In this study, we attempted to obtain an active ingredient that inhibits the production of matrix metalloproteinase-1 (MMP-1) that breaks down collagen in human skin fibroblasts. More than 50 plant extracts were screened, and Lonicera japonica was selected for this study. The stem of L. japonica was extracted with 70% ethanol and fractions with solvents in the order of hexane, ethyl acetate, and butanol. MMP-1 production were significantly inhibited at the concentration of 50 ㎍/mL of the ethyl acetate layer and 200 ㎍/mL of the butanol layer. To get a fraction containing all of these effective components, 80% ethanol fraction (LJ F80) was obtained through HP20 resin column chromatography. The reference substance, loganin and LJ F80 inhibited dose-dependently MMP-1 production. At the same concentration, LJ F80 showed a higher inhibitory effect than loganin. The stability of this fraction was analyzed with HPLC while kept storing at 4 ℃, room temperature, and 40 ℃, for 16 week. The stability was maintained as ± 10% of initial value with reference loganin. Therefore, it is thought that LJ F80 of L. japonica may be used to improve wrinkles.

Lateral torsional buckling causessafety accidentssuch as collapse accidents during erection. Therefore, anaccurate safety designshould be conducted. Lateral torsional buckling canbe prevented by reinforcing the end orreducing the unbraced length. The method ofreducing the unbraced length by installing a crossframe has high material and installation costs and low maintenance performance. In addition, structuralsafety may be deteriorated due to cracks. The end reinforcement method using Concrete Filled Half Pipe Stiffeneris a method ofreinforcing the end of a plate girder using a stiffenerin the form of a semi-circular column. This method increasesthewarping strength ofthe girder and increasesthe lateral torsional buckling strength. In thisstudy, the effect ofincreasing the warping strengthof plate girders with concrete filled half pipe stiffeners was confirmed. To verify the effect, the results ofthe designequationand the finite element analysis were compared and verified through a experiment. As a result, the plate girderwithCFHPS increased thewarping strengthand confirmed that the lateral torsional buckling strength was increased.

In this study, a series of experiments were conducted using a standard solution containing ${\alpha}$ and ${\beta}$-endosulfan to follow the removal effect of residual pesticides on soil and aqueous solution. An analytical method for residual pesticides was established by a gas chromatography equipped Ultra II[$(30m{\times}0.25mm(ID){\times}0.25{\mu}m$] capillary column and a ${\mu}$-electron capture detector(${\mu}$-ECD). Recovery rates of residual pesticides for soil samples were 96-100%. The amount of ${\alpha}$ and ${\beta}$-endosulfan that was spread in the soil was checked for various period of time. It indicated that the amount was reduced to 73 and 61%, respectively. When the water spread amount increased from 10 to 100 mL, ${\alpha}$-endosulfan was eliminated from 45 to 85% and while ${\beta}$-endosulfan from 44 to 88%. Removal rates of ${\alpha}$-endosulfan and ${\beta}$-endosulfan were 99% and 98% respectively within 30 minutes. It was assumed that the organic salts and strong alkali elements contained in the pesticide degradator hydrolyzed the residual pesticide.

Coinjection of alcohol and air or alcohol flooding only were evaluated with 3-D soil tank for removal of nonaqueous phase liquid (NAPL) contaminant from soil. 70%-ethanol and 40%-isopropanol were used for non-NAPL-swelling alcohol and NAPL-swelling alcohol, respectively. 729 ml-benzene was placed in the 37 liter soil tank. Alcohols were respectively injected from the injection well placed near the bottom of the tank and mobilized free phase NAPL and aqueous phases were then recovered from the extraction well placed in the upper part of the soil tank. Approximately 50% of removed NAPLs were free-phase in all experiments. The results were completely different to the previous soil column experiment results and also implied that alcohol properties did not affect the NAPL removal efficiency in the 3-D soil tank experiment. Air was also co-injected with alcohol to evaluate co-injection effects on NAPL removal. Enhanced NAPL removal effect of co-injection of 70%-ethanol and air was also found even in the 3-D soil tank evaluation. However, co-injection effect of 40%-iso-propanol and air was less apparent. This study determined that the most important parameter governing alcohol flooding for NAPL removal would be extraction capacity to recover NAPL and aqueous phase flowing in the soil. More researches are required for improving recovery efficiency of extraction well in real soil contamination conditions.

