Purpose: The purpose of this study was to analyze the colony count of airborne microbes contamination every hour in the Neurosurgical Intensive Care Unit (NSICU) in order to identify the relationship of colony count to person-visits. Method: Data were collected during from 11:00 a.m. September 5 to 11:00 a.m. September 6, 2002. This study used blood agar & nutrient agar and handtally counter (USA) for collection of airborne microbes and number of person-visits. Data was analyzed using the SPSSWIN 10.0 with means, Pearson correlation coefficient, and simple regression. Result: The result of this study are as follows. Total colony count of airborne microbes for 24 hours in the NSICU was 4,609. Total number of person-visits to the NSICU was 15,347. The highest scores fur the total colony count in different areas of the NSICU was the rear door, followed by the preparation room, and the front entrance, while the lowest count was in the isolation rooms. There was a statistically significant relationship between colony count and number of person-visits to the NSICU. The most frequently airborne microbes in the NSICU were Micrococcus, CNS, Staphylococcus Micrococcus, Aureus. Conclusion: These findings indicate that the number of person-visits in hospitals influences total colony count of airborne microbes. This study contributes to assessment of biological indoor air quality in hospital and in the development of an NSICU care plan.
The cell counting of colonial Microcystis spp. is a rather difficult and error-prone proposition, as this genus forms irregularly-shaped and irregularly-sized colonies, which are packed with cells. Thus, in order to facilitate a cell count, four methods of dividing the colonies into single cells were compared, including vortexing, sonication, $TiO_2$ treatment, and boiling. As a result, the boiling method was determined to generate the greatest number of single cells from a colony, and all colonies were found to have divided completely after only 6 min of treatment. Furthermore, no significant cell destruction, which might alter the actual cell density, was detected in conjunction with the boiling method (P=0.158). In order to compute the cell number more simply, the relationship between the colony size and the cell number was determined, via the boiling method. The colony volume, rather than the area or diameter was correlated more closely with the cell number ($r^2=0.727$), thereby suggesting that the cell numbers of colonial Microcystis sp. can also be estimated effectively from their volumes.

The purpose of this study was to evaluate the specificity, sensitivity, and diagnostic power of caries activity test using laser fluorescence. The subjects of this study were 50 children of $7\sim9$ years old. Fluorescence from initial carious lesion of teeth illuminated by an argon laser(480nm) was observed and photographed with barrier filter. Visual examination for the dDfFtT rate and Streptococcus mutans colony counting was done to evaluate correlation with caries activity test using laser fluorescence. Data analysis was accomplished by Axelsson's method. The results from the present study can be summarized as follows: 1. There was positive correlation $(\gamma=0.48)$ between laser fluorescence test and Streptococcus mutans count. And also positive correlation $(\gamma=0.39)$ exists between laser fluorescence test and dDfFtT rate (P<0.01). 2. Positive correlation $(\gamma=0.27)$ between Streptococcus mutans colony count and dDfFtT rate was found(P<0.05). 3. When dDfFtT rate was defined to standard testing method, the specificity, senstivity, and diagnostic power of laser fluorescence test were 44.4%, 85.7%, and 87.8%. 4. When dDfFtT rate was defined to standard testing method, the specificity, senstivity, and diagnostic power of S. mutans colony counting were 77.8%, 92.9%, 84.8%. 5. When S. mutans colony counting was defined to standard testing method, sensitivity, specificity and diagnostic power of laser fluorescence test were 40.0%, 84.8%, 95.1%. In regard to above results, laser fluorescence test considered to be accurate and reliable method for determining caries activity because of it's close relationship with caries susceptibility test and caries experience measurements. And it was also considered to be practical because it would be simple, inexpensive, and time saving method.

