산성조건하에서 시판요구르트의 유산균 생존률과 ${\beta}-galactosidase$의 활성도 (Survival Rate of Lactic Acid Bacteria and the Change of ${\beta}-Galactosidase$ Activity in Commercial Yogurts Under the Acidic Conditions)

  • 국내에서 시판되는 요구르트(plain)의 이화학적 특성 및 산성조건하에서의 유산균의 생존률과 ${\beta}-galactosidase$의 활성도를 측정하였다. pH는 $3.71{\sim}4.08$범위, 적정산도는 $0.990{\sim}1.045%$, 점도는 $256{\sim}3,164\;cps$ 범위였으며, 유산균수는 $10^{8-9}\;cfu/ml$로 규정보다 높았다. 완충능은 요구르트(50 ml)의 고유 pH값에서 -2의 pH값까지 적정하는 데 소비된 1.0 M-HCl의 량으로 $3.58{\sim}4.33\;ml$였다. 산성조건하에서 유산균의 생존율은 pH 3.5에서는 2시간 동안 거의 변화가 없었으며, pH 2.5, 1.5에서는 각각 $3.5{\times}10^{-2}{\sim}3.6{\times}10^{-1}%,\;8.3{\times}10^{-5}{\sim}4.2{\times}10^{-3}%$ 범위였다. ${\beta}-galactosidase$활성은 120분 후에 pH 3.5에서는 큰 변화가 없었으며, pH 2.5, 1.5에서 ${\beta}-galactosidase$의 잔존활성은 각각 $9.4{\sim}36.2%.\;4.2{\sim}19.0%$ 범위로 나타났다. 이상의 결과로 요구르트를 섭취하였을 때 유산균은 위산에 노출되어 극히 그 일부만이 장내에 도달할 수 있을 것이라고 판단되며, ${\beta}-galactosidase$는 유산균의 세포벽이나 세포막 및 요구르트에 포함된 무지고형분의 보호를 받아 그 활성을 어느정도 유지할 수 있으리라 사료된다.

The Effect of Milk Protein on the Biological and Rheological Properties of Probiotic Capsules

  • Probiotics are often infused into functional foods or encapsulated in a supplement form to maintain a healthy balance between the gut microbiota and their host. Because there are milk-based functional foods such as yogurt and cheese on the market, it has been suggested that milk-based probiotics could be incorporated into skim milk proteins in a liquid capsule. Skim milk is mainly composed of casein and whey protein, which create a strong natural barrier and can be used to encapsulate probiotics. In this study, we compared the encapsulated probiotics prepared with milk-based concentrated cell mixtures using commercial probiotics. Probiotic capsules were emulsified with skim milk proteins using vegetable oil to form a double coating layer. The product was heat-stable when tested using a rheometer. The survival rate of the milk-based probiotic cells in the lower gastric environment with bile was significantly higher than commercial probiotics. Thus, milk-encapsulated probiotics exhibited greater efficacy in the host than other types of probiotics, suggesting that the former could be more viable with a longer shelf life under harsh conditions than other form of probiotics. Our findings suggested that, compared with other types of probiotics, milk-based probiotics may be a better choice for producers and consumers.

밤 용액의 젖산발효 (Lactic Acid Fermentation of Chestnut Broth)

  • 분변과 요구르트에서 10주의 젖산균을 분리하여 이중 6주는 Bifidobactria로 4주는 Lactobacilli로 동정하였다. 이들은 이용하여 8% 증숙밤용액을 발효시켰을 때 발효액의 산도는 요구르트보다는 낮았으나 두유를 비롯한 식물성 요구르트보다는 2배이상 높았다. 산도를 증가시키기 위해 Yeast extract 와 tryptone peptone를 첨가할 때 0.2%와 0.4%의농도에서 가장 높았다. 포도당을 0.5-0.8% 범위의 첨가하였을 때 lactobacilli에 의한 발효액에서는 산생성이 촉진되었으나. bifidobacteria 에서는 그렇지 않았다. 시험된 과일과 채소즙 중에서는 당근즙의 첨가가 가장 큰 촉진 효과를 나타내었다. 25% 밤용액에서는 고지추출액에 의한 가수분해는 젖산생성을 크게 촉진하였으나 가수분해하지 않은 8% 밤용액에서는 젖산생성량과 비교하였을 때 산생성 순증가량은 그다지 크지 않았다.