This study was conducted to compare STQ method, multi-residue method in Korean food code and QuEChERS method for validated selected and accuracy, reproducibility and efficiency. A total of 45 selected and targeted pesticides were the analyzed by GC and 5 of them were crops (apple, potato, green pepper, rice, soy bean). $R^2$ values were calculated in the standard calibration curve was over 0.990. Recovery tests were performed by three replications in two levels and the relative standard deviation of the repeated experiments was less than 30%. The average percentage of recoveries in the multi-residue method in Korean food code was 89.13%, QuEChERS method was 92.45% and STQ method was 85.28%. In addition, matrix effects in multi-residue method in Korean food code was 24.61%, QuEChERS method was 23.98% and STQ method showed 11.24%. The STQ method is easy and showed high clean-up effect in extracting the sample solution than the QuEChERS method and clean-up with C18, PLS, PSA cartridge columns. A large volume of the sample was injected in order to compensable for the problem, that occurred due to high detection limit in the analyser. When the STQ method was applied using a large volume injector, the standard calibration curve showed a higher linearity $R^2=0.990$, and method detection limit was 0.01 mg/kg. It showed an average recovery of 91.84% and the relative standard deviations of three replications repeated in two level process was less than 30% and had an average matrix effect of 17.90%.
The purpose of this study was to investigate the effect of methanolic extracts of graviola, Annona muricate leaves (AMME) on antioxidant activity and melanin production. First of all, DPPH radical and reducing power were performed to determine the antioxidant effect of AMME and organic solvent fractions. AMME and organic solvent fractions showed antioxidative activity in a concentration dependent manner. The ethyl acetate fraction of AMME among organic solvent fractions showed the highest antioxidant activity. Moreover, tyrosinase activity was performed to confirm the effect of organic fractions on melanin production. AMME, ethyl acetate, and hexane fractions increased tyrosinase activity a dose dependent manner. Next, the hexane fraction with the best effect on melanin synthesis in AMME organic solvent fraction was divided into 12 fractions by silica column chromatography. Among them, the fraction 7 and 8 showed the highest DPPH radical scavenging activity and reducing power. In addition, the fraction 7 and 8 at $64{\mu}g/ml$ showed melanin synthesis by 260% and 184%, respectively. Finally, the fraction 8 at $4{\mu}g/ml$ showed melanin synthesis by 34% in B16F1 cells. LC-MS analysis showed that fraction 7 and fraction 8 have a molecular weight of 617 and 619, respectively. FT-IR analysis showed that fractions 7 and 8 is similar to bis(2-hydroxyethly)dimerate. Above results suggest that graviola leaves extracts could be applicable to the development of natural antioxidants or hair cosmetics which are related to the promoting effect of melanin production.
