  • This study was performed to determine the composition of phenolic substances contained in the leaves of Cirsium setidens (Compositae), validate the high-performance liquid chromatography (HPLC) method, and determine the in vivo sedative effect of the main component pectolinarin. Six phenolic compounds isolated from C. setidens were spectroscopically identified as chlorogenic acid (1), hyperoside (2), 3,4-di-O-caffeoylquinic acid (3), caffeic acid methyl ester (4), linarin (5), and pectolinarin (6) and then used as standard compounds for HPLC analysis. HPLC proved to be precise, accurate, and sensitive for the simultaneous analysis of the phenolic substances. In particular, six compounds showed good regression ($R^2$ > 0.999) within test ranges and recovery was in the range of 95.4 - 104.8%. The content of pectolinarin was considerably higher (156.48 mg/g) than those of other phenolic substances including the other flavone glycoside, linarin (18.99 mg/g). The contents of other phenolic substances, in order, were chlorogenic acid (8.41 mg/g), 3,4-di-O-caffeoylquinic acid (5.74 mg/g), hyperoside (4.33 mg/g), and caffeic acid methyl ester (0.51 mg/g). Oral administration with compound 6 (10 and 20 mg/kg) enhanced the sleeping time induced by pentobarbital in mice, indicating that it has a sedative effect.

  • Six caffeoylquinic acids of Cacalia firma (Komarov) Nakai (Compositae) leaves were identified using standard compounds by HPLC. Each content of those compounds in dried weight was determined as follows: 3,4-di-O-caffeoylquinic acid (1.44${\pm}$0.04 mg/g of dried weight), 3,5-di-O-caffeoyl-muce-quinic acid (2.47${\pm}$0.12 mg/g), 3,5-di-O-caffeoylquinic acid (3.74${\pm}$0.24 mg/g), 5-caffeoylquinic acid (chlorogenic acid, 5.20${\pm}$0.09 mg/g), 3-caffeoylquinic acid (1.35${\pm}$0.01 mg/g) and 3-Op-coumaroylquinic acid (3.84${\pm}$0.25 mg/g). The total content of six caffeoylquinic acids in the plant material was calculated as 18.05${\pm}$0.69 mg/g while the percentage of the six compounds in the MeOH extract was calculated as 30.85${\pm}$1.18%. The $IC_{50}$ value of the MeOH extract scavenging peroxynitrite ($ONOO^-$) was shown as 3.22${\pm}$0.57 ${\mu}g$/ml.

  • Disagreement among the various publications providing lists of Korean endemic plants makes confusion inevitable. We summarized the six previous reports providing comprehensive lists of endemic plants in Korea: 407 taxa in Lee (1982), 570 taxa in Paik (1994), 759 taxa in Kim (2004), 328 taxa in Korea National Arboretum (2005), 515 taxa in the Ministry of Environment (2005) and 289 taxa in Flora of Korea Editorial Committee (2007). The total number of endemic plants described in the previous reports was 970 taxa, including 89 families, 302 genera, 496 species, 3 subspecies, 218 varieties, and 253 formae. Endemic plants listed four times or more were collected to compare the data in terms of scientific names and synonyms (339 taxa in 59 families and 155 genera). If the varieties and formae were excluded, the resulting number of endemic plants was 252 taxa for the 339 purported taxa analyzed. Seven of the 155 genera analyzed were Korean endemic genera. Among the 339 taxa, the same scientific names were used in the original publications for 256 taxa (76%), while different scientific names were used for 83 taxa (24%). The four largest families were Compositae (42 taxa, 12.4%), Ranunculaceae (19 taxa, 5.6%), Rosaceae (19 taxa, 5.6%), and Scrophulariaceae (19 taxa, 5.6%). Saussurea (Compositae) had the highest number of taxa within one genus (17 taxa; 5% of total endemic taxa).

  • In Korea, the leaves of Sonchus brachyotus (Compositae), an edible mountainous vegetable, are traditionally used to treat hepatitis and hemorrhage and are known to have diuretic action. The aqueous ethanolic extract of this plant was selected in our screening experiment using the peroxynitrite ($ONO_2^-$)-scavenging assay, and the present study was performed to qualitatively and quantitatively identify the active compounds from S. brachyotus and validate the present high-permormance liquid chromatography (HPLC) coupled with ultraviolet absorption detection method based on accuracy, precision and repeatability. Five phenolic substances including the main compound, luteolin $7-O-{\beta}-D$-glucuronopyranoside, as well as chlorogenic acid, luteolin 7-O-rutinoside, luteolin $7-O-{\beta}-D$-glucopyranoside, and luteolin, were found in the aqueous ethanolic extract of S. brachyotus. In the HPLC validation experiment, the linearity of the four compounds was established by $R^2$ values of more than 0.999 within the test ranges, and the recovery rate ranged from 98.2 - 105.3%. Luteolin 7-O-glucuronide was a predominant compound (143 mg/g of extract and 18.3 mg/g of the dry weight of plant material) with a potent peroxynitrite-scavenging effect ($IC_{50}$, $1.02{\pm}0.08{\mu}M$). Luteolin and its three glycosides together with chlorogenic acid were qualitatively and quantitatively determined using an HPLC method validated in the present study.

  • Plants from the genus Centaurea (C.) (Family: Asteraceae alt. Compositae), widely distributed in Asia, Europe and North America, have traditionally been used in the treatment of various ailments. As a part of our on-going studies on the plants from the genus C. for their phytochemistry and biological activities, extracts of the seeds of Turkish endemic C. species, C. bornmuelleri and C. huber-morathii, were tested for their cytotoxicity towards the CaCo2 colon cancer cell line as well as for the toxicity towards the brine shrimps, using the MTT and the brine shrimp lethality assays, respectively. Among the extracts, the MeOH extract of these plants showed significant toxicity towards the brine shrimps ($LD_{50}=55.2{\times}10^{-2}\;and\;42.4{\times}10^{-2}mg/ml$, respectively). The MeOH extract of both C. species also inhibited the growth of CaCo2 colon cancer cells in the MTT assay ($IC_{50}$=29.9 and 33.0 g/ml, respectively). As the most prominent activities in both assays were observed with the MeOH extracts, it can be assumed that the compound(s) responsible for these activities are polar in nature.
