한국산 장고말속(녹조식물문) 6분류군의 추가 (An Addition of 6 Taxa of the Genus Cosmarium (Chlorophyta) in Korea)

  • Six taxa of unrecorded Korean species of genus Cosmarium were collected from several freshwater sites of South Korea in 2003. They were 3 species and 3 varieties of Genus Cosmarium. Genus Cosmarium of Korea was reported to totally 273 taxa 125 species, 113 varieties and 35 forms including this study.

  • In this study, phytoplanktons collected monthly at Paroho from January till April in 1968 and from May till August in 1973. The Diatom samples were identified and classified by Engler's Classification System. It resutled in 1 phyla, 1 class, 2 subclasses, 5 orders, 8 families, 18 genera, 41 species, 11 varieties and 2 forma. Of them, 8 species, 6 varieties and 2 formas are new to Korea.

  • As a continuation of part I, this paper deals with the classification of phytoplankton species collected from Chawol-do, Soya-do Pung-do of Kyonggi-Bay. One hundred and fortythree kinds of photoplankton from this area were identified and classified by Engler's system into 17 families, 47 genera, 129 species, 10 varieties and 4 forms, blonging to 2 phyla, 3 classes, 3 subclasses, 8 orders and 1 suborder. 3 families, 7 genera, 10 species, 3 varieties and 3 forms are added. To the list published in part I. Seven species and one form described in this paper are new to Korea.

대두의 품종에 관한 연구 -제1보 생태형과 성숙군의 분류- (Studies on the soybean varieties in Korea -1. Classificiatons of Ecotypes and Maturity groups-)

  • Experiments were carried out to classify the ecotypes and maturity groups of soybean varieties, and to make clear the relationships among these ecotypes, maturity groups and some characteristics in the growing and ripening process of soybean varieties in Korea. Soybean varieties used as the material were 138, 57 varieties collected from Japan, America and Canada and 81 local varieties of Korea. These varieties were grown in the Experimental Farms, College of Agriculture, Seoul National University, Suwon, Korea and Chinju Agricultural College, Kyungsang Namdo, Korea. Seed sowing was conducted at 8 times from April15 to July 29, at 15 day-intervals, in 1962. The classification methods of ecotypes and maturity groups applied in this study, and the results obtained are summarized as follows; 1. Ecotypes were classified from the standpoint of the periods from emergence to flowering, the shortened ratio of days from sowing to flowering by delayed sowing, and the periods from flowering to maturity(as shown in Table A). 2. All varieties wer classified into 10 ecotypes of Ia$_1$, Ib$_1$, IIa$_2$, IIb$_1$, IIb$_2$, IIIb$_1$IIIb$_2$, IIIc$_1$and IIIc$_2$ symbols. Korean local varieties, however, belong to 6 ecotypes of IIb$_1$, IIb$_2$, IIIb$_1$, IIIc$_2$, IIIc$_1$ and IIIc$_2$, respectively(table 1). 3. Durations from sowing to maturity were from 120 to 190 days in April sowing plots of all varieties. From the standpoint of the durations, maturity groups were classified into 9 groups from group I, extreme early, to group IX, extreme late, but our local varieties belong to 5 maturity groups of III, IV, V, VI and VII symbols, respectively(Table 2). 4. Kinds of ecotypes and maturity groups, and the number of soybean varieties belonging to those are as shown in Table B. 5. In this study, it was observed that there were some close relations between these ecotypes and maturity groups (Table 3,4), and among the ecotypes, maturity groups and some other characteristics in the growing and ripening process of the soybean varieties(Table 5). 6. Furthermore, it was also observed that the kinds of ecotypes and maturity groups of recommended varieties in the southern Korean provinces are more numerous than those of the northern provinces in Korea.

  • 전국에서 수집된 혼식용 유색대두 32품종에 대하여 조사된 21개 형질로서 분류적 거리와 Q 상관에 의하여 품종간의 유사정도를 계산하고 이들에 의하여 Single link clustering 방법에 의하여 dendrogram을 그리고 Q 상관에 의하여 10개의 품종군을 분류하였다. 각 품종군의 군내 Q상관은 군간 Q상관보다 높았으며, 각 품종군의 각 형질평균으로 보아 각 군이 특징있게 구분되었다.

