The purpose of this study was to investigate the effect of butanol (BuOH) fraction of Alisma canaliculatum (Ac) and/or selenium (Se) treatment on glycogen level, lipid metabolism and lipid peroxidation in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were assigned to one of the five groups: normal, STZ-control, and three experimental groups (Ac group, Ac-Se group, and Se group). Diabetes was experimentally induced by intravenous administration of 45 mg/kg of STZ in citrate buffer. The BuOH fraction of Ac (400 mg/kg bw) was orally administered for 3 weeks. The Se group were fed a AIN-93 recommended diet mixed with Na$_2$SeO$_3$ (2 mg/kg diet). The liver glycogen level of Ac and Ac-Se groups were significantly higher, when compared with the STZ-control groups. The muscle glycogen level was not significantly differ among all groups. The levels of liver triglyceride were higher in Ac-Se group than the STZ-control group. Pancreas protein levels were significantly increased in Ac-Se group than STZ-control group. The concentration of liver malondialdehyde (MDA) was significantly decreased in Ac and Se groups and decreased in Ac-Se group. Administration of BuOH fraction of Alisma canaliculatum and selenium supplementation increased the liver glycogen and triglyceride levels, and reduced peroxidative liver damage in STZ induced diabetic rats. These results suggest that treatment with a BuOH fraction of Alisma canaliculatum in combination with selenium has no synergistic antioxidative effect. Selenium supplementation may lead a decrease MDA of liver in diabetic rats.
This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the lipid peroxidation and mineral levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' liver and muscle glycogen, liver and kidney protein, cholesterol and triglyceride (TG) in liver, malondialdehyde (MDA) in liver and kidney values were measured, along with the hepatic of chromium (Cr), iron (Fe), and zinc (Zn) content. The liver glycogen levels was significantly affected in N-GW 20% group among all the experimental groups. The liver MDA levels of the STZ-GW 10% and STZ-GW 20% groups were significantly lower than for the STZ-control group. There were significant differences between the N-control group and the STZ-control group in the hepatic of Zn levels. The hepatic of Cr levels in the N-GW 20% and STZ-GW 10% and STZ-GW 20% groups were significanly higher than for the each control groups. These results exhibited dose related effect of glasswort and it may have favorable influence on lipid peroxidation in the liver.
This studies were to investigate the effects of low power Helium-Neon Infra-Red (He-Ne IR) laser on the healing of the peripheral neuropathy in diabetic rats. The twenty one Spraque-Dewley adult male rats were assigned to the 3 groups: the control group (7), the diabetic group (7) and the laser group (7). The diabetic group was induced with a single intravenous injection of 50mg/kg body weight streptozotocin dissolved to 0.01mol/L citrate buffer, pH 4.5. The experimental laser group was irradiated low power He-Ne IR laser for 5 minutes every day during 21 days to the diabetic group. The results were as follows: For on and two weeks with laser treatment, there were significantly increased the terminal latencies of the laser treatment group than those of diabetic group, but that was significantly decreased to be similar to the control group on the laser treatment group for three weeks. The change of the amplitude on the laser treatment groups for three weeks was significantly increased to be similar to normal group than that of the diabetic group. As the result, we thank that there were activately effected of lower power He-Ne IR laser on the heeling of the peripheral neurophathy in diabetic rats.
This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the plasma blood glucose and lipid metabolites in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. The rats' body weights, aminotransferase activities and hematocrit (Hct) values were measured, along with plasma levels of glucose, protein, cholesterol, HDL-cholesterol, triglyceride (TG) and free fatty acids (FFA). The non-diabetic rats gained weight, while the diabetic rats lost weight. There were significant differences between the control group and the diabetic groups in the weight of the kidney, liver and pancreas. Asparate aminotransferase activity was lower in the non-diabetic control group compared to diabetic experimental groups, even though the difference was not significant. The plasma protein of N-GW 20% group was lower among all experimental groups but it was not significantly different. The blood glucose levels of the STZ-GW 10% group and STZ-GW 20% group were significantly lower than for the diabetic-control group. There were no significant difference of cholesterol levels among diabetic groups. The normal rats of 20% glasswort group in FFA and TG levels showed significant changes among all groups. These results exhibited dose related effect of glasswort and it may contain antihypoglycemic compounds.
