Study on Acute Toxicity of Fermented Ohyaksungi-san (Wuyaoshunqi-san) Extracts (오약순기산 발효물의 급성 독성에 관한 연구)

  • Objectives: The purpose of this study was to investigate the acute toxicity with oral administration in ICR mice of Ohyaksungi-san fermented with Lactobacillus sp. Methods: In single oral administered toxic test, four groups were administrated different dosages(0, 1250, 2500, 5000 mg/kg) of fermented Ohyaksungi-san. After single oral administration, we observed number of death, clinical signs, body weight changes, hematological values and autopsy. Results: Compared with the control group, we could not find any toxic signs in the mortalities, clinical signs, body weight changes, necropsy findings and hematological values in all treated groups (1250, 2500 and 5000 mg/kg). Conclusions: These results indicate that the $LD_{50}$ value of fermented Ohyaksungi-san extract might be over 5000 mg/kg and it have no side toxic effect to ICR mice

  • Sixty non-vital primary molars of forty six children who were patients at the Pedodontic Department of Seoul University Hospital, aged from 2 years 7 months to 9 years 1 month were performed for coronal pulp therapy. For the evaluation of non-vital pulp therapy, the author observed those teeth both clinically and roentgenographically. The results were as follows: 1. Coronal therapy of non-vital tooth can be accepted as one of the ordinary methods at the view of the save of tooth, time and simplicity. 2. The results were satisfactory 78.3% clinically and 68.3% roentgenographically. 3. The dissatisfactory signs were a) Clinical view There were dissatisfactory signs on 13 cases. (1) 2 teeth had fistulous opening. (2) 11 teeth reacted to purcussion and mobility. b) Roentgenographical view There were dissatisfactory signs on 19 cases. (1) 13 teeth showed roentgenolucency at bifurcation area. (2) 2 teeth showed pathological root resorption. (3) 4 teeth showed periodontal thickening.

  • This study represented an endeavor to observe clinical signs and pathological lesions in broiler chicks suffering from experimental Infectious stunting syndrome(ISS). One hundred and twenty day old broiler chicks were divide randomly into two equal groups i.e. control (A) and inoculated (B). At day one of age each chick of the groups (A and B) was dosed with one ml of either tryptose phosphate broth or prepared inoculum respectively. Chicks of both the groups were housed separately under similar standard management. Inoculation induced characteristic clinical changes in birds of treatment group like of brownish diarrhea, lameness, feather developing problems and paleness of combs, wattles and shanks. By day-29 of the experiment all the stunted birds from group-B and an equal number of birds from group-A were slaughtered. These birds were examined thoroughly to record the gross changes in various structures and then the severely affected organs were processed for histopathological examination. The skeletons of affected birds were brittle, keel bones showed quite prominence while the muscles and subcutaneous tissues were almost devoid of fat. Grossly it was observed that pancreas, spleen and bursa of Fabricius were severely atrophied while the intestines were ballooned with undigested feed and gases. Histopathological examination of pancreas and spleen revealed a classical picture necessary for understanding the pathogenesis of the syndrome. The acivar cells of pancreas were atrophied and underwent vacuolation, degeneration and vecrosis. The zymogen granules were almost absent from the acinar cells. A characteristic change was an inflammatory reaction in one or more pancreatic ducts where the epithelium and fibrous tissues occluded the lumen of the ducts and led to the obstruction in pancreatic drainage.

Role of Multislice Computerized Tomographic Angiography in Vasospasm Following Aneurysmal Subarachnoid Hemorrhage

  • Objective : We evaluate the role of multislice computerized tomographic angiography[MCTA] in the diagnosis of intracranial vasospasm following subarachnoid hemorrhage[SAH] in patients suspected of having vasospasm on clinical ground. Methods : Between October 2003 and June 2005, patients with ruptured cerebral aneurysms of the anterior circulation clipped within 3 days of the onset were included. We performed follow-up MCTAs in patients who were suspected to have vasospasm on transcranial doppler sonography[TCD] findings and clinical grounds. Based on the clinical presentation of symptomatic vasospasm, we investigated the correlation between clinical, TCD, and MCTA signs of vasospasm and evaluated the role of MCTA in vasospasm. Results : One hundred one patients met the inclusion criteria and symptomatic vasospasm developed in 25 patients [24.8%]. We performed follow-up MCTAs in 28 patients. MCTA revealed spasm in the vessels of 26 patients. The sensitivity of MCTA was 100%. Among the 26 patients with MCTA evidence of vasospasm, 3 patients had TCD signs of vasospasm after symptomatic vasospasm presentation. Another 3 patients with symptomatic vasospasm had no TCD signs of vasospasm in daily serial recordings. Six other patients without symptomatic vasospasm showed MCTA evidence of vasospasm [false positive result] but these patients had also positive TCD signs of vasospasm. Volume rendering[VR] images tended to show significantly more exaggerated vasospasm than maximum intensity projection[MIP] images. The mean cerebral blood flow velocity of both proximal segment of the middle cerebral artery [M1] was significantly correlated with each reduced M1 diameter on MCTA [P<005]. Conclusion : MCTA could be a useful tool for evaluation and planning management of critically ill patients suspected of having vasospasm; however, more randomized controlled trials are necessary to assess these points definitively.

