Comparison of pathogens and antimicrobial susceptibility of Staphylococcus aureus isolated from conventional and robotic milking herds (일반목장과 로봇착유 목장에서 분리한 세균과 Staphylococcus aureus의 항생제 감수성 비교)

  • This study was conducted to compare the patterns of mastitic pathogens and the antimicrobial susceptibility of Staphylococcus (S.) aureus from conventional milking (CM) and robotic milking (RM) dairy herds. To accomplish this, the minimum inhibitory concentrations of 14 antimicrobial agents were tested against S. aureus by the microdilution method. Regardless of the milking system, S. aureus, coagulase negative staphylococcus, and Streptococcus uberis were isolated. Additionally, significant differences in the antimicrobial susceptibility of S. aureus isolates between RM and CM farms were only observed in response to tetracycline.

Species of Bacteria and Antimicrobial Susceptibility Isolated from Clinical Specimens in Jeon Buk Area (전북지역 임상가검물에서 분리된 세균의 종류와 항생제 감수성)

  • Considering many problems caused by the abuse of antibiotics recently, the appearance of antibiotic resistance bacteria is believed to help the cure of patients greatly. From Jan. 1st, 1996 to Dec. 31, 1996, 6135 strains were examined after being asked of and seperated from the clinical pathology departments of general hospitals, and the isolation frequency of identified bacteria and the susceptibility of antibiotics showed the following result. 1. The isolation frequency of strains was Escherichia coli, 1134 strains (18.4%), Pseudomonas aeruginosa, 856 strains (13.9%), coagulase negative Staphylococcus, 793 strains (12.89%), Staphylococcus aureus, 555 strains (9.02%), B. cepacia, 421 strains (6.84%), Enterobacter cloacae, 366 strains (5.95%), Enterobacter faecalis (4.86%), and Klebsiella pneumonia, 220 strains (3.85%). 2. The isolation rate of specimen was urine, 1, 969 strains, wound 1, 104 strains, sputum 701 strains, blood 643 strains, vaginal swab, 342 strains, and eye discharge, 192 strains, 40% of urine strains were E. coli 18% of wound strains were B. cepacia, 43.7% of sputum were P. aeruginosa, and in blood strains there were Enterobacter cloacae (25.8%), coagulase negative Staphylococcus (19.6%), and P. aeruginosa (8.7%). 3. The result of antibiotics susceptibility showed that, among gram negative bacilli, P. aeruginoas had resistance in almost all antibiotics except ceftazidme imipenem. But B. cepacia, the same glucose non-fermentation gram negative bacilli had more than 90% of sensitivity in aztreonam, ceftazidime, ciproflxacin, piperacillin, trimethoprim/sulfa and had resistance in the others. Enterococcus faecalis showed more than 85% of sensitivity in penicillin-G, ampicillin, ciprofloxacin. 4. In the case of specimen antibiotics susceptibility, Enterobacter cloacae was lower in specimen isolated from blood than in those isolated from others and p. aeruginosa was low in specimen isolated from urine, which showed that there was difference in specimen antibiotics susceptibility. The result of this study shows that there happen many resisitances in antibiotics used frequently and some countermeasure is necessary because many bacteria began to show new resistance. Also it is desirable that the choice of antibiotics for infection diagnosis and its cure should be made after the inspection of antibiotics.

A Study about Clinical Characteristics of Methicillin Resistant-Coagulase Negative Staphylococcus Infections in Neonatal Intensive Care Unit (신생아 중환자실에서 Methicillin Resistant-Coagulase Negative Staphylococcus 감염에 대한 임상적 고찰)

  • Purpose : Methicillin Resistant-Coagulase Negative Staphylococcus(MR-CNS) infection has become an increasingly important cause of morbidity in NICU infants. We investigated the c linical characteristics of MR-CNS sepsis. Methods : This study included 40 neonates with MR-CNS sepsis who were admitted to the neonatal intensive care unit of Kangnam Sacred Heart Hospital, Hallym University from January 1998 to July 2002. MR-CNS sepsis was defined as MR-CNS recovery from blood with clinical symptoms and signs of infection. Retrospective analyses of the medical records of patients with MR-CNS sepsis were performed. The analyses included demographic findings, clinical features, hospital courses, risk factors for infection including invasive procedures and mortality. Results : From 1998 to 2002, there were 40 cases of MR-CNS sepsis, comprising 17.7% of late onset infections in NICU of Kangnam Sacred Heart Hospital. The male/female ratio was 1.5 : 1. The mean gestational age of infected babies was $32.4{\pm}4.3$ weeks at birth. And the first positive MR-CNS culture was done in the day $10.6{\pm}9.3$ after birth. Clinical symptoms such as fever, dyspnea, cyanosis, grunting, bradycardia, vomiting and diarrhea were frequent in MR-CNS. Mechanical ventilation was applied in 12 cases and catheter was inserted in 11 cases. The mortality(12.5%) directly attributable to MR-CNS sepsis was similar to other late onset infections. Conclusion : MR-CNS is a pathogen responsible for most late onset and nosocomial infections. And it will be life-threatening in high-risk neonate. Awareness of increasing infections due to MR-CNS in NICU is important not only for infection control but also placing a great limit in use of antibiotics and invasive procedures, especially in premature infants.

