한국도시여성의 얼굴색과 의복색과의 배색이미지에 관한 연구 (A Study on Coordination Image of Korean city woman's Face Color (5YR 7/3) and Clothes Colors)

  • The purpose of present study was to examine how each clothes colors on the basis of 5YR 7/3 face color affect clothes colors images as follows : (1) what general consciousness of clothes colors in, (2) how the impression of the harmony of 5YR 7/3 face color and clothes colors is, (3) when we divide clothes colors according to the property of colors- chromatic color and achromatic color, cool color.neutral color.warm color, in tone, in color colume- if there is the difference of visual evaluation, (4) image analysis of 45 clothes colors with the view of each kind of adjectives. The result of this study is as the following: 1. As a result of the analysis of general consciousness on clothes colors, when subjects chose clothes, they most considered colors and they also considered their face colors. They would choose the color of clothes, which were becoming to their having clothes colors or their face colors when they bought clothes. 2. The impressions of coordination of 5YR 7/3 face color and clothes colors consisted of three dimensions - evaluation, activity and harmony. 3. It was known that as a result of the analysis of visual evalutional differences according to dividing the clothes colors by property of colors, there were such notable differences that they might effect the coordination images of face color and clothes colors differently. 4. After arranging 45 clothes colors on the graphs in 17 adjectives, gethering them thogether in each dimension and as the result of the analysis in the evaluation dimension, estimation of yellow, light green column were low and that of achromatic colors were high. That is, it was known that the evalution dimension was concerned with hue of the color properties. In activity dimension, there were different image according to each adjectives. 이온교환크로마토그래피에서 라이소자임 분리에 미치는 pH와 온도 영향 (The Effect of pH and Temperature on Lysozyme Separation in Ion-exchange Chromatography)

  • 라이소자임은 용균 작용, 조직 회복 과정의 촉진 등의 작용을 하며 난백 중에 0.3% 함유되어 있다. 난백에서 라이소자임을 분리하는 방법으로 친화성 크로마토그래피, 이온교환크로마토그래피, 한외여과법 등이 있는데 이 중 이온교환크로마토그래피가 가장 많이 사용된다. 라이소자임을 양이온 젤이 충전된 유리 칼럼에서 분리 정제할 때 최적의 pH와 온도 조건을 찾는 것을 실험 목표로 하고, ASPEN Chromatography 전산 모사의 결과와 비교하였다. 실험에 사용되는 완충용액은 인산완충용액이었고 pH를 5~8로 변화를 주어 상온에서 실험하였고, 가장 분리가 잘 이루어진 pH에서 온도를 $5^{\circ}C$ 간격으로 $25{\sim}40^{\circ}C$로 변화시켜 실험하였다. RP-HPLC (Reversed phase High Performance Liquid Chromatography) 분석을 통해 라이소자임의 체류 시간을 확인하였고, OriginPro 8을 이용해 용출 단계에서 크로마토그램의 면적을 비교하여 라이소자임의 양을 정량분석하였다. 결과를 분석한 결과, pH 5일 때, 온도가 $25^{\circ}C$에서 가장 많은 양의 라이소자임이 분리되었다.

AOPP-g-AN 섬유이온교환체를 이용한 간수로부터 우라늄 이온 흡착특성에 관한 연구 (A Study of Adsorption Characteristics of Uranium ion Using Amidoximated PP-g-AN Fibrous ion-exchanger in Brine Water)

