Nitrite Accumulation and Nitrite Oxidation Efficiency of High-Concentration Ammonia Nitrogen by SRT change (SRT 변화를 통한 고농도 암모니아성 질소의 아질산성 질소 축적 및 아질산화 효율 평가)

  • 축산물의 수요가 증가함에 따라 가축의 사육규모 및 두수도 증가하여 가축분뇨의 발생량이 증가한다. 가축분뇨는 일반하수에 비해 고농도의 유기물, 질소, 인 등의 오염물질이 포함되어 있다. 적절한 처리 없이 하수처리장 및 수계로 유입될 경우 하수처리장 처리 효율에 영향을 미치거나 부영양화 등 다양한 문제를 야기 시킬 수 있다. 이러한 문제를 해결하기 위해 아질산화 반응을 이용하는 다양한 공정들이 연구되고 있다. 아질산화 반응은 완전 질산화 반응에 비해 질산화 단계에서 약 25%의 산소요구량이 절감되고, 탈질 단계에서 약 40%의 탄소원이 절감되는 경제적 장점이 있기 때문이다. 본 연구에서는 부피 8L의 실험실 규모 아질산화 반응조 원통형 아크릴로 제작되었고, 서울 A하수처리장 미생물을 채취하여 사용했다. 또한 SRT의 영향을 살펴보기 위하여 35℃ 동일 온도를 유지했다. 반응조 슬러지 반송 및 폐기가 없는 완전 혼합 반응조로 SRT와 HRT가 동일하게 운전하는 방법을 사용하여 SRT를 조절하는 방식으로 운전했다. SRT의 경우 8일, 6일, 4일, 2일의 변경조건을 통해서 차이를 살펴보았다. Ammonia Removal Rate(%)의 경우 각각 86%, 86%, 87%, 24%의 효율을 보였고, Nitrite Conversion Rate(%)의 경우 각각 10%, 45%, 80%, 41%의 효율을 보였다. 35℃ 실험실 규모 반응조에서 가축분뇨 유입 원수의 아질산화 반응을 유도하기 위해서는 SRT운전 조건은 4~8일, 고효율의 아질산화 반응을 유도하기 위해서는 SRT 4일 조건이 적합하다고 판단된다. 본 연구는 실제 가축분뇨 처리 효율 상승을 위해서 아질산화 공법을 도입할 경우 중요한 자료로 이용 가능할 것으로 판단된다.

Effects of Kanghwalsokdan-tang on Dermal Fibroblast (강활속단탕(羌活續斷湯)이 인체피부 섬유아세포에 미치는 영향)

  • Objectives: This study was performed to elucidate the effects of Kanghwalsokdan-tang extract(KS) on hyper-plasy of collagen and cell damage in UVB-irradiated dermal fibroblast. Methods: To demonstrate the effects of KS on wound healing we used human dermal fibroblast(F6). We evaluated the amount of increased PICP, TIMP-1 in dermal fibroblast. PICP, TIMP-1 concentration was measured using EIA kit. Also, we measured the nitrite production, and LDH release in UVB-irradiated dermal fibroblast to elucidate the action-mechanism of KS. Results: 1. KS decreased the cell proliferation of dermal fibroblast. 2. KS decreased the biosynthesis of collagen in dermal fibroblast. 3. KS decreased the synthesis of TIMP-1 in dermal fibroblast. 4. KS had no effect on the LDH-release of UVB-irradiated dermal fibroblast. 5. KS inhibited nitrite production in UVB-irradiated dermal fibroblast. Conclusions: From the results, we concluded that KS has a protective effect on wound healing and photoaging.

Impact of Physiological Stresses on Nitric Oxide Formation by Green Alga, Scenedesmus obliquus

  • The rate of apparent nitric oxide (NO) release, as measured in the exhaust gas of green alga, Scenedesmus obliquus, depended on the light intensity and pH. It doubled after lowering the temperature from $25^{\circ}C{\;}to{\;}15^{\circ}C$ and strongly decreased from $35^{\circ}C{\;}to{\;}42^{\circ}C$. The Scenedesmus cells, deficient in nitrogen or phosphorus, demonstrated a significant increase in NO production following their transfer to nitrate- and phosphate-rich media. The addition of herbicides (DCMU and glyphosate) or toxic concentrations of $Cu^{2+}{\;}or{\;}Fe^{3+}$ produced strong NO peaks, resembling those that occurred after sudden darkening. An increase in the $Ni^{2+}$ concentration to 20 ppm resulted in a gradual increase of NO release from the initial ~1.5 ppbv to>20 ppbv, whereas $Cd^{2+}$ instantaneously suppressed the NO by the cultures of Scenedesmus was not altered by L-NNA, an inhibitor of nitric oxide synthase (NOS), or by its substrate, L-arginine. This seems to exclude the role of NOS in the NO formation under study. Accordingly, it can be assumed that the rate of NO formation is mainly a function of dynamic nitrite pool sizes and environmental factors significantly affect the NO production in algae.

