The Modification of X-ray induced Mutation in Paramecium by Pre-and Post-irradiation Treatment with Colchicine. (X-선에 의한 돌연변이 빈도에 미치는 Colchicine의 영향)

  • We have carried out a study on the modification of the frequency of X-ray induced lethal and slow growing mutations by colchincine treatment before and after X-ray irradiation in Paramecium aurelia. 1. Lethal and slow-growing mutation induced by X-ray in Paramecium aurelia were reduced by colchicine treatment. 2. The effects of colchicine on the X-ray induced mutations were remarkable in the radiosensitive stages of cell division. 3. The pre-irradiation treatment with colchicine showed no significant influence on the frequency of mutations. 4. It is believed that the reduction of mutation induced by X-ray after treatment with colchicine was due to the fact that the delay of the cell division allowed more time for the pre-mutational damage to recover.

  • Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Codonopsis lanceolata. Three tetraploid plants of C. lanceolata were produced from seeds which absorbed 0.1 % colchicine solution for 12 hours, and 0.5% colchicine solution for 1 and 6 hours from seedlings, respectively. But tetraploid was not produced from shoot tips treated by colchicine solution. Compared to diploid, tetraploid plants had larger stomata, but less number of stomata. Fresh weight of tetraploid plants was 1.4∼3.6 times heavier than diploid plants.

  • Several lines evidence indicate that microtubule depolymerization initiates DNA synthesis or enhances the effects of serum or purified growth factors in many types of fibroblasts. Yet little is known about the intracellular events responsible for the mitogenic effect of microtubule disrupting agents. The effects of antitubulin agents on DNA synthesis in sparse and dense cultures in the presence or absence of serum and possible involvement of G-proteins in their mitotic action were examined. In these studies, colchicine by itself appeared to be mitogenic only for confluent quiesecent human lung fibroblasts. In sparse culture, however, colchicine inhibited serum-stimulated DNA synthesis. Colcemid, another antitubulin agent, showed similar effects of growth inhibition and stimulation in sparse and confluent cultures while lumicolhicine, inactive colchicine, did not. The mitogenic effect of two antitubulin agents, colchicine and colcemid, was partially inhibited by pertussis toxin. These data suggest that microtubular integrity is associated with the expression of either negative or positive control on DNA synthesis and mitogenic effect of antitubulin agents may be partially mediated by pertussis toxin-sensitive G protein.

  • Inhibition of platelet aggregation by colchicine, thiocolchicine, O-demethylated and 3-glucosidic congeners was measured in vitro. 2-And 3-demethylated analogs of colchicine and thiocolchicine were found tobe as potent as colchicine and thiocolchicine, respectively, whereas 1-demethylated compounds were found to be considerably less active. Both glucosides, colchicoside and thiocolchicoside, were not very active in this assay. 2, 3-Didemethylcolchicine, which is much less toxic than colchicine, showed similar inhibitory effects on platelet aggregation as colchicine.

High Frequency Production of Doubled-haploid Plants by Colchicine Application in Anther Cultures of Rice (벼 약배양에서 콜히친이 식물체 배수성에 미치는 영양)

  • To optimize the in uitro chromosome doubling procedure in anther cultures of rice, anthers were cultured on callus induction medium with 0.001 to 0.1 mg/L colchicine for 30 days. The addition of colchicine slightly reduced callus formation and plant regeneration in comparison to the colchicine-free medium (control medium). This reduction was greater with higher concentration colchicine. Microspore-derived rice plants in control medium were found to be mainly haploid (50 to 58.8%) and doubled-haploid (31 to 40%) in anther culture of 3 Japonica and 1 Tonsil type cultivars. The application of 0.001 mg/L colchicine was increased to 54.3∼60.0% in the frequency of fertile doubled-haploid plants. These results indicate that the addition of colchicine to the callus induction medium is an efficient means to obtain doubled-haploid plants in anther cultures of rice.