국내 자생벼 계통의 탈립성과 이층조직의 특성 (Relationship between Degree of Grain Shedding and Histological Peculiarities of Abcission Region of Red Rice(Oryza sativa L.) Collected in Korea)

  • 우리나라 자생벼 수집 보존 계통들이 지니고 있는 우량 형질을 선발하여 벼 신품종 육종의 소재로 삼기 위하여, 우선 벼알과 소지경 사이의 인장강도 및 항곡강도를 측정하여 자생벼들의 탈립성정도를 분급함과 동시에 외국산 적미들 및 우리나라 재래종, 통일형 및 일본형 등 주요 품종들과 비교하였으며, 자생벼들의 탈립성이 강한 원인을 규명하기 위하여 벼알 탈락부위에 형성되는 이층조직의 유무 및 붕괴성과 벼알의 탈립성정도와 관련이 있을 것으로 생각되는 벼알 탈락부위의 몇 가지 해부형태학적 특성을 조사하였다. 그 결과를 요약하면 다음과 같다. 1. 우리나라 자생벼들의 인장강도 및 항곡강도는 외국산 적미들과 비슷한 분포를 보였으며, 통일형 품종과 재래종 및 일본형 주요 품종들 보다는 약한 것으로 나타났다. 자생벼들 중에서는 최근 수집된 '샤레'들이 가장 약했다. 2. 우리나라 자생벼를 비롯한 모든 공시 품종에서 항곡강도와 인장강도 사이에 높은 정의 상관관계가 인정되었다. 3. 우리나라 자생벼들 중 대부분은 소수축과 소지경 선단 돌출부 사이에 이층조직이 형성되어 있었는데, 단립적색종 및 단립갈색종들은 거의 모두 붕괴되지 않은 이층조직을 가지고 있었으며, 장립적색종 및 '샤레'들은 붕괴되는 이층조직을 가지고 있었고, 이층조직이 없는 자생벼들도 일부 포함되어 있었다. 4. 자생벼의 인장강도 및 항곡강도는 이층조직이 붕괴되는 것, 이층조직이 붕괴되지 않는 것 그리고 이층조직이 없는 것 순으로 강하게 나타났다. 5. 자생벼들 중 장립적색종들은 소지경 선단 돌출부의 직경, 중심 유관속의 직경, 벼알 부착부의 직경 중심 유관속 주변의 후벽조직 두께 및 소지경선단 돌출부의 길이 등 벼알 탈락부위의 해부형태학적 특성이 다른 자생벼들과 상이한 것으로 나타났다. 6. 자생벼에서 장ㆍ단위종 모두 인장강도 및 항곡강도와 벼알 부착부의 직경 및 중심 유관속 주변의 후벽조직 두께와의 사이에는 각각 높은 정의 상관관계가 인정되었다.

Authentication and classification of strawberry varieties by analysis of their leaves using near infrared spectroscopy.

  • It is well known now that near infrared spectroscopy (NIRS) is a fast, no destructive, and inexpensive analytical technique that could be used to classify, identify, and authenticate a wide range of foods and food items. Therefore, the main aims of this study were to provide a new insight into the authentication of five strawberry (Fragaria x ananassa) varieties and to correlate them with geographical zones and the propagating methods used. Three weeks plants of five different strawberry varieties (F. x ananassa Duch. cv Camarosa, Seascape, Chandler, F. Chiloensis, and F. Virginiana) were cultivated in vitro first and then transferred to pots with special soil, and grown in a greenhouse at CINVESTAV, all varieties were acquired from California (USA). After 18 months, ten leaves from each variety were collected. Transmission spectra from each leave were recorded over a range of 10, 000-4, 000 cm$-^{1}$, 32 scans of each strawberry leave were collected using a resolution of 4 cm$-^{1}$ with a Paragon IdentiCheck FT-NIR System Spectrometer. Triplicates of each strawberry leave were used. All spectra were analyzed using principal component analysis (PCA) and soft independent modeling class analogy (SIMCA). The optimum number of components to be used in the regression was automatically determined by the software. Camarosa was the only variety grown from the same shoot but propagated by a different method (direct or in vitro). Five different classes (varieties) or clusters were observed among samples, however, larger inter class distances were presented by the two wildtype samples (F. Chiloensis and F. Virginiana). Camarosa direct and Camarosa in vitro displayed a small overlapping region between them. On the other hand, Seascape variety presented the smallest rejection percentage among all varieties (more similarities with the rest of the samples). Therefore, it can be concluded that the application of NIRS technique allowed the authentication of all strawberry varieties and geographical origin as well. It was also possible to form subclasses of the same materials. The results presented here demonstrate that NIRS is a very powerful and promising analytical tool since all materials were authenticated and classified based on their variety, origin, and treatment. This is of a tremendous relevance since the variety and origin of a plant material can be established even before it gives its typical fruit or flower.

