A Clinical Study of Patients Developing the Other Psychiatric Disorder During the Course of Social Phobia (사회공포증 경과중 타 정신질환이 발병한 환자들에 대한 연구)

  • We have studied 16 patients diagnosed of social phobia among the hospitalized patients for 1 year, from September 1994 to August 1995, to evaluate whether serial phobic symptoms are the prodromal symptoms of the other psychiatric disorder or concomitant disorder. The results were as follows; 1) In the subject of this study, the mean age of onset of social phobic symptoms was 17.3, and the mean age of inital hospital visit was 27.4. 2) The proportion of offensive type was higher than the simple type : 7 cases of the subjects classified as simple type, 9 cases as offensive type. 3) According to the type of social phobia, they developed different type of psychiatric disorder. Majority of patients with simple type had final diagnosis of mood disorder. Patients with offensive type had schizophrenia spectrum disorder, such as schizophrenia, delusional disorder, and schizophrenia, personality disorder. 4) Social phobic symptoms have taken various courses according to the final diagnosis. In the patients who developed major depression and delusional disorder, social phobic symptoms have not been changed during the course. Patients with the final diagnosis of schizophrenia and schizotypal personality disorder have showed various social phobic symptoms. In the patients with the final diagnosis of bipolar disorder, their social phobic symptoms disappeared with improvement of depression.

Simultaneous Formation of Fructosyltransferase and Glucosyltransferase in Aureobasidium pullulans

  • Aureobasidium puliulans possesses the capacity for simultaneous formation of fructosyltransferase and glucosyltransferase in various sugar media including sucrose, maltose, glucose and fructose. Among them, sucrose (300 g/1) was the most suitable carbon source for fructosyltransferase production, while fructose (100 g/1) gave the maximal production of glucosyltransferase. There existed a critical concentration for the optimal formation of enzymes in sucrose, glucose and fructose media. By contrast, no effect of maltose concentrations up to 300 g/1 was observed. The specific activity of the glucosyltransferase on maltose medium was highest during the early period of fetmentation, after which a sharp decrease occurred, whereas fructosyltransferase activity on sucrose medium maintained a nearly constant rate for a given culture period. Concomitant production of fructosyltransferase and glucosyltransferase was investigated with different combinations of lower concentrations of sucrose and maltose. Maltose supplementation in sucrose media and sucrose addition to maltose media enhanced the activity ratios of fructosyltransferase to glucosyltransferase as compared to that of non-supplemented media. Several polymers and surfactants were added in an attempt to enhance enzyme production, and supplementation of polyoxyethylene-sorbitan monolaurate (Tween 20) promoted fructosyltransferase production by 20%.

Effects of Xylose Reductase Activity on Xylitol Production in Two-Substrate Fermentation of Recombinant Saccharomyces cerevisiae

  • Three recombinant Saccharomyces cerevisiae strains showing different levels of xylose reductase activity were constructed to investigate the effects of xylose reductase activity and glucose feed rate on xylitol production. Conversion of xylose to xylitol is catalyzed by xylose reductase of Pichia stipitis with cofactor NAD(P)H. A two-substrate fermentation strategy has been employed where glucose is used as an energy source for NADPH regeneration and xylose as substrate for xylitol production. All recombinant S. cerevisiae strains Yielded similar specific xylitol productivity, indicating that xylitol production in the recombinant S. cerevisiae was more profoundly affected by the glucose supply and concomitant It generation of cofactor than the xylose reductase activity itself. It was confirmed in a continuous culture that the elevation of the glucose feeding level in the xylose-conversion period enhanced the xylitol productivity in the recombinant S. cerevisiae.

