The aim of this study was to identify antigens for a vaccine or drug target to control rabbit coccidiosis. A combination of 2-dimensional electrophoresis, immunoblotting, and mass spectrometric analysis were used to identify novel antigens from the sporozoites of Eimeria stiedae. Protein spots were recognized by the sera of New Zealand rabbits infected artificially with E. stiedae. The proteins were characterized by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF/TOF-MS) analysis in combination with bioinformatics. Approximately 868 protein spots were detected by silver-staining, and a total of 41 immunoreactive protein spots were recognized by anti-E. stiedae sera. Finally, 23 protein spots were successfully identified. The proteins such as heat shock protein 70 and aspartyl protease may have potential as immunodiagnostic or vaccine antigens. The immunoreactive proteins were found to possess a wide range of biological functions. This study is the first to report the proteins recognized by sera of infected rabbits with E. stiedae, which might be helpful in identifying potential targets for vaccine development to control rabbit coccidiosis.
Poultry products including meat and eggs constitute a major protein source in the American diet and disease-causing pathogens represent major challenges to the poultry industry. More than 95% of pathogens enter the host through the mucosal surfaces of the respiratory, digestive and reproductive tracts and over the past few decades, the two main mechanisms used to control diseases have been the use of vaccines and antibiotics. However, in the poultry industry, there are mounting concerns over the ability of current vaccines to adequately protect against emerging hyper-virulent strains of pathogens and a lack of suitable, cost effective adjuvants. Thorough investigation of the immunogenetic responses involved in host-pathogen interactions will lead to the development of new and effective strategies for improving poultry health, food safety and the economic viability of the US poultry industry. In this paper, I describe the development of immunogenomic and proteomic tools to fundamentally determine and characterize the immunological mechanisms of the avian host to economically significant mucosal pathogens such as Eimeria. Recent completion of poultry genome sequencing and the development of several tissue-specific cDNA libraries in chickens are facilitating the rapid application of functional immunogenomics in the poultry disease research. Furthermore, research involving functional genomics, immunology and bioinformatics is providing novel insights into the processes of disease and immunity to microbial pathogens at mucosal surfaces. In this presentation, a new strategy of global gene expression using avian macrophage (AMM) to characterize the multiple pathways related to the variable immune responses of the host to Eimeria is described. This functional immunogenomics approach will increase current understanding of how mucosal immunity to infectious agents operates, and how it may be enhanced to enable the rational development of new and effective strategies against coccidiosis and other mucosal pathogens.

This study examined the efficacy of natural ingredient formulated herbal medicine as feed additives against coccidiosis in broiler chicken. Medicinal herbs used in this study were Artemisia annua, Pulsatilla chinensis, Clematis chinensis and Scutellaria baicalensis. Two week old chicks were administered with 1% ingredient formulated herbal medicine. Observations of the efficacy were confirmed by examining the lesion score, histopathological appearance of cecum, shedding of fecal oocyst and production index in farm. S. baicalensis of the most effective substance were analyzed by UPLC. The results are as follows; among various types of Chinese medicine, the most efficacious ingredient was found to be pure bicalein. The major index substance is bicalein and $13{\mu}g/ml$ per gram was found. Feeding of chickens with S. baicalensis induced the coccidiostat effect and reduced the fecal shedding of E. tenella oocyst by 14%. Experimental results on a farm, feed conversion ratio and weight gain and production rate in the group with natural ingredient formulated additives than in control group increased. The rate of weight gain (%) in each experimental group showed 108% and 126% increase while the mortality rate was 1.4% lower than the control group. The investigation showed that the most efficacious ingredient was found to be S. baicalensis among herbal medicine products for coccidiostats. However, as it was found in the farm experiment that pure S. baicalensis alone is economically unviable. The additives with cocktailed of efficacious ingredient are suggested to be implemented for effective protection against coccidiosis.
Coccidiosis of domestic fowl is a parasitic disease that infects poultry drastically and can cost the pastured poultry producer much in the course of a year. Two hundred forty, day-old-broiler chicks were purchased and were randomly divided in eight groups, containing thirty birds in each group. Decoquinate $6\%$, maduramicin ammonium $2\%$, monensin sodium $13.2\%$, salinomycin sodium $12\%$, a live attenuated vaccine, and trivalent live attenuated vaccine, was provided to the day chicks of six groups, respectively. The chicks of last two groups served as infected non-medicated and uninfected non-medicated, respectively. Feed consumption, weight gain, feed conversion ratio, mortality and oocyst count per gram feces were recorded during the conduction of the experiment. Among treated groups, performance of salinomycin group was significantly better (p<0.05) in all aspects. Salinomycin acts against the sporozoites, trophozoites and first generation schizonts and is highly effective against the economically important species of Eimeria. The present study confirms that use of salinomycin, as an anticoccidial, is a drug of choice.