Purpose : To assess the efficacy of recombinant human granulocyte-macrophage colony-stimulating factor(GM-CSF) in the neutropenia by radiotherapy. Materials and Methods : Eleven patients with various solid tumor were treated with a daily subcutaneous dose of GM-CSF(3-7microgram/kg) for 5days during the radiotherapy. Before and during the course of the study all the patients were monitored by the recording of physical examination, the complete blood count with differential and reticulocyte count and liver function test. Eight patients received prior or concurrent chemotherapy. Results : In 10 patients, the neutrophilic nadir was significantly elevated and the lenght of time that Patients had a neutrophil count below $10^3/mm^3$ a threshold known to be critical to acquiring infective complications was shortened following GM-CSF injection. A significant rise (two fold or greater) of neutrophil count was seen in 10 of 11 patients. In most patients, discontinuation of GM-CSF resulted in a prompt return of granulocyte counts toward baseline. However the neutrophil count remained elevated over $10^3/mm^3$ during radiation therapy, and radiotherapy delays were avoided. Other peripheral blood components including monocytes and platelets also increased after GM-CSF treatment. No significant toxicity was encountered with subcutaneous GM-CSF treatment. Conclusion : GM-CSF was well tolerated by subcutaneous route and induced improvement in the neutropenia caused by radiotherapy.
Administration of 3 type KGCs [recombinant human granulocyte colony-stimulating factor (rhG-CSF)] to mice with cyclophosphamide (CPA)-induced neutropenia for 4 consecutive days from the day after the CPA dosing (100 mg/kg) resulted in a dose dependent increase in the peripheral blood neutrophil count 6 hours after the final KGC injection. Within the KGC dose range of 0.1 to 40$\mu$g Per mouse Per day, there was a sigmoidal relationship between the logarithm of the dose and the peripheral blood neutrophil count (relative value for neutrophil count of the basal dose) in the treated mice. The sigmoidal relationship of test KGC preparations shows that there is a saturation point in terms of efficacy. Compared with e(fact of KGC-Orange, Green, and Blue, KGC-Orange recovers neutrophils more effectively than the others do.

This study was carried out to evaluate water quality based on physicochemical properties, including ammonium nitrogen ($NH_3-N^+$), nitrate nitrogen ($NO_3-N^-$), consumption of $KMnO_4$ and microbial indicators, such as total colony counts and total surveyed a group of 30 samples three times in 2005 (January to March, July to September, October to December). The mean concentration values of ammonium nitrogen ($NH_3-N^+$), nitrate nitrogen ($NO_3-N^-$) and the consumption of $KMnO_4$, were 0.01 mg/L (range 0.00~0.09 mg/L), 0.48 mg/L (range 0.00~3.31 mg/L) and 0.61 mg/L (0.0~3.42 mg/L), respectively. Based on the results, total colony count was detected in the ranges of 0~412 CFU/mL. In details, 84.4% of test samples was ${\leq}30CFU/mL$, 10.0% was 30~100 CFU/mL and 5.6% was ${\geq}100CFU/mL$. The detected rate of total coliforms was 6.6%. In conclusion, the ground water quality of 30 military facilities in Seoul, Kyunggi-do, and Inchon-city was acceptable for drinking except for a few detection of total colony count and total coliforms over standard.
The ground pork patties were made to add two level of sodium chloride(2.5%, 3.0%) and replace part(50%) of the sodium chloride(NaCl) with either potassium chloride(KCl), magnesium chloride(MgCl2) or calcium chloride(CaCl2). These samples were analyzed for their chemical composition, VBN value, TBA value, microbial counts, and cooking loss. The ground pork with NaCl 2.5% was more desirable in saltness than the ground pork with NaCl 3.0%. Replacing 50% of the sodium chloride with potassium chloride was more desirable to flavor, color, juiciness, and overall acceptability than replacing 50% of the sodium chloride with either magnesium chloride or calcium chloride. The ground pork with NaCl 2.5% or NaCl 1.25% +KCI 1.60% had higher pH value than the ground pork with NaCl 1.25% +MgCl2 0.67% or NaCl 1.25% +CaCl2 0.79%. The ground pork with the ground pork with NaCl 2.5% had lower VBN value than the ground pork with either NaCl 1.25%+KCI 1.60%, NaCl 1.25% + MgCl2 0.67%, or NaCl 1.25% +CaCl2 0.79%. The ground pork with NaCl 1.25% + CaCl2 0.67% had higher increase in total colony count than the ground pork with NaCl 2.5% or NaCl 1.25% + CaCl2 0.79%. Cooking loss of ground pork with NaCl 2.5% was lowest and cooking loss of ground pork with NaCl 1.25% + KCl 1.60% was highest. Potassium chloride would not be a substitute for sodium chloride in cooking loss and total colony count but potassium chloride more closely approximated the sensory properties of sodium chloride than either magnesium chloride or calcium chloride.
The purpose of this study was to investigate the safety of Houttuynia cordata Thunb extract (HCE) in Institute of Cancer Research (ICR) mice, to determine the effects of the extract on the growth performance and colony count, and to establish the optimal concentration of HCE. In total, 60 five-week-old male ICR mice with an average initial body weight (BW) of 27.24 ± 0.44 g were used in a four-week experiment. Mice were randomly allotted to four treatment groups (five replications per group, three mice per cage): 1) a control (CON) group fed with normal distilled water; 2) treatment group 1 (T1) fed with normal distilled water containing 0.05% HCE; 3) treatment group 3 (T3) fed with normal distilled water containing 0.1% HCE; and 4) treatment group 3 (T3) fed with normal distilled water containing 0.2% HCE. BW, feed intake (FI), and water intake were measured on the first, fourteenth, and eighteenth days. T2 showed a significant improvement (p < 0.05) in the feed conversion ratio (FCR) over the experimental period. However, water intake levels did not show significant differences among the groups. In the large intestine and feces, E. coli and Lactobacillus levels were significantly improved (p < 0.05) in the HCE treated group compared to the CON group. Supplying HCE via the drinking water improved the growth performance and colony count in ICR mice. Based on results of this study, utilizing HCE in livestock species is expected to be safe and feasible.