Study on the pathogenesis of the piglets experimentally infected with Korean isolate of Aujeszky′s disease virus I. Histopathologic and electron microscopic observation (Aujeszky's disease virus 국내분리주 접종자돈의 병리발생에 관한 연구 I. 병리학적 및 전자현미경적 관찰)

  • This study was conducted to elucidate the pathogenesis of Aujeszky's disease virus(ADV) by histopathologic examination. The first Korean ADV Isolate, which was isolated from piglets with clinical signs of Aujeszky's disease in Yangsan(YS) county, Kyungnam province, was inoculated into 32 days old piglets with a dose of $10^{5.9}$$TCID_{50}/ml$ through intranasal or intramuscular route. These piglets were sacrificed at intervals of every 24hrs for 8 days. The virulence of YS strain was determined by the observation of clinical signs, gross findings, and histopathologic changes in tissues. The virus recovery test was performed from brain, spleen, lung and tonsil in cell culture. The pathogenesis of YS strain was determined by the observation of histopathologlc lesions in CNS and neuronal tracts. The major clinical signs were fever, anorexia, dyspnea, constipation, tremor, ataxia, circling movement, hindleg paralysis and salivation. The clinical signs were more severe in piglets of the group inoculated intranasally than those of the intramuscularly inoculated gorup. Lymphocytopenia was detected on day 5 to day 6 postinoculation (PI). The ADV was recovered from the tissue homogenates of tonsil, lung, spleen and cerebrum in cell culture. The highest virus titer was detected from tonsil between day 6 and day 7 PI. Reddish sublobar consolidation foci were scattered in the apical and cardiac lobes of lung. Although yellowish necrotic foci were detected in tonsil and liver, hemorrhagic lesions were mainly observed in heart, kidney and lymph nodes. Histopathologically, degeneration and necrosis of nerve cells, nonsuppurative meningoe-ncephalitis, nodular gliosis and perivascular cuffings were observed in CNS. Multifocal fibronecrotic foci were observed in lung, liver, lymph nodes and spleen. The major pathologic changes were detected in the midbrain, pons and medulla oblongata. Eosinophilic intranuclear inclusion bodies were mainly observed in epithelia and /or macrophages of tonsil, liver, lung, spleen and submandibular lymph nodes, and neurons of brain, respectively. Observation of viral particles at various stages of replication were possible from the endothelial cells of the alveolar capillaries and tonsillar crypt epithelia by transmission electron microscope.

Histopathological Observations of the Natural Case and Experimental Occurence of Perirenal Edema in Pig (돼지의 Perirenal Edema의 자연발생예(自然發生例)와 실험적발생예(實驗的發生例)의 병리학적소견(病理學的所見))

  • This paper dealt with the histopathological observations of the perirenal edema in pigs and rabbits administered with Amaranthus retroflexus, based on the clinical and pathological observations of the porcine perirenal edema naturally occurred in Korea. The results observed are summarized as follows; In the natural case, clinical signs were trembling, weakness and incoordination of the hindquarters, followed by sternal recumbency, coma and death. Death usually occurred within 24 hours after the signs of illness appeared. In gross findings, the grayish yellow fluid in the perirenal area was observed in each case. In some cases, the amount of fluid in the thoracic and abdominal cavities was increased and the yellowish red color of the hydrothorax and ascites was seen. When the renal capsule was incised, the kidneys were enlarged and congested and petechiae on the cortical surface and blood clots on the capsule appeared. In microscopical findings, there were cloudy swelling, hyaline droplets and necrosis of the convoluted tubules containing proteinaceous casts and a few oxalate crystals. In addition, interstitial and perivascular edema, distention of the Bowman's space and the convluted tubules and hemorrhages were recognized. In the weanling pigs and the adult pig fed various weeds, including Amaranthus retroflexus, Euxolus blitum and Portulaca oleracea, the pigs fed Amaranthus retroflexus appeared clinical signs and pathological findings of perirenal edema usually seen in the pigs of natural cases. In the pigs fed Euxolus blitum or fed Portulaca oleracea, neither clinical signs nor pathological changes were seen. It was regarded that this disease was affected with Amaranthus retroflexus, but there was no sensitivity in the adult pigs.

  • Horner's syndrome is a group of clinical signs that results from interference with the sympathetic innervation of the globe and adnexa. Three dogs were presented with unilateral ptosis, miosis, enophthalmos and protrusion of the third eyelid. There were no other clinical signs on physical and neurological examination. On ophthalmic examination, the symptomatic eyes were diagnosed as Horner's syndrome. In order to localize the site of the lesion, pharmacological testing was performed through assessment of ocular response to the topical administration of 10% phenylephrine and clinical signs were resolved within 20 minutes. The pharmacological testing suggested that the deficit could be at the postganglionic neuron. Total resolution of clinical signs was observed within 6 months after their initial appearance without any treatment.