Even though the anticarcinogenic effect of dietary factors especially beta - carotene has been reported by various investigators, the mechanism of the action of ${\beta}-carotene$ has not yet been identified. We carried out the present study to determine the possibilities of relative cancer risk related to dietary intake of vitamin A ( both ${\beta}-carotene$ and retinol ) and blood levels of vitamin A among Koreans. The subjects were divided into two groups; cancer patients and controls. Blood levels for ${\beta}-carotene$ and retinol were analyzed by alumina column chromatography and colorimetry. Dietary intake was examined by food profile and convenient method for evaluating nutritional status through recalling 10 years of food habits. The results obtained are as follows : 1 ) Calorie, protein, fat, and carbohydrate intakes of cancer patient were lower than those of control. Calorie and carbohydrate intakes showed no significant difference but protein and fat intakes were significantly lower in cancer patients. According to cancer sites, in stomach cancer only fat intake was significantly lower than that of control. In lung and larynx cancer calorie, protein, fat and carbohydrate intakes showed similar trend as in control. 2 ) Vitamin A intake of cancer patient was significantly lower than that of control. It was estimated that 83.6% of total Vitamin A intake were provided by ${\beta}-carotene$ for control and cancer patient respectively. 3 ) The mean intake of dietary ${\beta}-carotene$ in cancer patient was significantly lower than that in control ( $7002\;\mu}g/day$ versus $10326\;{\mu}g/day$ ) According to cancer sites in mean intake of dietary ${\beta}-carotene$ was significantly lower in all but stomach cancer compared with that of control. Lung and larynx cancer showed lowest ${\beta}-carotene$ intake with mean value of $5855{\mu}g/day$ and $5492{\mu}g/day$ respectively. 4 ) The mean intake of dietary retinol in cancer patient was significantly lower than that in control ( $245{\mu}g/day$ versus $338{\mu}g/day$ ), but the difference was not significant. 5 ) The relative risk of all cancers in the first (lowest) to the forth quartile level of ${\beta}-carotene$ consumption such as $0-5999{\mu}g/day$. $6000-8999{\mu}g/day$, $9000-11999{\mu}g/day$/ day and $12000-20000{\mu}g/day$ was 85 : 1.7 : 20 : 1.0. The relative risk of all cancers in the first (lowest) to the forth quartile level of retinol consumption, such as $0-299{\mu}g/day$, $300-599{\mu}g/day$, 600-899${\mu}g/day$, and $900-1200{\mu}g/day$, was 1.14 : 067 : 0.21 : 1.0. 6 ) The various food group consumption of cancer patient were significantly lower than those of control in green leafy vegetables, fruits, sea weeds, milk and cheese and eggs. But the Kimchie consumption in cancer patient was three fold higher than that of control ( $1840\;{\mu}g/day$ versus $562\;{\mu}g/day$ ) and in the stomach cancer, Kimchie consumption was the highest, ( $1890\;{\mu}g/day$) According to cancer sites, the consumption of green leafy vegetables was significantly lower in all but stomach cancer compared to control and other vegetables showed no difference between two. In lung and larynx cancer, green leafy vegetables consumption was lowest ( $6094{\mu}g/day$$5921{\mu}g/day$) and milk and cheese consumption was also( $5\;{\mu}g/day$ and $11{\mu}g/day$) 7 ) The recovery of ${\beta}-carotene$ from human serum by alumina column chromatography was $94.4{\pm}2.3%$. 8 ) Cancer patients showed significantly lower serum retinol ($56.4{\pm}18.1\:{\mu}g/100ml$ versus $72.2{\pm}21.8\:{\mu}g/100ml$) and ${\beta}-carotene$ ($48.9{\pm}33.8\:{\mu}g/100ml$ versus $72.2{\pm}42.6\:{\mu}g/100ml$) concentrations than in controls. 9 ) But breast cancer patients were not significantly different from controls in their serum retinol and ${\beta}-carotene$ concentrations.

We estimate atmospheric radiative heating effect of aerosols, based on AErosol RObotic NETwork (AERONET) and lidar observations and radiative transfer calculations. The column radiation model (CRM) is modified to ingest the AERONET measured variables (aerosol optical depth, single scattering albedo, and asymmetric parameter) and subsequently calculate the optical parameters at the 19 bands from the data obtained at four wavelengths. The aerosol radiative forcing at the surface and the top of the atmosphere, and atmospheric absorption on pollution (April 15, 2001) and dust (April 17~18, 2001) days are 3~4 times greater than those on clear-sky days (April 14 and 16, 2001). The atmospheric radiative heating rate (${\Delta}H$) and heating rate by aerosols (${\Delta}H_{aerosol}$) are estimated to be about $3\;K\;day^{-1}$ and $1{\sim}3\;K\;day^{-1}$ for pollution and dust aerosol layers. The sensitivity test showed that a 10% uncertainty in the single scattering albedo results in 30% uncertainties in aerosol radiative forcing at the surface and at the top of the atmosphere and 60% uncertainties in atmospheric forcing, thereby translated to about 35% uncertainties in ${\Delta}H$. This result suggests that atmospheric radiative heating is largely determined by the amount of light-absorbing aerosols.