The purpose of this study was to investigate the effect of green tea catechins on the antioxidative defense enzyme activity of kidney in diabetic rats. Sprague-Dawley male rats weighting 100$\pm$10g were randomly assigned to one normal and three STZ-induced diabetic groups; catechin free diet(DM-0C group), 0.25% catechin diet(DM-0.25C group) and 0.5% catechin diet(DM-0.5C group). Diabetes was induced by intravenous of 55mg/Kg body weight of STZ in sodium citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Rats were sacrified at the 6th day of diabetic states. Superoxide dismutase(SOD) activity in kidney was decreased by 25% and 20% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. Glutathione peroxidase(GSHpx) activity in kidney was were no significant differences the diabetic groups compared to normal group. Xanthin oxidase(XOD) activity was increased by 110% and 63% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. The contents of superoxide radical(O$_2$)in kindney were 116% and 33%, respectively, higher in DM-0C and DM-0.25C groups than normal group. DM-0.5C group and normal groups were similar levels in their superoxide radical contents of kidneys. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 62% in DM-0C group, when compared with normal group, but those of DM-0.5C group were similar to that of normal groups. These results indicate that free radical generation system was weakened and free radical scavenger system was enhance in kidney of STZ-induced diabetics rats by dietary catechin. Thereby it may reduce renal disorders such as oxidative damage and aging of tissue.
Purpose: To investigate the histopathological changes in the temporomandibular joint in streptozotocin-induced diabetic rat following irradiation. Materials and Methods : Sprague-Dawley rats weighing about 250 gm were divided into three groups: control, diabetic, and diabetic-irradiated groups. Diabetes mellitus was induced in the rats by injecting streptozotocin. Rats in the control group were injected with citrate buffer only. After 5 days, the head and neck region of the rats in diabetic-irradiated group were irradiated with single absorbed dose of 10 Gy. The rats were killed at 1, 3, 7, 14, 21, and 28 days after irradiation. The specimen including the temporomandibular joint were sectioned and observed using a histopathological method. Results : In the diabetic group, severe bone resorption in the mandibular condyle was observed throughout the period of experiment. Necrosis of bone marrow and trabeculae was observed at 28 days after diabetic state. Atrophy and fibrosis in the retrodisca] tissue was gradually progressed during the time of the experiment. In the diabetic-irradiated group, severe bone resorption in the mandibular condyle was observed during the early experimental phases, but regeneration of bone marrow was initiated at ]4 days after diabetic state and irradiation. A]so, calcification of abnormal trabeculae was observed at 28 days after diabetic state and irradiation. The retrodisca] tissue was degenerated in the early experimental phases, but it had been gradually regenerated during the experimental time. Conclusion: This experiment suggests that bone resorption and degeneration in the mandibular condyle are caused by the induction of diabetes, and abnormal bone formation is induced after irradiation in diabetic state.
Using sodium hypophosphite as reducing agent, bath composition and plating condition of electroless copper plating on plating rate have been studied. The followings were determined as optimum, bath composition; $CuSO_4\;0.025M,\;NiSO_4\;0.002M,\;NaH_2PO_2\;0.4M$, sodium citrate 0.06M, $H_3BO_3$ 0.6M, thiourea or 2-MBT $0.2mg/{\ell}$, and operation conditions; pH $9{\sim}10$ at bath temperature rage of $60{\sim}70^{\circ}C$. A small amount of nickel ion($Ni^{2+}/Cu^{2+}$=0.002/0.025) to the hypophosphite reduced solution promotes autocatalysis and continuous plating. An additive such as thiourea or 2-MBT of a small amount($0.2mg/{\ell}$) can be used to stabilize the solution without changing plating rate much. The attivation energy between $20^{\circ}C\;and\;70^{\circ}C$ were calculated to be 11.3kcal/mol for deposition weight. Plating reaction had been ceased by the adjustment of pH above 13, temperature higher than $90^{\circ}C\;and\;under\;20^{\circ}C$. Deposited surface became worse in the case of increment of bath temperature above $80^{\circ}C$.
