Spatiotemporal characteristics of atrial $Ca^{2+}$ sparks: evidence from two-dimensional rapid confocal imaging

  • Atrial myocytes have two functionally separate $Ca^{2+}$ release sites: those in peripheral sarcoplasmic reticulum (SR) adjacent to the $Ca^{2+}$ channels of surface membrane and those in central SR not associated with $Ca^{2+}$ channels. Study on the spatio-temporal properties of focal $Ca^{2+}$ releases (“sparks”) occurring spontaneously in central and peripheral sites of voltage-clamped rat atrial myocytes, using rapid two-dimensional (2-D) confocal $Ca^{2+}$ imaging revealed that peripheral and central sparks were similar in size and release time (~300,000 $Ca^{2+}$ ions for=12 ms), but significantly larger and longer than ventricular sparks. Both sites were resistant to Cd$^{2+}$ and inhibited by ryanodine. Peripheral sparks were brighter and flattened against surface membrane, had ~5-fold higher frequency, ~2 times faster diffusion coefficient, and dissipated abruptly. Central sparks, in contrast, occurred less frequently, were elongated along the cellular longitudinal axis, and dissipated slowly. Compound sparks (composed of 2-5 unitary focal releases) aligned longitudinally, occurred more frequently at the center.at the center.

Detection of Mitochondrial Reactive Oxygen Species in Living Rat Trigeminal Caudal Neurons

  • Growing evidence suggests that mitochondrial reactive oxygen species (ROS) are involved in various pain states. This study was performed to investigate whether ROS-induced changes in neuronal excitability in trigeminal subnucleus caudalis are related to ROS generation in mitochondria. Confocal scanning laser microscopy was used to measure ROS-induced fluorescence intensity in live rat trigeminal caudalis slices. The ROS level increased during the perfusion of malate, a mitochondrial substrate, after loading of 2',7'-dichlorofluorescin diacetate ($H_2DCF-DA$), an indicator of the intracellular ROS; the ROS level recovered to the control condition after washout. When pre-treated with phenyl N-tert-butylnitrone (PBN) and 4-hydroxy-2,2,6,6-tetramethylpiperidene-1-oxyl (TEMPOL), malate-induced increase of ROS level was suppressed. To identify the direct relation between elevated ROS levels and mitochondria, we applied the malate after double-loading of $H_2DCF-DA$ and chloromethyl-X-rosamine (CMXRos; MitoTracker Red), which is a mitochondria-specific fluorescent probe. As a result, increase of both intracellular ROS and mitochondrial ROS were observed simultaneously. This study demonstrated that elevated ROS in trigeminal subnucleus caudalis neuron can be induced through mitochondrial-ROS pathway, primarily by the leakage of ROS from the mitochondrial electron transport chain.

Preconditions for High Speed Confocal Image Acquisition with DMD Scanning.

  • Digital image-projection and several modifications are the classical applications of Digital Micromirror Devices (DMD), however further applications in the field of optical metrology are also available. Operated with certain patterns, a DMD can function, for instance, as an array of pinholes that may substitute the Galvanic mirror or the stage scanning system presently used for 2 dimensional scanning in confocal microscopes. The various process parameters that influence the result of measurement (e.g. pinhole size, lateral scanning pitch and the number of pinholes used simultaneously, etc.) should be configured precisely for individual measurements by appropriately operating the DMD. This paper presents suitable conditions for the diffraction limited analysis between DMD-optics-CCD to achieve the best performance. Also sampling theorem that is necessary for the image acquisition by scanning system is simulated with OPTISCAN which is the simulator based on the diffraction theory.

Effect of three different irrigation solutions applied by passive ultrasonic irrigation

  • Objectives: This study evaluated the maximum depth and percentage of irrigant penetration into dentinal tubules by passive ultrasonic irrigation (PUI). Materials and Methods: Thirty extracted human teeth were instrumented and divided into three groups. According to final irrigation regimen, 5.25% sodium hypochlorite (Group A, NaOCl), 2% chlorhexidine (Group B, CHX) and saline solution (Group C, control group) were applied with Irrisafe 20 tips (Acteon) and PUI. Irrigant was mixed with 0.1% rhodamine B. Sections at 2 mm, 5 mm, and 8 mm from the apex were examined with confocal laser scanning microscopy (CLSM). The percentage and maximum depth of irrigant penetration were measured. Kruskal-Wallis test and Mann-Whitney test were performed for overall comparison between groups at each level and for pairwise comparison, respectively. Within a group, Wilcoxon test was performed among different levels. p values less than 0.05 were considered significant. Results: In all groups, highest penetration depth and percentage of penetration were observed at the 8 mm level. At 2 mm level, Groups A and B had significantly greater depths and percentages in penetration than Group C (p < 0.05), but there were no significant differences between Groups A and B. At 5 mm level, penetration depths and percentage of penetration was not significantly different among the groups. Conclusions: NaOCl and CHX applied by PUI showed similar depth and percentage of penetration at all evaluated levels.