This research was conducted to determine the effect of interruption layer for capillary rise on the sand based growing media when growing Kentucky bluegrass (Poa pratensis L.) on soil reclamation and saline water irrigation. Growing media profile consists of three layers as top soil of 30 cm, 20 cm of the interruption layer for capillary rise and 10 cm of reclaimed paddy soil. Growing media profile was packed in 30 cm diameter column pots. The top soil was a mixture of sand dredged up from Lake Bhunam Tae Ahn, Korea and peat at the ratio of 95:5 by volume. Bottom part of column was covered with plastic net and the pots were soaked into 5 cm depth saline water reservoir with salinity $3-5\;dS\;m^{-1}$. Kentucky bluegrass was established by sod and irrigated using $2\;dS\;m^{-1}$ saline water ($5.7\;mm\;day^{-1}$) in 3 days interval. The results showed that the largest accumulation of salt in the spring with electrical conductivity in saturated extract (ECe) of $5.4\;dS\;m^{-1}$ and sodium absorption ratio (SAR) 34.0 in growing media without the interruption layer for capillary rise and ECe of $4.6\;dS\;m^{-1}$ and SAR 8.24 at growing media using gravel as the interruption layer for capillary rise material. The interruption layer for capillary rise of gravel and coarse sand reduced the accumulation of Na by 16% and 25%, ECe by 7% and 13% in the growing media. Visual quality of Kentucky bluegrass was higher in growing media with the interruption layer for capillary rise of gravel than no interruption layer by 8.3 compared to 7.9 in rates. The interruption layer for capillary rise of gravel and coarse sand enhanced the visual quality by 4.1 and 4.0%, root length by 50 and 38%, and root dry weight by 35 and 17% of Kentucky bluegrass, and reduced the accumulation of Na by 16% and 25%, ECe by 7% and 13% in the growing media.
The objective of this research was to evaluate an sampling effect of fish species and individual number on sampling techniques of electrofishing and conventional capture methods of cast net and kick net in 38 stream sites sampled during June - September, 2005. For the study, sampling gears were categorized as three types of electrofishing method (EM), conventional sampling technique (CM), and the combined technique (CT) of the electrofishing and the conventional method to compare statistical differences. Major differences of species composition between the CM and EM method were found in some benthic species of Misgurnus mizolepis and Iksookimia koreensis along with lithophilic species of Pungtungia herzi. These species were predominated in the EM rather than the CM, indicating that conventional sampling can underestimate the abundance of benthic or lithophilic species. In contrast ,individual number of typical water column species such as Zacco platypus and Zacco temmincki were more sampled by the CM, so that these fish populations were community overestimated. Also, t-tests on the types of sampling gear from various watersheds of Chogang Stream, Yugu Stream, Daejeon Stream, and Gap Stream showed that total individual numbers and species number in each stream were significantly (t values = 2.806-6.896, p values<0.05, n = 5-14) greater in the CT than the CM. Similar statistical significance (p<1.001, n = (10-24) on sampling seasons were observed during the monsoon and postmonsoon. These results indicate that if the electrofishing is not added to the conventional gears, the abundance of fish population and community can be underestimated and some benthic or lithophilic species may be excluded from the analysis, resulting in overall errors including sampling, fish fauna, and final judgement of community abundance. Overall our results strongly suggest that new application of electrofishing method along with the conventional sampling gears reduce sampling bias on underestimation of the real fish populations and communities.