  • Acrylonitrile을 그라프트시킨 아미드옥심화 polypropylene 섬유이온교환체(AOPP-g-AN)의 충전 bed 높이를 변화시켜 간수 중 우라늄 이온 흡착 특성 및 흡착 공정 특성을 관찰하였다. 아미드옥심형 섬유이온교환체의 팽윤율은 그라프트율 100%, 물과 과산화수소 용매에 대하여 각각 8.54, 8.87 g/g을 나타내었다. 이온교환용량은 그라프트율이 증가함에 따라 증가하였으며, 그라프트율 100%에서 3.99 meq/g으로 최대를 나타내었다. 회분식 흡착에서 우라늄 흡착은 10분 이내에 초기 흡착 평형에 도달하며, 흡착 속도는 9.50 mg/min으로 나타났다. 최종 흡착 용량은 3.95 meq/g이었으며, pH에 따른 흡착 특성 변화는 관찰되지 않았다. 충전비에 따른 연속식 흡착시 흡착 용량은 L/D=1에서 3.92 meq/g으로 최대를 나타내었으며, L/D<2에서 편류 및 불균일 흡착에 의한 2단계 과정으로 파과가 나타남을 확인하였다. 실제 간수에 대한 우라늄 흡착 실험 결과, 흡착 용량 및 파과시간은 각각 3.63 meq/g, 26 min으로 모의용액과 비교 시 주목할만한 흡착능 저하는 관찰되지 않았다.

Triterpenoids from the Flower of Campsis grandiflora K. Schum. As Human Acyl-CoA: Cholesterol Acyltransferase Inhibitors

  • The flower of Campsis grandiflora K. Schum. Was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and H$_2$O. From the EtO Ac fraction, seven triterpenoids were isolated through the repeated silica gel, ODS column chromatographies and preparative HPLC. From the result of physico- chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as 3${\beta}$-hydroxyolean-12-en-28-oic acid (oleanolic acid, 1), 3${\beta}$-hydroxyurs-12-en-28-oic acid (ursolic acid, 2), 3${\beta}$-hydroxyurs-12-en-28-al (ursolic aldehyde, 3), 2${\alpha}$,3${\beta}$-dihydroxyolean-12-en-28-oic acid (maslinic acid, 4), 2${\alpha}$,3${\beta}$-dihydroxyurs-12-en-28-oic acid (corosolic acid, 5), 3${\beta}$,23-dihydroxyurs-12- en-28-oic acid (23-hydroxyursolic acid ,6) and 2${\alpha}$,3${\beta}$,23-trihydroxyolean-12-en-28- oic acid (arjunolic acid, 7). These teriterpenoids were isolated for the first time from this plant. Also, compounds 4, 5, 6, and 7 revealed relatively high hACAT-1 inhibitory activity with the value of 46.2${\pm}$1.1, 46.7${\pm}$0.9, 41.5${\pm}$1.3 and 60.8${\pm}$1.1% at the concentration of 100${\mu}$g/mL, respectively.

병포장 갓김치의 항산화 효과에 대한 연구 (Antioxidation Activities of Bottled Mustard Leaf Kimchi during Fermentation)

  • Mustard Leaf Kimchi (MLK) is a traditional fermented Korean vegetable food. This study was conducted to investigate the effects of part vacuum treatment on MLK packed in a glass bottle during fermentation. There have been a few previous studies that examined the chemical and microbial changes during MLK fermentation. However, the major object of this study was to investigate the antioxidative activities of vacuum treated MLK. In this study, the antioxidative activity of vacuum treated mustard leaf Kimchi (VM) and control mustard leaf Kimchi (CM) were examined. VM and CM were fermented at $5^{\circ}C$ for 8 weeks. A model system was designed to evaluate the antioxidative activity of crude chlorophylls and carotenoids (CCC) extracts from Mustard leaf Kimchi. The oxidative reaction of the linoleic acid mixture system at $30^{\circ}C$ in the dark was quantified determining the peroxide value and conjugated dienoic acid content. The effect of the CCC extracts on lipid peroxidation in a rat liver homogenate was examined. Formation of lipid peroxides was estimated by the TBA value, and the CCC extracts were found to inhibit the TBA value. Chlorophyll a and b, and carotenoids, Which are the major components in the CCC extracts of Kimchi were isolated on a DEAE-sepharose CL-6B and Sepharose CL-6B column and TLC. The effects of chlorophyll a and b, caroteins on linoleic acid autoxidation were measured by determining the peroxide value. In addition, their effects on free radical scavenging were investigated by DPPH. In this assay, chlorophyll a showed the greatest antioxidative activity followed by chlorophyll b, and carotenoids. MLK contains a sufficient content of chlorophyll a and b, and carotenoid which have strong antioxidative activities.