Mechanism of intragranular ferrite formation in heat-affected zone of titanium killed steel

  • A lot of work is carried out concerning to acicular ferrite formation in the weld metal of high strength and low-alloy steel. Those results are suggesting that oxides that contain titanium elements provides nucleation site of intragranular ferrite, referred as acicular ferrite. Thus, when intragranular ferrite is expected to form in heat-affected zone, oxide containing titanium element should be formed in the steel. However, normal steel is deoxidized by using aluminum element (Al-killed steel) with little oxygen content. It means almost oxygen is deoxidized with aluminum elements. In the present work, in order to form the acicular ferrite in the heat affected zone, with the same concept in the case of weld metal, the steel deoxidized with titanium element (titanium killed-steel) is prepared and the acicular ferrite formation is observed in detail by using laser-conforcal microscopy technique. The confocal technique makes it possible that the morphological change along the phase transformation from austenite to ferrite is in-situ tracked. Thus, the inclusion that stimulated the ferrite nucleation could be directly selected from the observed images, in the HAZ of the Ti-killed steel. The chemical composition of the selected inclusion is analyzed and the nucleation potential is discussed by changing the nucleation site with boron element. The potency for the ferrite nucleation is summarized and the existence of effective and ineffective manganese sulfide for nucleation is made clear.

Preliminary Study of the Measurement of Foreign Material in Galvanic Corrosion Using Laser Ultrasonic

  • A laser ultrasonic inspection system has the advantage of nondestructive testing. It is a non-contact mode using a laser interferometer to measure the vertical displacement of the surface of a material caused by the propagation of ultrasonic signals with the remote ultrasonic generated by laser. After raising the ultrasonic signal with a broadband frequency range using a pulsed laser beam, the laser beam is focused to a small point to measure the ultrasonic signal because it provides an excellent measurement resolution. In this paper, foreign materials are measured by a non-destructive and non-contact method using the laser ultrasonic inspection system. Mixed foreign material on the corroded part is assumed and the laser ultrasonic experiment is conducted. An ultrasonic wave is generated by pulse laser from the back of the specimen and an ultrasonic signal is acquired from the same location of the front side using continuous wave laser and Confocal Fabry-Perot Interferometer (CFPI). The characteristic of the ultrasonic signal of existing foreign material is analyzed and the location and size of foreign material is measured.

High-speed Three-dimensional Surface Profile Measurement with the HiLo Optical Imaging Technique

  • Various techniques to measure the three-dimensional (3D) surface profile of a 3D micro- or nanostructure have been proposed. However, it is difficult to apply such techniques directly to industrial uses because most of them are relatively slow, unreliable, and expensive. The HiLo optical imaging technique, which was recently introduced in the field of fluorescence imaging, is a promising wide-field imaging technique capable of high-speed imaging with a simple optical configuration. It has not been used in measuring a 3D surface profile although confocal microscopy originally developed for fluorescence imaging has been adapted to the field of 3D optical measurement for a long time. In this paper, to the best of our knowledge, the HiLo optical imaging technique for measuring a 3D surface profile is proposed for the first time. Its optical configuration and algorithm for a precisely detecting surface position are designed, optimized, and implemented. Optical performance for several 3D microscale structures is evaluated, and it is confirmed that the capability of measuring a 3D surface profile with HiLo optical imaging technique is comparable to that with confocal microscopy.

원전 고온 1차수 환경에서 응력부식균열의 실시간 마이크로 스케일 관찰 방법 개발 (Development of Method for In-situ Micro-Scale Observation of Stress Corrosion Cracking in High-Temperature Primary Water Environment)

  • The aim of this study was to develop a new in-situ observation method and instrument in micro-scale to investigate the mechanism of stress corrosion cracking (SCC) initiation of Ni-base alloys in a high temperature water environment of pressurized water reactors (PWRs). A laser confocal microscope (LCM), an autoclave with diamond window view port, and a slow strain-rate tester with primary water circulation loop system were components of the instrument. Diamond window, one of the core components of the instrument, was selected based on its optical, chemical, and mechanical properties. LCM was used to observe the specimen in micro-scale, considering the experimental condition of a high-temperature primary water environment. Using in-situ method and instrument, it is possible to observe oxidation and deformation of specimen surface in micro-scale through the diamond window in a high-temperature primary water in real-time. The in-situ method and instrument developed in this work can be utilized to investigate effects of various factors on SCC initiation in a high-temperature water environment.

