Potential Association between Insulin-like Growth Factor-1 Receptor Activity and Surf Clam Spisula sachalinensis Larvae Survival

  • We investigated the relationship between viability and IGF-1 receptor (IGF-1R) activity in D-shaped and umbo larvae of the surf clam Spisula sachalinensis after treatment with vitrification solution (VS) or freezing. In a toxicity assay, VS1, containing 5 M dimethyl sulfoxide (DMSO), was very harmful to D-shaped and umbo larvae. However, VS2, containing 5 M ethylene glycol (EG), was not harmful to either larval stage. Although VS2 had a promising toxicity test outcome, none of the larvae survived vitrification. After immersion into VSs and freezing, IGF-1R ${\beta}$-subunits were detected in all larvae; however, tyrosine phosphorylation of intracellular ${\beta}$-subunits was detected only in the control and live groups. These results suggest that activation of IGF-1R may influence surf clam larvae viability.

  • The Manila clam, Ruditapes philippinaMm (Pelecypoda: Veneridae) is present along the coasts of Korea, China and Japan. In particular, Recently, due to reclamation of tidal areas along the west coast, marine pollution, and reckless overharvesting of this clam, its standing stock has reduced for a decade. Therefore, it is necessary to manage the population of the clam with a proper fishing regime that will maintain an optimal population size in aqua farm. (omitted)

분쇄패각이 시멘트 모르터의 물성에 미치는 영향 (Effects of Crushed Shells on the Physical Properties of Cement Mortar)

  • 굴과 꼬막과 같은 패각의 일부는 채묘종과 비료 등으로 재활용 되기도 하나, 대부분 인근 해안에 적치 되거나 불법 매립되고 있는 실정이다. 본 연구에서는 패각의 기초물성을 측정하고 패각을 시멘트 모르터에 혼입할 수 있는 가능성을 평가하고자 하였다. 그 결과 패각을 25% 혼입한 시멘트 모르터의 경우 흡수율과 압축강도가 Plain과 유사하게 측정되었다. 반면 굴패각을 25% 혼입한 경우 Plain에 비해 압축강도가 35% 저하되었으며, 흡수율은 2배로 증가하였다. 따라서 굴을 제외한 나머지 패각을 25% 범위 내에서 시멘트 모르터에 혼입하여 사용하는 것은 가능할 것으로 사료된다. 하지만 굴의 경우 다양한 혼입율을 적용한 추가 실험이 요구될 것으로 판단된다.

Single-cell Gel Electrophoresis Assay에 의한 대합에서의 In Vivo 유전독성 평가 (Use of the In Vivo Single-cell Gel Electrophoresis Assay for Evaluating Genotoxicity in Clam)

  • The suitability of the single cell gel electrophoresis (SCGE) assay as a test for the monitoring of genotoxicity of aquatic environment was evaluated. The SCGE assay was employed to detect DNA damage induced in clam (Spisula sachalinensis) exposed to a direct mutagen, N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) or an indirect mutagen, benzo[a]pyrene (B[a]P). The cells of gill and digestive glands were isolated from clam by homogenization, which was the optimized cell dissociation method, and the level of DNA damage was assessed and expressed as mean tail length. In the gill cells, significant dose- and time-dependent increase was observed in the mean tail length at the concentration from 0.01 to 0.5 ppm MNNG for 96 h. The linear correlation between relative dam-age index (RDI) values was suggested to provide criteria of genotoxicity monitoring for direct acting mutagen. The dose- and time-dependent responses of the digestive glands cells were less sensitive than those of the gill cells. In contrast, the genotoxic response resulting from the exposure of 0.01~1.0 ppm B[a]P to clam revealed a higher sensitivity in the digestive glands cells than the gill cells. The comparison between the time profiles of genotoxic responses in clam and carp, the latter had been obtained in our previous study, indicated that the metabolism of genotoxic compounds in the two aquatic organisms were quite different each other. We conclude that the SCGE assay has the potential as a screening test for routine genotoxicity monitoring of aquatic organisms because of its higher sensitivity and simplicity.