불꽃원자 흡수 분광법으로 활성탄소에 흡착된 메틸티몰 블루로 납(II)의 예비농축, 분리 및 측정 (Preconcentration, Separation and Determination of lead(II) with Methyl Thymol Blue Adsorbed on Activated Carbon Using Flame Atomic Absorption Spectrometry)

  • 예비농축 및 분리를 위한 온라인 시스템을 소개하였다. 이 방법은 활성탄소에 흡착된 메틸티몰 블루로 납(II)의 착체형성을 기저로 한다. 간섭이온의 효과는 물론 수용액상의 산성도, 고상의 용량, 흐름변수와 같은 희석용액으로부터 납(II)의 정량회수와 고상 시약의 준비 상태에 대해 연구되었다. 예비농축 후에 금속이온은 0.5M HNO3 용액의 5 ml에 자동적으로 용출되고 납이온의 양은 불꽃원자 흡수 분광법로 측정하였다. 최적조건하에서, 수용액시료의 납이온은 컬럼에 의해 약 1000배 분리 및 농축되었다. 검출한계는 0.001mg mL-1였다. 납은 강과 수돗물시료에서 98에서 102% 회수율을 보였다.

Determination of Non-Steroidal Anti-Inflammatory Drugs in Human Urine Sample using HPLC/UV and Three Phase Hollow Fiber-Liquid Phase Microextraction (HF-LPME)

  • Three phase hollow fiber-liquid phase microextraction (HF-LPME), which is faster, simpler and uses a more environmentally friendly sample-preparation technique, was developed for the analysis of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) in human urine. For the effective simultaneous extraction/concentration of NSAIDs by three phase HF-LPME, parameters (such as extraction organic solvent, pH of donor/acceptor phase, stirring speed, salting-out effect, sample temperature, and extraction time) which influence the extraction efficiency were optimized. NSAIDs were extracted and concentrated from 4 mL of aqueous solution at pH 3 (donor phase) into dihexyl ether immobilized in the wall pores of a porous hollow fiber, and then extracted into the acceptor phase at pH 13 located in the lumen of the hollow fiber. After the extraction, 5 ${\mu}L$ of the acceptor phase was directly injected into the HPLC/UV system. Simultaneous chromatographic separation of seven NSAIDs was achieved on an Eclipse XDB-C18 (4.6 mm i.d. ${\times}$ 150 mm length, 5 ${\mu}m$ particle size) column using isocratic elution with 0.1% formic acid and methanol (30:70) at a HPLC-UV/Vis system. Under optimized conditions (extraction solvent, dihexyl ether; $pH_{donor}$, 3; $pH_{acceptor}$, 13; stirring speed, 1500 rpm; NaCl salt, 10%; sample temperature, $60^{\circ}C$; and extraction time, 45 min), enrichment factors (EF) were between 59 and 260. The limit of detection (LOD) and limit of quantitation (LOQ) in the spiked urine matrix were in the concentration range of 5-15 ng/mL and 15-45 ng/mL, respectively. The relative recovery and precision obtained were between 58 and 136% and below 15.7% RSD, respectively. The calibration curve was linear within the range of 0.015-0.96 ng/mL with the square of the correlation coefficient being more than 0.997. The established method can be used to analyse of NSAIDs of low concentration (ng/mL) in urine.

Pentafluorophenylprophyl Ligand-based Liquid Chromatography-Tandem Mass Spectrometric Method for Rapid and Reproducible Determination of Metformin in Human Plasma

