Parasitic protozoans of fish, such as Chilodonella, Costia, Trichodina, Ichthyophthirius are effectively eradicated by treating with a mixture of $50\~60\;ppm$ formalin, 0.2 ppm malachite green and 0.6 ppm dipterex in water recycling aquariums. In this case any histopathological changes in the gill of fish were not observed. The parasites began to disappear 6 hours after treatment. Upon increased concentrations of formalin of 130 to 140 ppm, a significant change on the epithelium of gills was observed in $24\~48\;hours $ after treatment. In this case gill epithelium degenerated and necrosis of the gill lamella supporting cells follows.

Environmental specimen bank (ESB) is a new tool to assess ecosystem in environmental impact assessment (EIA). ESB looks at changes in the concentration of pollutants in human and environmental specimens over long periods of time through retrospective analysis of archived samples. Korea started to design its National Environmental Specimen Bank (NESB) in 2007 and planned to launch an operational pilot project by 2010. NESB prepares five Standard Operating Procedures (SOPs) of shoots of Red and Korean pine, leaves of Mongolian oak, Pigeon's egg, and Common carp's muscle out of 14 planned specimens in 2009. ESB contributes to monitoring the effectiveness of EIA projects and policies by providing a time capsule through ecosystem assessment of representative species. This study reviews ecosystem assessment in EIA and the ESB establishment in Korea and probes NESB applications in ecosystem assessment.
To investigate the innate immune response involved in early stage of anti-viral defence, carps were injected with UV-inactivated spring viraemia of carp virus (SVCV), poly inosinic:cytidylic acid (Poly I:C) and concanavalin A (Con A), respectively and examined lysozyme activity, serum complement activity and chemiluminescent (CL) response of leucocytes isolated from head kidney at 3 days post-injection. There was no significant difference in plasma lysozyme activities among all experimental groups. However, lysozyme activities of head kidney in the groups injected with antiviral activity inducers were significantly higher than those of the control injected with physiological saline. Bactericidal activities of serum of the groups injected with antiviral activity inducers were not significantly different from control group. However, the CL responses were significantly higher at lower dose of Poly I:C and Con A, whilst dose-dependent increase was shown in UV-inactivated SVCV-injected group. In the challenge test with 1×104 TCID50/fish of SVCV at 4 days post-injection, UV-inactivated SVCV- and Poly I:C-injected groups showed higher relative percent survival (RPS) than Con A-injected group. Furthermore, strong protection was observed in the group injected higher dose of Poly I:C although showed lower activities in lysozyme and CL response. These results suggested that Poly I:C might stimulate other factors belonging to non-specific immune system have induced protective immunity against the SVCV challenged.

It has been reported that various anti-oxidant substances stimulate non-specific immune responses in fishes. In this study it was examined whether N-acetylcysteine (NAC), an anti-oxidant glutathione (GSH) precursor, can modulate non-specific immune parameters in 8 different fishes. NAC was intraperitoneally administered at 10 mg/kg to catfish (Silurus asotus), loach (Misgurnus mizolepis), common carp (Cyprinus carpio), crucian carp (Carassius carassius), eel (Anguilla japonica), snakehead (Channa argus), tilapia (Oreochromis niloticus) and mullet (Mugil cephalus). Forty-eight hours later, chemiluminescence (CL) response of head-kidney leukocytes and serum lysozyme activity were assessed. In all fishes except crusian carp and loach, CL responses were amplified by NAC. Lysozyme activity was increased by NAC in all fish species but not in tilapia. This result suggests that NAC stimulates non-specific immune responses in various species, and that such effects may have beneficial significance in aquaculture for practical utilization.
This is a comprehensive study for considering the effective treatment and control program of bacterial disease occurring in common carp, israel carp, color carp, crucian carp, eel and tilapia by clarifying the causes, mechanism of infection and onset and the diagnostic criteria. As a first step, the authors investigated the external views, gross and histopathologic findings of diseased fish using 450 infected fishes obtained from various farmer of Korea. This infection was characterized by hyperemia, hemorrhage and swelling of body surface and fins, congestion of liver, spleen, kidney, inflammation of intestine, hemorrhagic inflammation of various tissues, and necrosis and ulcer of various tissues were accompanied in serious cases. Bacteriologically, Aeromonas hydrophila and Edwardsiella tarda were isoiated from these fishes. Particularly in the regular check on 222 eels, 177 strains were isolated as 29.94% of Aeromonas hydrophila, 48.58% of Edwardsiella tarda and 21.47% of Flexibacter columnaris. Hexibacter columnaris was isolated from corroded gill of eels. The identical disease was occurred by innoculating the isolated Aeromonas hydrophila and Edwardsiella tarda and the identical strains were isolated from infected experimental fishes. The eels which were diagnosed Aeromonas disease from Kwangju, Pusan accompanied hemorrhage, swelling of body surface and fins, inflammation of stomach and intestine containing mucous fluids mixed with the pathogens. Color carp and crucian carp which were innoculated with the isolated 5 strins of Aeromomas hydrorphil died within 3 or 4 days accompanying with the characteristics of Aeromonas disease. Edward disease was characterized by abscesses of body surface, pus formation with concentration on phagocytes. The size of absecsses increased with progression elf disease. There were also various abscesses at internal organ and white nodules appeared in kidney. Histologically, various progressive granuloma were examined without inflammation of intestine. Columnaris disease of eels showed no hemorrhage except slight white body color. In autopsy, most of internal organs appeared normal and there were no septic odors. The only character was corrosion of gills. In order to treat these bacterial diseases, infected fishes must bathe in 20ppm chloramphenicol or kanamycin solution for 1 hour. Besides, medication program in oral ingestion of 75mg/kg chloramphenicol per day continuing for 5 to 7 days. After injecting the formalin treated Aermonas hydrophila antigen into carp, relatively high agglutination titer showed between 3 weeks and 6 weeks. Though this titer decreased from that time, it was continued for 18 weeks. In the case of injecting the formalin treated Edwardsiella tarda antigen into tilapia, the titer also increased. But tilapia which were immersed in the suspension fluid of the formalin treated Edwardsiella tarda showed no increase of the titer.