This study was carried out to evaluate the effects of fractions of methanol(MeOH) extracts of Lycopus lucidic Turcz on hyperglycemia and energy metabolites in streptozotocin(STZ) diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of STZ dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight, and the rats were divided into 7 groups, that is, one normal group and 6 diabetic groups: STZ-control, hexane, chloroform(CHCl$\sub$3/). ethylacetate(EtOAc), butanol(BuOH) and H$\sub$2/O fraction-fed groups. All groups were fed an AIN-93 diet and the fractions of Lycopus lucidic Turcz were administered orally with 2 % Tween 80 for 14 days after the STZ injection. Body weight, diet intake and organ weights were monitored. The plasma levels of blood glucose, insulin and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglycerides and free fatty acid were assayed. The plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase(ALT) were also measured. Body weight losses were observed by feeding the fractions of Lycopus lucidic Turcz in STZ experimental groups, and the kidney weight was increased. The extent of blood glucose decrement was significantly greater in the hexane and BuOH fraction-fed groups than STZ-control group. The plasma protein level was significantly lower in the H$\sub$2/O fraction-fed group. The plasma cholesterol level was decreased in BuOH and H$\sub$2/O fraction-fed groups compared with the STZ-control group. The levels of free fatty acids in the CHC1$\sub$3/ and H$\sub$2/O fraction-fed groups were significantly decreased(p<0.05). ALT activitiy of BuOH fraction-fed group was lower than control but it was not significantly different. These results suggest that the fractions of Lycopus lucidic Turcz are capable of lowering blood glucose and fat metabolites concentrations when administered to STZ-treated rats, and AST/ALT activity and insulin levels show the possibility of therapeutic use to diabetes mellitus.

This study was designed to test the effect of dandelion (Taraxacum officinale) extracts on the intestinal microorganisms of streptozotocin-induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively. Diabetes was induced by injecting streptozotocin (55 mg/kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45g of raw dandelion / kg diet for 50 days. The growth of Lactobacillus was more enhanced in dandelion leaf-water and leaf-ethylacetate extract group than that of the diabetic control group, whereas the growth of E. coli decreased. Results indicate that the dandelion extracts would be effective to improve the intestinal microorganisms.

The purpose of this study was to investigate the effects of green tea catechin o n free radical generation system and peroxidative damage in the liver of streptozotocin(STZ)-induced diabetic rats. Spragu-Dawley male rats weighing 150$\pm$10gm were randomly assigned to one normal and three STZ-induced diabetic groups; diabetic groups were classified to catechin free diet(DM-oC group), 0.5% catechin diet(DM-0.5C group) and 1% catechin diet(DM-1C group) according to the levels of dietary catechin supplementation. Diabetes was experimentally induced by intravenous injection of 55mg/kg of body wt of STZ in citrate buffer(pH 4.3) after feeding of three experimental diet for 4 weeks. Animals were sacrificed at the 6th day of diabetic states. Activities of serum glutamic oxaloacetic transaminase(GPT) in DM-oC groups were higher than those of the normal group, and those in catechin supplementation group were similar to those of the normal group. Liver lipid peroxide values increased by 153%, 49%, and 27% in Dm-oC, DM-0.5C and DM-0C and Dm-1C but was not significantly different in catechin supplementation groups compared with the normal group, and liver cytochrome $P_{450}$ contents was similar to result of XOD activity. In electron microscopic examination of liver, lysosome was relatively scattered in Dm-oC and Dm-0.5C group and preserved normal shapes in DM-1C group. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stress, leading to the acceleration of lipid peroxidation process, but this was reduced by anti-oxidative effect of high level of dietary catechin. It is concluded that dietary catechin serves as powerful antioxidant against lipid peroxidation in diabetic rats.