Morphological and Cytogenetic Analysis of Colchicine-induced Tetraploids of Fallopia multiflolra Haraldson (Colchicine 처리에 의해 유기된 4배체 하수오의 형태 및 세포유전학적 특성)

  • Background: For stable induction of tetraploidy in Fallopia multiflora Haraldson, colchicine was treated to establish the condition of induction and investigated the morphological and cytogenetic traits of the tetraploid plants obtained compared to those of diploid ones. Methods and Results: For the induction of tetraploidy, F. multiflora plants were soaked in aqueous solutions of colchicine at various concentration (0.1, 0.5, and 1.0%). After this, 2% dimethyl sulfoxide (DMSO) was added at room temperature on a shaker set at 150 rpm for periods of 12, 24, and 48 h. The induction rate of tetraploids appeared to be the highest in plants treated with 0.5% colchicine for 24 h. As the colchicine concentration and soaking time increased above these levels, the growing tip of the roots did not develop and they began to rot. When compared to diploid plants, tetraploids differed greatly in various characteristics, including the sizes and shapes of the leaves, fruits, flowers and roots. The induced tetraploid F. multiflora had larger guard cells, and chloroplasts, increased number of chloroplast in the guard cells and decreased stomatal densities. Conclusions: When colchicine induced plants for tetraploid, it can be distinguished from diploids, in various characteristics such as morphological changes as stomatal size, number of chloroplasts per guard cell, number of chromosomes and flow cytometry. Therefore, it proved that these methods are suitable, quick and easy methods for the identification of the ploidy level of F. multiflora.

A Study of the Classification of Different Types of the Golgi Apparatus around the Bile Canaliculus in the Rat Liver (담세관주위 Golgi장치의 유형 분류에 관한 전자현미경적 연구)

  • In this study, we have made morphological and cytochemical observations to investigate the type of Golgi apparatus around the bile canaliculus. The animal (Wister, $220{\sim}250gm$) were divided into 4 groups; normal, hydrochol, colchicine and hydrochol-colchicine. The Golgi apparatus is classified into 16 different types from 4 different groups. In the normal group, we could observe 12 different types of the sixteen. Type I which showed convexed cisterns facing the bile canaliculi was most abundant of the types. In the hydrochol group, 14 types were observed. Type VII and type I showed convexed cisterns facing the bile canaliculus and were abundant. In the colchicine group, 11 different types were viewed and type XIV which showed intensely dilated cisterns without the polarity was predominant. In the hydrochol-colchicine group, we observed 3 different types. Type XIV clearly showed the highest percentage, although that type was less numerous in this group than in the colchicine group. In the hydrochol group, the Golgi apparatus showed a tendency to increase in numbers, while in the hydrochol-colchicine group the Golgi apparatus showed a tendency to decrease in numbers. The reactive products of thiamine pyrophosphatase and acid phosphatase were apparent over the distal Golgi cistern in the normal and hydrochol groups, but were decreased or not observed in the colchicine and hydrochol-colchicine groups. From the results, it is assumed that with the presence of the microtubule, Golgi cisterns are dilated with polarity after stimulation of secretion. Without the microtubule, the cistern becomes more intensely dilated and none polaric. Also the enzymes within the cisternal membrane become decreaed or absent and the Golgi apparatus decreases in numbers after activation of secretion.

  • Colchicine is a drug used for the treatment of acute gouty arthritis or various autoimmune diseases. Gastrointestinal adverse effects such as abdominal pain and vomiting are the common side effects of the drug, but rarely myopathy has been reported to occur particularly in renal recipients who were treated concomitantly with immunosuppressants. Herein, we report a case who presented with acute myopathy after treated with colchicine for acute gouty arthritis.

  • Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Platycodon grandiflorum in Campanulaceae. The most successful colchicine treatment for tetraploid production in P. grandiflorum was soaking treatment using 0.01 and 0.5% colchicine solution for 1 hour and 12 hours, respectively. Morphological characteristics of both diploid and tetraploid were similar, but tetraploid plants had more leaves. Compared to diploid, tetraploid had the larger stomata, but less number of stomata. Fresh weight of tetraploids was 20∼40% heavier than that of diploid.

  • A 47-year-old woman ingested about 12 mg of colchicine with suicidal intent. Colchicine, a highly poisonous alkaloid, is a commonly used treatment for gout, Bechet's disease, and familial Mediterranean fever. Despite the knowledge of its side effects, the risk of a significant overdose is under-appreciated. She suffered from acute multisystem toxicity, including gastrointestinal disorders, bone marrow suppression, alopecia, and probable pancreatitis, but she ultimately recovered with supportive therapy. We report a case of acute colchicine toxicity from a single overdose with a review of the literature.