미백 처리된 치아의 표면처리법이 레진과의 결합에 미치는 영향 (EFFECTS OF TOOTH-WHITENING AGENT ON ENAMEL AND DENTIN - A CONFOCAL LASER SCANNING MICROSCOPY STUDY)

  • The purpose of this study was to estimate the effect of a bleaching agent on tooth surfaces and to evaluate the resin bond strength according to different surface treatments on bleached teeth. To prepare for the experimental samples, first, extracted human third molars were used and the body portions of the crowns were cut into four equal-sized specimens. Next, each specimen was mounted in an plastic bottle with self-cured resin and highly polished to have them reveal the enamel or dentin surfaces. Then, the enamel(E) and dentin(D) specimens were divided into four ; 1) non-bleached, laser-treated(NBLA) group 2) bleached, laser-treated(BLLA) group 3) non-bleached, acid-treated(NBAC) group and 4) bleached, acid-treated(BLAC) group. Here, $opalescence^{(R)}$ (10% carbamide peroxide) was used for bleaching agent. The treated specimens were observed by confocal laser scanning microscopy and bonded with composite resin for shear bond test. The following results were obtained from this experiment : 1. Compared with the ENB group, the EBL group was shown be dyed about $20{\mu}m$ deeper with rhodamine B. The DBL group appeared to be caved in at the entry part of the dentinal tubules, was dyed about $20{\mu}m$ deeper and $5{\mu}m$ wider in diameter, compared with the DNB group. 2. In comparison with the EBLAC group, the ENBAC group looked evenly bonded with the resin, while the DNBAC group, compared to DBLAC group, was observed to have its resin tags penetrated about $50{\mu}m$ deeper. Other than those, however, no observable differences between ENBLA and EBLLA group or between DNBLA and DBLLA group were found. 3, At the shear bond test, the ENBAC group was shown to have statistically significant higher shear bond strength than the EBLAC group(p<0.05). No statistically significant differences between the ENBLA and the EBLLA groups were observed(p>0.05). 4. At the shear bond test, the DNBAC group was shown to have statistically significant higher shear bond strength than the DBLAC group(p<0.05). No statistically significant differences between the DNBLA and the DBLLA groups were observed(p>0.05). The in vitro observations above suggest that tooth-bleaching procedure brings a certain change on enamel and dentin surfaces and it weakens the shear bond strength with composite resin when the bleached tooth was acid-treated.

압력에 따른 균열 간극변화와 수리전도도 변화 관찰 (The Changes of Aperture Variation and Hydraulic Conductivity for Compression Variability)

  • 압축력의 변화에 따른 균열 간극변화 양상을 측정하고 간극변화와 수리전도도와의 관계를 파악하기 위해 이 연구에서는 다섯 단계의 수직 일축압축력을 균열면에 연속적으로 가하면서 고해상도의 공초점 레이저 스캔 현미경 (confocal laser scanning microscope; CLSM)을 이용하여 간극의 크기를 측정하고 디지털 이미지를 획득하였다. 기존의 연구들과는 달리 이 연구의 측정방법은 압력변화에 대한 동일시료 간극의 반응을 연속적으로 파악할 수 있는 점이 특징이다. 측정결과는 간극크기가 일정하지 않은 불평탄한 균열형태를 매우 잘 나타내었다. 균열 조도(roughness)로 인해 압력에 따른 간극 변화량은 일정하지 않고 부분마다 다름을 보였다. 각 압력단계에서 간극변화에 따른 투수성 변화양상을 파악하고자 실내투수시험을 병행하여 실시한 결과, 각 압력단계에서의 투수성 변화도 일정한 감소율을 나타내지 않고 간극 변화율이 크더라도 투수율은 오히려 작은 경우도 관찰되었다. 현미경을 통해 측정한 물리적 간극과 실제 유체유동 경로가 되는 수리간극의 크기 차이를 파악하기 위해 계산을 한 결과, 미미한 갈이지만 물리적 간극보다 작은 크기의 수리간극이 구해졌다. 또한, 실내투수시험 결과를 이용하여 구한 투수계수는 삼승법칙을 따르지 않는 것으로 드러났으며, 이러한 사실들은 균열 양쪽이 서로 평행하지 않고 불평탄한 양상을 가짐을 의미하는 것으로서, 현미경을 통해 직접 관찰한 간극양상과 잘 일치하는 것이다.
