Determination of Gallium by differential Pulse Adsorptive Stripping Voltammetry (펄스차이 흡착법김전압전류법에 의한 갈륨 정량)

  • A very sensitive adsorptive stripping voltammetric method is studied on the gallium-morin complex at a hanging mercury drop electrode (HMDE) in 0.1 M acetate buffer solution. The effects of various analytical conditions are discussed on the reduction peak current of the adsorbed complex on the suface of HMDE. Interferences by other trace metals and surfactant are also discussed. Detection limit is 1.7 nM of gallium with 60 seconds deposition time, and the relative standard deviation (n = 7) at 4 ${\mu}$g/l gallium is 2.8%.

  • The hyperthermophilic archaebacterium Thermococcus profundus Isolated from a deep-sea hydrothermal vent system, produced several amylolytic enzymes such as extracellular amylase and pullulanase, intracellular a-1,4-91ucosidase in respone to the presence of complex carbohydrates In the growth medium. This strain showed high activities on 0.5% maltose than on complex carbohydrates One of the amylases was partially purified by ammonium sulfate precipitation, DEAE-Toyopearl chromatography. The amylase exhibited maximal activity at pH 5.5 and 80$^{\circ}C$, and was stable in the range of pH 5.5 to 9.5 and up to 80$^{\circ}C$ for 30 min. The enzyme activity was no dependence on Ca2+ and not inhibited by detergents. The amylase hydrolyzed soluble starch, amylose, amylopectin and glycogen to produce maltose and maltotriose with trace amounts of glucose, but not pullulan and ${\alpha}$-, ${\beta}$-, ${\gamma}$-cyclodextrin. Malto-oligosaccharides ranging from maltotetraose to maltoheptaose were hydrolyzed in an endo fashion.

  • Epoxidation of olefins has been studied using iodosylbenzene (PhIO) as the terminal oxidant and binuclear and mononuclear complexes of $Mn^{2+}$, $Co^{2+}$, and $Cu^{2+}$ as catalysts. Epoxides were the predominant products with trace amounts of allylic oxidation products, and the metal complexes were found to be effective catalysts in the epoxidation reactions. The reactivity of binuclear copper complexes was greater than that of the mononuclear copper complexes, whereas the binuclear and mononuclear complexes of $Mn^{2+}$ and $Co^{2+}$ showed similar reactivities. The nature of the ligands bound to copper did not influence the reactivity of the binuclear copper complexes so long as copper ions were held in close proximity. A metal-iodosylbenzene complex, such as suggested previously for Lewis acidic metal complex-catalyzed epoxidation by iodosylbenzene, is proposed as the active epoxidizing species. Some mechanistic aspects are discussed as well.

  • Reactions of alkaline metal carbonates with sulfur are investigated in detail. The evolution of CO and a trace of $SO_2$ were observed in the course of reaction with major component of polysulfides. Some evidences that the reaction proceeds with breaking of terminal sulfur-sulfur bond in the sulfur polymer, and forming CO, $SO_2$ and polysulfide are presented. Detection of CTX-M and Clonal Analyses using MLST in Cefotaxime Resistant $Escherichia$ $coli$ Isolated from the Han-River, Korea

  • Bacteria resistant to various antibiotics have recently become an issue of the utmost importance. Resistant strains are not uncommon, even in municipal drinking water sources. The health threat posed by resistant, pathogenic bacteria has serious ramifications for both public health and agriculture. In this study, we isolated antibiotic resistant bacteria from water samples from the Han River, Korea, which is contaminated by the wastewater from many industrial complexes, hospitals, agricultural and animal husbandry estates, and from wastewater treatment facilities. We determined the degrees of resistance to various antibiotics exhibited by the isolated strains. The similarities between the isolated $E.$ $coli$ strains were examined, using the pulsed field gel electrophoresis and multilocus sequence typing, in order to trace their origins and to explore the syntechnic adaptations and pathogenicity of the various strains and relate these to their genetic sequence. A total of 25 $E.$ $coli$ strains were isolated from six stations along the Han River. All the 25 strains exhibited resistance to ampicillin. We also investigated resistance to amoxicillin, clavulanic acid, cefazolin, cofoxitin, cefotaxime, cefpodoxime, ceftriaxone, cefepime, nalidixic acid, aztreonam, ciprofloxacin, streptomycin, gentamicin, chloramphenicol and imipenem. Based on the ESBL detection, 14 strains belonged to the ESBL producing strains. The number of the clonal complex producing strains was 5 among the 14 isolated strains. The 5 strains were included in the 168, 23, 38, 469, 156 clonal complex, respectively. The rest 9 strains were not included in the clonal complex, but showed independent STs.