In order to elucidate the taste compounds of wild common carp, Cyprinus carpio, and Korean snakehead, Channa argus, organic acids, sugars ana minerals were analyzed as a part of the study on the taste compounds of fresh-water fishes. Among organic acids, the contents of succinic acid, butyric acid, propionic acid and valeric acid were fairly high, while those of oxalic acid, fumaric acid, maleic acid, tartaric acid and citric acid were trace. As for the sugars, glucose was found to be the most predominant monosaccharide of the species. Extremely small amounts of fructose and arabinose were also detected and inositol and ribose were trace in content. Among minerals, $K^{+},\;Na^{+},\;PO_{4}^{3-}\;and\;Cl^{-}$ were found to be the major ions and small amounts of $Ca^{2+}\;and\;Mg^{2+}$ were detected.
To assess the adverse effects of suspended solids on fishes, the hatching rate of embryo, and survival and growth of larvae were measured with common fish species of river such as crusian carp (Carassius auratus), common carp (Cyprinus carpio) and medaka (Oryzias latipes). Bioassay on hatching rate of embryo was conducted with a range of turbidity as 10 levels from 0.2 to 8,080 NTU. Another bioassay on 4-day larval survival and 21-day larval growth were also conducted with a range of turbidity as 8 levels from 0.1 to 8,260 NTU, and 6 levels from 0.7 to 2,030 NTU, respectively. The hatching rate of C. auratus was not significantly different from that of control at turbidity below 4,040 NTU, whereas it decreased when turbidity was 8,080 NTU (p<0.05). The hatching rate of C. carpio was not affected by concentration of suspended solids. For O. latipes, the hatching rate significantly decreased from 254 NTU (p<0.001) and it was zero when turbidity was 8,080 NTU. The 4-day survival of larvae of all 3 species was not affected by turbidity, while the larval growth of all 3 species was significantly affected. The turbidity effect on the growth of C. auratus and C. carpio was observed from 7 days after the exposure at turbidity level of 145, 143 NTU, respectively. The turbidity effect on the growth of O. latipes was observed from 14 days after the exposure at turbidity level of 254 NTU. The results of this study will provide the basic information for the derivation of water quality criteria on suspended solids for the protection of aquatic ecosystem and the quantitative ecological risk assessment of freshwater environment.
This experiment was carried out to compare the change of muscle protein compositions, amino acid compositions of muscle protein, and free amino acid compositions by age of the Israeli strain of common carp, Cyprinus carpio nudus. Protein compositions of the muscle were: sarcoplasmic protein $25.8-27.2\%$, myofibrillar protein $62.3-56.2\%$, residual intracellular protein $9.6-13.2\%$ and stroma $2.3-2.9\%$. In between 1 year and 3 years, there were differences as follows; myofibrillar protein in 1 year was much than 3 years, and other proteins in 3 years were much than 1 year. By SDS-polyacrylamide gel electrophoresis, sarcoplasmic protein of the samples in 1 year an 3 years were composed of 11 subunits and 10 subunits, respectively. And appeared 210,000 dalton component in 1 year but did not appeared in 3 years. Myofibrillar protein was composed of 23 subunits in both 1 year and 3 years but the differences of subunits by age were not observed. No differences were observed by age in the composition of myofibrillar protein and residual intracellular protein. Amino acid composition of muscle protein in both 1 year and 3 years were no differences to each other, but the contents of glutamic acid, aspartic acid, lysine were higher than other amino acids. The amount of total free amino acid in 1 year was much than in 3 years.
The phototaxis of common carp Cyprinus carpio was studied tinder laboratory conditions to find out distributional pattern of the fish under light gradient and to find the light intensity which causes the maximum gathering rate. The optimum light intensities were determined, The fish tended to escape from tile light source when the light intensity was stronger or weaker than a certain optimum value. The mean illumination intensity which caused the maximum gathering rate was 3.813lux (2.99-4.76 lux) in the daytime and 6.292 lux (5.0-7.89 lux) at night. The gathering rate of the fish could be divided into two types: reflex gathering rate and equilibrium gathering rate. The reflex gathering rate appeared quickly soon after lighting and gradually changed as time elapsed, while the equilibrium gathering rate was almost always constant.
Natural-killer-cell-enhancing factor (NKEF) belongs to the newly defined peroxiredoxin (Prx) family. It was originally isolated from human erythroid cells. The black rockfish NKEF cDNA was identified through the expressed sequence tag (EST) analysis of PBLs libraries. The full-length NKEF cDNA was 1433 bp long and contained an open reading frame (ORF) of 594 bp that encoded 198 amino-acid residues. The 5' UTR had a length of 39 bp, and the 3’UTR 800 bp. The deduced amino-acid sequence of the black rockfish had a density 93.4, 92.9, 87.8, 85.8, 84.8, 83.8, 80.3, 79.7, 77.2, and 75.2% that of the pufferfish, olive flounder, channel catfish, zebrafish, chicken, common carp, Myotis lucifugus, cattle, human PrxI, rat PrxI, human NKEF-A, and Xenopus tropicalis, respectively. The NKEF gene was expressed in all the tissues of the black rockfish. The RT-PCR indicated that the NKEF transcripts were predominantly in the spleen and gill, less dominantly in the PBLs, head kidney, trunk kidney, and liver, and least in the intestine and muscles. This is the first report on the existence of the NKEF-A gene in black rockfish.