Role of Osmotic and Salt Stress in the Expression of Erythrose Reductase in Candida magnoliae

  • The osmotolerant yeast, Candida magnoliae, which was isolated from honeycomb, produces erythritol from sugars such as fructose, glucose, and sucrose. Erythrose reductase in C. magnoliae (CmER) reduces erythrose to erythritol with concomitant oxidation of NAD(P)H. Sequence analysis of the 5'-flanking region of the CmER gene indicated that one putative stress response element (STRE, 5'-AGGGG-3'), found in Saccharomyces cerevisiae, exists 72 nucleotides upstream of the translation initiation codon. An enzyme activity assay and semiquantitative reverse transcription polymerase chain reaction revealed that the expression of CmER is upregulated under osmotic and salt stress conditions caused by a high concentration of sugar, KCl, and NaCl. However, CmER was not affected by osmotic and oxidative stress induced by sorbitol and $H_2O_2$, respectively. The basal transcript level of CmER in the presence of sucrose was higher than that in cells treated with fructose and glucose, indicating that the response of CmER to sugar stress is different from that of GRE3 in S. cerevisiae, which expresses aldose reductase in a sugarindependent manner. It was concluded that regulation of CmER differs from that of other aldose reductases in S. cerevisiae.

Effects of Extacellular Divalent Cations on the Hyperpolarization-activated Currents in Rat Dorsal Root Ganglion Neurons (세포 밖 2가 양이온이 과분극에 의해 활성화되는 전류($I_h$)에 미치는 영향)

  • The hyperpolarization-activated current ($I_h$) is an inward cation current activated by hyperpolarization of the membrane potential and plays a role as an important modulator of action potential firing frequency in many excitable cells. In the present study we investigated the effects of extracellular divalent cations on $I_h$ in dorsal root ganglion (DRG) neurons using whole-cell voltage clamp technique. $I_h$ was slightly increased in $Ca^{2+}$-free bath solution. BAPTA-AM did not change the amplitudes of $I_h$. Amplitudes of $I_h$ were decreased by $Ca^{2+}$, $Mg^{2+}$ and $Ba^{2+}$ dose-dependently and voltage-independently. Inhibition magnitudes of $I_h$ by external divalent cations were partly reversed by the concomitant increase of extracellular $K^+$ concentration. Reversal potential of $I_h$ was significantly shifted by $Ba^{2+}$ and $V_{1/2}$ was significantly affected by the changes of extracellular $Ca^{2+}$ concentrations. These results suggest that $I_h$ is inhibited by extracellular divalent cations ($Ca^{2+}$, $Mg^{2+}$ and $Ba^{2+}$) by interfering ion influxes in cultured rat DRG neurons.

The Epoxy-metal Interphase and Its Incidence on Practical Adhesion

  • Epoxy-amine liquid prepolymers are extensively applied onto metallic substrates and cured to obtain painted materials or bonded joint structures. Overall performances of such systems depend on the created interphase between the organic layer and the substrate. When epoxy-amine liquid mixtures are applied onto more or less hydrated metallic oxide layer, concomitant amine chemical sorption and hydroxide dissolution appear lending to the chelate formation. As soon as the chelate concentration is higher than the solubility product, these species crystallize as sharp needles. Moreover, intrinsic and thermal residual stresses are developed within painted or bonded systems. When residual stresses are higher than the organic layer/substrate adhesion, buckling, blistering, debonding may occur leading to a catastrophic drop of system performances. Practical adhesion can be evaluated with either ultimate parameters (Fmax or Dmax) or the critical strain energy release rate, using the three point flexure test (ISO 14679-1997). We observe that, for the same system, the ultimate load decreases while residual stresses increase when the liquid/solid time increases. Ultimate loads and residual stresses depend on the metallic surface treatment. For these systems, the critical strain energy release rate which takes into account the residual stress profile and the Young's modulus gradient remains quite constant whatever the metallic surface treatment was. These variations will be discussed and correlate to the formation mechanisms of the interphase.