Prevalence of Methicillin-resistant Staphylococci Isolates from Horses and Horse-related Personnel in Korea (말 및 말관련 종사자의 methicillin 내성 포도상구균의 유병율 조사)

  • Methicillin-resistant staphylococci (MRS) are emerging as important pathogens in humans and animals worldwide. The aim of this study was to investigate the prevalence of MRS in the racehorse population and in horse-related personnel in Korea. A total of 195 horses and 18 humans (eight veterinarians, three veterinary hospital staff, and seven horse-handlers) from racehorse farms in Korea were included in the study. The samples were collected from nasal cavities using bacterial transport medium and were cultivated on tryptic soy agar with 5% sheep blood for 3 days at $37^{\circ}C$ to confirm the presence of Staphylococcus spp. Presumptive Staphylococcus spp. isolates were identified by 16S ribosomal RNA gene analysis. The coagulase test and oxacillin susceptibility tests were performed using the tube dilution and disk diffusion methods, respectively. The presence of the mecA gene was determined using a polymerase chain reaction assay. Of the 195 horses, 29 (15.6%) yielded 29 MRS isolates. Twelve (66.7%) of the 18 horse-related personnel yielded 12 MRS isolates. All of the MRS isolates from horses or horse-related personnel were identified as methicillin-resistant coagulase-negative staphylococci (MRCNS). The result of this study suggest that the prevalence of MRS increased with the duration of antibiotic use (p = 0.002). This study also provides evidence for the zoonotic transmission of MRCNS between horses and humans, although further investigations are needed.

Isolation and characterization of Staphylococcus hyicus subsp hyicus from chickens (닭으로부터 Staphylococcus hyicus subsp hyicus의 분리(分離) 및 그 특성(特性))

  • Isolation of Staphylococcus hyicus subsp. hyicus from the skin of healthy chickens was performed and biochemical characteristics of the isolates were examined. Staphylococcus hyicus subsp hyicus was isolated from 129(14.8%) of 874 healthy chickens. The rate of isolation of this organism from chickens was found to vary depending upon the poultry farms at the isolation rate of 0% to 38.7% and this organism was isolated more frequently from adult chickens of over 1 year old than young chickens. All of the isolates were negative for haemolysis on rabbit, sheep, pig and chicken blood agars, protein A and ${\beta}$-glucuronidase, but positive for coagulase using pig plasma, heat-stable DNase, phosphatase and Tween 80 hydrolysis. Hyaluronidase was positive in 97.7% of isolates, but their reaction was weaker than that of swine strains of Staphylococcus hyicus subsp hyicus. The isolates were biotyped on the basis of the reactions for protease, urease, nitrate, galactose, trehalose and lactose, and 11 biotypes were found among the 129 isolates of Staphylococcus hyicus subsp hyicus.

Antimicrobial drugs susceptibility of bacterial flora in horses with respiratory tract infections (경주마의 호흡기질환 유래균의 약제 감수성 시험)

  • Bacteria isolated from nasal cavity of 50 Thoroughbred horses with respiratory tract infection were examined. There were isolated Pseudomonas aeruginosa(33.5%), Escherichia coli(10.2%), Pseudomonas spp(7.6%), Klebsiella oxytoca(5.9%), Streptococcus equi subsp zooepidemious(6.2%), Klebsiella pneumoniae(3.4%), Acinetobacter spp(5.5%) and coagulase negative staphylococcus(2.1%). The majority of isolates were highly susceptible to amikacin, amoxicillin, aztreonam, cefotaxime, cefepime, cefotetan, ceftazidime, cefuroxime, chloramphenicol, ciprofloxacin, clindamycin, erythromycin, gentamicin, imipenem, tetracyclin and vancomycin. These results can provide basic information for the treatments of respiratory tract infections in Thoroughbred horses.

Prevalence of resistance to macrolide, lincosamide. streptogramin and ketolide antibiotics against Gram-positive bacteria in Korea

  • The purpose of this study is to investigate the prevalence of resistance to macrolide, lincosamide, streptogramin and ketolide antibiotics in Korea. The antibiotic susceptibility test was performed to the macrolide erythromycin, clarithromycin, azithromycin, josamycin, the lincosamide clindamycin, the streptogramin synercid and the ketolide ABT -773 against 337 clinical Staphylococcus aureus(SAU). Coagulase-negative Staphylococci (CNS) and Enterococci isolates exhibited an average percentage of 64%, 56%, and 81 % of resistance to erythromycin, respectively.