새싹인삼 분말 열수추출물의 유식품 적용 연구 (Suitability of Hot Water Extract from Panax ginseng Sprout Powder as a Dairy Additive)

  • This study aimed to determine the effect of extraction temperature and time on the antioxidant activity of hot water extract from Panax ginseng sprout powder and to evaluate the suitability of this extract for use in dairy products. Water-soluble fractions of commercial Panax ginseng sprout powder were obtained by hot water extraction at 25, 60, or 80℃ for 0.5, 2, 12, or 24 h. The antioxidant activity of each extract was evaluated by measuring its free radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS). DPPH and ABTS free radical scavenging activity increased with extraction temperature from 25 to 80℃. At 80℃, increasing the extraction time from 0.5 to 2 h led to increases in DPPH and ABTS free radical scavenging activity. Thus, the extract obtained under 2 h at 80℃ was selected for addition to milk and yogurt. After 16 days of storage, there were no significant changes in the pH of the milk or the antioxidant activity of the extract. With regard to yogurt fermentation, adding the extract did not affect the pH or the number of viable lactic acid bacteria. In conclusion, hot water extract from Panax ginseng sprout powder can be added to dairy products to enhance antioxidant activity.

우유발효에 이용되는 Starter Culture와 그 특성 (Starter Clutures for Milk Fermentation and Their Characteristics)

  • All over the world there is an increasing consumer awareness of the potential influence of various foodstuffs on our health. Today dairy products are expected to be more just food. They have to taste well, appeal and give pleasure, provide of well-being, provide specific health benefits and prevent disease. This paper reviews the different types of fermented milks and their microflora and includes recent work on yogurt, soft cheese and buttermilks, kefir and koumiss. There is considerable interest in the new health promoting products which are now available. Meanwhile during the last decade a new generation of fermented milk products containing selected intestinal bacteria has been introduced to the markets. These are discussed in the light of some recent findings on the ability to lower the blood cholesterol concentration and stimulate the immune response and also describes some fermented milk products available, selection criteria for commercial starter cultures.

시판 드링크 및 액상 요구르트 제품중 Lactobacilli 및 Bifidobacteria의 내산성 연구 (Gastric Acid Resistance of Lactobacilli and Bifidobacteria in Commercial Drink and Liquid Yogurts)

  • 국내 시판 19종의 드링크 및 18종의 액상 요구르트 제품에 함유된 2종의 유산균인 Lactobacilli 및 Bifidobacteria의 내산성을 인공위액(pH 1.5) 조건에서 비교 고찰되었다. 드링크 및 액상 요구르트 제품중 초기 Lactobacilli의 생존율은 각각 $10^8~10^{10}$ cfu/ml 및 $10^6~10^{10}$cfu/ml 범위 내에 있었다. 한편 드링크 요구르트 중 초기 Bifidobacteria의 생존율 $10^8~10^{10}$cfu/ml 이었다. 요구르트 제품중 Lactobacilli 및 Bifidobacteria의 생존율은 인공위액에 노출 시간이 증가함에 따라 점차 감소하는 경향을 나타내었으며 120분 노출시에 제품들간에 큰 차이를 나타내었다. (<$10^3~10^6$cfu/ml),캡슐을 함유한 요구르트 제품의 경우, 캡슐을 제외한 Mix는 인공위액에 30분에서 120분 노출시에 약$10^5$cfu/ml이상의 생존율을 나타내었으며 타사 제품과 유사한 거동을 보였다. 그러나 Bifidobacteria를 함유한 캡슐요구르트 제품의 경우 인공위액에 약 120문 노출시켰으때 타사 제품보다 생존율이 약10~737배 이상 증가하였다. 즉 Bifidobacteria의 내산성은 코팅물질을 사용한 캡슐로 봉입하였을 때 증가됨을 알 수 있었다.

성인으로부터 분리된 Lactobacillus casei CU2604의 우유배지에서의 생장 특성 및 생리적 특성 (Growth Characteristics and Physiological Properties in Milk of Lactobacillus casei CU2604 Isolated from Adult Feces)

