A coagulation/ultrafiltration membrane hybrid system was operated to treat river water with capacity of $0.06m^3/d$. The impact on membrane fouling by flux and linear velocity was investigated. It is known that pressure increase is proportional to flux increase. However, pressure increase was much faster than theoretical value in the pilot plant test. So it was suggested that flux was on important factor in ultrafiltration of continuous operation. Membrane fouling was decreased when linear velocity was increased. This phenomenon was found more obviously without coagulation. With the combination of coagulation and sedimentation, membrane fouling was not reduced conspicuously. Big particles formed during coagulation and sedimentation were destroyed by feed and circulation pumping, which resulted in little effect on membrane fouling reduction. The degree of destruction was similar at various linear velocities. In this study, the hollow fiber membrane was used and the system was operated in pressure type module. In case of the system used in this study, membrane fouling has been affected lightly by linear velocity variation when coagulation pretreatment was applied.
Purpose: The aim of this study was to assess the benefits of routine pre-endoscopy coagulation screening tests and platelet counts in Korean children. Methods: Between March 2004 and December 2009, children who underwent gastrointestinal endoscopy for the evaluation of various gastrointestinal symptoms were included. All of the subjects included in the study also underwent routine coagulation screening and platelet count determinations prior to endoscopy and biopsy. The clinical records and laboratory tests were retrospectively reviewed in all patients. Results: One hundred sixty-two of 1,476 (11%) patients who underwent endoscopy had abnormal results on pre-screening coagulation tests. Fourteen patients underwent coagulation factor assays due to abnormal clotting results in consecutive tests or due to clinical evidence of a bleeding tendency. Seven patients were diagnosed with factor XII deficiency, one patient was diagnosed with von Willebrand disease, one patient had von Willebrand disease and factor XII deficiency, and one patient was presumed to have mild hemophilia. The remaining 4 patients had normal results with the factor assays. The results of platelet counts were normal with the exception of 1 patient. No patient had significant bleeding during the endoscopic procedures, despite abnormal pre-endoscopic coagulation tests. Conclusion: Routine coagulation screening tests and platelet counts revealed abnormal results in some patients. Most of the patients with abnormal clotting were shown to have a factor XII deficiency, which had no significant associated bleeding tendencies; the other patients were diagnosed with hemophilia or von Willebrand disease. Therefore, although abnormal pre-endoscopic coagulation is not always related to significant bleeding complications, pre-endoscopic coagulation screening may be useful in some children in predicting the risk of bleeding tendency during endoscopic procedures.

This study presents a modeling method to predict fate of resuspended sediment in the development of deep-sea mineral resources. Resuspended deep-sea sediment during the development is considered a major environmental problem. In order to quantitatively analyze the resuspended sediment in the water column, particle size distribution (PSD) is considered an important factor. The model developed here includes PSD and coagulation process, as well as sedimentation process. Using the model, basic simulation was performed under representative environmental setting. The simulation showed the dynamics of change of particle size distribution for 50 m depth of water column up to 10 days of simulation time. Coagulation seemed an important factor in the fate of resuspended deep-sea sediment.
We report a case of polycythemia vera combined with coagulation disorder. The patient was 54 years old man who complained of continuous bleeding after incision of skin abscess 20days ago. Laboratory tests were revealed prolonged aPTT and slightly prolonged PT. Coagulation factor, I, VIII, IX, XI and fibrinogen decreased, however FDP did not increased. It appears that patient with polycythemia vera have chronic activation of coagulation system, probably initiated by activation of factor XII. Platelet aggregation test to ADP, collagen, epinephrine was also revealed poor response.
This study is about efficiency of pretreatment process and operating factor to membrane process at continuous coagulation/ultrafiltration process in water treatment. The capacity of pilot plant was $0.06{\beta}(C)/d$. The raw water used was from Nakdong stream which was characteristized by high organic matter and high turbidity. The result of the test was that coagulation is good process as to high removal rate to organic matter and turbidity but It caused problem to membrane pore blocking. This paper is to determine the membrane fouling potential under different membrane flux, backwash pressure and linear velocity. Backwash pressure and flux is important parameter on operation of membrane system. Those are directly affected on membrane system. When backwash pressure increased from 150 kPa to 200 kPa, the result showed that fouling (pressure increase rate) changed from 3.69 kPa/h to 0.93 kPa/h and the recovery rate changed from 90.7 % to 82.0 %. Linear velocity had slightly effect on fouling. Linear velocity increased from 0.2 m/s to 0.5 m/s, the corresponding pressure rate changed from 0.93 kPa/d to 0.77 kPa/d.