The present study was conducted to investigate the prevalence of Eimeria infection according to anti-coccidiosis drugs treatment in 1,300 slaughtered chickens from 130 farms in Gyeongnam, Jeonnam, Jeonbuk, Gyeongbuk and Chungnam during the period from October 2002 to August 2003. The prevalence of Eimeria infection by drug administration for treatment and prevention or non-administration was shown $71.4\%,\;82.6\%\;and\;72.3\%$, respectively, and so infection rate of chicken farms was similar independent on drug administration or not. The prevalence of Oocysts infection level of chicken farms by administrated drug for treatment was shown lower $(25.3\%)$ than for prevention $(35.4\%)$ and non-treatment $(36.2\%)$. The prevalence of Oocysts infection level by drug administration under 2 days for treatment was shown higher infection rate $(32.2\%)$ than over 3 days $(20\%)$ and also under 2 days for prevention has similar tendency of that of treatment and so, it was conclusion that administration of drug over 3 days has the effect for treatment and prevention of Eimeria infection. The prevalence of Oocysts infection level by age for prevention has various level from $48.8\%\;to\;22.9\%$ but on $22\~25$ days was shown the most higher ratio $(48.8\%)$ and on $19\~21$ days was shown the lowest rate $(22.9\%)$ among the all age groups.
A thirty-five week old elk was referred for Chronic Wasting Disease (CWD) confirmation from Chungnam province in Korea. Necropsy revealed much bloody contents in the lumen of small and large intestines, and reddening of its mucosa. Microscopically, few coccidia showing various developmental stages were infiltrated in lamina propria of small intestine with granulomatous inflammation and congestion. They were identified as Eimeria spp. according to their location and morphological characteristics. Because fecal examination and oocyst culture were not available, the species of the Eimeria could not be confirmed. There were no detection of pathogenic bacteria such as clostridia and virus in intestinal contents. CWD was shown to be negative in immunohistochemistry test. In conclusion, it was the first Eimeria case detected in small intestine of Korean domestic elk as far as we know although Eimeria spp. were not the main cause of death.
From January of 1991 to December of 1992, 42 chickens collected from 21 poultry farms and also diagnosed as necrotic enteritis(NE) was examined clinical signs, gross and histopathological findings. Main clinical signs were characterized by decreased appetite, mild to severe depression, reductance to move, ruffled feathers, greenish to yellow-browinish diarrhea sometimes including blood. As progressed, diseased chickens showed feces mixed with necrotic debris which detached from the intestinal mucosa and mostly resulted in the death. In chronic cases, there were dirty feathers around cloaca due to diarrhea and notably retarded growth. Principle gross lesions were usually confined to the jejunum and ileum, especially toward the lower part of Meckel's diverticulum. The part of small intestine was frequently distended with gas, and also showed mucosal congestion and hemorrhages with varying degrees. Sometimes, the intestinal mucosa was thickened, and also covered with fibronecrotic psuedomembrane. In addition, there were focal necrosis and severely multifocal ulcreation in the mucosa of small intestine. Major histopathological findings included villous necrosis and erosion of the small intestine covering with lots of bacterial colonies, inflammatory cell infiltration in the lamina propria, and dilatation and hyperplasia of crypts. Luminal exudate contained bacterial colonies, fibrin, erythrocytes, and desquamated epithelium. Thirteen(61.9%) out of 21 NE-occurring farms were complicated with intestinal coccidiosis.
Avian coccidiosis is a very important disease of poultry industry. In recent years, many researchers studied for the immunology of avian coccidiosis. Coccidial vaccines have been used at some poultry farms in many countries. In order to evaluate the anticoccidial effects of coccidial vaccine to chicken treated with anticoccidial drugs, 4 week-old chicken were administered anticoccidial drugs and challenged with avian coccidia at 2 weeks after vaccination of coccidia. The body weight gains, lesion scores, oocyst indices and anticoccidial indices of chicken were investigated at the time of vaccination, challenge and necropsy. The body weight gains and feed conversion rates of groups immunized with coccidial vaccine and treated with anticoccidial drugs were moderately higher and lower than those of groups treated with anticoccidial drugs, respectively. The lesion score and oocyst indices of all groups immunized with coccidial vaccine and/or treated with anticoccidial drugs were milder than those of infected control groups. The anticoccidial indices of groups immunized with coccidial vaccine and treated with anticoccidial drugs were moderately higher and lower than those of groups. treated with anticoccidial drugs. But all of the investigated items were not statistically significant.
