하였다. 양파의 추출액은 10.0% 희석농도에서는 60% 이상의 발아 억제 효과가 있었고, 대황의 추출액은 2.0% 그리고 황련의 추출액은 1.0%의 희석 농도에서도 포자 발아억제율이 90% 이상으로 나타나 황련추출액이 가장 우수함을 알 수 있었다. 양파와 대황의 추출액은 2.0%의 희석농도에서는 어느 정도 방제 효과가 있었으나 1.0% 이하의 희석 농도에서는 효과가 많이 떨어졌고, 황련의 추출액은 1.0%의 희석농도에서도 50%이상의 방제 효과가 있었으나, 3가지 식물체 추출액 모두 대조약제인 Mancozebwp에 비해서는 그 효과가 상당히 떨어지는 경향이 었다. 세가지 식물체 추출액 모두 10.0%의 희석농도에서는 인삼 잎의 엽육이나 엽맥이 갈변하는 등의 약해의 징후가 나타났고, 황련의 추출액은 2.0%의 희석 농도에서도 약간의 약해 징후가 관찰되었다.

Fluorescent Pseudomonas Induced Systemic Resistance to Powdery Mildew in Mulberry (Morus spp.)

  • Native fluorescent pseudomonas bacteria were isolated from rhizosphere soil of mulberry and were evaluated against powdery mildew. In vitro conidial germination study showed significant (P<0.05) variation in conidial germination by bacterial strains Pf1 and Pf3. Mildew incidence was significantly varied due to treatment with various pseudomonas strains in vivo. Significantly (P<0.05) less mildew incidence was in plants treated with the bacterial strain Pf1 (9.11%) followed by Pf3 (13.48%) controlling 69.40% and 54.75% respectively compared with untreated control. Similarly, mildew severity was least (8.51%) in plants treated with strain Pf1 followed by Pf5 (9.23%) and Pf3 (9.72%) controlling the severity by 84.51%, 77.01% and 71.96% respectively compared with control. The bacterial strains significantly influenced biochemical constituents such as chlorophyll, protein and soluble sugar content of the mulberry leaf. Similarly, bacterial strains significantly increased the activity of the peroxidase (PO) and Polyphenol oxydase (PPO) activity from $7^{th}$ day up to the $28^{th}$ day after treatment. The strain Pf1, Pf3 and Pf5 exhibited a marked enhancement in the peroxidase at different periods of infection. Significant (P<0.01) negative correlation was found between powdery mildew severity with phenol content ($R^2=0.67$) as well as peroxidase ($R^2=0.92$) and polyphenol oxidase ($R^2=0.72$) activity thus confirms induction of systemic resistance in mulberry by pseudomonas bacteria. The study shows scope for exploration of rhizosphere fluorescent pseudomonas bacteria for induction of systemic resistance in mulberry to contain powdery mildew disease effectively.

Antagonistic activity of Streptomyces halstedii and S. violaceusniger In pepper anthracnose fungus Colletotrichum gloeosporioides

  • More than 1200 microorganisms were isolated from soil samples collected from various sources and localities. Among the isolates, 2 actinomyces (TH-04 and BA313) and 1 Bacillus sp. (CJ3) were selected as antagonists to pepper anthracnose fungus Colletotrichum gloeosporioides. These 3 isolates inhibitied mycelial growth of C gloeosporioides and the inhibition rates were over 70% on PDA. When the isolates were co-cultured with conidia of C. gloeosporioides in potato dextrose broth, conidial germination was severely inhibited and the inhibition rates of TH-04, BA313, and CJ3 at 24 hours were 75%, 72%, and 68%, respectively. The inhibition rates at n hours incubation were not much different from the rates at 24 hours. To check the activity on the plant, each isolate was mixed with equal volume of conidial suspension of C. gloeosporioides and wound-inoculated on green pepper fruit. After 6 days, the anthracnose lesions on the fruits inoculated with the mixture were much smaller than the lesions caused by the C. gloeosporioides itself. The lesion areas of TH-04 or BA313 treated pepper were less than 30% of the check. TH-04 and BA313 also showed antagonistic activity to Phytophthora spp. and Botrytis cinerea. By scanning electron microscopy and fatty acid analyses (MIDI), TH-04 and BA313 were identified to Streptomyces halstedii and S. violaceusniger, repectively.
