Antibacterial Properties of Silver-alginate/PVP Nanofiber (은-알지네이트/PVP 나노섬유의 항균 특성)

  • In order to incorporate silver ions into the alginate, silver-alginate was prepared with aqueous solutions of silver nitrate. In the study, the silver-alginate was prepared by blending with poly vinylpyrrolidone solutions and the electrospinning was performed by using this blend solution. Antibacterial properties of silver-alginate/PVP solutions were estimated for Escherichia coli and Staphylococcus aureus by the colony counting test. Electrospinning conditions of silver-alginate/PVP solution were the tip-to-collector distance of 22 cm, the flow rate of the solution at 0.01 mL/min, and the voltage at 26 kV. The form and size of silver-alginate/PVP nanofibers were estimated by SEM and Image J. The average diameter of the electrospun SA5P15 fibers was 124 nm and showed a narrow diameter distribution. The reduction of bacteria for SA5P15 exhibited 99.9% after 24 h.

The ACC deaminase from rhizobateria promoted resistance of salininty stress in seedling and growth of plant

  • Rhizobacteria are a diverse group of free-living soil bacteria that live in plant rhizosphere and colonized the root system. Plant growth-promoting rhizobacteria (PGPR) possessing ACC deaminase (ACCD) can reduce ACC and ethylene in plant tissue and mediated the growth of plants under various stresses including salt stress. ACCD decrease ethylene levels in plant tissue that produce high levels of ethylene in tissue via elevated levels of ACC under salt stress. We selected strains of Pseudomonas sp. possessing ACCD activity for their ability to promote plant growth under salt stress from soil sample collected at Byeonsan, Jeonbuk, South Korea. The Pseudomonas strains possessing ACCD increased the rate of the seedling and growth of chinese cabbage seeds under salt stress. We cloned ACCD gene from P.fluorescens and expressed recombinant protein in Escherichia coli. The active form of recombinant ACCD converted ACC to a-ketobutyrate. The in vivo treatment of recombinant ACCD itself increase the rate of the seedling and growth of Chinese cabbage seeds under salt stress. The polyclonal P.fluorescens anti-ACCD antibody specifically reacted with ACCD originated from Pseudomonas. This indicates that the antibody might act as an important indicator for ACCD driven from Pseudomonas exhibiting plant growth-promoting activity. This study will be useful for identification of newly isolated PGPR containing ACCD and exploioting the ACCD activity from PGPR against various biotic and abiotic stresses.

Production of Multiple Probiotics and the Performance of Laying Hens by Proper Level of Dietary Supplementation (복합 생균제의 생산과 산란계에서 생균제의 적정 급여 수준에 의한 산란 효과)

  • This experiment was conducted to evaluate the dietary effects of multiple mixture of probiotics on laying performance and the faecal examination in laying hens (Hy-line Brown) at the early (21~40 wk) and middle (41~65 wk) laying term. Multiple probiotics were produced by developing products and the properties of microorganisms were examined for detecting of acid-resistance, bile salt-resistance and antibacterial activity against pathogenic enteric bacteria. Probiotics produced to the fermenting cultures of four selected organisms and soybean meal substrates by nine steps of NK proliferating system. The most microorganisms were shown higher resistance of acidity and bile salt. High antibacterial activities against Bacillus subtilis, Lactobacillus plantarum and Enterococcus faecium were observed, but was not against Saccharomyces cerevisiae. Total egg production of the treatment was significantly higher than control group but was not statistically different between 0.1% and 0.2% treatments (P>0.05). Average egg weight of the treatment in early laying term was also significantly higher than control but was not significantly different between 0.1% and 0.2% treatments (P>0.05). But the egg weight of the treatment in middle laying term was significantly higher than control and between 0.1% and 0.2% treatments (P>0.05). The mortality of 0.2% treatment was significantly lower than control (P<0.05), and 0.2% treatment in the early laying term was tended to decreased than 0.1% treatment and control. But there was not significantly between 0.1% and 0.2% treatments in middle laying term. In feed intake, 0.2% treatment in middle laying term was significantly increased than control and 0.1% treatment (P<0.05) but not in early laying term. In faecal examination, the total number of Lactobacillus of 0.1% treatment was significantly increased than control in whole laying term (P<0.05), but Coli form of the treatment was decreased than control in middle laying term. In conclusion, dietary long term supplementation of multiple probiotics improved performance of lay hens, egg weight and mortality drop by regulating enteric bacteria.