A novel inhibitory protein against blood coagulation factor Va (FVa) was purified from muscle protein of granulated ark (Tegillarca granosa, order Arcoida, marine bivalvia) by consecutive FPLC method using anion exchange and gel permeation chromatography. In the results of ESI-QTOF tandem mass analysis and database research, it was revealed that the purified T. granosa anticoagulant protein (TGAP) has 7.7 kDa of molecular mass and its partial sequence, HTHLQRAPHPNALGYHGK, has a high identity (64%) with serine/threonine kinase derived from Rhodopirellula baltica (order Planctomycetales, marine bacteria). TGAP could potently prolong thrombin time (TT), corresponding to inhibition of thrombin (FIIa) formation. Specific factor inhibitory assay showed that TGAP inhibits FVa among the major components of prothrombinase complex. In vitro assay for direct-binding affinity using surface plasmon resonance (SPR) spectrometer indicated that TGAP could be directly bound with FVa. In addition, the binding affinity of FVa to FII was decreased by addition of TGAP in dose-dependant manner ($IC_{50}$ value = 77.9 nM). These results illustrated that TGAP might interact with a heavy chain of FVa ($FVa_H$) bound to FII in prothrombin complex. The present study elucidated that non-cytotoxic T. granosa anticoagulant protein (TGAP) bound to FVa can prolong blood coagulation time by inhibiting conversion of FII to FIIa in blood coagulation cascade. In addition, TGAP did not significantly (P < 0.05) show fibrinolytic activity and cytotoxicity on venous endothelial cell line (ECV 304).
Inhibitory mechanism of the anticoagulant polysaccharide purified from the fruit body of Coriolus versicolor was investigated in this paper. The anticoagulant polysaccharide (CV-40-Va-1) was proposed to have functions of the inhibition of intrinsic pathway in the blood coagulation pathway together with the interuption of a human platelet aggregation induced by von Willebrand factor (vWF). CV-40-Va-I inhibited other factors of the coagulation cascade such as factor VIII, IX, and as well as thrombin. Especially, CV-40-Va-I inhibited the fibrin formation mediated by thromin, however the polysaccharide did not affect the fibrin formation directly but affected the anticoagulant activity through the activation of antithrombin III. The sulfation of the anticoagulant polysaccharide increased the anticoagulant activity, showing that the sulfate concentration of anticoagulant polysaccharide was important factor in the blood coagulation cascade. Low molecular weight subfraction (MW 1,000) obtained by partial hydrolysis of the CV-40-Va-1 generated potent antiplatelet activity, but showed decreased anticoagulant activity.
A study on relation of angiogenesis and blood stagnation in cancer was done, and the results were as follows. 1. Angiogenesis is a sequence of vascular proliferation and accomplished by regulation of anti-angiogenesis factor and indicating factor. These factors are secreted in the course of blood coagulation, inflammation, and regeneration. 2. Angiogenesis in cancer is a important action in growth of tumor and metastasis because it supply oxygen and nutrition. 3. The complicated processes, for example, platelet coagulation, action of coagulator factor & dissolution factor and interaction of variety factors are related to blood stasis and promoting blood circulation to remove blood stasis in oriental medicine. 4. Promoting blood circulation to remove blood stasis is expected to suppress angiogenesis, and we expect advanced study will be accomplished in future.
von Willebrand factor (vWF) is a multimeric glycoprotein which functions within the coagulation system. It colocalizes with factor VIII (FVIII) by non-covalent interaction and alters its intracellular trafficking. vWF is also instrumental in maintaining the stability of secreted FVIII. The principal objective of this study was to generate a lentivirus-based vWF expression vector for gene therapy of hemophilia A. We inserted a vWF of 8.8 Kb into a lentiviral vector thereby producing VSV-G-pseudotyped vEx52. However, its titer was quite low, presumably because the length of vWF gene exceeds the size limit of the lentiviral vector. In order to overcome the low-titer, we concentrated the vEx52 and thus increased the efficiency of transduction approximately 6-fold with $1/100^{th}$ of the volume. However, as concentration requires an additional laborious step, we attempted to enhance the transduction efficiency by deleting exons 24-46 and 29-46 in pRex52 to construct pRex23 and pRex28, and in pvEx52, yielding pvEx23 and pvEx28, respectively. The transfected pRex52 had a profound effect on the activity of secreted FVIII, and this activity declined as domains of vWF were deleted. However, when the domain-deleted vWF-lentiviruses were transduced into K562 cells, the vEx28 increased the activity of the secreted FVIII compared to what was observed with vEx52. This result is probably due to higher efficiencies of transduction and expression while retaining the essential domains required for proper interaction with FVIII.

Tissue factor (TF) is a cell surface receptor of coagulation factor Ⅶ and is the principal initiator of the vertebrate coagulation cascade. TF is found in high levels in some organs such as brain, lung and placenta, whereas blood monocytes, endothelial cells contain only trivial amount of TF when quiescent, and is stimulated to synthesize TF by infections or vascular lesions. TF is reported to be found in high levels in atherosclerotic plaques, cancer cells. TF activation in various cells in many infectious or immunologic diseases tells us the physiologic importance of TF. We screened many edible vegetables for TF inhibitor, by measuring the prothrombin time to detect the TF activity, and we picked Aster scaber to isolate the TF inhibitory substance. Aster scaber showed two kinds of anti thrombotic activity, one is TF inhibition and the other is elongation of plasma recalcification time. The anti thrombotic substances were found to be saponins which has echinocystic acid as aglycone.