Evaluation of Serum NT-proBNP and Cardiac Troponin I Concentrations in Dogs with Heartworm Disease

  • Biomarkers used in dogs with heartworm disease include N-terminal pro B-type natriuretic peptide (NT-proBNP) and cardiac troponin I (cTnI), which are associated with damage to the myocardium. Pulmonary hypertension is one of the clinical signs of canine heartworm disease. The purpose of this study is to investigate the change in the concentration of each biomarker, severity of pulmonary hypertension and the correlation between biomarkers according to the severity of clinical signs. Five healthy dogs and 10 heartworm-infected dogs were recruited for the study. The heartworm-infected group was classified based on the history, clinical signs, and blood assay, thoracic radiography, and echocardiography after confirming the infection according to the results of the commercial ELISA kit (SNAP test, IDEXX Laboratories, Maine, USA). NT-proBNP was higher in the severely infected group than the control group (p < 0.05); cTnI was also higher in the severely infected group than the control group (p < 0.05). The pressure gradient of pulmonary hypertension was higher in the severe group than the mild group (p < 0.05). The severity of pulmonary hypertension was correlated with NT-proBNP (r = 0.818, p < 0.01), cTnI (r = 0.894, p < 0.01). When the correlation of the two serum values for each group was examined, a correlation was not found in the mild group (r = 0.707, p = 0.182), but a correlation was found in the severe group (r = 0.9, p < 0.05). NT-proBNP and cTnI were significantly increased and correlated with severe clinical signs. Pulmonary hypertension was significant higher in the severe group than in the mild group (p < 0.05). Evaluation of blood biomarker concentration and severity of pulmonary hypertension and referring to each correlation between these indicators may be helpful to assess the severity of the heartworm disease.

Treatment of central diabetes insipidus with anemia in a dog

  • A 10-year-old, spayed female miniature schnauzer was referred to the Veterinary Medical Teaching Hospital of Chungnam National University due to evaluation of sudden polyuria (PU) and, polydipsia (PD) (540 mL/kg/day) with severe anemia and weight loss. Blood examination results were normal except for severe anemia (hematocrit, [HCT]: 11.8%). Urinalysis revealed a urine specific gravity (USG) of 1.003, whereas urine sediment was not specific. Urine osmolality was 90 mOsm (reference range: 800~2500 mOsm), and plasma osmolality was 303 mOsm. No specific lesions were found using diagnostic imaging including radiography, ultrasonography and magnetic resonance imaging (MRI). The serum cortisol level was normal in cosyntropin stimulation test. Plasma arginine vasopressin (AVP) concentration was <0.4 pg/mL (reference range: 3.49~5.45 pg/mL). Blood transfusion was initiated in addition to an oral prescription of desmopressin acetate (DDAVP, 0.1 mg/head) thrice a day for one week. The patient was rechecked for clinical signs, urine osmolality, and USG; the clinical signs of PU/PD were resolved, urine osmolality increased to 1106 mOsm, and, USG increased to 1.021. Considering the improved clinical signs, and increased urine osmolality, and USG after DDAVP treatment, the dog was diagnosed with central diabetes insipidus. USG and urine osmolality increased to >1.030 and 2200 mOsm, respectively. Anemia also gradually improved and HCT increased to >37%. DDAVP was tapered to 0.1 mg/head twice a day and all clinical signs in the patient have completely resolved.

Gold Beads Implants for The Treatment of Canine Chronic Recurrent Otitis Externa

  • A clinical trial was performed to assess clinical efficacy and/or reduction in relapses by gold-bead implantation into acupuncture points in dogs with canine chronic recurrent otitis externa (CCROE). Forty dog-patients randomly divided into two groups were diagnosed as suffering CCROE, having intact tympanic membranes and a history of recurrences. Treatments were: control group (CG), treated with commercially available antimicrobial otic droplets, dosed twice daily for 7 days; and experimental group (GBI- gold bead implants), treated as for CG plus the insertion of 13 gold-bead implants under light anesthesia. Overall per cent assessment of composite clinical progression and progression of individual clinical signs were recorded. Bilateral chronic external otitis was diagnosed in 60% of the cases and left or right otitis in 20% of the cases each. Logistic model for repeated measures analysis showed that GBI induced a better clinical recovery as far as lesion score of some clinical signs is concerned. The overall percent cures of each group showed statistically significant difference. A McNemar analysis revealed that higher number of relapses was observed in CG patients as compared to the GBI (P < 0.05). In particular during these days, lesion on the pinna showed in odd ratios analysis a 7:1 ratio (recurrence CG:GBI) and ear wax/pus (4:1) from D42 to D365. It is concluded that gold-bead implantation into acupuncture points improves resolution of some clinical signs and greatly reduce relapses in CCROE affected dogs after 1 year follow-up (98.75%).