  • 이 연구는 성인분변으로부터 발효유 제조에 이용되는 probiotic로서의 우수한 능력을 지닌 균주를 선발하는데 목적을 두었으며 선택배지에서의 성장, 그람 양성, 현미경 관찰을 통해서 간균인 총 204개의 sample 중에서 10% skim milk에서 24시간 배양 후 커드 형성 여부에 따라 19개를 선발하였고, 2차 계대하여 48시간 후에 커드 형성 여부를 통해 6개를 선발하였다. 그 중에서 산 생성 능력이 우수한 균주를 3개 선발하여 원유로부터 분리한 L. casei MCL과 L. casei YIT 9029의 특성을 비교하였다. 선발된 균주는 그람 양성이고, 간균이며, 당 발효 실험과 16S rDNA 분석결과 Lactobacillus casei로 판명되어 L. casei CU2604, L. casei CU3204, L. casei LC8로 명명하였다. 이중 L. casei YIT 9029과 동일한 당 발효패턴을 보인 L. casei CU2604는 pH 변화와 생균수의 변화가 L. casei YIT 9029와 비슷한 양상을 보이고 PFGE 패턴이 같았다. 그러나 세포벽 지방산조성 분석결과를 통하여 L. casei YIT 9029와는 다른 균주임을 알 수 있었으며, 담즙내성, pH내성은 더 우수한 결과를 보였고, 또한 L. casei CU2604는 병원성균에 대하여 우수한 생육억제 능력을 나타내었다. 이러한 결과를 토대로 L. casei CU2604는 상업용 균주에 못지 않은 스타터로서의 특징을 가지고 있는 것으로 판단되며, 이 균주의 probiotics 특성에 대한 추가적인 연구가 진행 중에 있다.

Lactobacillus acidophilus의 산업적 이용과 CLA 생성 (Production of Conjugated Linoleic Acid by Lactobacillus acidophilus and Their Industrial Application)

  • Application of lactic acid bacteria in the markets are divided into four categories: dairy industry, health food industry, animal feed industry and pharmaceutical industries. Recently, Lactobacillus acidophilus have been used in the food industry and have obtained great attention as key cultures for health benefit. Since commercial application of L. acidophilus has become a common practice, characterization of these cultures were made. Futhermore, the strains selected should produce a final dairy product possessing good taste and acceptable body and texture, a selection step that cannot be achieved unless the product is actually manufactured. Conjugated linoleic acid (CLA) have been recognized as antioxidants, cancer inhibitors, cholesterol depressing agents, and growth promoting factors. Food products from ruminants, particularly dairy products, are the major dietary source of CLA f3r humans. The CLA content in yogurt or cheese can be increased by action of the starter cultures. The finding of the production of CLA by food starter culture opens interesting perspectives far the future in producing fermented products enriched in CLA.

Studies of the Microbial and Physical Properties of Oriental Style Dairy Product Kou Woan Lao with Probiotics

  • The objective of this research was to combine the physiological functionality of probiotics (Lactobacillus acidophilus and Bifidobacterium longum) and the milk-clotting activity of culture filtrate from lao-chao to develop a new dairy product which was different from the commercial yogurt. Rhizopus javanicus and Saccharomyces cerevisiae were chosen as a mold and yeast starter for production of culture filtrate. The study results indicated that both probiotic counts increased with incubation time and maintained $10^7$-$10^8$ CFU/ml after 6 h incubation with 10-30% culture filtrates. By contrast, samples with 40% culture filtrates inhibited the growth of L. acidophilus and B. longum. The more culture filtrates were added, the lower titratable acidities and higher pH values in Kou Woan Lao were detected after 36 h fermentation. No significant differences (p>0.05) were found for both L. acidophilus and B. longum, when grown in differing concentrations of skim milk powders. Storage results showed both L. acidophilus and B. longum exhibited excellent stability for 14 days at $4^{\circ}C$ in the Kou Woan Lao.

유제품 섭취와 당뇨 예방 (Dairy Products Intake and Managing Diabetes)

  • Milk intake is widely recommended for healthy diet, not only for bone growth and maintenance, but also as a protein, calcium and magnesium sources as part of an adequate diet. Many research suggest that milk and dairy products are associated with a lower risk of type 2 diabetes mellitus (T2DM). Milk and dairy products are low Glycemic index (GI) and Glycemic load (GL) foods. The GI and GL are useful tools to choose foods to help control blood glucose levels in people with diabetes. The GI and GL of milk are 32~42 and 4~5, respectively, and which are about 1/2 and 1/5 of boiled rice. The mechanisms underlying the effects of dairy on T2DM development includes the calcium and vitamin D content in dairy foods and the possible positive effect of high milk and calcium intake on weight control. The role of dairy products on reducing the risk of diabetes can be inferred from the reports that lower serum IGF-1 levels were positively associated with diabetes and the girls with low milk intake had significantly lower IGF-1. Accumulating data from both patients and animal models suggest that microbial ecosystems associated with the human body, especially the gut microbiota, may be associated with several important diseases, such as inflammatory bowel disease, obesity, diabetes and cardiovascular disease. It was thought that fermented milk containing lots of probiotics can be useful for controling blood glucose levels and preventing complication of diabetes, but sucrose in commercial yogurt should be substituted. There are some reports of oligosaccharide, xylitol, and stevia as a potentially useful sweetener in the diabetic diet.