Development of SNP marker set for discriminating among Korean rice varieties and imported rice in Korea

  • In accordance with the opening of the Korean rice market, this study was focused on establishment of database for discriminating the Korean rice varieties and imported brand rices using DNA markers. In this study, the SNP markers were developed using single nucleotide polymorphisms between the reference sequences of japonica and them of 40 brand rices which collected in Australia, China, Thailand, United States and Vietnam. The developed SNP markers were screened to a total of 360 rices including 320 Korean rice varieties and 40 imported brand rices. We selected polymorphic markers among Korean bred rive varieties and imported brand rices. The selected markers were classified into 3 grades. The markers of A grade produced DNA band in 360 rices of 30~40%, B grades produced in 40~60%, and C grades produced bands over 60% rices. First, we tried to set-up the discriminating system using the minimum SNP markers of A grade. Especially, a set of sixteen SNP markers could identify among Korean bred rice varieties and imported brand rices. Additionally, some SNP markers like NSb for Pib gene, JJ80-T for Pi5 and YL155/YL87 for Pita which linked to resistance genes to blast were used to fingerprinting system. These markers were set-up as multiplex set for enhancing the identification efficiency among rice varieties. Finally, the selected SNP markers would be used to the fluidigm assay to construct the database for elaborate discrimination of rice varieties.

Identification of rice blast major resistance genes in Korean rice varieties using molecular marker

  • Rice blast caused by Magnaporthe oryzae is one of the most serious diseases that affect the quantity and quality of rice production. The use of resistant rice varieties would be the most effective way to control the rice blast. However R gene incorporation into the rice variety takes time and pathogen could overcome the R gene effects after for a while. For monitoring the rice blast resistance gene distribution in Korean varieties, the four major blast resistance genes against M. oryzae were screened in a number of Korean rice varieties using molecular markers. Of the 120 rice varieties tested, 40 were found to contain the Pi-5 gene, 25 for the Pi-9 gene, 79 for Pi-b and 40 for the Pi-ta gene. None of these rice varieties includes tested 4 R genes. 3 R genes combination, Pi-5/Pi-9/Pi-b, Pi-5, Pi-9.Pi-ta, or Pi-9/Pi-b/Pi-ta were found in 12 varieties, the rice blast disease severity were showed as resistant in the rice verities containing Pi-9/Pi-b/Pi-ta R genes combination, respectively. Also pathogenic diversity of M. oryzae isolates collected in the rice field from 2004 to 2015 in rice field in Korea were analyzed using rice blast monogenic lines, each harboring a single blast resistance gene. Compatibility of blast isolates against rice blast monogenic lines carrying the resistance genes Pi5, Pi9, Pib, and Piz showed dynamic changes by year. It indicates that pathogen has high evolutionary potential adapted host resistances to increase fitness and would lead to rice blast resistance bred into the cultivar becoming ineffective eventually.

마늘 유전자원의 주요 생리적 특성과 RAPD 페턴과의 관련성 (Relationship with major physiological characters and RAPD patterns of garlic (Allium sativum L.) germplasm.)

  • 세계 각지에서 수집된 마늘 유전자원의 주요 생리적 특성과 RAPD 페턴과의 관련성을 검토하기 위하여 분석한 결과는 다음과 같다. 84개의 수집마늘의 생리적 특성들에 의한 다변량 해석의 결과 10개의 품종군으로 구분되어졌고 어느 품종군에도 속하지 않은 계통이 9개였다. 조만성은 주로 중국, 일본, 한국에서 수집된 마늘들에서 조생종마늘이 많았고 유럽원산의 마늘들에서는 만생형 품종이 많았으며 RAPD 마커 WE6 $l_{1,630}$과 상당한 관련을 보였다. 이차생장은 거의 대부분의 마늘들에서 나타나는 경향을 보였으나 동유럽지역 수집종에서 발생율이 높은 경향을 보였고 네팔을 중심으로 한 지역종들은 이차생장이 발생하지 않았다. 인편미분화는 유럽원산의 마늘들에서 많은 발생율을 보였지만 표지인자를 찾기는 어려웠다. 추대성은 RAPD마커 WF70$_{1,400}$에서 벤드가 나타난 마늘은 추대종 마늘이었다. 마늘 임성과 관련 된 RAPD 마커 는 WF64$_{1,400}$이 었으며 임성마늘에서 벤드가 나타났다.