Selection and Characterization of Staphylococcus hominis subsp. hominis WiKim0113 Isolated from Kimchi as a Starter Culture for the Production of Natural Pre-converted Nitrite

  • Synthetic nitrite is considered an undesirable preservative for meat products; thus, controlling synthetic nitrite concentrations is important from the standpoint of food safety. We investigated 1,000 species of microorganisms from various kimchi preparations for their potential use as a starter culture for the production of nitrites. We used 16S rRNA gene sequence analysis to select a starter culture with excellent nitrite and nitric oxide productivity, which we subsequently identified as Staphylococcus hominis subspecies hominis WiKim0113. That starter culture was grown in NaCl (up to 9%; w/v) at 10℃-40℃; its optimum growth was observed at 30℃ at pH 4.0-10.0. It exhibited nonproteolytic activity and antibacterial activity against Clostridium perfringens, a bacterium that causes food poisoning symptoms. Analysis of Staphylococcus hominis subspecies hominis WiKim0113 with an API ZYM system did not reveal the presence of β-glucuronidase, and tests of the starter culture on 5% (v/v) sheep blood agar showed no hemolytic activity. Our results demonstrated the remarkable stability of coagulase-negative Staphylococcus hominis subspecies hominis WiKim0113, especially in strain negative for staphylococcal enterotoxins and sensitive to clinically relevant antibiotics. Moreover, Staphylococcus hominis subspecies hominis WiKim0113 exhibited a 45.5% conversion rate of nitrate to nitrite, with nitrate levels reduced to 25% after 36 h of culturing in the minimal medium supplemented with nitrate (200 ppm). The results clearly demonstrated the safety and utility of Staphylococcus hominis subspecies hominis WiKim0113, and therefore its suitability as a starter culture.

Detection of multidrug resistant patterns and associated-genes of Methicillin Rdsistant Staphylococcus aureus(MRSA) isolated from clinical specimens (임상검체에서 분리된 Methicillin Resistant Staphylococcus aureus의 다제내서양상과 내성 관련 유전자의 검색)

  • Methicillin Resistant Staphylococcus aureus (MRSA) was obtained from the clinical specimens at Pusan national university Hospital, Pusan, Korea. The sensitivities against various antibiotics were examined by using disc diffusion test and associated genes such as mecA, mecR1, mecI and femA were detected by polymerase chain reaction. Among Seventy-nine strains of MRSA, 38 strains(48.1%)were sensitive to streptomycin and 32 strains(40.5%) to cefoperazone, while one strain(1.3%) were resistant to vancomycin. In considering the result of this study, 7 strains showed resistance to 9 kinds of different antibiotics, 12 strains were to 8 kinds, 24 strains were to 7,25 strains were to 6, 9 strains were to 5, and 2 strains were to 4 antibiotics. Among 79 strains of MRSA, 67 strains were coagulase positive and 12 were coagulase negative. In the detection of MRSA associated genes by PCR method, mecA, mecR1, mecI, and femA genes were detected in 30 strains(44.8%), 28 strains(41.8%), 23 strains(34.3%) and 15 strains(22.4%), respectively. MecA type that is without femA were found in 21 strains(31.3%), femA type that is without regulator genes were shown in 4 strains(6.0%), while mecA-mecR1-mecI type with regulator genes were shown more to be 17 strains(25.4%). There was little statistical significance between multidrug resistance and MRSA associated genes. Considering these result, it is necessary to include moecular biological studies of related genes to the study drug resistance.

Isolation Frequency of Staphylococcus species from Clinical Materials (임상검체에서 포도알균의 분리 빈도)

  • The total number of clinical materials collected from Dec 1, 2005 to Nov 30, 2006 in C. hospital was 63,133. Among these materials, the isolation of Staphylococcus spp. was 5,252 strains. The proportions of Staphylococcus species infection were as follows ; 46.7% in S. aureus, 39.3% in S. epidermidis, 4.7% in S. hominis, 3.8% in S. haemolyticus, 2.9% in S. capitis and 2.5% in others. Seventeen strains of Staphylococcus species were identified with an isolation range of 0.02-46.7%. The isolation ratios of Staphylococcus species were 7.8% (2,134/27,362) in female, 8.7% (3,118/35,807) in male and 8.3% (5,252/63,133) in both. The age-related frequencies of Staphylococcus species were 22.1% in the 60-69 year group, 19.3% in the 70-79 year group and 17.1% in the 50-59 year group. The isolation ratio of the 0-49 year group to the 50-over 80 year group were 1.74 times in female, 1.92 in male and 1.85 in both. The isolation frequency of Staphylococcus spp. was at its highest in April (9.9%) and at its lowest in February (6.8%) according to monthly analysis. According to seasonal variation, the isolation frequencies of Staphylococcus spp. were 27.0% in Summer, 25.9% in Spring, 23.9% in Fall and 23.2% in Winter. The clinical departments showing over 9% isolation rate against Staphylococcus spp. were critical care medicine (12.9%), department of pediatrics (11.6%), department of urology (9.8%) and department of neuro-surgery (9.1%). On the other hand, the lowest number was observed in the department of family medicine. The clinical material showing over 15% isolation rate against Staphylococcus spp. were 32.1% in urine (random urine and catheterized urine), 20.6% in blood and 16.0% in sputum.