Effect of Dietary Supplementation of Silicate Mineral $(Vita2000^{(R)})$ on Growth Performance and Intestinal Microflora in Broiler Chicks (사료 내 규산염 광물질$(Vita2000^{(R)})$ 첨가 급여가 육계 생산성과 장내 균총에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary Vita2000 on growth performances and immune response in broiler chickens. One-day-old male chicks were fed diets containing 0, 0.5 and 1% Vita2000 (with or without antibiotics) for 5 wks. There were no significant differences in daily weight gain among the treatments, but feed intake in 1% Vita2000 groups (T3 and T4) were significantly lower than control (P<0.05). The relative abdominal fat weight and the level of crude fat in leg meat on groups fed diets containing 1% Vita2000 (T3 and T4) were significantly decreased as compared to those of control (P<0.05). The content of cholesterol in leg meat was not affect by dietary treatment. The intestinal total microbes, Coli form, Lactic acid bacteria and Salmonella spp. from bird fed 1% Vita2000 diets was significantly reduced compared to those of control. The production of IB antibody in chicks fed diet containing 0.5% Vita2000 was significantly higher than that of control groups. The overall results indicate that dietary Vita2000 may be a valuable alternative to antibiotics for optimizing growth performances, particularly for reducing the abdominal fat of broiler chicks.

Antioxidative and Antimicrobial Activities of Lilium Species Extracts Prepared from Different Aerial Parts (부위별 식용백합 추출물의 항산화 및 향균효과)

  • In this study, Lilium sp. were separated into bulbs, leaves, and flowers. Then, total polyphenol contents, electron donating ability (EDA), superoxide dismutase (SOD)-like activity, hydroxyl radical scavenging activity, and antimicrobial activity were measured from the extracts of each of the three aforementioned parts. The examination of physiologically active substances in the three parts revealed that Lilium davidii leaves had high total polyphenol contents, SOD-like activity, hydroxyl radical scavenging activity, and EDA, while the flowers of L. lancifolium showed high SOD-like activity, hydroxyl radical scavenging activity, and EDA, as well as a high level of total polyphenols in the bulb. Measurements of the antimicrobial activities of the extracts against Gram positive bacteria revealed that the leaves and flowers of L. davidii and L. lancifolium caused Bacillus subtilis and Salmonella enteritidis to form clear zones greater than 10 mm. Furthermore, the flowers of L. lancifolium showed particularly high antimicrobial activity against B. subtilis, and the flowers of L. davidii had high activity against S. enteritidis. For the Gram negative bacteria, the leaves and flowers of L. davidii and L. lancifolium caused Listeria monocytogenes and Escherichia coli to form clear zones greater than 10 mm, and finally, the flowers of L. davidii and L. lancifolium showed high antibacterial activity, with inhibition exceeding 12 mm.

Effects of Ecklonia cava as fucoidan-rich algae on growth performance, nutrient digestibility, intestinal morphology and caecal microflora in weanling pigs

  • Objective: In the present study, role of increasing levels of Ecklonia cava (seaweed) supplementation in diets was investigated on growth performance, coefficient of total tract apparent digestibility (CTTAD) of nutrients, serum immunoglobulins, cecal microflora and intestinal morphology of weanling pigs. Methods: A total of 200 weaned pigs (Landrace${\times}$Yorkshire${\times}$Duroc; initial body weight $7.08{\pm}0.15kg$) were randomly allotted to 4 treatments on the basis of body weight. There were 5 replicate pens in each treatment including 10 pigs of each. Treatments were divided by dietary Ecklonia cava supplementation levels (0%, 0.05%, 0.1%, or 0.15%) in growing-finishing diets. There were 2 diet formulation phases throughout the experiment. The pigs were offered the diets ad libitum for the entire period of experiment in meal form. Results: The pigs fed with increasing dietary concentrations of Ecklonia cava had linear increase (p<0.05) in the overall average daily gain, however, there were no significant differences in gain to feed ratio, CTTAD of dry matter and crude protein at both phase I and phase II. Digestibility of gross energy was linearly improved (p<0.05) in phase II. At day 28, pigs fed Ecklonia cava had greater (linear, p<0.05) Lactobacillus spp., fewer Escherichia coli (E. coli) spp. (linear, p<0.05) and a tendency to have fewer cecal Clostridium spp. (p = 0.077). The total anaerobic bacteria were not affected with supplementation of Ecklonia cava in diets. Polynomial contrasts analysis revealed that villus height of the ileum exhibited a linear increase (p<0.05) in response with the increase in the level of dietary Ecklonia cava. However, villus height of duodenum and jejunum, crypt depth, villus height to crypt depth ratio of different segments of the intestine were not affected. Conclusion: The results suggest that Ecklonia cava had beneficial effects on the growth performance, cecal microflora, and intestinal morphology of weanling pigs.