Chemical Composition of Pinus koraiensis Seed and Its Biological Activity

  • This study was to investigate the chemical composition and biological activities of Pinus Koraiensis seed. The oil, moisture, ash, crude protein, and carbohydrate contents of P. Koraiensis seed were 58.21, 7.84, 1.56, 14.26, and 18.13%, respectively. The ratios of essential amino acid and nonessential amino acid against total amino acids were 36.6 and 60.3%, respectively. The saponification value of seed oil was 166.8 mg KOH/g oil. Among various fatty acids, the linoleic acid content was the highest, 35.5%, which was approximately 72.6% of polyunsaturated fatty acid. The ethylacetate extract of P. Koraiensis seed had the highest DPPH radical scavenging activity (62.8%) at 7.0 mg/mL, followed by hexane extract, methanol extract, and hot water extract. The maximum nitrite scavenging activity was obtained 59.3% at pH 1.2. The total phenolic concentration of ethylacetate extract was 98.7 mg/g, approximately 4.8 folds higher than that of the hot water extract. The maximum inhibition activities of elastase using ethylacetate extract and collagenase using hexane extract were 58.8 and 40.7%, respectively. These results indicate that P. koraiensis seed extract could be applied to present the possibilities of industrial applications for the developments of cosmetics.

Nitrate Removal in a Packed Bed Reactor Using Volatile Fatty Acids from Anaerobic Acidogenesis of Food Wastes

  • A packed bed reactor (PBR) was fed with nitrate containing synthetic wastewater or effluent from a sequencing batch reactor used for nitrification. The C source introduced into the PBR consisted of volatile fatty acids (VFAs) produced from anaerobic acidogenesis of food wastes. When nitrate loading rates ranged from $0.50\;to\;1.01\;kg\;N/m^{3}{\cdot}d$, the PBR exhibited $100{\sim}98.8%\;NO_{3}^{-}-N$ removal efficiencies and nitrite concentrations in the effluent ranged from $0\;to\;0.6\;NO_{2}^{-}-N\;mg/L$. When the PBR was further investigated to determine nitrate removal activity along the bed height using a nitrate loading rate less than $1.01\;kg\;N/m^{3}{\cdot}d$, 100% nitrate removal efficiency was observed. Approximately 83.2% nitrate removal efficiency was observed in the lower 50% of the packed-bed height. When reactor performance at a C/N ratio of 4 and a C/N ratio of 5 was compared, the PBR showed better removal efficiency (96.5%) of nitrate and less nitrite concentration in the effluent at the C/N ratio of 5. VFAs were found to be a good alternative to methanol as a carbon source for denitrification of a municipal wastewater containing 40 mg-N/L.

The Nirite Scabenging and Electron Donating Ability of Potato Extracts (감자 추출물의 아질산염 소거 및 전자공여 작용)

  • This study was conducted to investigate functional properties of 70% acetone extract in different parts of two potato varieties. 'Superior' and 'Atlantic' potato were processed by following method. Potatoes are washed, peeled, sliced and steamed before hot air drying for flesh and peel powder. Hunter's L and b values of flesh powder from blanched 'Superior' potato were higher than flesh powder from fresh 'Superior' potato. Discoloration of 'Superior' potato was inhibited by steam blanching. Contents of total polyphenol and chlorogenic acid in the 70% acetone extract of 'Superior' potato were higher than those of 'Atlantic' potato paticularly in the blanched flesh and peel. But flavonoid was not detected in flesh extract. The phenolic concentration was not decreased by the heat treatment. The nitritescavenging and electron donating ability was greatest at the tuber peel including the skin and cortex tissue 1mm beneath the skin. From the above results browning reaction and polyphenol contents in different parts of potato slices were dependent on cutivars. Peels contained more polyphenols than those from the flesh. The results indicate that potato peel extract of steam blanched plus hot air dried potato tuber showed the effectiveness as a natural nitrite scavenger and antioxidant.

Selection of Mediators for Bioelectrochemical Nitrate Reduction

  • The bioelectrochemical reduction of nitrate in the presence of various mediators including methyl viologen and azure A was studied using a 3-electrode voltammetric system. The catalytic potential for the reduction of the mediators was observed in the reactor, which for methyl viologen and azure A were -0.74 V and -0.32 V, respectively, with respect to the potential of Ag/AgCl reference electrode. This potential was then applied to a working electrode to reduce each mediator for enzymatic nitrate reduction. Nitrite, the product of the reaction, was measured to observe the enzymatic nitrate reduction in the reaction media. Methyl viologen was observed as the most efficient mediator among those tested, while azure A showed the highest electron efficiency at the intrinsic reduction potential when the mediated enzyme reactions were carried out with the freely solubilized mediator. The electron transfer of azure A with respect to time was due to the adhesion of azure A to the hydrophilic surface during the reduction. In addition, the use of the adsorbed mediator on conductive activated carbon was proposed to inhibit the change in the electron transfer rate during the reaction by maintaining a constant mediator concentration and active surface area of the electrode. Azure A showed better than nitrite formation than methyl viologen when used with activated carbon.