Induced apoptosis in human Uterine Leiomyoma Cells by treatment with Chiljehyangbu-hwan (칠제향부환(七製香附丸)이 자궁근종세포의 성장억제와 세포자멸사에 미치는 영향)

  • Purpose : Uterine leiomyoma (fibroids) are benign smooth muscle tumors originating from the myometrium. These benign neoplasms of monoclonal origin are typically diagnosed during the reproductive years, occurring only after puberty and tending to regress after menopause. In the present study we used Chiljehyangbu-hwan to determine its growth inhibitory effect and apoptosis in human uterine leiomyoma cells. Methods : Primary cultured human uterine leiomyoma cells were treated with Chiljehyangbu-hwan. Cell viability analysis was analyzed by MTS assay and FACS was performed to ascertain the effects Chiljehyangbu-hwan. DNA fragmentation analysis and casapase-3 activity test were done. Expression of apoptosis related proteins were evaluated by Western blot analysis. Results : Cell viability was significantly influenced by Chiljehyangbu-hwan treatment in a dose-dependent manner in leiomyoma cells compare to normal myometrial cells. FACS showed that Chiljehyangbu-hwan induced Sub G1 arrest. DNA fragmentation assay was carried out and apoptosis was detected. Activation of caspase-3, down-regulation of Bcl-2, with concomitant increased expression in Bid and Bax were observed. Chiljehyangbu-hwan treatment of uterine leiomyoma cells resulted in a concentration-dependent cell death induced via the mitochondrial pathway.

Surgical treatment of the disease involving ascending aorta (상행 대동맥 질환의 외과적 치료)

  • From February 1985 to February 1993, 18 operations were performed in 17 patients for treatment of aneurysmal disease [n=12] and/or dissection of the ascending aorta [n=6]. The ages ranged from 26 to 69 years [mean 44.3 $\pm$ 11.0 years].The proposed operations include composite graft replacement of aortic valve and ascending aorta with coronary reimplantation in 11, graft replacement of ascending aorta alone in 5, aortic valve replacement and supracoronary graft replacement in 1 and ascending aorta to abdominal aorta bypass with thromboexclusion of descending aorta in one patient. Both Bentall [n=6] and Cabrol [n=5] technique were utilized for reimplantation of coronary arteries.Concomitant replacement of aortic arch and arch vessel reconstruction was necessary in two patients. Hypothermic circulatory arrest was utilized in 6 patients. Recently, four patients were managed on warm blood continuous cardioplegia via retrograde route. There were no operative deaths. No significant postoperative complications were noted. Postoperative follow up was complete in 15 patients from 1 month to 72 months. Redo operation was necessary in one patient who had suffered from distal recurrence of dissection 5 years after successful Bentall operation. The other patients are all in excellent clinical condition. From our early experience with those 17 cases, we assume that satisfactory operative result could be achieved with a variety of surgical technique including hypothermic circulatory arrest. In addition, continuous perfusion of warm blood cardioplegia via retrograde route is supposed to be beneficial in selected cases.

Clinical Study of Surgical Treatment of Acquired Tricuspid Regurgitation (후천성 삼첨판폐쇄부전증의 외과적 치료에 대한 연구)

  • Fourty-five cases of operation were performed for the correction of tricuspid regurgitation [TR] in Pusan National University Hospital between 1982 and 1991.The mean age of the patients was 32.6 years and female was dominant[M:F=1:2.2].Isolated tricuspid regurgitation was rare and 43 patients underwent concomitant other valvular operation including mitral valve replacement. Functional cause was in 39 cases and organic lesions were found in 6 cases. Operative methods were Kay annuloplasty[29], De Vega annuloplasty[12], and tricuspid valve replacement[4]. Ring annuloplasty was not performed. Operative mortality rate was 11.1%[5/45] and late mortality rate was 6.7%[2/30]. The tricuspid valve surgery itself was not a serious risk factor for hospital death and no heart block nor thrombosis was complicated. By echocardiogram early[within 30 days] and late [mean:4.9years] changes of postoperative TR were evaluated. De Vega annuloplasty seemed to bring better late result than Kay annuloplasty[p<0.05]. In four patients with late severe TR,previously replaced tissue valve degeneration[2], pulmonary hypertension[1] and rheumatic TR[1] were the possible causes. This experience suggests that De Vega annuloplasty can be a reliable method in the majority of patients with moderate-to-severe functional TR.
