최근 여러 종양에서 단백분해효소의 분비와 암의 악성도에 대한 연구가 이루어져 왔으며, 이를 신세포암종 환자의 예후 측정인자로 사용하려는 시도가 진행되고 있다. 이에 저자들은 제 4형 collagenase 중 대표적인 MMP-9의 발현정도를 정상 신조직과 신세포암종 조직에서 비교하였고, 또 암의 침윤 및 전이정도와의 관계와 다른 임상적 인자들과의 상관성을 분석하여 암의 단계적 진행과정에서의 MMP-9의 발현변화에 대하여 조사하였다. 그 결과 정상 신 조직에 비해 신세포암종 조직에서 MMP-9의 발현이 증가되며 암의 크기가 크고 혈관침범이 있으며 병기가 높을수록 MMP-9의 발현이 증가됨을 관찰할 수 있었다. 이는 MMP-9 발현의 증가가 신세포암종의 발생과정 및 암의 후기 진행에 관여함을 시사하므로 향후 신세포암종의 예후척도로 사용되어 치료방침을 결정하는데 도움을 줄 수 있을 것으로 생각한다.
Low-molecular weight peptides derived from fish collagen exhibit several bioactivities, including antioxidant, antiwrinkle, antimicrobial, antidiabetic, and antihypertension effects. These peptides are also involved in triglyceride suppression and memory improvement. This study aimed to investigate the optimal processing condition for preparing low-molecular weight peptides from flounder skin, and the properties of the hydrolysate. The optimal processing conditions for peptic hydrolysis were as follows: a ratio of pepsin to dried skin powder of 2% (w/w), pH of 2.0, and a temperature of 50℃. Peptic hydrolysate contains several low-molecular weight peptides below 300 Da. Gly-Pro-Hyp(GPHyp) peptide, a process control index, was detected only in peptic hydrolysate on matrix-assisted laser desorption/ionization-time-of-flight(MALDI-TOF) spectrum. 2,2'-azinobis-(3-3-ethylbenzothiazolline-6- sulfonic acid(ABTS) radical scavenging activity of the peptic hydrolysate was comparable to that of 1 mM ascorbic acid, which was used as a positive control at pH 5.5, whereas collagenase inhibition was five times higher with the peptic hydrolysate than with 1 mM ascorbic acid at pH 7.5. However, the tyrosinase inhibition ability of the peptic hydrolysate was lower than that of arbutin, which was used as a positive control. The antibacterial effect of the peptic hydrolysate against Propionibacterium acne was not observed. These results suggest that the peptic hydrolysate derived from a flounder skin is a promising antiwrinkle agent that can be used in various food and cosmetic products to prevent wrinkles caused by ultraviolet radiations.
Objectives : The present study was carried out to investigate the effect of Trichosanthis Radix extract (TRE) on the proliferation and activation of eosinophils which were prepared from lung cells of asthma-induced mice by ovalbumin (OVA) treatment. Methods : C57BL/6 mouse was exposed to OVA three times a week for 6 weeks. The mouse lung tissues were dissected out, chopped and dossiciated with collagenase (1 $\mu$g/ml). Eosinophils were activated by rmIL-3/rmIL-5 co-treatments. The lung cells were treated with TRE, incubated for 48 hr at 37$^{\circ}C$, and analyzed by flow cytometer, ELISA and RT-PCR methods Results : To measure cytotoxicity, mouse lung fibroblast cells (mLFCs) were pretreated with various concentrations of TRE. TRE at 100 $\mu$g/ml, the highest concentration, examined did not have any cytotoxic effects on mLFCs. In FACS analysis, number of granulocyte/lymphocyte, CD3e-/CCR3+, CD3e+/CD69+, CD4+/CD8+ T cells in asthma-induced lung cells were significantly decreased by TRE treatment compared to the control group. But CD4+/CD25+ T cells were not examined significant change in lung cells treated with TRE. In ELISA analysis, production levels of IL-3, IL-5, IL-13 and histamine in asthma-induced lung cells, which were induced by rIL-3 plus rmIL-5 co-treatment, were significantly decreased by TRE treatment. Conclusions : The present data suggested that Trichosanthis Radix on the inhibition of parameters associated with asthma responses in eosinpophils, and thus implicate the possibility for the clinical application of Trichosanthis Radix.
To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.
This research aimed to assess the effect of collagenolytic proteases from Bacillus subtilis B13 and Bacillus siamensis S6 for tenderizing goat meat during wet aging. Collagenolytic proteases B13 and S6 were prepared at 5 U/mL of collagenolytic activity before injecting into goat meat with 10% (v/w) of initial weight. The control sample was injected with distilled water and used as a negative control. The injected meats were placed in vacuum-sealed bags and wet aged at 4℃ for 0, 3, 5, 7, 14, and 21 days. Thereafter, total aerobic count and physicochemical quality were elucidated. Both enzyme-treated samples from B13 and S6 aged for 5 days showed an acceptable microbial quality with lower than 5.7 Log CFU/g. These conditions produced the tender meats by the reduction in shear force accounting for 30% for B13 and 26% for S6 as compared to the control. Moreover, the enzyme-treated samples showed lower values of hardness, gumminess, and chewiness, with higher springiness and trichloroacetic acid-soluble peptides than the control (p<0.05). The detrimental impact on cooking loss and lipid oxidation was not found. Enzyme-injected meat had a lower cooking loss than the control (p<0.05) with no significant difference in lipid oxidation (p>0.05). Notably, meats treated with B13 and S6 were lower in CIE L* value as compared to the control (p<0.05) with no significant impact on CIE a* and CIE b* (p>0.05). These results suggested that these two collagenolytic proteases could enhance the quality of goat meat in terms of tenderness and reduce the aging time for meat tenderization.