Genetic Polymorphism of Marsh Clam (Corbicula leana) Identified by RAPD- PCR

  • Genomic DNA from the muscle of marsh clam (Corbicula leana) from Gochang, Muan and a Chinese site was extracted to identify genetic differences and similarity by randomly amplified polymorphic DNAs-polymerase chain reaction (RAPD- PCR). Out of 20 primers, seven primers produced amplified fragments which were consistently polymorphic. A total of 1,246 amplified products were produced of which 530 were polymorphic $(42.5\%)$. The number of polymorphic bands produced per primer varied from 40 to 122 with an average of 75.7 in marsh clam from Gochang. 3.28 of the 23.0 polymorphic bands per lane were found to be polymorphic. Also, about $4.34\%$ of total polymorphic bands were specific to marsh clam from Gochang. The major common bands of 0.28 kb generated by primer OPB-15 (GGAGGGTGTT) were present in every individuals, which were polymorphic. This common bands in every individuals should be diagnostic of specific strains, species and/or their relatedness. Primer OPB-19 (ACCCCCGAAG) produced the highest number of 12 specific bands. The intra-population variation was revealed in the band patterns identified by this primer. The random primer OPB-12 (CCTTGACGCA) yielded the amplified fragments which were consistently polymorphic between the marsh clams from Gochang and from Muan. This primer produced a total of 77 polymorphic bands: 31 bands from Gochang, 14 from Muan and 32 from the Chinese populations. An average of polymorphic bands were 1.8 from Gochang and 2.5 from the Chinese populations. This value obtained from the Chinese population was higher than those from the two domestic populations. Generally, the RAPD polymorphism generated by these primers may be useful as a genetic marker for strain or population identification of marsh clam.

패육의 건조 및 저장중 아미노산 함량의 변화 (Changes in Amino Acid Contents during Drying and Storage of Shellfish Meat)

  • 건조온도, 저장온도 및 저장기간에 따른 홍합 및 바지락의 아미노산 함량 변화는 다음과 같다. 생시료의 경우 총 아미노산의 함량은 두시료 모두 6575.30mg% 와 6764.43mg%로 비슷하게 나타났으며, 건조에 의한 함량의 감소는 홍합 보다 바지락의 경우가 더 많이 감소하였다. 아미노산 함량은 두시료 모두 Glx가 790.55 및 990.89mg%로 가장 많았으며 , 기타 아미노산의 함량은 시료에 따라 조금씩 달랐다. 시료를 건조함에 따라 아미노산의 함량은 감소하는 경향이였으며, 건조온도가 높을수록 감소하는 폭이 컸다. 그리고 저장온도가 높을수록 저장기간이 길수록 아미노산의 함량이 감소하는 폭이 더 크게 나타났다.

Tin-free 방오제인 Sea-Nine 211에 노출된 북방대합에서 MFO 효소계의 반응 (Responses of MFO System in Surf Clam, Pseudocardium sachalinensis, Injected with Sea-Nine 211 Antifoulant)