  • This paper describes first development and validation of pentafluorophenylprophyl ligand-based liquid chromatography coupled to tandem mass spectrometry (PFPLC-MS/MS) method to determine metformin, a highly polar compound, in human plasma. Metformin and Phenformin (internal standard) were extracted from human plasma 50 ${\mu}L$ with a single-step protein precipitation. The chromatographic separation was performed using a linear gradient elution of mobile phase involving 5.0 mM ammonium formate solution with 0.1% formic acid (A) and acetonitrile (B) over 3.0 min of run time on a Phenomenex Luna PFP column. The detection was performed using a triple-quadrupole tandem mass spectrometer (Waters Quattro micro) with electrospray ionization in the mode of positive ionization and multiple-reaction monitoring (MRM). The developed method was validated with 5.0 ng/mL of lower limit of quantification (LLOQ). The calibration curve was linear over 5-3000 ng/mL of the concentration range ($R^2$ > 0.99). The specificity, selectivity, carry-over effect, precision, accuracy and stability of the method met the acceptance criteria. The method developed in this study had had rapidness, simplicity and ruggedness. The reliable method was successfully applied to high throughput analysis of real samples for a practical purpose of a pharmacokinetic study.

치환크로마토그래피에서 온도와 용리액의 흐름속도가 란탄족 원소들 (La : Ce, Ce : Pr, Pr : Nd) 의 분리에 미치는 영향 (The Effect of Temperature and Flow Rate of Eluent on the Separation of Adjacent Lanthanides (La : Ce, Ce : Pr, Pr : Nd) with Displacement Chromatography)

  • 이웃하는 란탄족 원소의 분리에서 온도와 흐름속도의 영향을 치환크로마토그래피로 연구하였다. 일렬로 연결된 두 컬럼에 Amberlite 120 양이온 교환수지를 채워서 충전용과 분리용으로 나누어 사용하였다. 보유이온은 $H^+$을, 흐름용액은 0.012M과 0.015M의 EDTA 용액을 사용하였다. 실험중에는 관의 온도를 90$^{\circ}C$로 유지하였으며 이온교환수지 관 속에 생기는 기포를 줄이기 위해 일정한 압력을 걸었다. 용출액은 ICP-AES로 직접 분석하였다. 이 실험에서 란탄족 원소쌍 La: Ce, Ce :Pr, Pr: Nd에 대한 분리인자는 각각 4.6, 2.8, 1.9이었으며 이것은 25$^{\circ}C$에서 이론적으로 얻은 것보다 큰 값이다. 흐름속도를 2.5 ml/min에서 1.5 ml/min로 줄이면 이론단해당높이(HETP)도 1.60 cm에서 0.88 cm로 줄었다. 따라서 용리액의 흐름속도를 줄이고 온도를 높이므로써 분리효율을 증가시킬 수 있었다. 이 실험에서 99.9% 이상의 순수 란탄족 원소에 대한 회수율은 49∼77%이었다.

사람 섬유아세포에서 전호(Anthriscus sylvestris Hoffmann)추출물이 콜라겐 생합성에 미치는 영향 (Effects of Anthriscus sylvestris Hoffmann Extract on the Biosynthesis of Collagen in Human Dermal Fibroblasts)

  • 본 연구에서는 전호추출물의 주름개선에 대한 효능을 측정하기 위하여 인간의 섬유아세포에 전호추출물을 처리하여 총콜라겐과 type I procollagen, MMP-1의 생합성에 미치는 영향을 조사하였다. 전호추출물을 인간 섬유아세포에 처리 시 콜라겐의 생합성은 증가시켰지만, MMP-1의 발현에 대해서는 영향을 미치지 못하였다. 이는 전호추출물이 자외선에 의해 영향을 받는 광노화보다는, 노화에 의해 콜라겐 생합성율이 감소되는 내인성노화에 훨씬 유효함을 시사한다. 전호추출물을 처리시 95% EtOH 추출물의 경우 25%까지 총콜라겐의 생합성을 증가시켰고, EtOAc층은 28%, EtOAc의 E6 소분획의 경우 50% 증가시켰으므로, 콜라겐 생합성에 영향을 미치는 유효성분은 EtOAc층에 대부분 함유되어 있을 것으로 사료되고, 이는 확인되지 않은 미지의 효능 성분이 존재하거나, 다양한 활성 성분들의 상호 작용에 의한 것으로 판단된다. 결론적으로 전호추출물은 콜라겐 생합성을 촉진함으로 내인성노화에 의한 주름생성 및 탄력저하를 개선할 수 있는 천연 유용자원으로 이용할 수 있을 것으로 사료된다.