Background : Even though it has been suggested that low-colony, scotochromogen nontuberculous mycobacteria (NTM) are usually contaminants and not true pathogens, evidence for this hypothesis has not been provided. This study investigated the colony characteristics, organism identification, and clinical significance of low-colony scotochromogen. Methods : The laboratory cultured 6,898 respiratory clinical specimens for an examination of mycobacteria over a three-month period. A low-colony count was arbitrarily defined as ${\leq}20$ colonies. This study analyzed the recovery rate of the mycobacteria, the number of colonies and their gross characteristics, and their clinical significance. PCR-restriction fragment length polymorphism analysis was carried out to identify the NTM species. NTM pulmonary disease was defined according to the American Thoracic Society. Results : A total of 6,898 respiratory specimens for mycobacterium were cultured. Of these, 263 (3.8%) grew NTM, and 382 (5.5%) grew M. tuberculosis. Of the 263 cultured NTM specimens, 124 (47.1%) were scotochromogens. The smear-positive rate was significantly lower in these scotochromogens (4.8%) than in the non-scotochromogens (23.7%) (p<0.05). The most common isolates were M. gordonae (83/102, 81.4%) in the scotochromogens, and MAC (52/121, 43.0%) in the non-scotochromogens. Even though three out of 113 patients with a low-colony scotochromogen has been diagnosed with NTM pulmonary disease, the isolated scotochromogen was not considered to be the cause of the NTM disease but was just a contaminant. Conclusion : In this study, the most common isolate of a low-colony count scotochromogen was M. gordonae, which appeared to be contaminants and not true pathogens. Greater efforts in the quality control of a mycobacterium laboratory are needed in cases where there is a high recovery rate of low-colony count scotochromogen.

The purpose of this study is to investigate the relationship between oral health behavior and dietary habits and caries occurrence test and to provide basic data necessary for prevention of dental caries. The average salivary salivary fraction was 8.35ml and the non - amorphous salivary fraction was 2.41ml. The salivary buffering ability was 9.29 and the mean glucose retention time was 15.81 minutes. In the Streptococcus mutans colony count test, low risk group <105 was 80.5% and low risk group <105 was 84.4% in Lactobacillus colony count test. Correlation between caries test scores showed a positive correlation between salivary ratio and non - salivary ratio, saliva buffering ability, non - salivary fraction and glucose retention time, and Lactobacillus. Factors affecting the irritant saliva fraction were alcohol as a factor affecting the ratio of nuts, pork, and non - irritant saliva. In order to prevent dental caries, proper dietary control education should be combined with dental caries activity test and oral health education for proper eating habits.