Extracellular vesicles (EVs) are nano-sized lipid bilayer vesicles released by cells. EVs act as messengers for cell-to-cell communication. Inside, it contains various substances that show biological activity, such as proteins, lipids, nucleic acids, and metabolites. The study of EVs extracted from terrestrial organisms and stem cells on inflammatory environments and tissue regeneration have been actively conducted. However, marine organisms-derived EVs are limited. Therefore, we have extracted EVs from sea urchins belonging to the Echinoderm group with their excellent regenerative ability. First, we extracted extracellular matrix (ECM) from sea urchin gonads treated with hypotonic buffer, followed by collagenase treatment, and filtration to collect ECM-bounded EVs. The size of sea urchin gonad-derived EVs (UGEVs) is about 20-100 nm and has a round shape. The protein content was higher after EVs burst than before, which is evidence that proteins are contained inside. In addition, proteins of various sizes are distributed inside. PKH-26 was combined with UGEVs, which means that UGEVs have a lipid membrane. PHK-26-labeled UGEVs were successfully uptaken by cells. UGEVs can be confirmed to have the same characteristics as traditional EVs. Finally, it was confirmed that Schwann cells were not toxic by increasing proliferation after treatment.
류마티스 관절염의 쥐의 활액에서 단백분해효소, 산화제와 유리기에 대한 녹용약침의 비특이적 면역억제효과를 연구하였다. 일련의 실험표본으로서 여러가지 세포질, 리소좀, 기질 백분해효소의 제 활성을 RA대조군과 녹용약침군의 활액에서 카르보닐기 유도로 생성되는 유리기-유발 단백질손상과 항산화를 비교하였다. 전반적으로 단백분해효소활성이 정상군과 비교하여 RA대조군에서 유의성 있게 증가하였다. 세포질 단백분해효소들은 정상군과 RA군의 차이에서는 유의성이 없었다. 녹용약침처리($100{\mu}g/kg$)결과 세포질, 리소좀, 기질 단백분해효소생성을 억제하였으며, RA군과 녹용약침군 또는 정산군 사이에 활액 또는 세포질 항산화에서 유의성 있는 차이가 없음에도 불구하고, RA군 활액의 단백질손상을 유발하는 유리기는 녹용약침군과 정산군에 비교하여 약 2배 정도 높았다. 이상의 결과에서 단백분해효소와 유리기는 RA유발시 단백질손상을 유도하는 물질로 밝혀졌으며, 따라서 단백분해효소 저해와 유리기소거능을 갖는 치료법개발이 새로운 RA예방치료법으로 제시되었다. 나아가서 여러가지 기질특이성을 갖는 활액내 단백분해효소류(cysteine, serine, metallo proteinases와 peptidases)에 대한 효과적인 저해제개발이 필요한 것으로 보인다. 따라서 본 녹용약침은 이와 같은 새로운 개념의 2가지(유리기제거, 단백분해활성) 관절염치료 요소를 충족하는 약리활성을 포함하는 훌륭한 제제로 평가된다.
쇠비름(Portulaca oleracea.L)은 쇠비름과에 속하는 한해살이풀로서 리놀렌산과 같은 불포화지방산, 페놀성 화합물, 플라보노이드, 비타민 C, 미네랄 함량이 높은 것으로 보고되어 있다. 본 연구에서는 쇠비름 추출물을 이용하여 UVB를 조사한 인간각질형성세포에서 광노화 억제능을 확인하였다. Matrix metalloproteinases는 세포의 기질을 분해하는 효소로 MMP-1는 collagenase, MMP-2와 MMP-9는 gelatinases로 피부 진피층을 구성하는 type I collagen을 분해시키는데 영향을 미친다. UVB를 조사한 인간각질형성세포에서 쇠비름 추출물을 처리했을 때 MMP-1, -2, -9의 발현이 감소하였으며, type I procollagen의 발현은 증가하는 것으로 나타났다. 또한 쇠비름 추출물을 처리한 군에서 UV에 의한 ROS 생성이 감소하였는데 이는 Nrf-2의 활성화를 통한 항산화 인자 SOD-1과 OH-1의 발현 증가로 인해 세포내 ROS 생성이 감소한 것으로 사료된다. 따라서 본 연구 결과를 통해 쇠비름 추출물이 UVB를 조사한 인간각질형성세포에서 MMP 인자 및 항산화 인자의 발현 조절을 통해 광노화로부터의 세포 보호 능을 가지는 것을 확인하였으며 나아가 화장품 소재로서의 개발 가능성을 확인하였다.
Cancer cells, characterized by local invasion and distant metastasis, are very much dependant on the extracellular matrix. The expression of matrix metalloproteinases (MMPs) has been implicated in the invasion and metastasis of cancer cells. In this study, we reported the effects of disulfiram, a clinically used anti-alcoholism drug, on tumor invasion suppression, as well as its effects on the activity of MMP-2 and MMP-9 in human osteosarcoma cells (U2OS). Disulfiram has been used for alcohol aversion therapy. However, recent reports have shown that disulfiram may have potential in the treatment of human cancers. Herewith, we showed that the anti-tumor effects of disulfiram, in an invasion assay using U2OS cells and that disulfiram has a type IV collagenase inhibitory activity that inhibits expression of genes and proteins responsible for both cell and non-cell mediated invasion on pathways. In conclusion, disulfiram inhibited expression of MMP-2 and MMP-9 and it regulated the invasion of human osteosarcoma cells. These observations raise the possibility of disulfiram being used clinical for the inhibition of cancer invasion.