  • 방오도료로 많이 쓰인 유기주석화합물은 일반생물에게 미치는 독성이 매우 강하고 또한 내분비계 장애물질임이 밝혀지면서 이를 대체할 화합물들의 개발이 요구되고 있다. 그 가운데 이런 목적으로 만들어진 화합물인 Sea-Nine 211을 사용하여 이것이 해양생물 특히 저서생물인 패류에게 얼마나 영향을 미치는지를 살펴보고자 북방대합(P. sachalinensis)에게 강제 주사하여 생존율과 중장선의 미크로좀 중 I상 약물대사효소의 변화를 4일째가지 조사하였으며, 또한 비교를 위해 tributyltin chloride (TBTC)의 주사실험을 병행하였다. 생존율에서는 sham구나 Sea-Nine 211 실험구(5, 25 및 50 mg kg$^{-1}$)모두 생존하였으나 TBTC(1, 2 및 5 mg kg$^{-1}$) 실험구의 생존율은 4일째에 각각 70%, 30% 및 0%였다. 한편, 약물대사효소계 중 I상효소인 CYP는 Sea-Nine 211주사 후 시간이 지나면서 유도되었지만, TBTC는 반대로 저해되는 경향을 보였다. 그리고 P450R 활성은 Sea-Nine 211 실험구에서 시간 경과와 더불어 약간 증가하는 추세를 보였지만 sham구와는 유의적인 차이가 없었으나, TBTC실험구는 주사농도와 비례하면서 저해되었다. b5R활성도 마찬가지로 Sea-Nine 211 실험구는 큰 변화가 없었으나, TBTC실험 구에서는 유의적으로 저해되는 경향을 관찰할 수가 있었다. 이처럼 비주석계 방오도료 화합물인 Sea-Nine 211은 북방대합에 대한 독성이 TBTC에 비해 크게 낮았으며, 약물대사효소계에 미치는 영향도 TBTC는 저해하는데 반해서 P450R이나 b5R에는 별다른 변화를 일으키지 않았으나 CYP는 유도하였다.

바지락치패의 폐사에 관한 연구-II 바지락 치패의 온도, 염분, 노출에 대한 저항성 (Studies on the Natural Mortality of the Young Short Necked Clam, Tapes Japonica-II . Resistance to Temperature , Salinity and Exposure for the Young Short Necked Clam)

  • In a previous paper, the dirunal and monthly changes of the tidal flat temperature and chlorinity were observed. In this paper the resistance of the young short necked clam in various growth stages to the temperature, to the chlorinity and to the exposure were studies. The young clam used were in early (E), medium (M) and late (L) young stages whose shell lengths were 2.0-3.5mm, 9.0-11.0mm. and 14.0-16.0mm., respectively. The results were : 1. At various sea water temperatures , the chlorinity resistance of the young clam was directly proportional to the shell length. 2. When both inadequate sea water temperature and abnormal chlorinity are simultaneously applied, the resistance of these young clams was more markedly reduced than the case of applying either one of these conditions. 3. In clams of M and L, no lethal effect were observed when daily immersion of four to eight hours for a week into the sea water of any concentration of chloriity at 26-34$^{\circ}C$, whereas in E, 37-90% of mortalities were obtained by immersion of eight hours daily into both the fresh water and the sea water of higher chloriniites (more than 23.5$\textperthousand$ Cl) at the same temperature above. 4. The lower critical thermal maximum for lethal to the young clam was 38$^{\circ}C$. With four hours immersion daily at the water temperature of 38$^{\circ}C$, the mortalities of E, M and L to the lower chlorinities (less than 6.7$\textperthousand$Cl) were 100, 70-100 and 27-37% respectively ; to the higher chlorinities (more than 23.5$\textperthousand$Cl) 10-70, 10-37 and 3 % respectively ; to the normal range of chlorinities (13.4-16.8$\textperthousand$Cl) 0-13, 3 and 0 % respectively. 5. No lethal effects were observed in E and M clams immersed continuously for seven days in sea water with chlornities of 7.2 -21.7$\textperthousand$Cl at 18-24$^{\circ}C$, while notable mortalities were observed in E which had been kept at lower (less than 4.8$\textperthousand$ Cl) and higher (more than 24.1$\textperthousand$ Cl) chlorinites. 6. Although the resistance of the young clam to the chlorinity may have to be related closely to the life history of the clam prior subject to the experiment, the adapted chlorinity range was 7.2-19.3$\textperthousand$ Cl and the optimum range was 13.4-16.8$\textperthousand$Cl. 7. Remarkable lethal effects were observed for the E and M clams to the exposure temperature of 38$^{\circ}C$ whereas the L and had no such fatal results.