Characteristics of Respirable Particulate Matter from the Third and Fourth Industrial Complex Area of Daejeon City Using Instrumental Neutron Activation Analysis (기기중성자방사화분석을 이용한 대전 3, 4 공단지역의 호흡성입자의 특성)

  • Instrumental neutron activation analysis was used to measure the concentrations of 27 elements associated with respirable particulate matter (PM10) that were collected from the Third and Fourth Industrial Complex Area of Daejeon City. The distribution patterns of elements were clearly distinguished with their concentrations varying across more than four orders of magnitude. The mean for Al were recorded to be the highest value of 1,527 $ng/m^3$, while that for In showed the lowest value of 0.1 $ng/m^3$. If compared in terms of enrichment factors, it was found that certain elements (e.g., As, Br, Cl, Cr, I, In, Sb, Se, and Zn) are enriched in PM10 samples of the study site. The results of factor analysis indicated six factors with statistical significance, which may exert dominant controls on regulating the elemental concentration levels in the study area.

The adsorption phenomena were observed by differential-pulse voltammetry. The effects of various analytical conditions were discussed on the reduction peak current of the adsorbed complex on the surface of HMDE. Interferences by other trace metals and surfactant were also discussed. Detection limit was 2.6 nM of indium after 90 second deposition time, and the relative standard deviation (n = 7) at 4TEX>${\mu}g$/l indium was 2.0%.

Determination of Heparin Using Norfloxacin-cerium Complex as a Fluorescence Probe by Spectrofluorimetry

  • A simple, rapid, practical and sensitive spectofluorimetric method was developed for the determination of trace amount of heparin (Hep). Under the Optimum conditions, we studied the interaction between NFLX-Ce$^{3+}$-Hep complex by using absorption and fluorescence spectra. It was observed that Hep remarkably enhance the fluorescence intensity of the NFLX-Ce$^{3+}$ complex at ${\lambda}$= 356 nm in the buffer solution of pH = 7.60 and the enhancement effect is shown to relate with the concentration of Hep. The linear range and detection limit for the determination of Hep was obtained. By the Rosenthal graphic method, the association constant (K) and binding numbers (N) of Hep with probe were investigated. This method is relatively free of interference from coexisting substances and successfully applied for the determination of heparin in heparin sodium injection samples. A suitable mechanism of fluorescence enhancement between NFLX-Ce$^{3+}$ and the NFLX-Ce$^{3+}$-Hep systems were proposed and discussed.

  • To produce 4N grade high-purity silica powder from natural ore, the mineralogical characteristics of natural silica ore were investigated and their effects on the purification process were revealed. The Chinese silica mineral ore used was composed of iron and aluminum as main impurities and calcium, magnesium, potassium, sodium, and titanium as trace impurities; these trace impurities generally exist as either single oxides or complex oxides. It was confirmed that liberation and acidic washing of the impurities were highly dependent on the particle size of the ground silica ore and on its mineralogical characteristics such as the distribution and phases of existing impurities. It is suggested that appropriate size reduction of silica ore should be realized for optimized purification according to the origin of the natural silica ore. A single step purification process, the mechano-chemical washing (MCW) process, was proposed and verified in comparison with the conventional multi step washing process.

  • A simulation program SIMULTSONIC is under development at CNDE to help determine and/or help optimize ultrasonic probe locations for inspection of complex components. SIMULTSONIC provides a ray-trace based assessment for immersion and contact modes of inspection. The code written in Visual C++ operating in Microsoft Windows environment provides an interactive user interface. In this paper, a description of the various features of SIMULTSONIC is given followed by examples illustrating the capability of SIMULTSONIC to deal with inspection of canonical objects such as pipes. In particular, the use of SIMULTSONIC in the inspection of very thin-walled pipes (with 450 urn wall thickness) is described. Ray trace based assessment was done using SIMULTSONIC to determine the standoff distance and the angle of oblique incidence for an immersion mode focused transducer. A 3-cycle Hanning window pulse was chosen for simulations. Experiments were carried out to validate the simulations. The A-scans and the associated B-Scan images obtained through simulations show good correlation with experimental results, both with the arrival time of the signal as well as with the signal amplitudes.