The Microbes and Protein Extractability of Hanwoo and Japanese Wagyu (한우 및 일본 화우육의 미생물 오염도와 단백질 추출성)

  • This study was carried out to get the informations on microbes and protein extractability through comparing the quality attributes of Hanwoo fed in Korea, Hanwoo fed in Japan and Japanese Wagyu. The fresh beefs were stored at 4±l℃ for 13 days. In microbiological test, the total plate counts were higher in rump than in other beef portion as loin, chuck (p<0.000l). The number of psychrotrobes in the rump were maintained high levels (p>0.0001) for storage period, whereas the loin from Hanwoo fed in Korea, Hanwoo fed in Japan and Wagyu were lowest levels. The number of E. coli were no significantly different among the samples. In lactic acid bacteria, the loin form 3 grade Hanwoo (K3) had highest levels (p<0.0001). Comparing to the protein extractability, water soluble proteins were high in chuck (p<0.001). In the case of loin, water soluble proteins of K3 (3 grade Hanwoo) and Wagyu were high as 3.010 mg/g and 2.977 mg/g, respectively (p<0.001). Salt soluble protein of K1 (1 grade Hanwoo) was high as 7.437 mg/g (p<0.0001).

Synthesis, Characterization and Antibacterial Activity of Silver Nanoparticles in Poly(vinyl alcohol) Prepared by Gamma-Ray Irradiation (감마선에 의해 제조된 Poly(vinyl alcohol) 하이드로젤에서 Silver Nanoparticle의 제조 및 항균 특성)

  • In this study, silver nanoparticles (AgNPs) have been prepared by using aqueous $AgNO_3$ solution in the poly(vinyl alcohol) (PVA) hydrogels. PVA powders were dissolved in deionized water, and then irradiated by gamma-ray with a radiation dose of 50 kGy to make hydrogels. PVA hydrogels were dipped into 0.01 and 0.05 M $AgNO_3$ solution for 1 h respectively. After that, the swollen hydrogels were irradiated by gamma-ray at various doses to form AgNPs. UV-vis analysis indicated that the concentration of Ag NPs was enhanced by increasing absorbed dose and the concentration of $AgNO_3$. FE-SEM measurements provided further evidence for the successful formation of Ag NPs in PVA hydrogels. Also, the antibacterial effect of PVA hydrogels stabilized AgNPs against Gram-negative bacteria (S.aureus and E.coli) in liquid as well as on solid growth media has been investigated. The AgNPs consolidated in PVA hydrogel networks have an excellent antibacterial effect.

The Use of Fermented Soybean Meals during Early Phase Affects Subsequent Growth and Physiological Response in Broiler Chicks

  • The objectives of this experiment was to evaluate the subsequent growth and organ weights, blood profiles and cecal microbiota of broiler chicks fed pre-starter diets containing fermented soybean meal products during early phase. A total of nine hundred 1-d-old chicks were randomly assigned into six groups with six replicates of 25 chicks each. The chicks were fed control pre-starter diet with dehulled soybean meal (SBM) or one of five experimental diets containing fermented SBM products (Bacillus fermented SBM [BF-SBM], yeast by product and Bacillus fermented SBM [YBF-SBM]; Lactobacillus fermented SBM 1 [LF-SBM 1]; Lactobacillus fermented SBM 2 [LF-SBM 2]) or soy protein concentrate (SPC) for 7 d after hatching, followed by 4 wk feeding of commercial diets without fermented SBMs or SPC. The fermented SBMs and SPC were substituted at the expense of dehulled SBM at 3% level on fresh weight basis. The body weight (BW) during the starter period was not affected by dietary treatments, but BW at 14 d onwards was significantly higher (p<0.05) in chicks that had been fed BF-SBM and YBF-SBM during the early phase compared with the control group. The feed intake during grower and finisher phases was not affected (p>0.05) by dietary treatments. During total rearing period, the daily weight gains in six groups were 52.0 (control), 57.7 (BF-SBM), 58.5 (YBF-SBM), 52.0 (LF-SBM 1), 56.7 (LF-SBM 2), and 53.3 g/d (SPC), respectively. The daily weight gain in chicks fed diet containing BF-SBM, YBF-SBM, and LF-SBM 2 were significantly higher values (p<0.001) than that of the control group. Chicks fed BF-SBM, YBF-SBM, and LF-SBM 2 had significantly lower (p<0.01) feed conversion ratio compared with the control group. There were no significant differences in the relative weight of various organs and blood profiles among groups. Cecal microbiota was altered by dietary treatments. At 35 d, chicks fed on the pre-starter diets containing BF-SBM and YBF-SBM had significantly increased (p<0.001) lactic acid bacteria, but lowered Coli-form bacteria in cecal contents compared with those fed the control diet. The number of Bacillus spp. was higher (p<0.001) in all groups except for LF-SBM 1 compared with control diet-fed chicks. At 7 d, jejunal villi were significantly lengthened (p<0.001) in chicks fed the fermented SBMs vs control diet. Collectively, the results indicate that feeding of fermented SBMs during early phase are beneficial to the subsequent growth performance in broiler chicks. BF-SBM and YBF-SBM showed superior overall growth performance as compared with unfermented SBM and SPC.