The Biological Activity of Deer Antler Extract in vitro (In vitro에 의한 녹용 추출물의 생리 활성 효과)

  • Our research objective was to examine the in vitro biological activity of deer antler(Nogyong in Korean) extract, including the antioxidative, nitrite scavenging, and tyrosinase inhibitory effects, as well as the antithrombotic, and angiotensin I converting enzyme(ACE) inhibitory activities. The carbohydrate, protein, fat, and mineral contents of the deer antler were 7.6%, 65.3%, 3.2% and 23.9%, respectively. The electron donating ability(EDA) by the reduction of 2,2'-diphenyl-1-picrylhydrazyl(DPPH) was 67.1%, and the inhibition rate of lipid peroxidation by the thiocyanate method using linoleic acid was 92.1% in 100 mg/ml of extract. The nitrite scavenging effects were pH dependent, and were highest at pH 1.2 and lowest at pH 6.0. The sample inhibition rate against tyrosinase was above 64.0%. The platelet aggregation induced by ADP(adenosine-5'diphosphate) was inhibited up to 51.7%, and the inhibitory effect was dependent on the sample concentration. Lastly, the inhibition rate of ACE was 47.5% in 100 mg/ml of deer antler extract.

Lipocortin 1 Mediates the Suppressive Effects of Dexamethasone on ConA-induced Proliferative Response and Nitric Oxide Production in Rat Splenic Leukocytes

  • Lipocortin 1 has been proposed as a putative mediator of anti-inflammatory actions of glucocorticoids. We investigated the role of lipocortin 1 in the effect of dexamethasone using rat splenic leukocytes. Concanavalin A(ConA; 1 ${\mu}g/ml$) increased the leukocyte proliferation and nitric oxide(NO) generation, which were measured as $[^3H]-thymidine$ uptake by the cells and nitrite accumulation in the culture media, respectively. Dexamethasone suppressed ConA-induced cell proliferation, in a concentration-dependent manner with $EC_{50}$ around 50nM. The addition of anti-lipocortin l(Anti-LCl) reversed dexamethasone effects: 0.24, 1.2, 6 ${\mu}g/ml$ of Anti-LC1 reversed dexamethasone(50 nM)-induced suppression of thymidine uptake by $9{\pm}3%$, $16{\pm}3%$, $36{\pm}5%$, respectively; 0.24, 1.2, and 6 ${\mu}g/ml$ of Anti-LCI reversed dexa-methasone-induced decrease of nitrite concentration by $49{\pm}16%$, $61{\pm}20%$, $77{\pm}19%$, respectively. The present data indicate that lipocortin 1 mediates, at least in part, glucocorticoids-induced suppression of leukocyte proliferation and blockade of NO generation.

Relationship between Vegetation Composition and Dissolved Nitrogen in Wetlands of Higashi-Hiroshima, West Japan

  • Twenty-four wetlands located in Higashi-Hiroshima City in West Japan were selected for this study in order to investigate both the relationship between aquatic plant composition and environmental conditions; and the relationship between changing land use patterns in the catchments and the concentration of different forms of nitrogen in the wetlands. The dominant and subdominant species which comprised the principal vegetation were determined based on a vegetation census conducted in each wetland during the growing season from June to August, 2006. The seasonal variations of water quality factors (pH, electrical conductivity, turbidity, dissolved oxygen, total dissolved solid, and temperature) and different forms of nitrogen such as nitrite, nitrate, ammonium, total nitrogen, dissolved organic nitrogen and dissolved inorganic nitrogen concentrations were analyzed as important indicators of water quality for the surface water of the wetlands. The surveyed wetlands were classified into three types (non-disturbed wetlands, moderately-disturbed wetlands and highly-disturbed wetlands), based on the degree of human disturbance to their catchment areas. An analysis of variance indicated that there was a significant difference among the wetland groups in the annual mean values of electrical conductivity, total dissolved solids, total nitrogen, nitrite, dissolved inorganic nitrogen and dissolved organic nitrogen. Classification of the wetlands into three groups has revealed a pattern of changes in the composition of plant species in the wetlands and a pattern of changes in nitrogen concentrations. A majority of the non-disturbed wetlands were characterized by Brasenia schrebi and Trapa bispinosa as dominant; with Potamogeton fryeri and Iris pesudacorus as sub-dominant species. For most of the moderately-disturbed wetlands, Brasenia schrebi were shown to be a dominant species; Elocheriss kuriguwai and Phragmites australis were observed as sub-dominant species. For a majority of the highly-disturbed wetlands, Typha latifolia and T. angustifolia were observed as dominant species, and Nymphea tetragona as the sub-dominant species in the study area. An analysis of land use and water quality factors indicated that forest area played a considerable role in reducing the concentration of nutrients, and can act as a sink for surface/subsurface nutrient inputs flowing into wetland water, anchor the soil, and lower erosion rates into wetlands.
